Bevacizumab biosimilar LY01008 compared with bevacizumab(Avastin)as first-line treatment for Chinese patients with unresectable, metastatic,or recurrent non-squamous non-small-cell lung cancer:A multicenter,randomized,double-blinded,phase Ⅲ trial显示文摘Background:Previous studies have demonstrated the preclinical pharmacological and toxicological consistency, and clinical pharmacokinetic equivalence of bevacizumab biosimilar LY01008 with reference bevacizumab(Avastin). This randomized controlled trial aimed to compare the efficacy and safety of LY01008 with Avastin in first-line treatment of Chinese patients with advanced or recurrent non-squamous non-small cell lung cancer(NSCLC).Methods:StageⅢB-ⅣNSCLC patients with evaluable lesions, good physical status, and adequate organ functions from 67 centers across China were randomized in a ratio of 1:1 to receive LY01008 or Avastin 15 mg/kg intravenously in combination with paclitaxel/carboplatin(combined treatment)for 4-6 cycles, followed by maintenance monotherapy with LY01008 until disease progression, intolerable toxicity,or death. The primary endpoint was objective response rate(ORR)in accordance with Response Evaluation Criteria in Solid Tumors(RECIST)version 1.1 confirmed by independent radiological review committees(IRRC).Secondary endpoints included disease control rate(DCR),duration of response(DoR),progression-free survival(PFS),overall survival(OS),and safety. This study was registered in Clinical Trials.gov(NCT03533127).Results:Between December 15^(th),2017,and May 15^(th),2019,a total of 649 patients were randomized to the LY01008(n=324)or Avastin(n=325)group.As of September 25th,2019 for primary endpoint analysis,589 patients received ORR evaluation, with a median number of combined treatment cycles of 5(range 1-6)andmedian duration of treatment of 3.0(range 0.0-5.1)months.ORRof responseevaluable patients in the LY01008 and Avastin groups were 48.5% and 53.0%,respectively.The stratified ORR ratio was 0.91(90%CI 0.80-1.04,within the prespecified equivalence margin of 0.75-1.33).Up to May 15^(th),2020,with a median follow-up of 13.6(range 0.8-28.4)months,no notable differences in DCR,median DoR,median PFS,median OS,and 1-year OS rate were observed between the LY01008 and Avastin groups. There were no clinically meaningful differences in safety and immunogenicity across treatment groups.Conclusions:LY01008 demonstrated similarity to Avastin in terms of efficacy and safety in Chinese patients with advanced or recurrent non-squamous NSCLC.LY01008 combined with paclitaxel/carboplatin is expected to become a new treatment option for unresectable,metastatic,LY01008 and Avastin groups. There were no clinically meaningful differences in safety and immunogenicity across treatment groups.Conclusions:LY01008 demonstrated similarity to Avastin in terms of efficacy and safety in Chinese patients with advanced or recurrent non-squamous NSCLC.LY01008 combined with paclitaxel/carboplatin is expected to become a new treatment option for unresectable, metastatic,or recurrent non-squamous NSCLC patients in the first-line setting. | Yuankai Shi Kaijian Lei Yuming Jia Bingqiang Ni Zhiyong He Minghong Bi Xicheng Wang Jianhua Shi Ming Zhou Qian Sun Guolei Wang Dongji Chen Yongqian Shu Lianke Liu Zhongliang Guo Yong Liu Junquan Yang Ke Wang Ke Xiao LinWu Tienan Yi Debin Sun Mafei Kang Tianjiang Ma Yimin Mao Jinsheng Shi Tiegang Tang Yan Wang Puyuan Xing Dongqing Lv Wangjun Liao Zhiguo Luo Bin Wang Xiaohong Wu Xiaoli Zhu Shuhua Han Qisen Guo Rongyu Liu Zhiwei Lu Jianyong Zhang Jian Fang Changlu Hu Yinghua Ji Guolong Liu Hong Lu Dedong Wu Junhong Zhang Shuyang Zhu Zheng Liu Wensheng Qiu Feng Ye Yan Yu Yanqiu Zhao Qinhong Zheng Jun Chen Zhanyu Pan Yiping Zhang Wenjuan Lian Bo Jiang Bo Qiu Guojun Zhang Hua Zhang Yanju Chen Yuan Chen Hongbing Duan Manxiang Li Shengming Liu Lijun Ma Hongming Pan Xia Yuan Xueli Yuan Yulong Zheng Emei Gao Li Zhao Shumin Wang Can Wu | 2021 | Cancer Communications2021,41,9: | 5
COVID-19-associated monocytic encephalitis(CAME):histological and proteomic evidence from autopsy显示文摘Severe neurological symptoms are associated with Coronavirus disease 2019(COVID-19).However,the morphologic features, pathological nature and their potential mechanisms in patient brains have not been revealed despite evidence of neurotropic infection. In this study, neuropathological damages and infiltrating inflammatory cells were quantitatively evaluated by immunohistochemical staining, ultrastructural examination under electron microscopy, and an image threshold method,in postmortem brains from nine critically ill COVID-19 patients and nine age-matched cadavers of healthy individuals.Differentially expressed proteins were identified by quantitative proteomic assays.Histopathological findings included neurophagocytosis, microglia nodules, satellite phenomena, extensive edema, focal hemorrhage, and infarction,as well as infiltrating mononuclear cells.Immunostaining of COVID-19 brains revealed extensive activation of both microglia and astrocytes, severe damage of the blood-brain barrier(BBB)and various degrees of perivascular infiltration by predominantly CD14+/CD16+/CD141+/CCR7+/CD11c+monocytes and occasionally CD4+/CD8+T lymphocytes.Quantitative proteomic assays combined with bioinformatics analysis identified upregulated proteins predominantly involved in immune responses, autophagy and cellular metabolism in COVID-19 patient brains compared with control brains.Proteins involved in brain development, neuroprotection, and extracellular matrix proteins of the basement membrane were downregulated, potentially caused by the activation of transforming growth factorβreceptor and vascular endothelial growth factor signaling pathways. Thus, our results define histopathological and molecular profiles of COVID-19-associated monocytic encephalitis(CAME)and suggest potential therapeutic targets. | Pei-Pei Zhang Zhi-Cheng He Xiao-Hong Yao Rui Tang Jie Ma Tao Luo Chuhong Zhu Tian-Ran Li Xindong Liu Dingyu Zhang Shuyang Zhang Yi-Fang Ping Ling Leng Xiu-Wu Bian | 2023 | Signal Transduction and Targeted Therapy2023,8,2: | 3
Influence of Spatial-Temporal and Environmental Factors on Larimichthys polyactis, Octopus variabilis, and Species Aggregated Set-Net CPUEs in Haizhou Bay, China显示文摘Set-nets are common alongshore fishing gear used in Haizhou Bay, which rely on flow to catch fish. The catch per unit effort(CPUE) of set-net is affected by spatial-temporal and environmental factors but no research has been conducted on this subject. In this study, we used generalized additive models(GAMs) to explore the influence of spatial-temporal and environmental factors on CPUEs of species aggregated, small yellow croaker(Larimichthys polyactis), and octopus(Octopus variabilis) based on logbooks investigations conducted at 4 stations in an alongshore area of Haizhou Bay from 2011 to 2012. The results showed that all CPUEs exhibited significant spatial-temporal differences at various scales. Aggregated CPUE was high when the sea surface temperature(SST) was 15-18℃ and 20-23℃, which was mainly determined by life history traits of the octopus and small yellow croaker(optimal SSTs 14-17℃ and 19-24℃, respectively). Chlorophyll-a concentration had significant influences on the aggregated, small yellow croaker and octopus CPUEs at optimal ranges of 3.8-6.2 mg m^(-3), 4.2-4.8 mg m^(-3) and 4.5-5.5 mg m^(-3), respectively. Flow through the net had positive relationships with CPUEs. The approximate logarithmic trends in regression curves had a critical point of 2.5 Mm^3 d^(-1), which was the dividing point that differentiated whether the major factor affecting CPUEs was the flow velocity or the fishery resource. Our results from this study will help guide fishery production and improve catch rate of set-net fishing in Haizhou Bay. | TANG Yanli MA Shuyang LIU Changdong WANG Xinmeng CHENG Shasha | 2018 | Journal of Ocean University of China2018,17,4: | 2
Incorporating mesopelagic fish into the evaluation of conservation areas for marine living resources under climate change scenarios显示文摘Mesopelagic fish(meso-fish)are central species within the Southern Ocean(SO).However, their ecosystem role and adaptive capacity to climate change are rarely integrated into protected areas assessments. This is a pity given their importance as crucial prey and predators in food webs, coupled with the impacts of climate change. Here,we estimate the habitat distribution of nine meso-fish using an ensemble model approach(MAXENT,random forest, and boosted regression tree).Four climate model simulations were used to project their distribution under two representative concentration pathways(RCP4.5 and RCP8.5)for short-term(2006–2055)and long-term(2050–2099)periods.In addition,we assess the ecological representativeness of protected areas under climate change scenarios using meso-fish as indicator species. Our models show that all species shift poleward in the future.Lanternfishes(family Myctophidae)are predicted to migrate poleward more than other families(Paralepididae,Nototheniidae,Bathylagidae, and Gonostomatidae). In comparison, lanternfishes were projected to increase habitat area in the eastern SO but lose area in the western SO;the opposite was projected for species in other families.Important areas(IAs)of meso-fish are mainly distributed near the Antarctic Peninsula and East Antarctica.Negotiated protected area cover 23% of IAs at present and 38%of IAs in the future(RCP8.5,long-term future). Many IAs of meso-fish still need to be included in protected areas, such as the Prydz Bay and the seas around the Antarctic Peninsula. Our results provide a framework for evaluating protected areas incorporating climate change adaptation strategies for protected areas management. | Shuhao Liu Yang Liu Katharina Teschke Mark AHindell Rachel Downey Briannyn Woods Bin Kang Shuyang Ma Chi Zhang Jianchao Li Zhenjiang Ye Peng Sun Jianfeng He Yongjun Tian | 2024 | Marine Life Science & Technology2024,6,1: | 0

Effects of oxygen vacancy on bond ionicity, lattice energy, and microwave dielectric properties of CeO_(2) ceramics with Yb^(3+)substitution显示文摘Novel Yb_(x)Ce_(1-x)O_(2-0.5x)(x=0-0.8)ceramics,designed by replacing Ce^(4+)with Yb^(3+)ions were prepared by conventional oxide reaction, and the structural stability of the cubic fluorite structure was assessed using lattice energy and ionic properties of Ce/Yb-O bonds. The oxygen vacancy caused by unequal substitution, which played a decisive role in bond ionicity and lattice energy, was analyzed experimentally by XPS and also theoretically by first principles. The Yb_(x)Ce_(1-x)O_(2-0.5x) ceramics maintain a stable cubic fluorite structure when x≤0.47,corresponding to the minimum lattice energy of 4142 kJ/mol with the lowest ionicity as ƒ_(i)=87.57%.For microwave dielectric properties, when the Yb_(x)Ce_(1-x)O_(2-0.5x)(x=0-0.4)ceramics are pure phase, the porosity-corrected permittivity is dependent on the bond ionicity. The Q׃ values are related to the lattice energy and grain distribution. The temperature coefficient of resonance frequency has been analyzed using bond valence. When the Yb_(x)Ce_(1-x)O_(2-0.5x)(x=0.5-0.8)ceramics are multiple phases, the microwave dielectric properties are associated with the phase composition and grain growth. | Mingan Shi Shuyang Ma Wangsuo Xia Ying Wang Jinzi Yu Haitao Wu | 2024 | Journal of Advanced Ceramics2024,13,2: | 0
