3HP0953-hypothetical virulence factor overexpresion and localization during Helicobacter pylori infection of gastric epithelium显示文摘BACKGROUND The high prevalence and persistence of Helicobacter pylori(H. pylori) infection, as well as the diversity of pathologies related to it, suggest that the virulence factors used by this microorganism are varied. Moreover, as its proteome contains 340hypothetical proteins, it is important to investigate them to completely understand the mechanisms of its virulence and survival. We have previously reported that the hypothetical protein HP0953 is overexpressed during the first hours of adhesion to inert surfaces, under stress conditions, suggesting its role in the environmental survival of this bacterium and perhaps as a virulence factor.AIM To investigate the expression and localization of HP0953 during adhesion to an inert surface and against gastric(AGS) cells.METHODS Expression analysis was performed for HP0953 during H. pylori adhesion. HP0953 expression at 0,3, 12, 24, and 48 h was evaluated and compared using the Kruskal-Wallis equality-of-populations rank test. Recombinant protein was produced and used to obtain polyclonal antibodies for immunolocalization. Immunogold technique was performed on bacterial sections during adherence to inert surfaces and AGS cells, which was analyzed by transmission electron microscopy. HP0953 protein sequence was analyzed to predict the presence of a signal peptide and transmembrane helices, both provided by the ExPASy platform, and using the GLYCOPP platform for glycosylation sites. Different programs, via, I-TASSER, RaptorX, and HHalign-Kbest, were used to perform three-dimensional modeling.RESULTS HP0953 exhibited its maximum expression at 12 h of infection in gastric epithelium cells.Immunogold technique revealed HP0953 localization in the cytoplasm and accumulation in some peripheral areas of the bacterial body, with greater expression when it is close to AGS cells.Bioinformatics analysis revealed the presence of a signal peptide that interacts with the transmembrane region and then allows the release of the protein to the external environment. The programs also showed a similarity with the Tip-alpha protein of H. pylori. Tip-alpha is an exotoxin that penetrates cells and induces tumor necrosis factor alpha production, and HP0953 could have a similar function as posttranslational modification sites were found;modifications in turn require enzymes located in eukaryotic cells. 4The relationship between transport-to-school habits and physical activity in a sample of New Zealand adolescents显示文摘Objectives:Adolescents using active transport(AT)to school have higher levels of physical activity(PA)compared with motorized transport(MT)users.This study compared school day and weekend day PA in adolescents using AT,MT,or combined AT and MT(AT + MT)to travel to school.Methods:Adolescents(n= 314;age:14.7±1.4 years;32.8% boys)from Dunedin(New Zealand)wore an accelerometer for 7 days and completed a self-reported survey regarding mode of transport to school(73 AT,56 AT + MT,and 185 MT).Data were analyzed using t tests,analysis of variance,and χ2 tests.Results:Although the proportion of adolescents meeting PA guidelines significantly differed among transport groups(AT,47.9%;AT + MT,46.4%;MT,33.5%;p=0.048;overall,39.2%),the observed differences were due mainly to girls.Compared with MT,AT and AT+MT engaged in more moderate-to-vigorous PA(MVPA)per day(AT:61.2 ± 23.2 min;AT+MT:59.6 ± 21.7 min;MT:52.5 ± 19.6 min;p = 0.004;p<0.001,adjusted for gender),per school day and before school.Immediately after school(15:00-16:00),AT engaged in significantly more MVPA compared with AT + MT and MT.No differences in MVPA between the groups were observed in the late afternoon/early evening period during school days or on weekend days.Conclusion:Compared with MT users,adolescent girls using AT or AT + MT accumulated more MVPA during school commute time.AT + MT to school is also a plausible way to increase adolescent girls’ PA when AT only is not feasible.Chiew Ching Kek Enrique García Bengoechea John C.Spence Sandra Mandic 2019Journal of Sport and Health Science2019,8,5:1

12Sarcoma stratification by combined pH2AX and MAP17(PDZK1IP1)levels for a better outcome on doxorubicin plus olaparib treatment显示文摘Sarcomas constitute a rare heterogeneous group of tumors,including a wide variety of histological subtypes.Despite advances in our understanding of the pathophysiology of the disease,first-line sarcoma treatment options are still limited and new treatment approaches are needed.Histone H2AX phosphorylation is a sensitive marker for double strand breaks and has recently emerged as biomarker of DNA damage for new drug development.In this study,we explored the role of H2AX phosphorylation at Ser139 alone or in combination with MAP17 protein,an inducer of DNA damage through ROS increase,as prognostic biomarkers in sarcoma tumors.Next,we proposed doxorubicin and olaparib combination as potential therapeutic strategies against sarcomas displaying high level of both markers.We evaluate retrospectively the levels of pH2AX(Ser139)and MAP17 in a cohort of 69 patients with different sarcoma types and its relationship with clinical and pathological features.We found that the levels of pH2AX and MAP17 were related to clinical features and poor survival.Next,we pursued PARP1 inhibition with olaparib to potentiate the antitumor effect of DNA damaging effect of the DNA damaging agent doxorubicin to achieve an optimal synergy in sarcoma.We demonstrated that the combination of olaparib and doxorubicin was synergistic in vitro,inhibiting cell proliferation and enhancing pH2AX intranuclear accumulation,as a result of DNA damage.The synergism was corroborated in patient-derived xenografts(PDX)where the combination was effective in tumors with high levels of pH2AX and MAP17,suggesting that both biomarkers might potentially identify patients who better benefit from this combined therapy.Marco Perez JoséManuel García-Heredia Blanca Felipe-Abrio Sandra Muñoz-Galván Javier Martín-Broto Amancio Carnero 2020Signal Transduction and Targeted Therapy2020,5,1:1

14Production of Volatile Moth Sex Pheromones in Transgenic Nicotiana benthamiana Plants显示文摘Plant-based bioproduction of insect sex pheromones has been proposed as an innovative strategy to increase the sustainability of pest control in agriculture.Here,we describe the engineering of transgenic plants producing(Z)-11-hexadecenol(Z11-16OH)and(Z)-11-hexadecenyl acetate(Z11-16OAc),two main volatile components in many Lepidoptera sex pheromone blends.We assembled multigene DNA constructs encoding the pheromone biosynthetic pathway and stably transformed them into Nicotiana benthamiana plants.The constructs contained the Amyelois transitella AtrΔ11 desaturase gene,the Helicoverpa armigera fatty acyl reductase HarFAR gene,and the Euonymus alatus diacylglycerol acetyltransferase EaDAct gene in different configurations.All the pheromone-producing plants showed dwarf phenotypes,the severity of which correlated with pheromone levels.All but one of the recovered lines produced high levels of Z11-16OH,but very low levels of Z11-16OAc,probably as a result of recurrent truncations at the level of the EaDAct gene.Only one plant line(SxPv1.2)was recovered that harboured an intact pheromone pathway and which produced moderate levels of Z11-16OAc(11.8μg g^(-1) FW)and high levels of Z11-16OH(111.4μg g^(-1)).Z11-16OAc production was accompanied in SxPv1.2 by a partial recovery of the dwarf phenotype.SxPv1.2 was used to estimate the rates of volatile pheromone release,which resulted in 8.48 ng g-1 FW per day for Z11-16OH and 9.44 ng g^(-1 )FW per day for Z11-16OAc.Our results suggest that pheromone release acts as a limiting factor in pheromone biodispenser strategies and establish a roadmap for biotechnological improvements.Rubén Mateos-Fernández Elena Moreno-Giménez Silvia Gianoglio Alfredo Quijano-Rubio Jose Gavaldá-García Lucía Estellés Alba Rubert JoséLuis Rambla Marta Vazquez-Vilar Estefanía Huet Asunción Fernández-del-Carmen Ana Espinosa-Ruiz Mojca Juteršek Sandra Vacas Ismael Navarro Vicente Navarro-Llopis Jaime Primo Diego Orzáez 2021BioDesign Research2021,,1:0
