| 1 | Chinese Tuina Downregulates the Elevated Levels of Tissue Plasminogen Activator in Sciatic Nerve Injured Sprague-Dawley Rats显示文摘Objective:To elucidate the mechanism of Chinese tuina in treating sciatic nerve crush injury,and to detect the levels of tissue plasminogen activator(t PA)and plasminogen activator inhibitor-1(PAI-1),which is thought to play an important role in nerve regeneration.Methods:Thirty-two adult male Sprague-Dawley rats were subjected to sciatic nerve crush injury and 16 rats(sham-operated group)went through a sham operation.Control group was given no treatment while tuina group received tuina therapy since day 7 post-surgery.Tuina treatment was performed once a day and lasted for 20 days.The sciatic functional index was examined every 5days during the treatment session.The rats'gastrocnemius muscles were evaluated for changes in mass and immunohistochemistry techniques were performed to detect the levels of tP A and PAI-1.Results:Tuina therapy improved the motor function of sciatic nerve injured rats(P<0.05),however,it did not increase muscle volume(P<0.05).Tuina downregulated the levels of t PA and PAI-1(P<0.05).Conclusions:The present study implies that tuina treatment could accelerate rehabilitation of peripheral nerve injury. | PAN Fan YU Tian-yuan Steven Wong XIAN Si-tong LU Meng-qian WU Jian-cong GAO Yu-feng LI Xiao-qin GENG Nan YAO Bin-bin | 2017 | Chinese Journal of Integrative Medicine2017，23，8： | 16 |
| 2 | “Three Methods and Three Points” regulates p38 mitogen-activated protein kinase in the dorsal horn of the spinal cord in a rat model of sciatic nerve injury显示文摘Tuina is a traditional Chinese treatment for sensory disturbances caused by peripheral nerve injury and related diseases. Our previous studies showed that tuina regulates relevant regions and indices of the spinal dorsal horn using the Dian, Bo, and Rou method in Yinmen(BL37), Yanglingquan(GB34), and Weizhong(BL40). Treatment prevents muscle atrophy, protects spinal cord neurons, and promotes sciatic nerve repair. The mechanisms of action of tuina for treating peripheral nerve injury remain poorly understood. This study established rat models of sciatic nerve injury using the crushing method. Rats received Chinese tuina in accordance with the principle of 'Three Methods and Three Points,' once daily for 20 days. Tuina intervention reduced paw withdrawal latency and improved wet weight of the gastrocnemius muscle, as well as promoting morphological recovery of sciatic nerve fibers, Schwann cells, and axons. The protein expression levels of phospho-p38 mitogen-activated protein kinase, tumor necrosis factor-α, and interleukin-1β also decreased. | 5 | Nkx6.2 synergizes with Cdx-2 in stimulating proglucagon gene expression显示文摘AIM: To investigate whether the transactivator of the proglucagon gene (Gcg), Cdx-2, synergizes with other transcription factors in stimulating Gcg expression and the trans-differentiation of Gcg-expressing cells. METHODS: We conducted affinity chromatography to identify proteins that interact with Cdx-2, using GST-tagged Cdx-2 against cell lysates from pancreatic InR1-G9 and intestinal GLUTag cell lines. This was followed by a mass-spectrometry analysis. From a potential Cdx-2 interaction protein identified, we examined its expression in pancreatic and gut endocrine cells, confirmed its interaction with Cdx-2 by GST-pull down and determined its effect in provoking Gcg expression in cell lines that do not express endogenous Gcg.RESULTS: We identified 18 potential Cdx-2 interacting proteins. One of them is Nkx6.2. This homeodomain (HD) protein is expressed in pancreatic α and intestinal endocrine L cells but not in insulin producing cell lines, inclu ding In111. Nkx6.2, but not Nkx6.1, was shown to intera ct with Cdx-2, detected by GST-pull down. Furthermore, Nkx6.2 was found to synergize with Cdx-2 in provoking Gcg expression when they were ectopically expressed in the In111 cell line. Finally, when Cdx-2 and Nkx6.2 were co-transfected into the undifferentiated rat intestinal IEC-6 cell line, it produced detectable amount of Gcg mRNA. CONCLUSION: Cdx-2 recruits Nkx6.2 in exerting its ef fect in stimulating Gcg expression.
