| 1 | Plasma and aqueous humor levels of adiponutrin and pannexin 1 in patients with and without diabetic retinopathy显示文摘AIM: To evaluate plasma and aqueous levels of adiponutrin and pannexin 1 in patients with and without diabetic retinopathy.METHODS: The study included three age and gendermatched groups of 20 cataract patients with no diabetes or additional disease(Group C), 20 cataract patients with diabetes and no retinopathy(Group DM+C), and 20 cataract patients with diabetic retinopathy(Group DR+C).All the patients were examined with respect to body mass index(BMI), fasting plasma glucose, hemoglobin A1c(HbA1c), and lipid profile.Phacoemulsification and intraocular lens(Phaco+IOL) implantation were performed to all patients in all the groups, and aqueous samples were taken during the operation.The plasma and aqueous adiponutrin and pannexin 1 levels were analyzed using enzyme-linked immunosorbent assays.RESULTS: A statistically significant difference was determined between the groups with respect to BMI, fasting plasma glucose, and HbA1c levels(P<0.05 for all parameters tested).The plasma adiponutrin levels of Group DR+C were statistically significantly lower than those of Group C and Group DM+C(P<0.001, P=0.004).No statistically significant difference was determined in the aqueous adiponutrin levels in three groups.The plasma pannexin 1 levels of Groups DM+C and DR+C were statistically significantly lower than those of Group C(both P=0.001).The aqueous pannexin 1 levels of Group DR+C were statistically significantly higher than those of Group C and Group DM+C(P=0.001, P<0.001).CONCLUSION: Adiponutrin and pannexin 1, which play an important role in the pathophysiology of diabetes and obesity, and have a regulatory role in hyperglycemia and insulin resistance.The measurement of adiponutrin and pannexin 1 levels may support clinicians in determining the risk of DR development. | Fatih Cem Gül Sabiha Güngör Kobat FatihÇelik Süleyman Aydin Ramazan Fazil Akkoç | 2022 | International Journal of Ophthalmology(English edition)2022,15,3: | 3 |
| 2 | The effect on wound healing of pazopanib and bevacizumab compared with corticosteroid in experimental glaucoma filtration surgery显示文摘AIM: To compare the effects of bevacizumab and pazopanib with corticosteroids on wound healing after trabeculectomy. METHODS: In the study, 35 New Zealand white rabbits were randomly divided into five groups. Apart from the first group, limbus-based trabeculectomy was performed for the eyes of rabbits. No postoperative treatment was administered for group Ⅰ. Topically administered saline, prednisolone acetate(1%), bevacizumab 5 mg/mL, pazopanib 5 mg/mL for group Ⅱ, Ⅲ, Ⅳ and Ⅴ respectively were applied for groups 6 h daily for 28 d. On day 28 of the experiment, eyes were enucleated and histologically and immunohistochemically analyzed.RESULTS: The fibroblast counts of groups Ⅳ and Ⅴ were determined to be lower than those of groups Ⅱ and Ⅲ(P<0.05). In the mononuclear cell(MNC) count evaluation, no statistically significant difference was determined between the treatment groups(P>0.05). The immunohistochemical staining intensity of fibroblast growth factor β(FGF-β) and vascular endothelial growth factor(VEGF) was determined to be lower in groups Ⅳ and Ⅴ than in groups Ⅱ and Ⅲ(P<0.05). No statistically significant difference was determined between groups Ⅳ and Ⅴ in respect of fibroblast count, MNC count, FGF-β and VEGF staining intensity(P>0.05). The platelet derived growth factor β(PDGF-β) intensity was lower in group Ⅴ than in groups Ⅱ, Ⅲ and Ⅳ(P<0.05). While the PDGF-β staining intensity was significantly lower in group Ⅳ thanin group Ⅱ, the difference compared with group Ⅲ was not statistically significant(P>0.05). | 3 | Seroprevalence of hepatitis B surface antigen in pregnant women attending antenatal clinic in Honiara Solomon Islands,2015显示文摘AIM To determine the seroprevalence of hepatitis B surface antigen(HBsAg) among pregnant women attending antenatal clinic in Honiara, Solomon Islands. METHODS This descriptive cross-sectional study was carried out in seven area health centers in Honiara. From March to June 2015, identification of eligible pregnant women in each site was conducted using systematic random sampling technique. A total of 243 pregnant women who gave written informed consent were enrolled. Standardized tool was used to record demographics, obstetric history and serology results. HBsAg and hepatitis B e antigen(HBeAg) were tested using point-of-care rapid diagnostic test. All HBsAg positive samples were verified using enzyme-linked immunosorbent assay. RESULTS The mean age of participants was 26 ± 6 years. The overall hepatitis HBsAg prevalence was 13.8% with higher rate(22%) reported in women between 30-34 years of age. Majority of HBsAg positive participants were Melanesians(29 out for 33). None of the pregnant women in the 15-19 years and ≥ 40 years tested positive for HBsAg. There was no statistically significant difference in HBsAg prevalence by age, ethnicity, education and residential location. The overall HBeAg seroprevalence was 36.7%. Women between 20-24 years of age had the highest rate of 54.5%. Low level of knowledge about hepatitis B vaccination was reputed. Overall, 54.6% of participants were not aware of their hepatitis B vaccination status and only 65.2% of mothers reported their child had been vaccinated. | 5 | Vasculitis with renal involvement in essential mixed cryoglobulinemia:Case report and mini-review显示文摘The discovery of a strong association between hepatitis C virus(HCV) infection and mixed cryoglobulinemia(MC) has led to an increasingly rare diagnosis of idiopathic essential MC(EMC).The incidence of EMC is high in regions where there is a comparatively low HCV infection burden and low in areas of high infection prevalence,including HCV.The diagnosis of EMC requires an extensive laboratory investigation to exclude all possible causes of cryoglobulin formation.In addition,although cryoglobulin testing is simple,improper testing conditions will result in false negative results.Here,we present a 46-year-old female patient with a case of EMC with dermatological and renal manifestations,highlighting the importance of extensive investigation to reacha proper diagnosis.We review the need for appropriate laboratory testing,which is often neglected in clinical practice and which can result in false negative results.This review also emphasizes the significance of an extended testing repertoire necessary for better patient management.Despite a strong association of MC with HCV infection and other causes that lead to cryoglobulin formation,EMC remains a separate entity.Correct diagnosis requires proper temperature regulation during sample handling,as well as characterization and quantification of the cryoprecipitate.Inclusion of rheumatoid factor activity and complement levels in the cryoglobulin test-panel promotes better patient management and monitoring.Consensus guidelines should be developed and implemented for cryoglobulin detection and the diagnosis of cryoglobulinemic syndrome,which will reduce variability in inter-laboratory reporting. | Sabiha Anis Khawar Abbas Mohammad Mubarak Ejaz Ahmed Sajid Bhatti Rana Muzaffar | 2014 | World Journal of Clinical Cases2014,2,5: | 2 |
| 6 | Phytochemical screening and evaluation of cytotoxic and hypoglycemic properties of Mangifera indica peels显示文摘Objective: To investigate the presence of different phytoconstituents in Mangifera indica(M. indica) peel and evaluate its cytotoxicity to Artemia salina and hypoglycemic potential in Swiss albino mice.Methods: The methanolic extract of M. indica peel was used to determine the presence of phytoconstituents. Brine shrimp lethality bioassay method was followed to determine the cytotoxic potential of plant extract. In the case of hypoglycemic activity, oral administration of extract at 200 and 400 mg/kg and standard glibenclamide at 10 mg/kg was done, followed by determining the percentage of reduction of plasma glucose from the initial level.Results: The methanolic extract of M. indica peel showed the presence of flavonoid,saponin, steroid, tannins, terpenoids, glycosides and alkaloids. In brine shrimp lethality bioassay, the LC_(50) of the extract and standard vincristine sulfate was found to be 2.04 and0.41 mg/mL, respectively. After 90 and 150 min, the methanolic extract at 200 and 400 mg/kg showed prominent plasma glucose reduction of 13.95%, 22.48% and 14.16%,26.18% respectively compared to standard glibenclamide showing 14.90% and 20.67% plasma glucose reduction.Conclusions: This current research affirms prominent cytotoxic and moderate hypoglycemic potential of M. indica peel. | 18 | Early detection and classification of live bacteria using time-lapse coherent imaging and deep learning显示文摘Early identification of pathogenic bacteria in food,water,and bodily fluids is very important and yet challenging,owing to sample complexities and large sample volumes that need to be rapidly screened.Existing screening methods based on plate counting or molecular analysis present various tradeoffs with regard to the detection time,accuracy/sensitivity,cost,and sample preparation complexity.Here,we present a computational live bacteria detection system that periodically captures coherent microscopy images of bacterial growth inside a 60-mm-diameter agar plate and analyses these time-lapsed holograms using deep neural networks for the rapid detection of bacterial growth and the classification of the corresponding species.The performance of our system was demonstrated by the rapid detection of Escherichia coli and total coliform bacteria(i.e.,Klebsiella aerogenes and Klebsiella pneumoniae subsp.pneumoniae)in water samples,shortening the detection time by >12 h compared to the Environmental Protection Agency(EPA)-approved methods.Using the preincubation of samples in growth media,our system achieved a limit of detection(LOD)of ~1 colony forming unit(CFU)/L in≤9 h of total test time.This platform is highly cost-effective(~$0.6/test)and has high-throughput with a scanning speed of 24 cm2/min over the entire plate surface,making it highly suitable for integration with the existing methods currently used for bacteria detection on agar plates.Powered by deep learning,this automated and cost-effective live bacteria detection platform can be transformative for a wide range of applications in microbiology by significantly reducing the detection time and automating the identification of colonies without labelling or the need for an expert. | Hongda Wang Hatice Ceylan Koydemir Yunzhe Qiu Bijie Bai Yibo Zhang Yiyin Jin Sabiha Tok Enis Cagatay Yilmaz Esin Gumustekin Yair Rivenson Aydogan Ozcan | 2020 | Light(Science & Applications)2020,9,1: | 1 |
