| 2 | Novel frameshift mutation in the SACS gene causing spastic ataxia of charlevoix-saguenay in a consanguineous family from the Arabian Peninsula:A case report and review of literature显示文摘BACKGROUND Familial cases of autosomal recessive spastic ataxia of charlevoix-saguenay have not been reported in the Arabian Peninsula,although the consanguineous marriage rate is very high.We report the first family from the Arabian Peninsula harboring a novel frameshift mutation in the SACS gene.CASE SUMMARY A 33-year-old man presented to our neurology clinic with balance problems and weakness of distal upper and lower limbs.He was previously clinically diagnosed with Friedreich's ataxia.However,the severity of polyneuropathy and the electrodiagnostic studies(EDX)findings are atypical features of Friedreich’s ataxia,and the deterioration was attributed to diabetic neuropathy.Close examination of other family members identified cerebellar ataxia,lower-limb pyramidal signs,peripheral neuropathy,and magnetic resonance imaging findings characterized by pontine linear hypointensities.Genetic testing for Friedreich’s ataxia did not yield a diagnosis.Whole exome sequencing identified a novel frameshift germline mutation in the SACS gene termed c.5824_5827delTACT using the transcript NM_014363.5,which is predicted to cause premature termination of the sacsin protein at amino acid position 1942(p.Tyr1942Metfs*9)and disrupts the sacsin SRR3 and domains downstream from it.The mutation segregated with the disease in the family.CONCLUSION Our data add to the spectrum of mutations in the SACS gene and argues for a need to implement suitably integrated clinical and diagnostic services,including next generation sequencing technology,to better classify ataxia in this area of the world. | Abdullah Al-Ajmi Sarah Shamsah Aleksandar Janicijevic Michayla Williams Fahd Al-Mulla | 2020 | World Journal of Clinical Cases2020,8,8: | 2 |

| 11 | Ensemble Machine Learning Based Identification of Pediatric Epilepsy显示文摘Epilepsy is a type of brain disorder that causes recurrent seizures.It is the second most common neurological disease after Alzheimer’s.The effects of epilepsy in children are serious,since it causes a slower growth rate and a failure to develop certain skills.In the medical field,specialists record brain activity using an Electroencephalogram(EEG)to observe the epileptic seizures.The detection of these seizures is performed by specialists,but the results might not be accurate due to human errors;therefore,automated detection of epileptic pediatric seizures might be the optimal solution.This paper investigates the detection of epileptic seizures by applying supervised machine learning techniques.The techniques applied on the data of patients with ages seven years and below from children’s hospital boston massachusetts institute of technology(CHB-MIT)scalp EEG database of epileptic pediatric signals.A group of Naïve Bayes(NB),Support vector machine(SVM),Logistic regression(LR),k-nearest neighbor(KNN),Linear discernment(LD),Decision tree(DT),and ensemble learning methods were applied to the classification process.The results demonstrated the outperformance of the present study by achieving 100%for all parameters using the Ensemble learning model in contrast to state-of-the-art studies in the literature.Similarly,the SVM model achieved performance with 98.3%for sensitivity,97.7%for specificity,and 98%for accuracy.The results of the LD and LR models reveal the lower performance i.e.,the sensitivity at 66.9%–68.9%,specificity at 73.5%–77.1%,and accuracy at 70.2%–73%. | Shamsah Majed Alotaibi Atta-ur-Rahman Mohammed Imran Basheer Muhammad Adnan Khan | 2021 | Computers, Materials & Continua2021,,7: | 0 |
