| 3 | Interleukin-33 drives hepatic fibrosis through activation of hepatic stellate cells显示文摘Liver fibrosis is a consequence of chronic liver disease,causing morbidity and mortality.Interleukin-33(IL-33)is a critical mediator of inflammation,which may be involved in the development of liver fibrosis.Here,we investigated the role of IL-33 in human patients and experimental bile-duct ligation(BDL)-induced fibrosis in mice.We report increased hepatic IL-33 expression in the murine BDL model of fibrosis and in surgical samples obtained from patients with liver fibrosis.Liver injury,inflammatory cell infiltration and fibrosis were reduced in the absence of the IL-33/ST2 receptor,and the activation of hepatic stellate cells(HSCs)was decreased in ST2-deficient mice.Recombinant IL-33 activated HSCs isolated from C57BL/6 mice,leading to the expression of IL-6,TGF-β,α-SMA and collagen,which was abrogated in the absence of ST2 or by pharmacological inhibition of MAPK signaling.Finally,administration of recombinant IL-33 significantly increased hepatic inflammation in sham-operated BL6 mice but did not enhance BDL-induced hepatic inflammation and fibrosis.In conclusion,BDL-induced liver inflammation and fibrosis are dependent on ST2 signaling in HSCs,and therefore,the IL-33/ST2 pathway may be a potential therapeutic target in human patients with chronic hepatitis and liver fibrosis. | Zhongming Tan Qianghui Liu Runqiu Jiang Long Lv Siamak S Shoto Isabelle Maillet Valerie Quesniaux Junwei Tang Wenjie Zhang Beicheng Sun Bernhard Ryffel | 2018 | Cellular & Molecular Immunology2018,15,4: | 24 |
| 4 | Banana Fusarium Wilt(Fusarium oxysporum f. sp. cubense)Control and Resistance, in the Context of Developing Wilt-resistant Bananas Within Sustainable Production Systems显示文摘Banana(Musa spp.) is seriously threatened by the soil-borne fungus Fusarium oxysporum f. sp. cubense(Foc), also known as Panama disease.Attempts to control Fusarium wilt with fungicides damage soil health and have limited efficiency due to pathogenic variability. Elucidating the mechanism of infection and molecular basis of host defense through banana genome sequencing, genome editing and proteomic profile analysis will help formulate strategies to develop resistant cultivars. This will include research to better understand the functions of Fusarium wilt-resistance proteins. Transgenic approaches and protoplast fusion could be employed as tools for transferring resistance genes from wild relatives to commercial banana varieties, and may serve as a new strategy in solving the problems faced by banana breeding programmes.Evaluation of banana germplasm for resistance to Fusarium wilt using in vitro mutation and selection, along with somaclonal variation and somatic hybridization, could improve banana breeding efficiency for resistance against Foc. Plant hormones could also play an important role in regulating plant growth and defense by mediating developmental processes and signaling networks involved in banana responses to Foc. A complementary approach for managing Fusarium wilt, such as exclusion, surveillance and biological control as important components of integrated disease management programs must be considered to prevent and contain contagion. This includes studies on banana plant-microbe interactions, embracing both plant growth promoting rhizobacteria(PGPR) to induce Foc resistance, and exploring Foc-derived elicitors for inducing defense-related enzymes in bananas. The role of Silicon and crop and livestock integration must also be included in the Fusarium control toolbox. | 5 | Effect of Silicon on Growth and Development of Strawberry under Water Deficit Conditions显示文摘Water stress is a major factor that limits agricultural crop production. Silicon(Si) is generally considered as a beneficial element for the growth of higher plants, especially for those grown under stressful environment. This study was conducted to examine the effects of Si on growth and development of strawberry(Fragaria × ananassa ‘Camarosa') under water stress conditions. A factorial experiment, in a completely randomized design, was used to investigate the effects of three irrigation levels and four Si treatments consisting of 0, 5, 10, and 15 mmol·L^(-1) potassium silicate(K_2 SiO_3). The results showed that an increase in the levels of water stress caused a decrease in most of the quantitative characteristics such as specific leaf area, chlorophyll fluorescence, net photosynthesis rate, and stomata conductance; addition of Si significantly increased most of the mentioned factors; water stress increased electrolyte leakage, proline, and water use efficiency(WUE); Si treatment significantly decreased transpiration rate and improved chlorophyll content and WUE. Water stress stimulated mineral nutrient absorption whereas Si application decreased it under water stress. In conclusion, it was found that in most of the investigated factors, 10 mmol·L^(-1) potassium silicate had the best effect on growth and development of strawberry. Furthermore, TLRs are also able to recognize endogenous alarmins released by damaged tissue and necrosis and/or apoptotic cells and are present in numerous autoimmune diseases. Therefore, the release of endogenous TLR ligands plays an important role in initiating and driving inflammatory diseases. Increasing data suggest a role for TLR signaling in rheumatoid arthritis, which is an autoimmune disease. | 10 | Studying thin film damping in a micro-beam resonator based on non-classical theories显示文摘In this paper, a mathematical model is presented for studying thin film damping of the surrounding fluid in an in-plane oscillating micro-beam resonator. The proposed model for this study is made up of a clamped-clamped micro-beam bound between two fixed layers. The microgap between the micro-beam and fixed layers is filled with air. As classical theories are not properly capable of predicting the size dependence behaviors of the micro-beam,and also behavior of micro-scale fluid media, hence in the presented model, equation of motion governing longitudinal displacement of the micro-beam has been extracted based on non-local elasticity theory. Furthermore, the fluid field has been modeled based on micro-polar theory. These coupled equations have been simplified using Newton-Laplace and continuity equations. After transforming to non-dimensional form and linearizing, the equations have been discretized and solved simultaneously using a Galerkin-based reduced order model. Considering slip boundary conditions and applying a complex frequency approach, the equivalent damping ratio and quality factor of the micro-beam resonator have been obtained. The obtained values for the quality factor have been compared to those based on classical theories. We have shown that applying non-classical theories underestimate the values of the quality factor obtained based on classical theo-ries. | 14 | Hospital readmissions in decompensated cirrhotics: Factors pointing toward a prevention strategy显示文摘AIM To reduce readmissions and improve patient outcomes in cirrhotic patients through better understanding of readmission predictors.METHODS We performed a single-center retrospective study of patients admitted with decompensated cirrhosis from January 1, 2011 to December 31, 2013(n = 222). Primary outcomes were time to first readmission and 30-d readmission rate due to complications of cirrhosis. Clinical and demographic data were collected to help describe predictors of readmission, along with care coordination measures such as post-discharge status and outpatient follow-up. Univariate and multivariateanalyses were performed to describe variables associated with readmission.RESULTS One hundred thirty-two patients(59.4%) were readmitted at least once during the study period. Median time to first and second readmissions were 54 and 93 d, respectively. Thirty and 90-d readmission rates were 20.7 and 30.1 percent, respectively. Predictors of 30-d readmission included education level, hepatic encephalopathy at index, ALT more than upper normal limit and Medicare coverage. There were no statistically significant differences in readmission rates when stratified by discharge disposition, outpatient follow-up provider or time to first outpatient visit.CONCLUSION Readmissions are challenging aspect of care for cirrhotic patients and risk continues beyond 30 d. | 18 | Simulation of proppant transport at intersection of hydraulic fracture and natural fracture of wellbores using CFD-DEM显示文摘Proppants transport is an advanced technique to improve the hydraulic fracture phenomenon,in order to promote the versatility of gas/oil reservoirs.A numerical simulation of proppants transport at both hydraulic fracture(HF)and natural fracture(NF)intersection is performed to provide a better understand-ing of key factors which cause,or contribute to proppants transport in HF-NF intersection.Computational fluid dynamics(CFD)in association with discrete element method(DEM)is used to model the complex interactions between proppant particles,host fluid medium and fractured walls.The effect of non-spherical geometry of particles is considered in this model,using the multi-sphere method.All interaction forces between fluid flow and particles are considered in the computational model.Moreover,the inter-actions of particle-particle and particle-wall are taken into account via Hertz-Mindlin model.The results of the CFD-DEM simulations are compared to the experimental data.It is found that the CFD-DEM sim-ulation is capable of predicting proppant transport and deposition quality at intersections which are in agreement with experimental data.The results indicate that the HF-NF intersection type,fluid velocity and NF aperture affect the quality of blockage occurrence,presenting a new index,called the blockage coefficient which indicates the severity of the blockage. | Siamak Akhshik Majid Rajabi | 2022 | Particuology2022,20,4: | 2 |
