2Study of Shaking Table Test on Dynamic Response Characteristics and Failure Mechanism of the Loess Slope显示文摘The natural loess that covers the ground surface has good stability due to its low water content.However,when violent earthquakes occur,the strong dynamic stress generated in the slope may induce landslide disasters with different sizes.In this paper,a large-scale shaking table model test is used to reveal the dynamic response and instability failure process of the loess slope.The test results show that different parts of the slope have different vibration characteristics and the first natural frequency in the model increases with the increase of the slope height.The response acceleration of different parts may change due to the coupling relationship between the spectral characteristics of input wave and the natural frequencies of different parts of slope,suggesting the characteristics of regional differential dynamic response.Under the condition of different dynamic response,stress state and boundary conditions of different parts of slope,a rapid microstructural damage,cumulative residual deformation evolution,and tension-shear coupling instability failure process may appear at the top of the slope with the strong dynamic response associated with the increase of dynamic loading intensity.The Sd values presented in this paper may reflect soil damage and slope instability and failure.PU Xiaowu WANG Lanmin WANG Ping CHAI Shaofeng XU Shiyang 2020Earthquake Research in China2020,34,1:3

5Tamoxifen attenuates reactive astrocyte-induced brain metastasis and drug resistance through the IL-6/STAT3 signaling pathway显示文摘Brain metastasis affects approxim ately 20%-30%of patients w ith triple-negative breast cancers(TNBCs).Even sm all metastatic lesions in the brain can trigg er severe neurological im pairm ents and result in extrem ely short survival time.Recently,active astrocytes were reported to be associated w ith brain metastases.However,how activated astrocytes regulate the behaviors of disseminated breast cancer cells in the brain remains unknown.In this study,human prim ary astrocytes were stim ulated w ith IL-1β to form active astrocytes to study the cross-talk between strom al cells(astrocytes)and TNBC cells in brain metastases.Our results showed that active astrocytes significantly increase the m alignancy of TNBC cells and prevent them from undergoing apoptosis caused by doxorubicin.We also found that the high level of IL-6 secreted by activated astrocytes was responsible for the drug resistance of breast cancer,which could be abolished by treatm ent of astrocytes with tam oxifen(TAM).The blockage of active astrocyte-derived IL-6 by a neutralizing antibody resulted in the attenuation of drug resistance,consequently enhancing the sensitivity of breast cancer cells to doxorubicin.Furthermore,the possible involved TAM-m odulated drug resistance mechanism may be associated with a decrease in IL-6 expression in astrocytes and the downregulation of MAPK and JAK2/STAT3 signaling in cancer cells.Our data suggested that TAMs might reduce drug resistance through the IL-6/JAK2/STAT3 signaling pathway,providing a possible therapy to treat brain metastasis in TNBCs,as estrogen receptor inhibitors(TAMs,etc.)can cross the blood-brain barrier.Yongming Xu Yanrong Zhu Yong Yue Shaofeng Pu Junzhen Wu Yingying Lv Dongping Du 2020Acta Biochimica et Biophysica Sinica2020,52,12:1
6Combined nanosuspensions from two natural active ingredients for cancer therapy with reduced side effects显示文摘Tumor drug resistance and systemic side effects of chemotherapeutic drugs are the main reasons for the failure of cancer treatment.In recent years,it was found that some natural active ingredients can reverse MDR and regulate body immunity to enhance the efficacy and reduce toxicity of chemotherapeutic drugs.In this paper,a new nanosuspensions,HCPT and QUR hybrid nanosuspensions(HQ-NPs),was prepared by the microprecipitation-high pressure homogenization method to reverse tumor drug resistance,reduce toxicity,and increase therapeutic efficacy.The in vitro investigation results showed that HQ-NPs had a unique shape(particle size was about 216.3±5.9 nm),changed crystalline,and different dissolution rates compared with HCPT-NPs and QUR-NPs,which is attributed to the strong intermolecular forces between HCPT and QUR as indicated by the results of the molecule dock.It was verified that the HQ-NPs could double the retention of HCPT in cells and enhance the cytotoxicity to A549/PTX cells in vitro tests compared with HCPT-NPs.We also found that HQ-NPs can significantly enhance the accumulation of HCPT in tumor sites,improve the antitumor activity of HCPT,and protect the immune organs and other normal tissues(P<0.01),compared with HCPT-NPs.Therefore,hybrid nanosuspensions can offer promising potential as the drug delivery system for HCPT and QUR to increase the therapeutic efficacy and reduce the toxicity of HCPT.Yonghui Qiao Zhihao Wei Tingting Qin Rufeng Song Zhiqiang Yu Qi Yuan Juan Du Qingbing Zeng Lanlan Zong Shaofeng Duan Xiaohui Pu 2021Chinese Chemical Letters2021,32,9:1
7Mechanosensitive Ion Channel TMEM63A Gangs Up with Local Macrophages to Modulate Chronic Post-amputation Pain显示文摘Post-amputation pain causes great sufering to amputees,but still no efective drugs are available due to its elusive mechanisms.Our previous clinical studies found that surgical removal or radiofrequency treatment of the neuroma at the axotomized nerve stump efectively relieves the phantom pain aficting patients after amputation.This indicated an essential role of the residual nerve stump in the formation of chronic post-amputation pain(CPAP).However,the molecular mechanism by which the residual nerve stump or neuroma is involved and regulates CPAP is still a mystery.In this study,we found that nociceptors expressed the mechanosensitive ion channel TMEM63A and macrophages infltrated into the dorsal root ganglion(DRG)neurons worked synergistically to promote CPAP.Histology and qRT-PCR showed that TMEM63A was mainly expressed in mechanical pain-producing non-peptidergic nociceptors in the DRG,and the expression of TMEM63A increased signifcantly both in the neuroma from amputated patients and the DRG in a mouse model of tibial nerve transfer(TNT).Behavioral tests showed that the mechanical,heat,and cold sensitivity were not afected in the Tmem63a-/-mice in the naïve state,suggesting the basal pain was not afected.In the infammatory and post-amputation state,the mechanical allodynia but not the heat hyperalgesia or cold allodynia was signifcantly decreased in Tmem63a-/-mice.Further study showed that there was severe neuronal injury and macrophage infltration in the DRG,tibial nerve,residual stump,and the neuromalike structure of the TNT mouse model,Consistent with this,expression of the pro-infammatory cytokines TNFα,IL-6,and IL-1βall increased dramatically in the DRG.Interestingly,the deletion of Tmem63a signifcantly reduced the macrophage infltration in the DRG but not in the tibial nerve stump.Furthermore,the ablation of macrophages signifcantly reduced both the expression of Tmem63a and the mechanical allodynia in the TNT mouse model,indicating an interaction between nociceptors and macrophages,and that these two factors gang up together to regulate the formation of CPAP.This provides a new insight into the mechanisms underlying CPAP and potential drug targets its treatment.Shaofeng Pu Yiyang Wu Fang Tong Wan-Jie Du Shuai Liu Huan Yang Chen Zhang Bin Zhou Ziyue Chen Xiaomeng Zhou Qingjian Han Dongping Du 2023Neuroscience Bulletin2023,39,2:1
8The response law of far-field seismic ground motion of the Wenchuan earthquake and its damaging mechanism in the Loess Plateau显示文摘A series of housing collapses and other serious damage was caused by the 2008 Wenchuan M_(S)8.0 earthquake in the seismic intensity Ⅵ areas of the Loess Plateau, which is hundreds of kilometers away from the epicenter, and which showed a remarkable seismic intensity anomaly. The seismic disasters are closely related to the seismic response characteristics of the site, therefore, the systematic study of the far-field seismic response law of the Wenchuan earthquake in the Loess Plateau is of great significance to prevent the far-field disaster of great earthquake. In this paper, the seismic acceleration records of several bedrock stations and loess stations from the seismogenic fault of the Wenchuan earthquake to the Loess Plateau were collected, and the attenuation law of ground motion along the propagation path and the characteristics of seismic response on the loess site are studied,and the mechanism of amplification effect of ground motion is analyzed based on the dynamic feature parameters of the loess site obtained through the HVSR method. Taking a typical loess site of thick deposit as the prototype, a series of shaking table tests of dynamic response of loess site models with different thicknesses were carried out.Amplification effect, spectral characteristics of acceleration in model sites were analyzed under the action of a farfield seismic wave of the Wenchuan earthquake. The results show that seismic attenuation on the propagation path along the NE strike of the seismogenic fault to the Loess Plateau is slower than that in other directions, and the predominant period range of ground motion on bedrock site of the Loess Plateau presents broadband characteristics. Because the natural periods of loess sites with thick deposits are within the predominant period range of bedrock input wave, loess sites appear significant amplification effect of ground motion, the horizontal acceleration of ground motion exceeds 0.1 g, the seismic intensity reaches 7°. The thicker the loess deposit is, the more significant the change of spectral characteristics of ground motion on loess sites, and the narrower the predominant period range of ground motion becomes, and the closer it is to the natural period of loess sites.Therefore, for some old houses on thick loess sites, the poor seismic performance and strong seismic response eventually led to their collapses and damages because their natural periods are very close to the predominant period of ground motion of the Wenchuan earthquake on thick loess sites;For these damaged high-rise buildings,the resonance effect might be the main reason for their damages because their natural periods are included in the predominant period range of ground motion of the Wenchuan earthquake on thick loess sites.These research results would provide a basis for seismic disasters prediction and evaluation and seismic design of construction engineering in the Loess Plateau.Xiaowu Pu Lanming Wang Ping Wang Xiufeng Tian Shiyang Xu Shaofeng Chai Haitao Guo 2022Earthquake Research Advances2022,2,3:0
9Experimental study on the influence of seismic subsidence of loess under bidirectional loading modes显示文摘Water content,dry density,void ratio,depth of soil layer,and seismic loading may all exert influence on the seismic subsidence of loess.Many scholars have carried out seismic subsidence tests of loess to simulate the stress on soil using unidirectional(axial)vibration instead of bidirectional vibration.We conduct seismic subsidence tests of loess using two different dynamic stress loading methods,unidirectional and bidirectional dynamic stress.In addition,the effects of different dynamic stress loading modes on the development of seismic subsidence of loess are compared for a case-study in northwestern China.The results show that(1)the increasing ratio of radial stress to axial stress has exerted significant influence on the seismic subsidence coefficient of loess under the loading mode of bidirectional dynamic stress(2)there is a critical ratio of radial stress to axial stress for seismic subsidence of loess,ranging from 0.6 to 0.8;when the ratio of radial load to axial load is greater than the critical value,the effect of bidirectional load on the development of seismic subsidence is more remarkable(3)when the ratio of radial load to axial load is smaller,the seismic subsidence of loess calculated by the existing unidirectional stress loading method is safe for engineering projects.However,if the value exceeds the safety ratio range it is dangerous to conduct safety evaluations using the seismic loess subsidence.The prediction value of seismic subsidence at engineering sites directly affects the later foundation treatment and the safety of the overlying structures.The seismic subsidence calculation and evaluation method in this study may provide a scientific basis for safety evaluations of loess sites in northwestern China.Gaofeng Che Ping Wang Qian Wang Xiaowu Pu Jinlian Ma Shaofeng Chai 2021Earthquake Research Advances2021,1,4:0
