| 1 | Novel DPY19L2 variants in globozoospermic patients and the overcoming this male infertility显示文摘Globozoospermia has been reported to be a rare but severe causation of male infertility,which results from the failure of acrosome biogenesis and sperm head shaping.Variants of dpy-19-like 2(DPY19L2)are highly related to globozoospermia,but related investigations have been mainly performed in patients from Western countries.Here,we performed a screening of DPY19L2 variants in a cohort of Chinese globozoospermic patients and found that five of nine patients carried DPY19L2 deletions and the other four patients contained novel DPY19L2 point mutations,as revealed by whole-exome sequencing.Patient 3(P3)contained a heterozygous variant(c.2126+5G>A),P6 contained a homozygous nonsense mutation(c.1720C>T,p.Arg574*),P8 contained compound heterozygous variants(c.H82-1184delATCf p.Leu394_Ser395deIinsPhe;c.368A>T,p.Hisl23Arg),and P9 contained a heterozygous variant(c.H82-1184delATCTTI frameshift).We also reported intracytoplasmic sperm injection(ICSI)outcomes in the related patients,finding that ICSI followed by assisted oocyte activation(AOA)with calcium ionophore achieved high rates of live births.In summary,the infertility of these patients results from DPY19L2 dysfunction and can be treated by ICSI together with AOA. | Yong-Liang Shang Fu-Xi Zhu Jie Yan Liang Chen Wen-Hao Tang Sai Xiao Wei-Ke Mo Zhi-Guo Zhang Xiao-Jin He Jie Qiao Yun-Xia Cao Wei Li | 2019 | Asian Journal of Andrology2019,21,2: | 9 |
| 2 | Respective IL-17A production by γδ T and Th17 cells and its implication in host defense against chlamydial lung infection显示文摘The role of IL-17A is important in protection against lung infection with Chlamydiae,an obligate intracellular bacterial pathogen.In this study,we explored the producers of IL-17A in chlamydial lung infection and specifically tested the role of major IL-17A producers in protective immunity.We found thatγδT cells and Th17 cells are the major producers of IL-17A at the early and later stages of chlamydial infection,respectively.Depletion ofγδT cells in vivo at the early postinfection(p.i.)stage,when mostγδT cells produce IL-17A,failed to alter Th1 responses and bacterial clearance.In contrast,the blockade of IL-17A at the time when IL-17A was mainly produced by Th17(day 7 p.i.)markedly reduced the Th1 response and increased chlamydial growth.The data suggest that theγδT cell is the highest producer of IL-17A in the very early stages of infection,but the protection conferred by IL-17A is mainly mediated by Th17 cells.In addition,we found that depletion ofγδT cells reduced IL-1αproduction by dendritic cells,which was associated with a reduced Th17 response.This finding is helpful to understand the variable role of IL-17A in different infections and to develop preventive and therapeutic approaches against infectious diseases by targeting IL-17A. | Hong Bai Xiaoling Gao Lei Zhao Ying Peng Jie Yang Sai Qiao Huili Zhao Shuhe Wang YiJun Fan Antony George Joyee Zhi Yao Xi Yang | 2017 | Cellular & Molecular Immunology2017,14,10: | 7 |
| 3 | Reduced protein diet with near ideal amino acid profile improves energy efficiency and mitigate heat production associated with lactation in sows显示文摘Background:The study objective was to test the hypothesis that 1)lowering dietary crude protein(CP)increases dietary energetic efficiency and reduces metabolic heat associated with lactation,and 2)excessive dietary leucine(Leu)supplementation in a low CP diet decreases dietary energetic efficiency and increases metabolic heat associated with lactation.Methods:Fifty-four lactating multiparous Yorkshire sows were allotted to 1 of 3 isocaloric diets(10.80 MJ/kg net energy):1)control(CON;18.75%CP),2)reduced CP with a near ideal or optimal AA profile(OPT;13.75%CP)and 3)diet OPT with excessive Leu(OPTLEU;14.25%CP).Sow body weight and backfat were recorded on day 1 and 21 of lactation and piglets were weighed on day 1,4,8,14,18,and 21 of lactation.Energy balance was measured on sows during early(day 4 to 8)and peak(day 14 to18)lactation,and milk was sampled on day 8 and 18.Results:Over 21-day lactation,sows fed OPT lost body weight and body lipid(P<0.05).In peak lactation,sows fed OPT had higher milk energy output(P<0.05)than CON.Sows fed OPTLEU tended(P=0.07)to have less milk energy output than OPT and did not differ from CON.Maternal energy retention was lower(P<0.05)in OPT and OPTLEU compared to CON sows,and did not differ between OPTLEU and OPT sows.Sows fed OPT had higher(P<0.05)apparent energy efficiency for milk production compared to CON.Heat production associated with lactation was lower(P<0.05)or tended to be lower(P=0.082),respectively,in OPT and OPTLEU compared to CON sows.Conclusion:The OPT diet,in peak lactation,improved dietary energy utilization for lactation due to less urinary energy and metabolic heat loss,and triggered dietary energy deposition into milk at the expense of maternal lipid mobilization.Leucine supplementation above requirement may reduce dietary energy utilization for lactation by decreasing the energy partitioning towards milk,partially explaining the effectiveness of OPT diet over CON diets. | Sai Zhang Jay S.Johnson Mu Qiao Nathalie L.Trottier | 2020 | Journal of Animal Science and Biotechnology2020,11,2: | 3 |
| 4 | Analysis of Factors Influencing Insomnia and Construction of a Prediction Model:A Cross-sectional Survey on Rescuers显示文摘Objective To determine the factors influencing insomnia and construct early insomnia warning tools for rescuers to informbest practices for early screening and intervention.Methods Cluster sampling was used to conduct a cross-sectional survey of 1,133 rescuers from one unit in Beijing,China.Logistic regression modeling and R software were used to analyze insomniarelated factors and construct a PRISM model,respectively.Results The positive rate of insomnia among rescuers was 2.74%.Accounting for participants’age,education,systolic pressure,smoking,per capita family monthly income,psychological resilience,and cognitive emotion regulation,logistic regression analysis revealed that,compared with families with an average monthly income less than 3,000 yuan,the odds ratio(OR)values and the[95%confidence interval(CI)]for participants of the following categories were as follows:average monthly family income greater than 5,000 yuan:2.998(1.307–6.879),smoking:4.124(1.954–8.706),and psychological resilience:0.960(0.933–0.988).The ROC curve area of the PRISM model(AUC)=0.7650,specificity=0.7169,and sensitivity=0.7419.Conclusion Insomnia was related to the participants’per capita family monthly income,smoking habits,and psychological resilience on rescue workers.The PRISM model’s good diagnostic value advises its use to screen rescuer early sleep quality.Further,advisable interventions to optimize sleep quality and battle effectiveness include psychological resilience training and smoking cessation. | SAI Xiao Yong CHEN Qiao LUO Ting Gang SUN Yuan Yuan SONG Yu Jian CHEN Juan | 2020 | Biomedical and Environmental Sciences2020,33,7: | 3 |
| 5 | Factors Affecting Patient Satisfaction with Community Health Service under the Gatekeeper System:A Cross-sectional Study in Nanjing,China显示文摘The gatekeeper policy has been implemented for approximately ten years on a pilot population in China. It is necessary to assess the satisfaction of patients utilizing community health service(CHS) under the gatekeeper system. Our study showed that the cognition of gatekeeper policy was associated with four dimensions including doctor‐patient relationships, information and support, organization of care, and accessibility(P < 0.001). One or more factors such as gender and self‐perceived health scores also affected their satisfaction. General practitioners must be prepared to focus on these aspects of information and support, organization of care, and accessibility as indicators of potential opportunities for improvement.
