| 1 | Seasonal variation of Microcystis in Lake Taihu and its relationships with environmental factors显示文摘In order to monitor the changes of Microcystis along with temporal and spatial variations, seasonal variation of Microcystis in Lake Taihu was investigated by 16S-23S rRNA internal transcribed spacer denaturing gradient gel electrophoresis (16S-23S rRNA-ITS DGGE) and microscopic evaluation. Samples were collected quarterly at four sites (River Mouth, Meiliang Bay, Cross Area, and Lake Center) from August 2006 to April 2007. Results showed that Microcystis dominated total phytoplankton abundance at the four sites in all seasons except winter. The average annual abundance of Microcystis was relatively high at River Mouth and Meiliang Bay, reaching 81.22 × 106 and 61.32 × 106 cells/L, respectively. For temporal variations, Shannon-Wiener diversity index (H ) according to DGGE profile revealed the richness of Microcystis in summer (H = 1.375 ± 0.034) and winter (H = 1.650 ± 0.032) was lower than that in spring (H = 2.078 ± 0.031) and autumn (H = 2.365 ± 0.032) (P < 0.05). While for spatial variations, the richness of Microcystis at River Mouth (H = 2.015 ± 0.074) was higher than at other sites during four seasons (P < 0.01). Very few dierences of Microcystis diversity in the same season were observed among the other three sites (P > 0.05). Canonical correspondence analysis (CCA) was performed to elucidate the relationships between Microcystis operational taxonomic units (OTUs) composition and the environmental factors. Results of CCA revealed that temperature was strongly positively correlated with the first axis (r = 0.963), while TSS was negative correlated with the second axis (r = -0.716). | 2 | Cloning of HBsAg-encoded genes in different vectors and their expression in eukaryotic cells显示文摘AIM: To compare the efficiency of different plasmids as DNA vectors by cloning three HBsAg-encoded genes into two eukaryotic expression vectors, pRc/CMV and pSG5UTPL/Flag, and to express HBsAg S, MS, and LS proteins in SP2/0 cells, and to establish monoclone SP2/0 cell strains that are capable of expressing S or S2S proteins stably.METHODS: Segments of S, preS2-S, preS1-preS2-S genes of Hepatitis B virus were amplified by routine PCR and preS1S fragment was amplified by Over-Lap Extension PCR. The amplified segments were cleaved with restricted endonuclease Hind Ⅲ/Not Ⅰ followed by ligation with pRc/CMV, or BamHI/EcoR Ⅰ followed by ligation with pSG5UTPL/Flag. After the plasmid vectors were cleaved with the correspond enzymes, the amplified segments were inserted into pRc/CMV or pSGSUTPL/Flag plasmid vectors with T4DNA ligase. KOZAK sequence was added before the initial ATG code of each fragment using specific primer. The inserted segments in the recombinant plasmids were sequenced after subcloning. BALB/c mice myeloma cells (SP2/0 cell line) were transfected with the recombinant plasmids. The expressions of the different recombinants were compared by Western-blot, using a monoclonal anti-HBs antibody as the primary antibody and peroxidase-labeled multi-linker as the secondary. Stable SP2/0-pRc/CMV-S or SP2/0- pRc/CMV-MS clones were established through clone screening with G418.RESULTS: Fragments with anticipated size were harvested after PCR. After recombination and screening, the sequences of the inserted segments in the recombinants were confirmed to be S, preS2S, preSl-preS2S and preSlS encoding genes,determined by sequencing. The results of Western-blot hybridization were positive for the anticipated proteins.Among them, pRc/CMV-S or pRc/CMV-MS demonstrated the highest expressing their respective antigen.CONCLUSION: Eight recombinant plasmids expressing S,M, L or preSlS proteins are obtained. For hepatitis surface antigen expression in eukaryotic cells, the vector pRc/CMV is superior to pSG5UTPL/Flag, and pRc/CMV-S and pRc/CMVMS are the most efficient in the pRc/CMV clones. | 3 | Enrichment of heavy metal elements and their adsorption on iron oxides during carbonate rock weathering process显示文摘Analysis of trace elements in the Lower Palaeozoic carbonate rock strata of the Beijing area shows that the Cr, Zn, Cd and Pb contents increase from primary carbonate rocks to weathered carbonate rocks and from primary carbonate rocks to the soil coexisting with carbonate rock. Distributions of Cr, Zn, Cd and Pb have the same trends as that of Fe2O3. Iron oxides play different roles at different stages of weathering of carbonate rock. The adsorption of Cr, Zn, Cd and Pb on Fe2O3 is greater than on FeO in soil. However, FeO, as a main component of clay mineral, is a better adsorbent of Cr, Zn, Cd and Pb than Fe2O3 in weathered and primary carbonate rocks. According to R-type clustering analysis, the main components of carbonate rock are divided into marine and continental sediments. | 4 | Heavy metal content in scalp hair of the inhabitants near Dexing Copper Mine,Jiangxi Province,China显示文摘Levels of seven metals(Mn,Cu,Zn,Pb,Cr,Cd,and Se) in the scalp hair of Daicun and Xiangtian inhabitants were determined by ICP-MS/AFS.Daicun,as an experimental site,is near Dexing Copper Mine,Jiangxi Province,China,and exposed to mining pollution;Xiangtian,as a control site,is free from any mining and other industrial pollution.The heavy metal distribution in the scalp hair of the two diverse population segments exposed to different environments was discussed against the background of the heavy metal content of local plants,waters,and soils.The results show:1) Levels of Cu,Zn,Pb,Cr,and Cd in the scalp hair of Daicun inhabitants were higher compared with Xiangtian counterparts which showed higher comparative levels of Se and Mn.But there were no significant differences of the levels of Cu and Zn in the hair samples between the two villages.In the hair of the inhabitants in the two villages,the levels for Se,Zn,and Cu were lower,while the level for Mn was higher,than the corresponding levels from other regions of the world.Compared with the standard issued by the Trace Element Research Council of China for Chinese inhabitants there were serious Se,Zn,Cu and Cr-deficiency phenomena in the hair samples from Daicun and Xiangtian.2) Against the geological background of high levels of Zn,Cu,Se and Cr,there were deficiencies of Zn,Cu,Se and Cr in the hair samples of the Daicun population.This may have been caused by the distribution forms of the heavy metals in the soils,and antagonism among the elements.3) Principal Component Analysis(PCA) and Cluster Analysis(CA) indicated that the origins of the heavy metals in the hair samples from the two villages had some differences as well as some similarities,this should be contributed mainly to the influence of the mining activities. | NI ShanQin LI RuiPing WANG AnJian | 2011 | Science China Earth Sciences2011,54,5: | 6 |
| 5 | Effectiveness and safety of traditional Chinese herbs in children with cough variant asthma: a systematic review and Meta-analysis显示文摘OBJECTIVE:To systematically investigate the clinical effectiveness and safety of traditional Chinese herbs(TCHs)as an alternative to conventional medicine(CM)in children with cough variant asthma(CVA).METHODS:Randomized controlled trial(RCT)studies that were published from their inceptions to March 31,2020,were identified from the electronic databases of China National Knowledge Infrastructure,Wangfang,Pub Med,and Cochrane Central Library.The primary outcome of the review was the total effective rate(TER),and the secondary outcomes were immunoglobulin E(Ig E),peak expiratory flow(PEF),adverse drug reactions,and relapse rates of interventions.RESULTS:For the Meta-analysis,13 studies involving 992 children with CVA were included.In terms of TER and Ig E,the experimental interventions of TCH,when compared with the control interventions of CM,on pediatric CVA were found to be significantly effective(P<0.0001),whereas for spirometry,PEF was not significantly improved in the TCH group(P=0.48).The incident rates of adverse drug reaction and relapse were found to be significantly lower in the TCH group than those in the CM group(P=0.02 and P<0.0001,respectively).CONCLUSION:Compared with CM therapy,the effects of TCH therapy on pediatric CVA were significantly beneficial in terms of TER and Ig E,but not for PEF,and the methodological quality of included studies was poor.Therefore,the results should be interpreted with caution.More randomized controlled trials with rigorous experimental methodologies are required for objectivity in the future. | Kyou-Hwan Han Ki Haeng Cho CUI Shanqin Lily Lin Jaejong Kim | 2021 | Journal of Traditional Chinese Medicine2021,41,5: | 6 |

| 7 | Development of a reverse transcription quantitative polymerase chain reaction-based assay for broad coverage detection of African and Asian Zika virus lineages显示文摘The Zika virus(ZIKV) is an arbovirus that has spread rapidly worldwide within recent times. There is accumulating evidence that associates ZIKV infections with Guillain-Barré Syndrome(GBS) and microcephaly in humans. The ZIKV is genetically diverse and can be separated into Asian and African lineages. A rapid, sensitive, and specific assay is needed for the detection of ZIKV across various pandemic regions. So far, the available primers and probes do not cover the genetic diversity and geographic distribution of all ZIKV strains. To this end, we have developed a one-step quantitative reverse transcription polymerase chain reaction(qRT-PCR) assay based on conserved sequences in the ZIKV envelope(E) gene. The detection limit of the assay was determined to be five RNA transcript copies and 2.94 × 10^(–3) 50% tissue culture infectious doses(TCID50) of live ZIKV per reaction. The assay was highly specific and able to detect five different ZIKV strains covering the Asian and African lineages without nonspecific amplification, when tested against other flaviviruses. The assay was also successful in testing for ZIKV in clinical samples. | 18 | Migration and enrichment of trace elements of Lower Palaeozoic carbonate rock strata in Beijing显示文摘Analyses of trace elements of the Lower Palaeozoic carbonate rock strata in Beijing show that the contents of As, Hg, F increase from primary carbonate rocks to weathered carbonate rocks and from primary carbonate rocks to the soil coexisting with carbonate rocks, but the distribution regularity of S is not obvious. In the whole weathered stages, the sorption of As is mainly affected by Fe2O3. In soil Fe2O3 is also the main affecting factor of Hg enrichment. The main existing forms of Hg in primary carbonate rocks should simply be physical adsorption, coprecipitation and false isomorphous form between surface of carbonate rock and Hg. In soil the enrichment of F has little relationship with sul-fides and Fe2O3. In primary carbonate rocks, F is mainly absorbed by sulfides and clay minerals, etc. Weathered samples have closer genetic relationships with primary carbonate rocks. This also implies that weathered carbonate rocks have the close existing forms to that of primary carbonate rocks.
