| 3 | Boceprevir or telaprevir in hepatitis C virus chronic infection:The Italian real life experience显示文摘AIM: To check the safety and efficacy of boceprevir/telaprevir with peginterferon/ribavirin for hepatitis C virus(HCV) genotype 1 in the real-world settings. METHODS: This study was a non-randomized, observational, prospective, multicenter. This study involved 47 centers in Italy. A database was prepared for the homogenous collection of the data, was used by all of the centers for data collection, and was updated continuously. All of the patients enrolled in this study were older than 18 years of age and were diagnosed with chronic infection due to HCV genotype 1. The HCV RNA testing was performed using COBAS-Taq Man2.0(Roche, LLQ 25 IU/m L). RESULTS: All consecutively treated patients were included. Forty-seven centers enrolled 834 patients as follows: Male 64%; median age 57(range 18-78), of whom 18.3% were over 65; mean body mass index 25.6(range 16-39); genotype 1b(79.4%); diagnosis of cirrhosis(38.2%); and fibrosis F3/4(71.2%). The following drugs were used: Telaprevir(66.2%) and PEG-IFN-alpha2a(67.6%). Patients were na?ve(24.4%), relapsers(30.5%), partial responders(14.8%) and null responders(30.3%). Overall, adverse events(AEs) occurred in 617 patients(73.9%) during the treatment. Anemia was the most frequent AE(52.9% of cases), especially in cirrhotic. The therapy was stopped for 14.6% of the patients because of adverse events or virological failure(15%). Sustained virological response was achieved in 62.7% of the cases, but was 43.8% in cirrhotic patients over 65 years of age. | 6 | CNTs tuning and vertical alignment in anodic aluminium oxide membrane显示文摘Anodic aluminium oxide (AAOM) membranes were used for template growth of carbon nanotubes (CNT) inside their pores by chemical vapour deposition (CVD) of different hydrocarbons, in the absence of transition metal catalyst. A composite material, containing one nanotube for each channel, having the same length as the membrane thickness and the external diameter close to the diameter of the membrane holes, was obtained. Yield, selectivity, and quality of CNTs in terms of diameter (up to very thin CNT), carbon order, length, arrangement (i.e. number of tubes for each channel), purity, that are critical requisites for several applications were optimized by investigating the effect of changing the hydrocarbon feedstock gas, also in the presence of hydrogen. The samples produced using methane as a feedstock have a well ordered structure. | 7 | Protein kinase CK2: a potential therapeutic target for diverse human diseases显示文摘CK2 is a constitutively active Ser/Thr protein kinase,which phosphorylates hundreds of substrates,controls several signaling pathways,and is implicated in a plethora of human diseases.Its best documented role is in cancer,where it regulates practically all malignant hallmarks.Other well-known functions of CK2 are in human infections;in particular,several viruses exploit host cell CK2 for their life cycle.Very recently,also SARS-CoV-2,the virus responsible for the COVID-19 pandemic,has been found to enhance CK2 activity and to induce the phosphorylation of several CK2 substrates(either viral and host proteins).CK2 is also considered an emerging target for neurological diseases,inflammation and autoimmune disorders,diverse ophthalmic pathologies,diabetes,and obesity.In addition,CK2 activity has been associated with cardiovascular diseases,as cardiac ischemia-reperfusion injury,atherosclerosis,and cardiac hypertrophy.The hypothesis of considering CK2 inhibition for cystic fibrosis therapies has been also entertained for many years.Moreover,psychiatric disorders and syndromes due to CK2 mutations have been recently identified.On these bases,CK2 is emerging as an increasingly attractive target in various fields of human medicine,with the advantage that several very specific and effective inhibitors are already available.Here,we review the literature on CK2 implication in different human pathologies and evaluate its potential as a pharmacological target in the light of the most recent findings. | Christian Borgo Claudio D'Amore Stefania Sarno Mauro Salvi Maria Ruzzene | 2021 | Signal Transduction and Targeted Therapy2021,6,6: | 0 |
