1再生障碍性贫血患者外周血单核细胞microRNA的差异表达及生物信息学分析显示文摘目的探讨不同分型的再生障碍性贫血(aplastic anemia,AA)患者外周血单核细胞(peripheral blood mononuclear cell,PBMNC)microRNA(miRNA)的差异表达情况。方法采用miRNA表达谱芯片筛查患者PBMNCmiRNA的差异表达谱,用实时定量PCR对差异表达的miRNA进行验证,对候选miRNA进行生物信息学分析。结果与正常对照相比,重型再生障碍性贫血(severe aplastic anemia,SAA)患者表达上调的miRNAs有6个,表达下调的miRNAs有10个;慢性非重型再生障碍性贫血(chronic aplastic anemia,CAA)患者表达上调的miRNAs有24个,表达下调的miRNAs有12个。SAA与CAA相比,表达上调的miRNAs有4个,表达下调的miRNAs有11个。与正常对照相比,sAA、CAA均表达上调的miRNAs有3个,均表达下调的miRNAs有4个。实时定量PCR检测提示,与正常对照相比,CAA、SAA患者miR-155-5p、miR-1260b表达均升高(P〈0.01),CAA患者miR-155-5p、miR-1260b表达均高于SAA患者(P〈0.01)。生物信息学分析提示,miR-155-5p、miR-1260b作用靶基因可参与调控细胞代谢、基因表达转录等生物过程及TNF信号通路、B细胞受体信号通路、FcγR-介导吞噬等信号通路过程。结论CAA和SAA患者具有特征性的miRNA差异表达谱,其中miRNA-155-5p和miRNA-1260b均表达上调。两者具有共同的靶基因ETS1,因此可能存在协同作用,通过抑制ETS1的表达,进而促进Th17的分化,在再生障碍性贫血的发病中具有重要的作用。李峻 陈劼 刘月皎 孙雪梅 朱学军 Shouguo Gao 2018中华医学遗传学杂志2018,35,5:8
2Antiferromagnetic element Mn modified PtCo truncated octahedral nanoparticles with enhanced activity and durability for direct methanol fuel cells显示文摘Pt-based magnetic nano catalysts are one of the most suitable cand idates for electrocatalytic materials due to their high electrochemistry activity and retrievability.Unfortunately,the inferior durability prevents them from being scaled-up,limiting their commercial applications.Herein,an antiferromagnetic element Mn was introduced into PtCo nanostructured alloy to synthesize uniform Mn-PtCo truncated octahedral nanoparticles(TONPs)by one-pot method.Our results show that Mn can tune the blocking temperature of Mn-PtCo TONPs due to its an tiferromag netism.At low temperatures,Mn-PtCo TONPs are ferromag netic,and the coercivity in creases gradually with in creasi ng Mn contents.At room temperature,the Mn-PtCo TONPs display superparamag netic behavior,which is greatly helpful for in dustrial recycling.Mn doping can not only modify the electronic structure of PtCo TONPs but also enhanee electrocatalytic performance for methanol oxidation reaction.The maximum specific activity of Mn-PtCo-3 reaches 8.1 A`m^-2,3.6 times of commercial Pt/C(2.2 A·m^-2)and 1.4 times of PtCo TONPs(5.6 A`m^-2),respectively.The mass activity decreases by only 30%after 2,000 cycles,while it is 45%and 99%(nearly inactive)for PtCo TONPs and commercial Pt/C catalysts,respectively.Qiqi Zhang Jialong Liu Tianyu Xia Jie Qi Haochang Lyu Baoyuan Luo Rongming Wang Yizhong Guo Lihua Wang and Shouguo Wang 2019Nano Research2019,12,10:5
3Gas emission quantity prediction and drainage technology of steeply inclined and extremely thick coal seams显示文摘Gas emissions of workfaces in steeply inclined and extremely thick coal seams differ from those under normal geological conditions, which usually feature a high gas concentration and a large emission quantity. This study took the Wudong coal mine in Xinjiang province of China as a typical case. The gas occurrence of the coal seam and the pressure-relief range of the surrounding rock(coal) were studied by experiments and numerical simulations. Then, a new method to calculate the gas emission quantity for this special geological condition was provided. Based on the calculated quantity, a further gas drainage plan, as well as the evaluation of it with field drainage data, was finally given. 4Suppression of microRNA-155 exerts an anti-inflammatory effect on CD4^+ T cell-mediated inflammatory response in the pathogenesis of atherosclerosis显示文摘In the current study,we aimed to investigate the effects of miR-155 on CD4^+T cell-mediated immune response in the pathogenesis of atherosclerosis.CD34+hematopoietic stem cells,CD4^+T lymphocytes,endothelial cells(ECs),and vascular smooth muscle cells(VSMCs)were harvested from the same donor.Knockdown of miR-155 in the CD4^+T cells was achieved by lentiviral transfection,whereas control RNA-transfected or untransfected lymphocytes were used as controls.The transfected CD4^+T cells were activated by incubating with oxidized low-density lipoproteiretreated dendritic cells.The proliferative capacities,phenotype distribution,and cytokine secretion profiles of the activated CD4^+T cells from different groups were evaluated.The activated lymphocytes were used to treat ECs co-cultivated with VSMCs.The ability of the CD4^+T cells to induce the apoptosis of the ECs and to promote the proliferation of the VSMCs was investigated.Inhibition of miR-155 was found to significantly reduce the proliferation rate of the transfected CD4^+T cells.CD4^+T lymphocytes transfected with the miR-155 inhibitor showed increased populations of T helper type 2 and regulatory T cells,as well as more production of anti-inflammatory cytokines.MiR-155 knockdown was also shown to significantly hamper the ability to CD4^+T cells to induce EC apoptosis and to promote the growth of VSMCs.Our data suggested that inhibition of miR-155 in CD4^+T cells could slow down the formation of atherosclerotic plaques.These results lay the groundwork for future research on the therapeutic potential of miR-155 against atherosclerosisassociated cardiovascular diseases.Jiayu Zheng Wenshuo Wang Tao Hong Shouguo Yang Jinqiang Shen Chen Liu 2020Acta Biochimica et Biophysica Sinica2020,52,6:3

7Preliminary Screening of Plant Ethanol Extracts Inhibiting IBV Proliferation显示文摘[Objective] The paper was to study the inhibition effects of ethanol extracts from different plants on infectious bronchitis virus( IBV). [Method]Certain amount of plant ethanol extract was mixed with the recombinant virus IBV-3ab-luc inosculating luciferase gene at room temperature for 20 min,and then added in H1299 cell culture system together. The activity of luciferase was detected after 24 h,to compare the inhibition effects of ethanol extracts from different plants on the virus. [Result] The logarithms of Gynura segetum leaf,Prunella vulgaris powder,Plantago asiatica leaf,Ophiopogon japonicus root,Lycium barbarum fruit and Citrus reticulata were 4,3,3,2,0 and 0,respectively.
