| 4 | Real world effectiveness of PCSK-9 inhibitors combined with statins versus statins-based therapy among patients with very high risk of atherosclerotic cardiovascular disease in China(RWE-PCSK study)显示文摘BACKGROUND The efficacy and safety of proprotein convertase subtilisin/kexin type 9(PCSK-9) inhibitors were confirmed by several clinical trials, but its effectiveness in routine clinical practice in China has not been evaluated. This study aims to describe the real world effectiveness of PCSK-9 inhibitors combined with statins compared with statins-based therapy among patients with very high risk of atherosclerotic cardiovascular disease(ASCVD).METHODS This is a multi-center observational study, enrolled patients from 32 hospitals who underwent percutaneous coronary intervention(PCI) from January to June in 2019. There are 453 patients treated with PCSK-9 inhibitors combined with statins in PCSK-9 inhibitor group and 2,610 patients treated with statins-based lipid lowering therapies in statins-based group. The lipid control rate and incidence of major adverse cardiovascular events(MACE) over six months were compared between two groups.A propensity score-matched(PSM) analysis was used to balance two groups on confounding factors. Survival analysis using Kaplan-Meier methods was applied for MACE.RESULTS In a total of 3,063 patients, 89.91% of patients had received moderate or high-intensity statins-based therapy before PCI, but only 9.47% of patients had low-density lipoprotein cholesterol(LDL-C) levels below 1.4 mmol/L at baseline. In the PSM selected patients, LDL-C level was reduced by 42.57% in PCSK-9 inhibitor group and 30.81%(P < 0.001) in statins-based group after six months. The proportion of LDL-C ≤ 1.0 mmol/L increased from 5.29% to 29.26% in PCSK-9 inhibitor group and 0.23% to 6.11% in statins-based group, and the proportion of LDL-C ≤ 1.4 mmol/L increased from 10.36% to 47.69% in PCSK-9 inhibitor group and 2.99% to 18.43% in statins-based group(P < 0.001 for both). There was no significant difference between PCSK-9 inhibitor and statins-based treatment in reducing the risk of MACE(hazard ratio = 2.52, 95% CI: 0.49-12.97, P = 0.250).CONCLUSIONS In the real world, PCSK-9 inhibitors combined with statins could significantly reduce LDL-C levels among patients with very high risk of ASCVD in China. | 6 | All in one theranostic nanoplatform enables efficient anti-tumor peptide I delivery for triple-modal imaging guided cancer therapy显示文摘Developing a reliable system to efficiently and safely deliver peptide drugs into tumor tissues still remains a great challenge since the instability of peptide drugs and low ability to traverse the cell membrane. Herein, we constructed a multifunctional nanoplatform based on porous europium/gadolinium (Eu/Gd)-doped NaLa(MoO4)2 nanoparticles (NLM NPs) to deliver antitumor peptide of B-cell lymphoma/leukemia-2-like protein 11 (BIM) for cancer therapy. The porous NLM NPs exhibited inherent photoluminescent, magnetic and X-ray absorbable properties, which enable them for triple-modal bioimaging, including fluorescence, magnetic resonance imaging (MRI) and computed tomography (CT). This triple-modal bioimaging can contribute to monitoring NLM NPs biodistribution and guiding therapy in vitro and in vivo. Furthermore, the NLM NPs showed negligible cytotoxicity in vitro and tissue toxicity in vivo. Importantly, NLM NPs could load the antitumor peptide of BIM and efficiently improve the resistance of peptide drugs to proteolysis. The BIM peptide was efficiently delivered into the tumor cells by NLM NPs, which can inhibit the growth and promote the apoptosis of cancer cells in vitro, significantly inhibit the tumor growth in vivo. Notably, NLM-BIM theranostic nanoplatform exhibits low systemic toxicity and fewer side effects in vivo. Results showed that the steel exhibited temperature-dependent cyclic softening characteristic and obvious ratcheting behavior, and dynamic strain aging was observed in the range of 250–350 ℃. Based on experimental observations, a temperature-dependent cyclic plastic constitutive model was proposed by introducing the nonlinear cyclic softening and kinematic hardening rules, and the dynamic strain aging was also considered into the constitutive model. | 12 | A palaeoearthquake event and its age revealed by the travertine layer along the Litang fault in the southeastern margin of the Qinghai-Tibetan plateau显示文摘The Litang fault(LTF),located in the southeast of the Qinghai-Tibetan Plateau,is known for its high level of present-day seismicity,whereas its Pleistocene activity has been scarcely documented.This study focused on a tract of banded travertine deposits precipitated from thermal waters along the NW–SE-trending LTF trace.The role of travertine deposits in recording neotectonic activity has been studied by identifying their internal structure.Typical soft-sediment deformation structures observed within the banded travertines include micro folds,liquefied breccia,and liquefied diapirs.These deformed structures,which are restricted to a single unit separated unconformably by undeformed layers,can be traced for tens of meters,indicating that they were formed by seismic shaking triggered by LTF activity.The deformation of the banded travertine layers is attributed to the combined effects of seismic shaking,liquefaction,and fluidization,and it can be related to a paleo earthquake event with a magnitude of MS>5.The U-series ages obtained from the banded travertine deposits perturbed by the earthquakes are in the range of 130.59–112.94 ka,indicating an important fault-assisted neotectonic activity that occurred during the Middle–Late Pleistocene.Analysis of such structures,in combination with the use of U-series dating methods,can yield a reliable timing of neotectonic activity and provide new evidence for under-standing the seismotectonic setting of the Litang area. | Yao Yang Mingjian Liang Chao Ma Jun Li Hualiang Shen Fang Du Song Luo Shao Liu Xuelian Rui | 2023 | Earthquake Research Advances2023,3,3: | 0 |
| 13 | Effect of subsurface impurity defects on laser damage resistance of beam splitter coatings显示文摘The laser-induced damage threshold(LIDT)of plate laser beam splitter(PLBS)coatings is closely related to the subsurface absorption defects of the substrate.Herein,a two-step deposition temperature method is proposed to understand the effect of substrate subsurface impurity defects on the LIDT of PLBS coatings.Firstly,BK7 substrates are heat-treated at three different temperatures.The surface morphology and subsurface impurity defect distribution of the substrate before and after the heat treatment are compared.Then,a PLBS coating consisting of alternating HfO2–Al2O3 mixture and SiO2 layers is designed to achieve a beam-splitting ratio(transmittance to reflectance,s-polarized light)of approximately 50:50 at 1053 nm and an angle of incidence of 45◦,and it is prepared under four different deposition processes.The experimental and simulation results show that the subsurface impurity defects of the substrate migrate to the surface and accumulate on the surface during the heat treatment,and become absorption defect sources or nodule defect seeds in the coating,reducing the LIDT of the coating.The higher the heat treatment temperature,the more evident the migration and accumulation of impurity defects.A lower deposition temperature(at which the coating can be fully oxidized)helps to improve the LIDT of the PLBS coating.When the deposition temperature is 140◦C,the LIDT(s-polarized light,wavelength:1064 nm,pulse width:9 ns,incident angle:45◦)of the PLBS coating is 26.2 J/cm2,which is approximately 6.7 times that of the PLBS coating deposited at 200◦C.We believe that the investigation into the laser damage mechanism of PLBS coatings will help to improve the LIDT of coatings with partial or high transmittance at laser wavelengths. | Wenyun Du Meiping Zhu Jun Shi Tianbao Liu Jian Sun Kui Yi Jianda Shao | 2023 | High Power Laser Science and Engineering2023,11,5: | 0 |
