1Experimental Study on Interaction Between Degrees of Freedom in a Wave Buoy显示文摘Increasing degrees of freedom(DOFs)is a useful way to raise the power capture efficiency of oscillating wave energy converters.Thus,this study proposes a buoy with three DOFs,which are surge,heave,and pitch.The hydrodynamic performance and power capture efficiency of the buoy is physically modeled.Amplitudes of unidirectional and coupled motions are compared to analyze the interaction effect between freedoms under conditions with and without power take-off damping.The capture width ratio and corresponding growth rates are also calculated.Results show that the buoy makes a periodic sinusoidal(or approximate)movement in every DOF.Coupling effect can cause an increase in the amplitude in one DOF and a decrease in the amplitudes of the others.This phenomenon shows that the kinematic energy of the buoy redistributes to all DOFs compared with the unidirectional conditions.Adding DOFs can improve the power absorption of the buoy in most cases,but the number of DOFs is not the more the better.HUANG Shuting SHI Hongda CAO Feifei TAN Junzhe CHENG Haixun LI Demin LIU Shangze GONG Haoxiang TAO Ji 2019Journal of Ocean University of China2019,18,6:5
2A novel selective autophagy receptor,CCDC50,delivers K63 polyubiquitination-activated RIG-I/MDA5 for degradation during viral infection显示文摘Autophagy is a con served process that delivers cytosolic substa nces to the lysosome for degradatio n,but its direct role in the regulation of antiviral inn ate immu nity remains poorly un derstood.Here,through high-throughput screeni ng,we discovered that CCDC50 functions as a previously unknown autophagy receptor that negatively regulates the type I interferon(IFN)signaling pathway initiated by RIG-I-like receptors(RLRs),the sensors for RNA viruses.The expression of CCDC50 is enhanced by viral infection,and CCDC50 specifically recognizes K63-polyubiquitinated RLRs,thus delivering the activated RIG-I/MDA5 for autophagic degradation.The associatio n of CCDC50 with phagophore membrane protei n LC3 is con firmed by crystal structure analysis.In contrast to other known autophagic cargo receptors that associate with either the LIR-docking site(LDS)or the UIMdocking site(UDS)of LC3,CCDC50 can bind to both LDS and UDS,representing a new type of cargo receptor.In mouse models with RNA virus infection,CCDC50 deficiency reduces the autophagic degradation of RIG-I/MDA5 and promotes type I IFN responses,resulting in enhaneed viral resistance and improved survival rates.These results reveal a new link between autophagy and antiviral innate immune resp on ses and provide additional in sights into the regulatory mecha nisms of RLR-mediated antiviral signaling.Panpan Hou Kongxiang Yang Penghui Jia Lan Liu Yuxin Lin Zibo Li Jun Li Shuliang Chen Shuting Guo Ji'An Pan Junyu Wu Hong Peng Weijie Zeng Chunmei Li Yingfang Liu Deyin Guo 2021Cell Research2021,31,1:5
3Physicochemical properties and structure of fluid at nano-/micro-interface:Progress in simulation and experimental study显示文摘In modern chemical engineering processes, the involvement of solid/fluid interface is the most important component of process intensification techniques, such as confined membrane separation and catalysis. In the review, we summarized the research progress of the latest theoretical and experimental works to elucidate the contribution of interface to the fluid properties and structures at nano-and micro-scale. We mainly focused on water, alcohol aqueous solution, and ionic liquids, because they are classical systems in interfacial science and/or widely involved in the industrialization process. Surface-induced fluids were observed in all reviewed systems and played a critical role in physicochemical properties and structures of outside fluid. It can even be regarded as a new interface, when the adsorption layer has a strong interaction with the solid surface. 5stAPAminer:Mining Spatial Patterns of Alternative Polyadenylation for Spatially Resolved Transcriptomic Studies显示文摘Alternative polyadenylation(APA)contributes to transcriptome complexity and gene expression regulation and has been implicated in various cellular processes and diseases.Singlecell RNA sequencing(scRNA-seq)has enabled the profiling of APA at the single-cell level;however,the spatial information of cells is not preserved in scRNA-seq.Alternatively,spatial transcriptomics(ST)technologies provide opportunities to decipher the spatial context of the transcriptomic landscape.Pioneering studies have revealed potential spatially variable genes and/or splice isoforms;however,the pattern of APA usage in spatial contexts remains unappreciated.In this study,we developed a toolkit called stAPAminer for mining spatial patterns of APA from spatially barcoded ST data.APA sites were identified and quantified from the ST data.In particular,an imputation model based on the k-nearest neighbors algorithm was designed to recover APA signals,and then APA genes with spatial patterns of APA usage variation were identified.By analyzing wellestablished ST data of the mouse olfactory bulb(MOB),we presented a detailed view of spatial APA usage across morphological layers of the MOB.We compiled a comprehensive list of genes with spatial APA dynamics and obtained several major spatial expression patterns that represent spatial APA dynamics in different morphological layers.By extending this analysis to two additional replicates of the MOB ST data,we observed that the spatial APA patterns of several genes were reproducible among replicates.stAPAminer employs the power of ST to explore the transcriptional atlas of spatial APA patterns with spatial resolution.This toolkit is available at https://github.com/BMILAB/stAPAminer and https://ngdc.cncb.ac.cn/biocode/tools/BT007320.Guoli Ji Qi Tang Sheng Zhu Junyi Zhu Pengchao Ye Shuting Xia Xiaohui Wu 2023Genomics, Proteomics & Bioinformatics2023,21,3:1
6A Strategy for Searching Antigenic Regions in the SARS-CoV Spike Protein显示文摘In the face of the worldwide threat of severe acute respiratory syndrome (SARS)to human life, some of the most urgent challenges are to develop fast and accurate analytical methods for early diagnosis of this disease as well as to create a safe anti-viral vaccine for prevention. To these ends, we investigated the antigenicity of the spike protein (S protein), a major structural protein in the SARS-coronavirus (SARS-CoV). Based upon the theoretical analysis for hydrophobicity of the S protein, 18 peptides were synthesized. Using Enzyme-Linked Immunosorbent Assay (ELISA), these peptides were screened in the sera from SARS patients. According to these results, two fragments of the S gene were amplified by PCR and cloned into pET-32a. Both S fragments were expressed in the BL-21 strain and further purified with an affinity chromatography. These recombinant S fragments were confirmed to have positive cross-reactions with SARS sera, either by Western blot or by ELISA. Our results demonstrated that the potential epitope regions were located at Codons 469-882 in the S protein, and one epitope site was located at Codons 599-620. 9DOPA-IGF-1 Coated HA/PLGA Microspheres Promoting Proliferation and Osteoclastic Differentiation of Rabbit Bone Mesenchymal Stem Cells显示文摘To explore the ability of dihydroxyphenylalanine-insulin-like growth factor-1 (DOPA-IGF-1) coated hydroxyapatite/poly(lactic-co-glycolic acid)(HA/PLGA) microspheres to promote the proliferation and osteoclastic differentiation of rabbit bone mesenchymal stem cells(rBMSCs), HA/PLGA microspheres with different HA content (10%, 30%, 50%, mass fraction) were prepared by electrospinning method and HA/PLGA microspheres with 50% HA were coated with IGF-1 and DOPA-IGF-1, respectively. They were co-cultured with rBMSCs, respctively. Cell counting kit-8(CCK-8) detection, confocal laser scanning microscopy(CLSM), alkaline phosphatase(ALP) detection and osteogenesis related genes COL IA1, Runx2 and bone morphogenetic protein-2(BMP-2) detection were conducted to detect the proliferation activity, cell morphology, differentiation ability and the expression level of osteogenesis-related genes of cells cultured on all microspheres groups. The results showed that rBMSCs proliferation increased in an HA content dependent manner, and cells proliferated more in the IGF-1 coated and DOPA-IGF-1 coated groups, in particular in DOPA-IGF-1 coated group, and the differences were more remarkable over time (P<0.05). 10Passively-targeted mitochondrial tungsten-based nanodots for efficient acute kidney injury treatment显示文摘Acute kidney injury(AKI)can lead to loss of kidney function and a substantial increase in mortality.The burst of reactive oxygen species(ROS)plays a key role in the pathological progression of AKI.Mitochondrial-targeted antioxidant therapy is very promising because mitochondria are the main source of ROS in AKI.Antioxidant nanodrugs with actively targeted mitochondria have achieved encouraging success in many oxidative stress-induced diseases.However,most strategies to actively target mitochondria make the size of nanodrugs too large to pass through the glomerular system to reach the renal tubules,the main damage site of AKI.Here,an ultra-small Tungsten-based nanodots(TWNDs)with strong ROS scavenging can be very effective for treatment of AKI.TWNDs can reach the tubular site after crossing the glomerular barrier,and enter the mitochondria of the renal tubule without resorting to complex active targeting strategies.To our knowledge,this is the first time that ultra-small negatively charged nanodots can be used to passively target mitochondrial therapy for AKI.Through in-depth study of the therapeutic mechanism,such passive mitochondria-targeted TWNDs are highly effective in protecting mitochondria by reducing mitochondrial ROS and increasing mitophagy.In addition,TWNDs can also reduce the infiltration of inflammatory cells.This work provides a new way to passively target mitochondria for AKI,and give inspiration for the treatment of many major diseases closely related to mitochondria,such as myocardial infarction and cerebral infarction.Qiong Huang Yuqi Yang Tianjiao Zhao Qiaohui Chen Min Liu Shuting Ji Yan Zhu Yunrong Yang Jinping Zhang Haixin Zhao Yayun Nan Kelong Ai 2023Bioactive Materials2023,,3:0
11Few-Atom Copper Catalyst for the Electrochemical Reduction of CO to Acetate:Synergetic Catalysis between Neighboring Cu Atoms显示文摘Single-atom catalysts(SACs)are gaining increasing recognition because of their superior catalytic properties for various reactions.However,the performance of SACs is often limited by the lack of neighboring metal centers to cooperate in catalysis.Herein,a synergetic interaction between neighboring Cu atoms of a few-atom catalyst(FAC)on graphdiyne(GDY)is found to greatly enhance the production of acetate in the CO electroreduction reaction relative to Cu SACs.In a 1.0 M KOH electrolyte,this Cu FAC exhibits an acetate Faradaic efficiency of 53.8±1.5%,an ultrahigh relative purity of up to 97 wt%for liquid products,and excellent stability over 23 h continuous electrolysis at–0.8 V versus reversible hydrogen electrode.Theoretical studies suggest that the intersite catalytic communication between two neighboring metal atoms confined in each pore of GDY facilitates the formation of acetic acid through either stepwise hydrogenation of CH_(2)CO^(*)or the direct reaction of H_(2)Owith CH_(2)CO^(*).Our study demonstrates the unprecedented synergetic catalysis of Cu FAC in promoting the selective CO electroreduction toward acetate production.Weifeng Rong Haiyuan Zou Sha Tan Enyuan Hu Fan Li Chao Tang Hao Dai Shuting Wei Yongfei Ji Lele Duan 2023CCS Chemistry2023,5,5:0
12Polyoxometalate-Based Porous Metal-Organic Gel for High-Efficiency Ultrasound-Assisted Oxidative Desulfurization of Model Fuel显示文摘A novel heterogeneous catalyst for ultrasound-assisted oxidative desulfurization(UAODS)was designed and synthesized by confining polyoxometalates(POMs)clusters within porous metal-organic gel.The catalyst microstructures were characterized by XRD,SEM-EDX,FT-IR,N_(2)adsorption-desorption,and XPS.Effects of POM loading,catalyst dosage,and ultrasonic power were also investigated.Results indicate that 60%-PMA@MOX(Al)exhibits optimal catalytic activity,with a sulfur removal rate of 98.61%and excellent reusability.Moreover,a kinetic study of different desulfurization approaches demonstrates that the unique thermal and cavitation effects of ultrasound can effectively improve the efficiency of oxidative desulfurization.Ji Haifeng Liu Shuting Shi Hongfei Wang Weidong 2022China Petroleum Processing & Petrochemical Technology2022,24,3:0
