1Essential oil and aromatic plants as feed additives in non-ruminant nutrition:a review显示文摘This paper summarizes the current knowledge regarding the possible modes of action and nutritional factors involved in the use of essential oils(EOs) for swine and poultry.EOs have recently attracted increased interest as feed additives to be fed to swine and poultry,possibly replacing the use of antibiotic growth promoters which have been prohibited in the European Union since 2006.In general,EOs enhance the production of digestive secretions and nutrient absorption,reduce pathogenic stress in the gut,exert antioxidant properties and reinforce the animal's immune status,which help to explain the enhanced performance observed in swine and poultry.However,the mechanisms involved in causing this growth promotion are far from being elucidated,since data on the complex gut ecosystem,gut function,in vivo oxidative status and immune system are still lacking.In addition,limited information is available regarding the interaction between EOs and feed ingredients or other feed additives(especially pro- or prebiotics and organic acids).This knowledge may help feed formulators to better utilize EOs when they formulate diets for poultry and swine.Zhikai Zeng Sai Zhang Hongliang Wang Xiangshu Piao 2016Journal of Animal Science and Biotechnology2016,7,1:19

3Epstein-Barr virus infection and persistence in nasopharyngeal epithelial cells显示文摘Epstein-Barr virus(EBV) infection is closely associated with undifferentiated nasopharyngeal carcinoma(NPC), strongly implicating a role for EBV in NPC pathogenesis; conversely, EBV infection is rarely detected in normal nasopharyngeal epithelial tissues. In general, EBV does not show a strong tropism for infecting human epithelial cells, and EBV infection in oropharyngeal epithelial cells is believed to be lytic in nature. To establish life-long infection in humans, EBV has evolved efficient strategies to infect B cells and hijack their cellular machinery for latent infection. Lytic EBV infection in oropharyngeal epithelial cells, though an infrequent event, is believed to be a major source of infectious EBV particles for salivary transmission. The biological events associated with nasopharyngeal epithelial cells are only beginning to be understood with the advancement of EBV infection methods and the availability of nasopharyngeal epithelial cell models for EBV infection studies. EBV infection in human epithelial cells is a highly inefficient process compared to that in B cells, which express the complement receptor type 2(CR2) to mediate EBV infection. Although receptor(s) on the epithelial cell surface for EBV infection remain(s) to be identified, EBV infection in epithelial cells could be achieved via the interaction of glycoproteins on the viral envelope with surface integrins on epithelial cells, which might trigger membrane fusion to internalize EBV in cells. Normal nasopharyngeal epithelial cells are not permissive for latent EBV infection, and EBV infection in normal nasopharyngeal epithelial cells usually results in growth arrest. However, genetic alterations in premalignant nasopharyngeal epithelial cells, including p16 deletion and cyclin D1 overexpression, could override the growth inhibitory effect of EBV infection to support stable and latent EBV infection in nasopharyngeal epithelial cells. 4Integrin-linked kinase overexpression promotes epithelial-mesenchymal transition via nuclear factor-κB signaling in colorectal cancer cells显示文摘AIM: To investigate the effect of integrin-linked kinase(ILK) on proliferation, metastasis, and invasion of the colorectal cancer cell line SW480. METHODS: In this study, the colorectal cancer cell line SW480 was stably transfected with ILK plasmids, and small interfering RNA(si RNA) was used to knockdown expression of nuclear factor(NF)-κB/p65. Methylthiazole tetrazolium(MTT) assay was performed to measure proliferation, and the wound healing migration assay and matrigel invasion assay were used to test the metastasis and invasion ability of SW480 cells. To explore the epithelial-mesenchymal transition(EMT) process, embryonic development, and the invasion and metastasis of tumors, the protein level of E-cadherin, vimentin, snail, and slug was detected by western blot. Immunofluorescence was also used to detect E-cadherin expression. Western blot was used to determine the level of phosphorylated-inhibitor of kappa B(IκB)a, inhibitor of gamma B(IγB)a, and nuclear factor kappa B(NF-κB) expressions and toexplore the ILK signaling pathway. RESULTS: Western blot results revealed that ILK expression significantly increased when ILK was overexpressed in SW480 cells(P < 0.05). Proliferation, metastasis, and invasion ability were improved in the vector-ILK group compared to the vector group(P < 0.05). Immunofluorescence results revealed that E-cadherin fluorescence intensity decreased after ILK was overexpressed(P < 0.05). Western blot results revealed that the protein expression of E-cadherin was reduced, while vimentin, snail, and slug were upregulated when ILK was overexpressed in SW480 cells(P < 0.05). In order to determine the role of the NF-κB signaling pathway in ILK overexpression promoted EMT occurrence, we overexpressed ILK in SW480 cells and found that levels of NF-κB/p65 and cytoplasmic phosphorylated-IκBa were increased and that cytoplasmic IкBa levels were decreased compared to the control group(P < 0.05). Furthermore, NF-κB/p65 knockout revealed that E-cadherin was increased in the overexpressed ILK group. 5Molecular architecture of the luminal ring of the Xenopus laevis nuclear pore complex显示文摘The nuclear pore complex(NPC)mediates the flow of substances between the nucleus and cytoplasm in eukaryotic cells.Here we report the cryo-electron tomography(cryo-ET)structure of the luminal ring(LR)of the NPC from Xenopus laevis oocyte.The observed key structural features of the LR are independently confirmed by single-particle cryo-electron microscopy(cryo-EM)analysis.The LR comprises eight butterfly-shaped subunits,each containing two symmetric wings.Each wing consists of four elongated,tubular protomers.Within the LR subunit,the eight protomers form a Finger domain,which directly contacts the fusion between the inner and outer nuclear membranes and a Grid domain,which serves as a rigid base for the Finger domain.Two neighboring LR subunits interact with each other through the lateral edges of their wings to constitute a Bumper domain,which displays two major conformations and appears to cushion neighboring NPCs.Our study reveals previously unknown features of the LR and potentially explains the elastic property of the NPC.Yanqing Zhang Sai Li Chao Zeng Gaoxingyu Huang Xuechen Zhu Qifan Wang Kunpeng Wang Qiang Zhou Chuangye Yan Wusheng Zhang Guangwen Yang Minhao Liu Qinghua Tao Jianlin Lei Yigong Shi 2020Cell Research2020,30,6:4

7Hierarchical Digital Modulation Classification Using Cascaded Convolutional Neural Network显示文摘Automatic modulation classification(AMC)aims to identify the modulation format of the received signals corrupted by the noise,which plays a major role in radio monitoring.In this paper,we propose a novel cascaded convolutional neural network(CasCNN)-based hierarchical digital modulation classification scheme,where M-ary phase shift keying(PSK)and M-ary quadrature amplitude modulation(QAM)modulation formats are considered to be classified.In CasCNN,two-block convolutional neural networks are cascaded.The first block network is utilized to classify the different classes of modulation formats,namely PSK and QAM.The second block is designed to identify the indexes of the modulations in the same PSK or QAM class.Moreover,it is noted that the gird constellation diagram extracted from the received signal is utilized as the inputs to the CasCNN.Extensive simulations demonstrate that CasCNN yields performance gain and performs stronger robustness to frequency offset compared with other recent methods.Specifically,CasCNN achieves 90%classification accuracy at 4 dB signal-to-noise ratio when the symbol length is set as 256.Juanjuan Huang Sai Huang Yuqi Zeng Hao Chen Shuo Chang Yifan Zhang 2021Journal of Communications and Information Networks2021,6,1:1

12The propensity for tumorigenesis in human induced pluripotent stem cells is related with genomic instability显示文摘The discovery of induced pluripotent stem cells (iPSCs) is a promising advancement in the field of regenerative medicine. Previous studies have indicated that the teratoma-forming propensity of iPSCs is variable; however, the relationship between tumorigenic potential and genomic instability in human iPSCs (HiPSCs) remains to be fully elucidated. Here, we evaluated the malignant potential of HiPSCs by using both colony formation assays and tumorigenicity tests. We demonstrated that HiPSCs formed tumorigenic colonies when grown in cancer cell culture medium and produced malignancies in immunodeficient mice. Furthermore, we analyzed genomic instability in HiPSCs using whole-genome copy number variation analysis and determined that the extent of genomic instability was related with both the cells′ propensity to form colonies and their potential for tumorigenesis. These findings indicate a risk for potential malignancy of HiPSCs derived from genomic instability and suggest that quality control tests, including comprehensive tumorigenicity assays and genomic integrity validation, should be rigorously executed before the clinical application of HiPSCs. 13Characterizing RNA Pseudouridylation by Convolutional Neural Networks显示文摘Pseudouridine(Ψ)is the most prevalent post-transcriptional RNA modification and is widespread in small cellular RNAs and m RNAs.However,the functions,mechanisms,and precise distribution ofΨs(especially in m RNAs)still remain largely unclear.The landscape ofΨs across the transcriptome has not yet been fully delineated.Here,we present a highly effective model based on a convolutional neural network(CNN),called Pseudo Uridy Lation Site Estimator(PULSE),to analyze large-scale profiling data ofΨsites and characterize the contextual sequence features of pseudouridylation.PULSE,consisting of two alternatively-stacked convolution and pooling layers followed by a fully-connected neural network,can automatically learn the hidden patterns of pseudouridylation from the local sequence information.Extensive validation tests demonstrated that PULSE can outperform other state-of-the-art prediction methods and achieve high prediction accuracy,thus enabling us to further characterize the transcriptome-wide landscape ofΨsites.We further showed that the prediction results derived from PULSE can provide novel insights into understanding the functional roles of pseudouridylation,such as the regulations of RNA secondary structure,codon usage,translation,and RNA stability,and the connection to single nucleotide variants.The source code and final model for PULSE are available at https://github.com/mlcb-thu/PULSE.Xuan He Sai Zhang Yanqing Zhang Zhixin Lei Tao Jiang Jianyang Zeng 2021Genomics, Proteomics & Bioinformatics2021,19,5:1

163-O-(Z)-coumaroyloleanolic acid overcomes Cks1b-induced chemoresistance in lung cancer by inhibiting Hsp90 and MEK pathways显示文摘OBJECTIVE To investigate 3-O-(Z)-coumaroyloleanolic acid(3-COA),an active ingredient of oleanolic acid in the TAIWANG leaves of E.oldhamii Maxim,that has been shown to have antitumor activity against A549 lung cancer cells,overcomes Cks1b-induced chemoresistance in lung cancer by inhibiting Hsp90 and MEK pathways.METHODS Cisplatin(CDDP)and doxorubicin(DOX)sensitivity was assessed through proliferation,viability,and clonogenic potential induction in cells overexpressing Cks1b(Cks1b-OE).The mechanism for resistance and 3-COA sensitivity were elucidated by immunoblot analysis of Hsp90 and MEK,and confirmed by sh RNA knockdown.Inhibition of 3-COA or3-COA combined with CDDP(3-COA&CDDP)was assayed in early primary lung cancer(IPH),late primary lung cancer(RFH)cells,and tumor-burdened immunodeficiency mice in vivo.RESULTS The ectopic overexpression of Cks1b in human lung cancer cells induces chemoresistance of the cells to CDDP and DOX,but not3-COA,through mechanisms independent of its canonical Skp2-p27 pathway.Further dissection with application of shR NA and selective inhibitors reveals that Hsp90 and MEK1/2 are the critical components of the non-canonical pathways responsible for the Cks1b-induced chemoresistance.Interestingly,inhibition of either Hsp90 or MEK1/2rendered a similar magnitude of antitumor activity by resensitization of the chemoresistant Cks1b-OE cells to CDDP and DOX,suggesting that both Hsp90 and MEK1/2are essentialto Cks1b for induction of chemoresistance.IC50of 3-COA is 6.82μmol·L-1in H358 Cks1b and8.22μmol·L-1in H226 Cks1b,which were not significantly higher than those in H358 EV and H226 EV,respectively.Furthermore,3-COA mimicked PU-H71,a Hsp90-specific inhibitor,targeted Hsp90 and MEK to reduce the expression of their downstream,respectively.Importantly,compared with CDDP treatment,3-COA or 3-COA&CDDP signifi-cantly inhibited RFH cells in vitro.Moreover,3-COA or3-COA&CDDP significantly prolonged more survival for a H358 Cks1b-OE inducing tumor-burdened mice than PD98059(a MEK-specific inhibitor)in vivo.CONCLUSION Our data report for the first time that Cks1b employs Hsp90 and MEK1/2 pathways in lung cancer cells to develop chemoresistance and identify 3-COA as a potential antitumor drug for clinical treatment of chemoresistant lung cancer.WANG He GUO Jia-yi MA Lei DENG Sai LIAO Si-yan SUN Ming-na DU Ling-ran PENG Wei WEI Min-yan LIU Bin CHEN Jing-qi ZENG Bo-hang LI Yue-shan YU Xi-yong FENG Ying-hong YI Zhou 2016中国药理学与毒理学杂志2016,30,10:0

18Effect of Home Blood Pressure Telemonitoring Plus Additional Support on Blood Pressure Control:A Randomized Clinical Trial显示文摘Objective Current clinical evidence on the effects of home blood pressure telemonitoring(HBPT)on improving blood pressure control comes entirely from developed countries.Thus,we performed this randomized controlled trial to evaluate whether HBPT plus support(patient education and clinician remote hypertension management)improves blood pressure control more than usual care(UC)in the Chinese population.Methods This single-center,randomized controlled study was conducted in Beijing,China.Patients aged 30-75 years were eligible for enrolment if they had blood pressure[systolic(SBP)≥140 mmHg and/or diastolic(DBP)≥90 mmHg;or SBP≥130 mmHg and/or DBP≥80 mmHg with diabetes].We recruited 190 patients randomized to either the HBPT or the UC groups for 12 weeks.The primary endpoints were blood pressure reduction and the proportion of patients achieving the target blood pressure.Results Totally,172 patients completed the study,the HBPT plus support group(n=84),and the UC group(n=88).Patients in the plus support group showed a greater reduction in mean ambulatory blood pressure than those in the UC group.The plus support group had a significantly higher proportion of patients who achieved the target blood pressure and maintained a dipper blood pressure pattern at the12th week of follow-up.Additionally,the patients in the plus support group showed lower blood pressure variability and higher drug adherence than those in the UC group.Conclusion HBPT plus additional support results in greater blood pressure reduction,better blood pressure control,a higher proportion of dipper blood pressure patterns,lower blood pressure variability,and higher drug adherence than UC.The development of telemedicine may be the cornerstone of hypertension management in primary care.MENG Wen Wen BAI Yong Yi YAN Li ZHENG Wei ZENG Qiang ZHENG Yan Song ZHA Lin PI Hong Ying SAI Xiao Yong 2023Biomedical and Environmental Sciences2023,36,6:0
19Efficient Strip-Mode SAR Raw-Data Simulator of Extended Scenes Included Moving Targets Based on Reversion of Series显示文摘The Synthetic Aperture Radar(SAR)raw data generator is required to the evaluation of focusing algorithms,moving target analysis,and hardware design.The time-domain SAR simulator can generate the accurate raw data but it needs much time.The frequency-domain simulator not only increases the efficiency but also considers the trajectory deviations of the radar.In addition,the raw signal of the extended scene included static and moving targets can be generated by some frequency-domain simulators.However,the existing simulators concentrate on the raw signal simulation of the static extended scene and moving targets at uniform speed mostly.As for the issue,the two-dimensional signal spectrum of moving targets with constant acceleration can be derived accurately based on the geometric model of a side-looking SAR and reversion of series.And a frequency-domain algorithm for SAR echo signal simulation is presented based on the two-dimensional signal spectrum.The raw data generated with proposed method is verified by several simulation experiments.In addition to reveal the efficiency of the presented frequency-domain SAR scene simulator,the computational complexity of the proposed method is compared with the time-domain approach using the complex multiplication.Numerical results demonstrate that the present method can reduce the computational time significantly without accuracy loss while simulating SAR raw data.Liang Yang Daojian Zeng Jianhua Yan Yaozhang Sai 2020Computers, Materials & Continua2020,,7:0
20Election Procedure for Rural Two Committees显示文摘In order to further strengthen the construction and improvement of villager autonomy system with democratic election,democratic decision making,democratic management,and democratic supervision of rural two committees with Chinese characteristics,experience of work team of Longyang District Agricultural Technology Extension Station dispatched to villager committees( or offices) and their guidance on election of rural two committees in 2007-2016 were used,and exploration was made for election of rural two committees from the third to the sixth committees.
