1Global-Scale Diversity and Distribution Characteristics of Reef-Associated Symbiodiniaceae via the Cluster-Based Parsimony of Internal Transcribed Spacer 2 Sequences显示文摘Photosynthetic species belonging to Symbiodiniaceae play important ecological roles in coral reef ecosystems. Nine phylogenetically supported Symbiodiniaceae clades(each containing multiple genetically distinct molecular types) have been detected in reef-associated animals and in seawater or sediments near coral reef areas. Although major advances in research on Symbiodiniaceae diversity have been made, information on the global-scale diversity and distribution characteristics of Symbiodiniaceae clades/types is still limited or must be expanded because most studies on Symbiodiniaceae have mainly focused on a limited number of hosts associated with Symbiodiniaceae or coral reef areas. Here, the diversity and distribution characteristics of Symbiodiniaceae data derived from reef-associated animals and seawater or sediments near coral reef areas were analyzed using 3899 qualified Symbiodiniaceae internal transcribed spacer 2(ITS2) sequences retrieved from the GenBank database in May 2017. These sequences were annotated as Symbiodiniaceae ITS2 types via sequence alignment against database of Symbiodiniaceae ITS2 types. Results showed that photosynthetic Symbiodiniaceae ITS2 sequences could be annotated as 119 Symbiodiniaceae ITS2 types within nine clades(A–I), and Symbiodiniaceae ITS2 types mainly inhabited coral reefs between 30?N and 30?S. Symbiodiniaceae ITS2 types could be associated with multiple host taxa. Interestingly, these results implied that the distribution of Symbiodiniaceae ITS2 types was distinct in the Atlantic Ocean, the Red Sea, the Mediterranean Sea, and the Indo-Pacific Ocean. 2Linking coral fluorescence phenotypes to thermal bleaching in the reef-building Galaxea fascicularis from the northern South China Sea显示文摘Coral fluorescence phenotypes have been suggested as an adaptation to a broad range of environmental conditions,yet the mechanisms linking thermal bleaching tolerance in reef-building coral populations,associated with fluorescence phenotypes due to GFP-like proteins,remains unclear.In this study,the relationship between the thermal sensitivity and phenotypic plasticity of corals was investigated using two phenotypes of Galaxea fascicularis,green and brown.The results reveal that brown G.fascicularis was more susceptible to bleaching than green G.fascicularis when exposed to a higher growth temperature of 32℃.Both phenotypes of G.fascicularis were associated with the thermotolerant Symbiodiniaceae symbiont,Durusdinium trenchii.However,the brown G.fascicularis showed a significant decrease in Symbiodiniaceae cell density and a significant increase in pathogenic bacteria abundance when the growth temperature was raised from 29 to 32℃.The physiological traits and transcriptomic profiles of Symbiodiniaceae were not notably affected,but there were differences in the transcriptional levels of certain genes between the two phenotype hosts of G.fascicularis.Under heat stress of 32℃,the gene encoding green fluorescent protein(GFP)-like and chromosome-associated proteins,as well as genes related to oxidative phosphorylation,cell growth and death showed lower transcriptional levels in the brown G.fascicularis compared to the green G.fascicularis.Overall,the results demonstrate that the green form of G.fascicularis is better able to tolerate ocean warming and defend against pathogenic bacteria,likely due to higher gene transcription levels and defense ability.Sanqiang Gong Jiayuan Liang Gang Li Lijia Xu Yehui Tan Xinqing Zheng Xuejie Jin Kefu Yu Xiaomin Xia 2024Marine Life Science & Technology2024,6,1:0
3Chip-based visual detection of micro RNA using DNA-functionalized gold nanoparticles显示文摘In the present study, we developed a highly sensitive and convenient biosensor consisting of gold nanoparticle(Au NP) probes and a gene chip to detect micro RNAs(mi RNAs). Specific oligonucleotides were attached to the glass surface as capture probes for the target mi RNAs, which were then detected via hybridization to the Au NP probes. The signal was amplified via the reduction of HAu Cl4 by H2O2. The use of a single Au NP probe detected 10 pmol L?1 of target mi RNA. The recovery rate for mi R-126 from fetal bovine serum was 81.5%–109.1%. The biosensor detection of mi R-126 in total RNA extracted from lung cancer tissues was consistent with the quantitative PCR(q PCR) results. The use of two Au NP probes further improved the detection sensitivity such that even 1 fmol L?1 of target mi R-125a-5p was detectable. This assay takes less than 1 h to complete and the results can be observed by the naked eye. The platform simultaneously detected lung cancer related mi R-126 and mi R-125a-5p. 4Genotype and Phylogenetic Diversity of Symbiodinium ITS2 Sequences Within Clade C in Three Typical Coral Species from Luhuitou Fringing Reef of the South China Sea显示文摘Dinoflagellates in the genus Symbiodinium, including nine clades(A–I), mainly form mutualistic symbioses with corals. More than 100 Symbiodinium molecular types have been identified by the ITS2-based genotype method within any given clade, and specifically within Symbiodinium clade C. However, the genotype identification method using the ITS2 sequence is likely to lead to high diversity estimates due to the intra-genomic variations in the ITS2 space; thus, further validation is essential for a correct identification. In this study, the molecular diversity of Symbiodinium ITS2 sequences cloned from two stone corals, Acropora sp. SY-01 and Pocillopora sp. SY-05, and one soft coral, Sarcophyton sp. SY-07, living in the northern part of South China Sea(SCS), were analyzed and compared using the ITS2-based genotype identification method, coupled with ITS2-based secondary structural and phylogenetic analyses. As the result, 12 Symbiodinium ITS2 genotypes were identified, while only six and three Symbiodinium ITS2 genotypes were supported by ITS2-based secondary structural and phylogenetic analyses, respectively. In addition, no shared Symbiodinium ITS2 genotypes were observed among the three coral species, suggesting coral species-dependent Symbiodinium genotypes were within clade C. 6A national survey of gross anatomy education in China during the Covid-19 pandemic显示文摘The Covid-19 pandemic launched the full use of online courses in Chinese medical schools.To evaluate the state of gross anatomy education in China during the pandemic,a nationwide survey was conducted through convenience sampling.Total 359 questionnaires were received.The first response from a given school was included to represent that school,thus 77 questionnaires were used for analyses.Schools represented were from all provinces in China's Mainland as well as Hong Kong and Macao.The survey found that before the pandemic,74.0%and 33.8%of the 77 schools conducted online theoretical and practical sessions of gross anatomy,respectively,36 schools had temporarily suspended practical sessions at the time the survey was performed. Body donation programs wereaffected with 26.0% and 27.3% of the 77 schools having suspended donation programs or saw a decreased numberof donations. During the pandemic, 40.3% of the 77 schools kept or initiated the implementation of activelearning, and online assessment was continued in 49.4% of the 77 schools.
