1Intrahepatic cholestasis of pregnancy:When should you look further?显示文摘Pruritis with abnormal liver function tests is the classical presentation of intrahepatic cholestasis of pregnancy(ICP),a condition associated with significant fetal complications.Although the etiology of ICP is unclear in many cases,certain features of the clinical presentation should alert the practitioner to the possibility of an underlying metabolic defect, which may not only affect subsequent pregnancies, but may be an indicator of more serious subsequent liver disease.We report a kindred of Anglo-Celtic descent,among whom many members present with ICP,gallstones or cholestasis related to use of oral contraception.Genetic studies revealed a novel mutation in the ABCB4 gene,which codes for a phospholipid transport protein.The clinical significance of this mutation and the importance of identifying such patients are discussed.Winita Hardikar Shivani Kansal Ronald P J Oude Elferink Peter Angus 2009World Journal of Gastroenterology2009,15,9:22
2Sensitivity of Labile Soil Organic Carbon Pools to Long-Term Fertilizer, Straw and Manure Management in Rice-Wheat System显示文摘Labile soil organic carbon(SOC) pools, estimated through chemical fractionation techniques, are considered sensitive indicators of management-induced changes in quality and composition of soil organic matter. Although the impacts of organic manure and crop residue applications on C sequestration in rice-wheat system are fairly well documented, their influence on labile SOC pools is relatively less known. Impacts of organic manure, rice straw, and inorganic fertilizer nitrogen(N) applications on soil total organic carbon(TOC)and SOC pools including water-extractable organic C(WEOC), hot water-soluble organic C(HWOC), potassium permanganateoxidizable organic C(KMnO 4-C), microbial biomass C(MBC), mineralizable organic C(Cmin), and the oxidizable fractions of decreasing oxidizability(easily-oxidizable, oxidizable, and weakly-oxidizable) were investigated in an 11-year field experiment under rice-wheat system. The field experiment included treatments of different combinations of farmyard manure, rice straw, and fertilizer N application rates, with C inputs estimated to be in the range from 23 to 127 Mg ha-1. After 11 years of experiment, WEOC,HWOC, and KMnO 4-C were 0.32%–0.50%, 2.2%–3.3%, and 15.0%–20.6% of TOC, respectively. The easily-oxidizable, oxidizable,and weakly-oxidizable fractions were 43%–57%, 22%–27%, and 10%–19% of TOC, respectively. The applications of farmyard manure and rice straw improved WEOC, HWOC, KMnO 4-C, easily-oxidizable fraction, Cmin, and MBC, though the rates of change varied considerably from-14% to 145% and-11% to 83% of TOC, respectively. At the C input levels between 29 and 78 Mg C ha-1during the 11-year period, the greatest increase was observed in WEOC and the minimum in KMnO 4-C. Water-extractable organic C exhibited a relatively greater sensitivity to management than TOC, suggesting that it may be used as a sensitive indicator of management-induced changes in soil organic matter under rice-wheat system. All the other labile SOC pools exhibited almost the same sensitivity to management as TOC. Most of the SOC pools investigated were positively correlated to each other though their amounts differed considerably. 4Retinal structural-vascular-functional relationship using optical coherence tomography and optical coherence tomography -angiography in myopia显示文摘Background:To examine the retinal structure–vascular-function relationship using optical coherence tomography(OCT)and optical coherence tomography angiography(OCTA)in myopia.Methods:This was a prospective cross-sectional study comprising 86 eyes of 45 individuals with varying axial lengths and spherical equivalents and no posterior segment abnormalities.All eyes underwent optical coherence tomography with the Spectralis SD-OCT and OCTA with RTVue-XR Avanti;Optovue.Individual macular retinal layer thicknesses and flow areas and vessel densities were measured on OCT and OCTA,respectively.Linear correlations were made between the macular layer thicknesses,flow areas and vessel densities with axial length,spherical equivalent and visual acuity.Results:The participants’mean ages were 33.34±14.45 years,mean spherical equivalent refractions were−7.17±5.71 D and axial lengths were 25.95±2.41 mm.There were significant positive correlations of foveal angle(r=0.757,p=0.001),inner retinal(r=0.764,p=0.001)and outer plexiform layer(r=0.771,p=0.001)thickness on OCT and vessel densities in deep capillary plexus(r=0.313,p=0.003)on OCTA with axial length and negative correlations with spherical equivalents and visual acuity.Significant negative correlations of outer nuclear layer(r=−0.560,p=0.03)and photoreceptor outer segment layer thickness(r=−0.856,p<0.001)were noted on OCT with axial length and positive correlations with spherical equivalents and visual acuity.Conclusion:The lateral retinal stretching in myopia could possibly explain the correlation between retinal layer thickness,vascular density and visual acuity in these eyes.Further research is required to investigate this.Ramesh Venkatesh Shivani Sinha Deepika Gangadharaiah Santosh G.K.Gadde Ashwin Mohan Rohit Shetty Naresh Kumar Yadav 2019Eye and Vision2019,6,1:7
5Novel targeting approaches and signaling pathways of colorectal cancer: An insight显示文摘Colorectal cancer(CRC) is the third most common cancer of mortality in the world. Chemotherapy based treatment leads to innumerable side effects as it delivers the anticancer drug to both normal cells besides cancer cells. Sonic Hedgehog(SHH), Wnt wingless-type mouse mammary tumor virus/β-catenin, transforming growth factor-β/SMAD, epidermal growth factor receptor and Notch are the main signaling pathways involved in the progression of CRC. Targeted therapies necessitate information regarding the particular aberrant pathways. Advancements in gene therapies have resulted in the recognition of novel therapeutic targets related with these signal-transduction cascades. CRC is a stepwise process where mutations occur over the time and activation of oncogenes and deactivation of tissue suppressor genes takes place. Genetic changes which are responsible for the induction of carcinogenesis include loss of heterozygosity in tumor suppressor genes such as adenomatous polyposis coli, mutation or deletion of genes like p53 and K-ras. Therefore, many gene-therapy approaches like gene correction, virusdirected enzyme-prodrug therapy, immunogenetic manipulation and virotherapy are currently being explored. Development of novel strategies for the safe and effective delivery of drugs to the cancerous site is the need of the hour. 7Top 100 cited articles in ophthalmic epidemiology between 2006 and 2016显示文摘AIM: To identify the most-cited articles in ophthalmic epidemiology over the last decade.METHODS: We performed a cited reference search on articles that were included in the ISI Web of Science database using the terms 'Epidemi*' AND 'ophthalm*' AND 'population*' during year 2006 to 2016. TOP 100 most cited articles(T100) in ophthalmic epidemiology were short listed and analysed using bibliometrics.RESULTS: These top 100 articles in ophthalmic epidemiology were cited between 61 to 333 times. Of these T100 articles, 36% originated from United States, and 34% were published in the Ophthalmology journal. The three major topics identified were age-related macular degeneration(AMD, n=23), glaucoma(n=16) and visual impairment(n=12). The top-cited article was a study on outdoor activities and its association with the prevalence of myopia in school-aged children, published in 2008. 8Molecular characterization of antimicrobial multi-drug resistance in non-typhoidal Salmonellae from chicken and clam in Mangalore, India显示文摘Salmonella enterica has been documented as one of the leading causes of salmonellosis throughout the world and is most commonly associated with the consumption of contaminated food products. Thus, this research was aimed at studying the antimicrobial susceptibility pattern and detection of quinolone resistance in Salmonella spp isolated from food of animal origin. Thirty-six Salmonella isolates comprising 8 from poultry and 28 from seafood(clams) were identified, serotyped and characterized for their antimicrobial susceptibility against 10 different antibiotics. Plasmid DNA was isolated from all the isolates by alkaline lysis, quinolone resistant non-typhoidal S. Weltevreden were examined for mutation in the DNA gyrase coding gene. Among the 36 Salmonella isolates, 20 were S. weltevreden(8 from poultry and 12 from seafood) and 16 were S. Typhimurium(from seafood). All the isolates showed multiple resistance to nalidixic acid, tetracycline, co-trimoxazole and nitrofurantoin, but, interestingly, the isolates were 100% susceptible to ampicillin, chloramphenicol and gentamicin. Resistant isolates from the study carried the genes responsible for resistance to respective antibiotics. The strain S130 isolated in the study showed single point mutation,Asp87Gly, at position 87 in quinolone resistance determining region. It revealed mutation in quinolone resistance determining region as a cause for quinolone resistance in non-typhoidal Salmonellae. 10Atomic force microscopy correlates antimetastatic potentials of HepG2 cell line with its redox/energy status: effects of curcumin and Khaya senegalensis显示文摘OBJECTIVE: The fatality of cancer is mostly dependent on the possibility of occurrence of metastasis. Thus, if the development of metastasis can be prevented through novel therapeutic strategies targeted against this process, then the success of cancer treatment will drastically increase. In this study, therefore, we evaluated the antimetastatic potentials of an extract of Khaya senegalensis and curcumin on the metastatic liver cell line HepG2, and also assessed the anticancer property of the extract.METHODS: Cells were cultured and treated with graded concentrations of test substances for 24, 48, or 72 h with provisions made for negative controls. Treated cells were assessed as follows: nanotechnologically—atomic force microscopy(AFM) was used to determine cell stiffness; biochemically—cell cytotoxicity, glutathione level and adenosine triphosphate status, caspase activation and mitochondrial toxicity were considered; and microbiologically—a carrot disk assay was used to assess the anticancer property of the extract of K. senegalensis.RESULTS: Curcumin and K. senegalensis increased the cell stiffness by 2.6-and 4.0-fold respectively, indicating their antimetastatic effects. Corresponding changes in redox(glutathione level) and energy(adenosine triphosphate) status of the cells were also demonstrated. Further mechanistic studies indicated that curcumin was not mitotoxic in HepG2 cells unlike the K. senegalensis extract. In addition, the extract potently inhibited the Agrobacterium tumefaciens-induced genetic transformation based on carrot disk assay.CONCLUSION: Cell elasticity measurement data, using AFM, strongly suggested, for the first time, that both curcumin and the extract of K. senegalensis exhibited antimetastatic properties on HepG2 cells.Jeremiah Olorunjuwon Olugbami Robert Damoiseaux Bryan France Michael A. 11Quantum chemical analysis for the formation of glycine in the interstellar medium显示文摘Glycine (C2H5NO2) was the first amino acid to be detected in space by the stardust space probe in Comet Wild2, and is used by living organisms to make proteins. We discuss three different reaction paths for the formation of glycine in interstellar space from some simpler molecules detected in the interstellar medium. The possibility of the formation of glycine in interstellar space is considered by radical-radical and radical-molecule interaction schemes using quantum chemical calculations with density functional theory at the B3LYP/6-31G (d,p) level. 13Vesicular nanocarrier based treatment of skin fungal infections: Potential and emerging trends in nanoscale pharmacotherapy显示文摘Occurrence of skin fungal infections is increasing nowadays and their presence is more prominent in patients suffering from immunocompromised diseases like AIDS. Skin fungal infections are a major cause of visits by patients to dermatology clinics. Although, a large number of antifungal agents are available for treatment of skin fungal infections, but, their toxic profile and physicochemical characteristics reduce therapeutic outcome. When these antifungal agents are delivered topically using conventional formulations like creams and gels, they may cause various side effects like redness, burning, and swelling at the site of application. Therefore, various vesicular formulations(phospholipid based or non phospholipid based) have been explored by pharmaceutical scientists to treat skin fungal infections topically. Vesicular formulation explored for the purpose are liposomes, ethosomes, transfersomes, transethosomes, niosomes, spanlastics, oleic acid vesicles, and nanoparticles.These formulations show various advantages like bioavailability enhancement of bioactives,high skin permeation power, no side effects at application site, dosing frequency reduction,and sustained drug release.
