3Safety and efficacy of Yupingfeng granules in children with recurrent respiratory tract infection:A randomized clinical trial显示文摘Importance:Recurrent respiratory tract infection(RRTI)is common in children.Inappropriate RRTI treatment will lead to asthma and other diseases,thereby seriously affecting the growth and physical health of children.Immune function modulation can prevent and alleviate childhood RRTI.Yupingfeng(YPF),a patented traditional Chinese medicine(TCM),has immunomodulatory effects and is widely used in China to treat children with RRTI.Objective:To evaluate the safety and efficacy of YPF monotherapy in treating children with RRTI.Methods:This multicenter,randomized,double-blind,double-simulation,noninferiority clinical trial was conducted from January 2015 to August 2017,with an 8-week treatment period and 52-week follow-up after the drug withdrawal.Children aged 2–6 years with RRTI meeting the inclusion and exclusion criteria were enrolled in 13 hospitals in China and divided randomly into three groups(2:2:1 ratio)to receive YPF,pidotimod,or placebo.The primary outcome was the proportion of RRTI returning to normal standard level during the follow-up.The secondary outcomes were reduction in the number of RRTI recurrences,effect on clinical symptoms(in accord with TCM practice),effect per symptom,and safety.The trial was registered at the Chinese Clinical Trials Registry(www.chictr.org.cn)under the unique identifier ChiCTR-IPR-15006847.Results:Three hundred and fifty-one children were enrolled and randomly assigned to 3 groups;124,125,and 61 children in the YPF,pidotimod,and placebo groups,respectively,had completed the trial.During the follow-up,the proportion of RRTI returning to normal standard level was 73.13%,67.15%,and 38.81%with YPF,pidotimod,and placebo,respectively(P<0.0001).The proportion of cases who returned to normal standard level in the YPF group was 34.32%higher than that in the placebo group.The safety profile did not significantly differ among the groups.Interpretation:YPF granules were noninferior to the active control drug pidotimod oral solution for the treatment of RRTI in children,and were superior to placebo,with a high safety profile.Baoping Xu Xinmin Li Siyuan Hu Yixiao Bao Fengmei Chen Zhimin Chen Yonggang Du Enmei Liu Yufeng Liu Qinghui Mou Baoling Su Bo Wang Jianwen Xu Guiping Xu Qiaozhi Yang Liwei Gao Xiaohui Liu Lei Li Rong Ma Kunling Shen 2022Pediatric Investigation2022,6,2:3

5Analytical computation of support characteristic curve for circumferential yielding lining in tunnel design显示文摘Circumferential yielding lining is able to tolerate controlled displacements without failure,which has been proven to be an effective solution to large deformation problem in squeezing tunnels.However,up to now,there has not been a well-established design method for it.This paper aims to present a detailed analytical computation of support characteristic curve(SCC)for circumferential yielding lining,which is a significant aspect of the implementation of convergence-confinement method(CCM)in tunnel support design.Circumferential yielding lining consists of segmental shotcrete linings and highly deformable elements,and its superior performance mainly depends on the mechanical characteristic of highly deformable element.The deformation behavior of highly deformable element is firstly investigated.Its whole deforming process can be divided into three stages including elastic,yielding and compaction stages.Especially in the compaction stage of highly deformable element,a nonlinear stress-strain relationship can be observed.For mathematical convenience,the stress-strain curve in this period is processed as several linear sub-curves.Then,the reasons for closure of circumferential yielding lining in different stages are explained,and the corresponding accurate equations required for constructing the SCC are provided.Furthermore,this paper carries out two case studies illustrating the application of all equations needed to construct the SCC for circumferential yielding lining,where the reliability and feasibility of theoretical derivation are also well verified.Finally,this paper discusses the sensitivity of sub-division in element compaction stage and the influence of element length on SCC.The outcome of this paper could be used in the design of proper circumferential yielding lining.Kui Wu Zhushan Shao Mostafa Sharifzadeh Siyuan Hong Su Qin 2022Journal of Rock Mechanics and Geotechnical Engineering2022,14,1:2

12Structure elucidation,immunomodulatory activity,antitumor activity and its molecular mechanism of a novel polysaccharide from Boletus reticulatus Schaeff显示文摘A new water-soluble heteropolysaccharide with a molecular weight of 15 k Da was isolated from the fruiting bodies of Boletus reticulatus Schaeff.Structural characterization results revealed that B.reticulatus Schaeff polysaccharide(BRS-X)had a backbone of 1,6-linkedα-D-galactose and 1,2,6-linkedα-D-galactose which branches were mainly composed of a terminal 4-linkedβ-D-glucose and the ratio of D-galactose and D-glucose was 5:1.Bioactivity assays indicated that BRS-X displayed a strong proliferative activity in T cells and B cells and promoted the secretion of immunoglobulin G(Ig G),Ig E,Ig D and Ig M.In addition,BRS-X could facilitate the proliferation and phagocytosis of RAW264.7 cells and could significantly inhibit the growth of tumors in S180-bearing mice.The results of transcriptome sequencing analysis illustrated that total 46 genes enriched in MAPK and total 34 genes enriched in PI3 K/Akt signaling pathways in BRS-X group.The protein VEGF and VEGFR expression were significantly reduced under the treatment with BRS-X.These findings provide a scientific basis for the edible and medicinal value of BRS-X.Siyuan Su Xiang Ding Yiling Hou Binbin Liu Zhouhe Du Junfeng Liu 2023Food Science and Human Wellness2023,12,2:1

14A de novo missense mutation in MPP2 confers an increased risk of Vogt–Koyanagi–Harada disease as shown by trio-based whole-exome sequencing显示文摘Vogt–Koyanagi–Harada(VKH)disease is a leading cause of blindness in young and middle-aged people.However,the etiology of VKH disease remains unclear.Here,we performed the first trio-based whole-exome sequencing study,which enrolled 25 VKH patients and 50 controls,followed by a study of 2081 VKH patients from a Han Chinese population to uncover detrimental mutations.A total of 15 de novo mutations in VKH patients were identified,with one of the most important being the membrane palmitoylated protein 2(MPP2)p.K315N(MPP2-N315)mutation.The MPP2-N315 mutation was highly deleterious according to bioinformatic predictions.Additionally,this mutation appears rare,being absent from the 1000 Genome Project and Genome Aggregation Database,and it is highly conserved in 10 species,including humans and mice.Subsequent studies showed that pathological phenotypes and retinal vascular leakage were aggravated in MPP2-N315 mutation knock-in or MPP2-N315 adeno-associated virus-treated mice with experimental autoimmune uveitis(EAU).In vitro,we used clustered regularly interspaced short palindromic repeats(CRISPR‒Cas9)gene editing technology to delete intrinsic MPP2 before overexpressing wild-type MPP2 or MPP2-N315.Levels of cytokines,such as IL-1β,IL-17E,and vascular endothelial growth factor A,were increased,and barrier function was destroyed in the MPP2-N315 mutant ARPE19 cells.Mechanistically,the MPP2-N315 mutation had a stronger ability to directly bind to ANXA2 than MPP2-K315,as shown by LC‒MS/MS and Co-IP,and resulted in activation of the ERK3/IL-17E pathway.Overall,our results demonstrated that the MPP2-K315N mutation may increase susceptibility to VKH disease.Xianyang Liu Jiayu Meng Xingyun Liao Yusen Liu Qian Zhou Zongren Xu Shuming Yin Qingfeng Cao Guannan Su Siyuan He Wanqian Li Xiaotang Wang Guoqing Wang Dali Li Peizeng Yang Shengping Hou 2023Cellular & Molecular Immunology2023,20,11:0
15Effect of Shale Reservoir Characteristics on Shale Oil Movability in the Lower Third Member of the Shahejie Formation, Zhanhua Sag显示文摘To reveal the effect of shale reservoir characteristics on the movability of shale oil and its action mechanism in the lower third member of the Shahejie Formation(Es3l), samples with different features were selected and analyzed using N2 adsorption, high-pressure mercury injection capillary pressure(MICP), nuclear magnetic resonance(NMR), high-speed centrifugation, and displacement image techniques. The results show that shale pore structure characteristics control shale oil movability directly. Movable oil saturation has a positive relationship with pore volume for radius > 2 μm, as larger pores often have higher movable oil saturation, indicating that movable oil is present in relatively larger pores. The main reasons for this are as follows. The relatively smaller pores often have oil-wetting properties because of organic matter, which has an unfavorable effect on the flow of oil, while the relatively larger pores are often wetted by water, which is helpful to shale oil movability. The rich surface provided by the relatively smaller pores is beneficial to the adsorption of immovable oil. Meanwhile, the relatively larger pores create significant pore volume for movable oil. Moreover, the larger pores often have good pore connectivity. Pores and fractures are interconnected to form a complex fracture network, which provides a good permeability channel for shale oil flow. The smaller pores are mostly distributed separately;thus, they are not conducive to the flow of shale oil. The mineral composition and fabric macroscopically affect the movability of shale oil. Calcite plays an active role in shale oil movability by increasing the brittleness of shale and is more likely to form micro-cracks under the same stress background. Clay does not utilize shale oil flow because of its large specific surface area and its block effect. 17Photooligomerization Determines Photosensitivity and Photoreactivity of Plant Cryptochromes显示文摘Plant and non-plant species possess cryptochrome(CRY)photoreceptors to mediate blue light regulation of development or the circadian clock.The blue light-dependent homooligomerization of Arabidopsis CRY2 is a known early photoreaction necessary for its functions,but the photobiochemistry and function of light-dependent homooligomerization and heterooligomerization of cryptochromes,collectively referred to as CRY photooligomerization,have not been well established.Here,we show that photooligomerization is an evolutionarily conserved photoreaction characteristic of CRY photoreceptors in plants and some non-plant species.Our analyses of the kinetics of the forward and reverse reactions of photooligomerization of Arabidopsis CRY1 and CRY2 provide a previously unrecognized mechanism underlying the different photosensitivity and photoreactivity of these two closely related photoreceptors.We found that photooligomerization is necessary but not sufficient for the functions of CRY2,implying that CRY photooligomerization is presumably accompanied by additional function-empowering conformational changes.We further demonstrated that the CRY2-CRY1 heterooligomerization plays roles in regulating functions of Arabidopsis CRYs in vivo.Taken together,these results suggest that photooligomerization is an evolutionarily conserved mechanism determining the photosensitivity and photoreactivity of plant CRYs.Qing Liu Tiantian Su Wenjin He Huibo Ren Siyuan Liu Yadi Chen Lin Gao Xiaohua Hu Haoyue Lu Shijiang Cao Ying Huang Xu Wang Qin Wang Chentao Lin 2020Molecular Plant2020,13,3:0

19Integrin β2 and β3:Two plasmatocyte markers deepen our understanding of the development of plasmatocytes in the silkworm Bombyx mori显示文摘Insect hemocytes play important biological roles at developmental stages,metamorphosis,and innate immunity.As one of the most abundant cell types,plasmatocytes can participate in various innate immune responses,especially in encapsulation and node formation.Here,2 molecular markers of plasmatocytes,consisting of integrinβ2 andβ3,were identified and used to understand the development of plasmatocytes.Plasmatocytes are widely distributed in the hematopoietic system,including circulating hemolymph and hematopoietic organs(HPOs).HPOs constantly release plasmatocytes with high proliferative activity in vitro;removal of HPOs leads to a dramatic reduction in the circulating plasmatocytes,and the remaining plasmatocytes gradually lose their ability to proliferate in vivo.Our results demonstrated that the release of plasmatocytes from HPOs is regulated by insulin-mediated signals and their downstream pathways,including PI3K/Akt and MAPK/Erk signals.The insulin/PI3K/Akt signaling pathway can significantly irritate the hematopoiesis,and its inhibitor LY294002 could inhibit the hemocytes discharged from HPOs.While the insulin/MAPK/Erk signaling pathway plays a negative regulatory role,inhibiting its activity with U0126 can markedly promote the discharge of plasmatocytes from HPOs.Our results indicate that the circulating plasmatocytes are mainly generated and discharged by HPOs.This process is co-regulated by the PI3K/Akt and MAPK/Erk signals in an antagonistic manner to adjust the dynamic balance of the hemocytes.These findings can enhance our understanding of insect hematopoiesis.Kui Zhang Jingjing Su Xin Hu Xiaomin Yan Siyuan Chen Chongyang Li Guangzhao Pan Hongbo Chang Wenli Tian Muhammad Nadeem Abbas Hongjuan Cui 2022Insect Science2022,29,6:0
