SPDEF suppresses head and neck squamous cell carcinoma progression by transcriptionally activating NR4A1显示文摘SAM pointed domain containing E26 transformation-specific transcription factor(SPDEF)plays dual roles in the initiation and development of human malignancies. However, the biological role of SPDEF in head and neck squamous cell carcinoma(HNSCC)remains unclear. In this study, the expression level of SPDEF and its correlation with the clinical parameters of patients with HNSCC were determined using TCGA-HNSC,GSE65858,and our own clinical cohorts.CCK8,colony formation, cell cycle analysis, and a xenograft tumor growth model were used to determine the molecular functions of SPDEF in HNSCC.ChIP-qPCR,dual luciferase reporter assay, and rescue experiments were conducted to explore the potential molecular mechanism of SPDEF in HNSCC.Compared with normal epithelial tissues,SPDEF was significantly downregulated in HNSCC tissues.Patients with HNSCC with low SPDEF mRNA levels exhibited poor clinical outcomes.Restoring SPDEF inhibited HNSCC cell viability and colony formation and induced G0/G1 cell cycle arrest, while silencing SPDEF promoted cell proliferation in vitro. The xenograft tumor growth model showed that tumors with SPDEF overexpression had slower growth rates, smaller volumes, and lower weights.SPDEF could directly bind to the promoter region of NR4A1 and promoted its transcription, inducing the suppression of AKT,MAPK,and NF-κB signaling pathways. Moreover, silencing NR4A1 blocked the suppressive effect of SPDEF in HNSCC cells. Here,we demonstrate that SPDEF acts as a tumor suppressor by transcriptionally activating NR4A1 in HNSCC.Our findings provide novel insights into the molecular mechanism of SPDEF in tumorigenesis and a novel potential therapeutic target for HNSCC. | Yanting Wang Xianyue Ren Weiyu Li Ruoyan Cao Suyang Liu Laibo Jiang Bin Cheng Juan Xia | 2021 | International Journal of Oral Science2021,13,4: | 3
Experimental and Modelling Study on Emission of Volatile Nitrogen Derived NO during Pressured Oxy-fuel Combustion under Wet Flue Gas Environment显示文摘Pressurised oxy-fuel combustion(POFC)is a clean and efficient combustion technology with great potential.Due to the recycling of flue gas,the concentration of steam in the flue gas is higher than that of conventional combustion, which enriches the free radical pool in the flue gas and thus affects the emission of gaseous pollutants. Therefore, further research into the effect of high steam concentrations on NO_(x)emission mechanisms in POFC is necessary. In this work,a fixed-bed reactor was used to conduct combustion experiments of volatiles and combined with chemical kinetic models to study the NO release characteristics for different pressures and steam concentrations in an O_(2)/CO_(2)atmosphere at 800℃/900℃temperature.The results of the study indicated that the volatile nitrogen comes from the pyrolysis of part of pyrrole, pyridine, and all quaternary nitrogen in coal. The increase in temperature promoted the formation of NO during combustion.Higher pressure affects the main reaction pathway for NO formation, promoting NO consumption by HCCO and C_(2)O groups while enhancing the overall NO reduction.Steam promoted NO consumption by NCO.In addition, steam increased the amount of H/OH groups during the reaction, which affected both NO formation and consumption. However, from the overall effect, the steam still inhibits the emission of NO. | ZAN Haifeng CHEN Xiaoping PAN Suyang GENG Pengfei LIU Daoyin MA Jiliang LIANG Cai | 2023 | Journal of Thermal Science2023,32,5: | 0
Reynolds-number scaling analysis on lift generation of a flapping and passive rotating wing with an inhomogeneous mass distribution显示文摘Insects usually fly by passively rotating wings, which has been applied to the design of flapping-wing Micro-Air Vehicles(MAVs)to reduce mechanical complexity. In this paper,a robotic passive rotating-wing model is designed to investigate wing kinematics and lift generation, which are measured by a high-speed camera and a force transducer, respectively. In addition, flow fields are measured using the Particle Image Velocimetry(PIV).Experimental results demonstrate that passive rotating motion has a coordinative relationship with actively stroking motion. As the stroke amplitude or frequency increases, the rotating amplitude is enlarged. To characterize the active stroking motion,a driving Reynolds number Redrivingis defined, which varies from 68 to 366 in this study.Moving the gravity center of the wing towards trailing ed ge induces the increase of additional torque M,which decreases the wing rotating amplitude and promotes the advance of wing rotation. We find that the timing of wing rotation is gradually delayed and the mean lift coefficient C^(-)_(L)monotonously decreases as Redrivingincreases.By increasing the additional torque M,C^(-)_(L)is slightly improved and approaches to the lift coefficient of a real fruit fly at driving Re approximately equal to 230.The instantaneous lifts combined with the vortical structures further demonstrate that the lift generation associated with wing rotation is mainly attributed to the growth of the LeadingEdge Vortex(LEV)and the passive wake capture mechanism.Passive wake capture is influenced by LEV,reversal stroke motion and wing additional torque together, which can only maintain the lift at a high level for a considerable period. The high-lift generation mechanisms of flapping and passive rotating flight could shed light on the simplified design of MAVs and the improvement of their aerodynamic performance. | Suyang QIN Haotian HANG Yang XIANG Hong LIU | 2024 | Chinese Journal of Aeronautics2024,37,2: | 0
An RNA-RNA crosstalk network involving HMGB1 and RICTOR facilitates hepatocellular carcinoma tumorigenesis by promoting glutamine metabolism and impedes immunotherapy by PD-L1+ exosomes activity显示文摘Hepatocellular carcinoma(HCC)is the global leading cause of cancer-related deaths due to the deficiency of targets for precision therapy.A new modality of epigenetic regulation has emerged involving RNA-RNA crosstalk networks where two or more competing endogenous RNAs(ceRNAs)bind to the same microRNAs.However, the contribution of such mechanisms in HCC has not been well studied.Herein, potential HMGB1-driven RNA-RNA crosstalk networks were evaluated at different HCC stages, identifying the mT0RC2 component RICTOR as a potential HMGB1 ceRNA in HBV^(+)early stage HCC.Indeed, elevated HMGB1 mRNA was found to promote the expressio n of RICTOR mRNA through competitively bin ding with the miR-200 family, especially miR-429.Functio nal assays emplo ying overexpressi on or in terference strategies dem on strated that the HMGB1 and RICTOR 3zuntra nslated regions(UTR)epigenetically promoted the malignant proliferation,self-renewal,and tumorigenesis in HCC cells.Intriguingly,in terference agai nst HMGB1 and RICTOR in HCC cells promoted a stron ger an ti-PD-L1 immuno therapy resp on se,which appeared to associate with the production of PD-L1^(+)exosomes.Mechanistically,the HMGB1-driven RNA-RNA crosstalk network facilitated HCC cell glutamine metabolism via dual mechanisms, activating a positive feedback loop involving mT0RC2-AKT-C-MYC to upregulate glutamine synthetase(GS)expression, and inducing mTORCI signaling to derepress SIRT4 on glutamate dehydrogenase(GDH).Meanwhile, this crosstalk network could impede the efficacy of immunotherapy through mTORCI-P70S6K dependent PD-L1 production and PD-L1^(+)exosomes activity. In conclusion, our study highlights the non-coding regulatory role of HMGB1 with implicatio ns for RNA-based therapeutic targeting together with a predictio n of an ti-PD-L1 immuno therapy in HCC. | Yanping Wei Xuewu Tang Yibin Ren Yun Yang Fengliang Song Jingbo Fu Shuowu Liu Miao Yu Jing Chen Suyang Wang Kecheng Zhang Yexiong Tan Zhipeng Han Lixjn Wei Baohua Zhang Zhangjun Cheng Liang Li Hongyang Wang | 2022 | Signal Transduction and Targeted Therapy2022,7,1: | 0

Subsurface Damage of Monocrystalline Germanium Wafers by Fixed and Free Abrasive Lappings显示文摘The subsurface damage(SSD)layers of monocrystalline germanium wafers lapped by three different ways were measured and compared by the method of nanoindentation and micro morphology.Three ways such as ice-fixed abrasive, thermosetting fixed abrasive and free abrasive lappings are adopted to lap monocrystalline germanium wafers. The SSD depth was measured by a nanoindenter, and the morphology of SSD layer was observed by an atomic force microscopy(AFM).The results show that the SSD layer of monocrystalline germanium wafer is mainly composed of soft corrosion layer and plastic scratch and crack growth layer.Compared with thermosetting fixed abrasive and free abrasive lappings, the SSD depth lapped with ice-fixed abrasive is shallower. Moreover, the SSD morphology of monocrystalline germanium wafer lapped with ice-fixed abrasive is superior to those of two other processing ways. | Tang Suyang Sun Yuli Wang gong Li Jun Xu gang Liu Zhigang Zhu Yongwei Zuo Dunwen | 2017 | Transactions of Nanjing University of Aeronautics and Astronautics2017,34,5: | 0
Optimal Micro-PMU Placement for Improving State Estimation Accuracy via Mixed-integer Semidefinite Programming显示文摘Micro-phasor measurement units(μPMUs)with a micro-second resolution and milli-degree accuracy capability are expected to play an important role in improving the state estimation accuracy in the distribution network with increasing penetration of distributed generations. Therefore, this paper investigates the problem of how to place a limited number ofμPMUs to improve the state estimation accuracy.Combined with pseudo-measurements and supervisory control and data acquisition(SCADA)measurements,an optimalμPMU placement model is proposed based on a two-step state estimation method. The E-optimal experimental criterion is utilized to measure the state estimation accuracy. The nonlinear optimization problem is transformed into a mixed-integer semidefinite programming(MISDP)problem, whose optimal solution can be obtained by using the improved Benders decomposition method.Simulations on several systems are carried out to evaluate the effective performance of the proposed model. | Yang Peng Zhi Wu Wei Gu Suyang Zhou Pengxiang Liu | 2023 | Journal of Modern Power Systems and Clean Energy2023,11,2: | 0

Scaling analysis of the circulation growth of leading-edge vortex in flapping flight显示文摘In this paper,an experiment of a robotic model at Reynolds number of approximately 240 is performed with the aim of establishing a scaling law for describing the circulation growth of the leading-edge vortex(LEV)on a flapping wing.Three typical modes of wing rotation,i.e.,advanced,symmetric,and delayed modes, are considered to examine the effects of wing rotation on the scaling formation of LEV.The streamwise velocity fields of the LEV along the span of the wing are measured by particle image velocimetry technique.Experimental results demonstrated that a spirally three dimensional(3D)LEV with spanwise distribution of circulation rolls up on the upper surface of wing and the circulation of LEV usually obtains the peak before the end of wing stroke.Based on the concept of vortex formation time, the formation time of the 3D LEV are defined in two distinct manners. One(denoted by T∗LEV)is defined based on the LEV circulation, and the other(denoted by T∗∗LEV)is defined based on the wing kinematics. It is found that T∗∗LEV increases monotonously during the upstroke and downstroke, whereas T∗LEV generally arrives peaks and then decreases. The peak value of T∗LEV indicates the formation number of LEV,which stays in the range of 2.5–5.5,agrees with the scaling formation number predicted by other vortices. Moreover, the mode of wing rotation plays a controllable role in the formation number of LEV by modulating the characteristic length scale that feeds the formation of LEV.After reaching the formation number, the LEVs stably remain attached on the flapping wing and even further grow at some spanwise locations because of vorticity transport.Furthermore, the linear relationship between T∗LEV and T∗∗LEV before reaching the formation number can suggest a potential model for predicting the circulation growth of LEV based on wing kinematics. | Yang Xiang Haotian Hang Suyang Qin Hong Liu | 2021 | Acta Mechanica Sinica2021,37,10: | 0
