1Reproduction Diversity of Enteromorpha prolifera显示文摘Enteromorpha prolifera(Muell.) J.Agardh(Chlorophyta,Ulvophyceae),which is distributed widely in the inter-tidal zone of the ocean,is one of the most common fouling green algae.However,the present understandings of the life history of E.prolifera have been insufficient to explain their seasonal abundances.Thus it is essential to investigate how many reproductive strategies are likely to contribute to the successful colonization and flourishing of the green alga.In the present study the reproduction diversity of E.prolifera was observed and studied systematically by culturing chopped tissues.Our results showed that there are in total seven pathways of reproduction for E.prolifera including sexual,asexual and vegetative reproduction.It was indicated that the variety of the reproductive ways and the large quantity of reproductive cells produced and released during the reproductive season are the two key factors that facilitate colonization of E.prolifera.The reproduction of the alga E.prolifera mainly depends on asexual methods.The results presented here contribute to increasing our understanding about how the opportunistic macroalgae successfully maintain colonization and excessive growth.Apeng Lin Songdong Shen Jianwei Wang Binlun Yan 2008Journal of Integrative Plant Biology2008,50,5:46
2Comparison of Chlorophyll and Photosynthesis Parameters of Floating and Attached Ulva prolifera显示文摘In mid-May 2008 a serious green tide caused mainly by floating Ulva prolifera(Mller) J.Agardh(Chlorophyta,Ulvales) thalli struck the coastal area of Qingdao,China.To understand the present physiological conditions of the floating alga,in this work both laboratory and field investigations were conducted on the floating U.prolifera thalli in comparison with the attached U.prolifera thalli collected from the area.The floating thalli of three distinctively different colors and attached thalli at three different stages of sporangium formation process were characterized under a microscope,while their photosynthetic parameters were determined with chlorophyll fluorescence technology.On the other hand,the sporangium formation status of the floating U.prolifera thalli was surveyed both in the laboratory and in the field.Comparisons showed that both of the paired morphological characteristics and the paired physiological parameters of the floating and attached U.prolifera thalli were consistent.Furthermore,some spores were confirmed in the field and some motile particles were found within the floating thalli.These results suggest that the floating U.prolifera thalli with different colors could be at different stages of sporangium formation.However,our results also showed that the floating alga thalli have only a limited reproductive potential.This might limit the duration and the further geographic expansion of the green algal bloom.Apeng Lin Songdong Shen Guangce Wang Qianqian Yi Hongjin Qiao Jianfeng Niu Guanghua Pan 2011Journal of Integrative Plant Biology2011,53,1:20
3A New Molecular Label Applied to the Study of the Yellow Sea Green Tide显示文摘From 2007 to 2017,large-scale green tides occurred every year in the Yellow Sea of China,and Ulva prolifera was the main species leading to the green tide.In this study,we used the Polymerase chain reaction and 3'Rapid-amplification of c DNA ends technique to amplify the nr DNA-LSU and IGS sequences in U.prolifera and one species of Blidingia.These techniques showed 3259 bp of nrD NA-LSU and 3388 bp of IGS in U.prolifera and 3282 bp nr DNA-LSU and 3059 bp IGS in Blidingia sp.At the same time,tandem repeats,short dyads,palindromic and multiple simple repeat sequences in the IGS sequence were found by analyzing the structure of the IGS sequence in U.prolifera and Blidingia sp.G+C contents of the IGS sequence in U.prolifera and Blidingia sp.were 52.42% and 53.09%,respectively.We divided the U.prolifera into two types according to the morphological characteristics.Although the specimens of U.prolifera from the Qingdao coastal area,Jiangsu coastal area and the Yellow Sea have different morphologies,their ITS and IGS sequences are almost identical.Therefore,the main species of the green tides in the Qingdao coastal area,Jiangsu coastal area and the Yellow Sea are the same and have the same origin.SHEN Weijie HE Yuan SHEN Songdong 2019Journal of Ocean University of China2019,18,6:4
4Isolated Cells of Porphyra yezoensis Cultured on Solid Medium显示文摘Vegetative cells of Porphyra yezoensis are isolated with sea snail enzyme and cultured on the solidified agar medium. The results of experiments show that the isolated cells can survive,divide and regenerate well on the medium solidified with agar. The first division on the solid medium starts after 7 days' culture, 4 days later than the liquid culture. The survival rate of isolated cells is 71.3% on the solid medium, lower than the 86.2% of that in seawater.Thalli, thalloids,conchocelis, spermatangia and multicellular masses are developed on the solid/medium in the first month, slowly but normally. Spermatangia sacs disappear within 4 weeks. Without adding nutrient liquid onto the surface of solid medium or injecting seawater under the agar layer in order to keep moisture, the thalli and cell groups release monospores to form new thalli instead of enlarging their areas after 5 weeks' culturing. Some monospores regenerate new thalli. Other monospores lose their pigments and minimize their volume and divide quickly to form light pink calli. After 16 weeks, numerous calli can be seen on the solid medium and after 24 weeks' culturing, almost only calli and conchocelis can be seen. 5A ^(13)CO isotopic study on the CO promotion effect in methane dehydroaromatization reaction over a Mo/HMCM-49 catalyst显示文摘The promotion effect of CO in methane dehydroaromatization was investigated using 13CO probe molecules. 8The Ulva prolifera genome reveals the mechanism of green tides显示文摘The genome of green microalgae has rarely been reported.Ulva prolifera is a green microalga that has received much attention.Despite research articles about U.prolifera in recent years,we know very little about its genome.Therefore,the 87.9-Mb haploid genome(containing 10311 protein-coding genes)of U.prolifera was studied,and the genome was compared with that of U.mutabilis,which is the only published Ulva species.Results showed that the two species are closely related.A phylogenetic tree was constructed among U.prolifera and other green algae available in GenBank,revealing the evolutionary status of U.prolifera in Chlorophyta.To understand why U.prolifera could grow rapidly,we identified some genes related to growth,such as those involved in cell division,phosphorylation,and cell proliferation.In addition,genes related to stress resistance were found,which supports the notion that U.prolifera grows vigorously in nature.These results help to characterize green tides from a new perspective and reveal some important insight into the biology of U.prolifera.Yuan HE Songdong SHEN Dachun YU Yehua WANG Jiao YIN Zongling WANG Yuantu YE 2021Journal of Oceanology and Limnology2021,39,4:1

10Expression analyses of miRNA Up-MIR-843 and its target genes in Ulva prolifera显示文摘microRNAs(miRNA)families play a critical role in plant growth,development,and responses to abiotic stress.In this study,we characterized Up-miR-843 and its targets genes in Ulva prolifera responses to nitrogen depravation and heat stress.The data demonstrated that 184 target genes of Up-miR-843 could be successfully validated.N deficiency not heat stress stimulus induced increase in abundance of the Up-miR-843 while exhibited reverse expression of target genes,including cyclin A3 and cyclin L,which were strictly required for cell cycle progression.In addition,U.prolifera with highly expression of Up-miR-843 showed improved biomass,and photosynthesis compared with that under normal growth conditions.Thus,the N deprivation and heat responsive miRNAs might be a possible member mediating the expression of these target genes,which further regulated the growth of U.prolifera.Juanjuan Yang Dachun Yu Songdong Shen 2020Acta Oceanologica Sinica2020,39,10:1
11Effects of light and salinity on carotenoid biosynthesis in Ulva prolifera显示文摘Ulva prolifera is a green alga that plays an important role in green tides.Carotenoid biosynthesis is a basic terpenoid metabolism that is very important for maintaining normal life activities in algae.In this study,we first reported the complete sequences of all genes in the 2-C-methyl-D-erythritol 4-phosphate(MEP)pathway,which is the only carotenoid synthesis pathway in U.prolifera.Then,we compared these genes with those of other species.Additionally,by detecting the carotenoid contents and expression levels of key genes participating in carotenoid biosynthesis in U.prolifera under three different light(1000 lx,5000 lx and 12000 lx)and salinity(12,24 and 40)regimes,we found that carotenoid synthesis could be influenced by light and salinity,such that low light and high salinity could promote the synthesis of carotenoids.The results showed that the expression levels of genes involved in the MEP and the downstream pathway could affect the biosynthesis of carotenoids at the molecular level.This study contributes to a better understanding of the roles of genes participating in carotenoid biosynthesis in U.prolifera and the environmental regulation of these genes.Yuan He Yuantu Ye Songdong Shen 2020Acta Oceanologica Sinica2020,39,10:1
12Sequences and Phylogeny Analysis of rbcL Gene in Marine Chlorophyta显示文摘The rbcL gene of Ulva pertusa,Enteromorpha prolifera and Monostroma grevillei was amplified,sequenced and analyzed.By comparing the rbcL sequences with seven other Ulvales species retrieved from GenBank,the sequence divergences and thephyletic evolution were analyzed and the phylogenetic tree was constructed.From the phylogenetic tree,it can be found that U.per-tusa,E.prolifera and U.californica group in one branch,while E.compressa,U.rigida and U.fenestrata cluster in another clade.Obviously,unlike the Enteomorpha species,the Ulva species do not gather in one branch.Therefore Ulva and Enteomorpha might beaffiliates of one genus.E.compressa and E.intestinalis gathered together,which coincided with the morphological characters.However,the thallus of U.pertusa is thick and with many holes,which is different from E.prolifera in morphology.They cluster togetherin the phylogenetic tree with a genetic distance of 0.005.The results indicate that Ulva and Enteromorpha are not distinguishedstrictly.SHEN Songdong LI Yanyan WU Xunjian DING Lanping 2010Journal of Ocean University of China2010,9,2:1
13Characterization of SARS-COV-2 main protease inhibitory peptides from Ulva prolifera proteins显示文摘The main protease(M^(pro))is essential for the replication of SARS-COV-2 and therefore represents a promising anti-viral target.In this study,we screened M^(pro)inhibitory peptides from Ulva prolifera protein on in-silico proteolysis.Cytotoxicity analysis using the online toxic prediction tool ToxinPred revealed that all the peptides were non-cytotoxic.The hexapeptide(SSGFID)exhibited high M^(pro)inhibitory activity in molecular docking and its IC_(50)value was 139.40±0.82μmol/L in vitro according to fluorescence resonance energy transfer assay(FRET).Quantitative real-time(qRT-)PCR results show that SSGFID could stimulate the expression of mitosis-related factors,including nuclear factor-κB,cyclin D1,and cyclin-dependent kinase 4,to promote the proliferation of mice splenocytes.Stability study revealed that SSGFID showed resistance against pepsin and trypsin but lost D(Asp)after pretreatment at121℃ for 15 min.Besides,SSGFID was mainly transported through the Caco-2 cell monolayer by the peptide transporter PepT1 and passive-mediated transport during the transport study.Unfortunately,the peptide was also degraded by Caco-2 intracellular enzymes,and the transfer rate of intact peptide was4.2%.Furthermore,Lineweaver–Burk plots demonstrated that SSGFID possessed a mixed inhibitory characteristic with M^(pro).Our study indicated the potential of Ulva prolifera as antiviral and immuneenhancing functional food ingredients and nutraceuticals.Zhiyong LI Yehua WANG Caiwei FU Dongren ZHANG Tuanjie CHE Songdong SHEN 2023Journal of Oceanology and Limnology2023,41,5:0
14Effects of Methyl Jasmonate on the Composition of Volatile Compounds in Pyropia yezoensis显示文摘Volatile organic compounds in marine algae have been reported to comprise characteristic flavor of algae and play an important role in their growth, development and defensive response. Yet their biogeneration remain largely unknown. Here we studied the composition of volatile compouds in Pyropia yezoensis and their variations in response to methyl jasmonate(MeJA) and diethyldithiocarbamic acid(DIECA) treatment using gas chromatography-mass spectrometry(GC-MS). A total of 44 compounds belonging to the following chemical classes(n) were identified, including aldehydes(11), alcohols(8), acids and esters(6), alkanes(5), ketones(5), alkenes(3), and S-or N-containing miscellaneous compounds(6). External treatment with plant hormone MeJA increased the content of 1-dodecanol, 4-heptenal, and 2-propenoic acid-2-methyl dodecylester, but decreased the content of phytol, 3-heptadecene, 2-pentadecanone, and isophytol. When pretreated with DIECA, an inhibitor of the octadecanoid pathway leading to the biosynthesis of endogeneous jasmonates and some secondary metabolites, phytol and isophytol were increased, while 4-heptenal, 1-dodecanol, and 2-propenoic acid-2-methyl dodecylester were decreased, both of which were negatively correlated with their variations under MeJA treatment. 15Study of screening,transport pathway,and vasodilation mechanisms on angiotensin-Ⅰconverting enzyme inhibitory peptide from Ulva prolifera proteins显示文摘In this study,Ulva prolifera protein was used for preparing angiotensin-I converting enzyme(ACE)-inhibitory peptide via virtual gastrointestinal digestion and in silico screening.Some parameters of the obtained peptide,such as inhibition kinetics,docking mechanism,stability,transport pathway,were explored by Lineweaver-Burk plots,molecular docking,in vitro stimulate gastrointestinal(GI)digestion and Caco-2 cells monolayer model,respectively.Then,a novel anti-ACE peptide LDF(IC_(50),(1.66±0.34)μmol/L)was screened and synthesized by chemical synthesis.It was a no-competitive inhibitor and its anti-ACE inhibitory effect mainly attributable to four Conventional Hydrogen Bonds and Zn701 interactions.It could keep activity during simulated GI digestion in vitro and was transported by peptide transporter PepT1 and passive-mediated mode.Besides,it could activate Endothelial nitric oxide synthase(eNOS)activity to promote the production of NO and reduce Endothelin-1(ET-1)secretion induced by AngiotensinⅡ(AngⅡ)in Human Umbilical Vein Endothelial Cells(HUVECs).Meanwhile,it could promote mice splenocytes proliferation in a concentration-dependent manner.Our study indicated that this peptide was a potential ingredient functioning on vasodilation and enhancing immunity.Zhiyong Li Yuan He Hongyan He Caiwei Fu Mengru Li Aiming Lu Dongren Zhang Tuanjie Che Songdong Shen 2023Acta Oceanologica Sinica2023,42,11:0
