| 1 | Identification, occurrence, and validation of DRE and ABRE Cis-regulatory motifs in the promoter regions of genes of Arabidopsis thaliana显示文摘Plants posses a complex co-regulatory network Researchwhich helps them to elicit a response under diverse adverse conditions.We used an in silico approach to identify the genes with both DRE and ABRE motifs in their promoter regions in Arabidopsis thaliana.Our results showed that Arabidopsis contains a set of 2,052 genes with ABRE and DRE motifs in their promoter regions.Approximately 72%or more of the total predicted 2,052 genes had a gap distance of less than 400 bp between DRE and ABRE motifs.For positional orientation of the DRE and ABRE motifs,we found that the DR form(one in direct and the other one in reverse orientation)was more prevalent than other forms.These predicted 2,052 genes include 155 transcription factors.Using microarray data from The Arabidopsis Information Resource(TAIR)database,we present 44 transcription factors out of 155 which are upregulated by more than twofold in response to osmotic stress and ABA treatment.Fifty-one transcripts from the onepredicted above were validated using semiquantitative expression analysis to support the microarray data in TAIR.Taken together,we report a set of genes containing both DRE and ABRE motifs in their promoter regions in A.thaliana,which can be useful to understand the role of ABA under osmotic stress condition. | Sonal Mishra Aparna Shukla Swati Upadhyay Sanchita Pooja Sharma Seema Singh Ujjal J.Phukan Abha Meena Feroz Khan Vineeta Tripathi Rakesh Kumar Shukla Ashok Shrama | 2014 | Journal of Integrative Plant Biology2014,56,4: | 5 |
| 2 | Treatment of wheat straw using tannase and white-rot fungus to improve feed utilization by ruminants显示文摘Background: Current research to enrich cattle feed has primarily focused on treatment using white rot fungi, while there are scarce reports using the enzyme tannase, which is discussed only in reviews or in the form of a hypothesis. In this context, the aim of the present study was to evaluate the effect of tannase on wheat straw(WS)and also the effect of lyophilized tannase at concentrations of 0.1%, 0.2%, and 0.3%(w/w) on WS followed by fermentation with Ganoderma sp. for 10 d and compared in relation to biochemical parameters, crude protein(CP)content, and nutritional value by calculating the C/N ratio in order to improve the nutritional value of cattle feed.Results: Penicillium chorlesii, a tannase-producing microorganism, produced 61.4 lU/mL of tannase in 54 h when 2%(w/v) tannic acid(TA) was initially used as a substrate in medium containing(% w/v) sucrose(1.0), NaN03(1.0), and MgS〇4(0.08 pH, 5.0) in a 300-L fermentor(working volume 220 L), and concomitantly fed with 1.0%(w/v) TA after24 h. The yield of partially purified and lyophilized tannase was 5.8 lU/mg. The tannin-free myco-straw at 0.1%(w/w) tannase showed 37.8%(w/w) lignin degradation with only a 20.4%(w/w) decrease in cellulose content and the in vitro feed digestibility was 32.2%. | 4 | Dengue:epidemiology,prevention and pressing need for vaccine development显示文摘Dengue fever is a mosquito born viral infection,and the complicated form of dengue is dengue hemorrhagic fever(DHF).In the recent decades incidence and distribution of dengue has increased dramatically.Dengue viruses belong to family flaviviridae with four serotypes and are transmitted mainly by mosquito Aedes aegypti.Today almost two-fifth of world’s population (2.5 million) is at risk of dengue and no specific antiviral drug or vaccine is available against it.Uncontrolled population growth in Africa and South East Asia has increased number of susceptible hosts in urban and semi urban areas.About 40%of world population resides in the high risk area for dengue transmission.According to latest estimates by WHO,yearly 50 to 100 million infections occur globally,this includes around 500 000 DHF and 22 000 deaths,mostly among children.Only symptomatic treatment in the form of analgesic,antipyretics and body fluid management is provided to the patient.Prevention strategies mainly focus on two approaches, firsdy on activities to control vector and secondly on activities to protect human from mosquito bite but there is always concerns regarding their sustainably and effectiveness.Theoretically development of an effective dengue vaccine is feasible and production of an effective and affordable vaccine could be a viable option to save humans from this dreadful disease. | 5 | CEACAM-1 Induced CSF3-receptor Downregulation in Bone Marrow Associated With Refractory Neutropenia in Advanced Cirrhosis显示文摘Background and Aims:Cirrhosis patients exhibit cyto-penia,and,at times refractory neutropenia to granulocyte colony-stimulating factor(G-CSF),which acts through the CSF3-receptor(CSF3R),and changes in CSF3R can affect the response.We conducted this study to assess the CSF3R status and its relevance in cirrhotic patients.Methods:Cirrhotic patients(n=127)and controls(n=26)with clini-cally indicated bone marrow(BM)examination were stud-ied.BM assessment was done by qRT-PCR and immunohis-tochemistry(IHC)for CSF3R.Circulating G-CSF,CSF3R,and carcinoembryonic antigen cell adhesion molecule-1(CEACAM1)were measured.BM hematopoietic precursor cells and their alterations were examined by flow cytom-etry.The findings were validated in liver cirrhosis patients who received G-CSF for severe neutropenia.Results:The mean age was 48.6±13.4 years,and 80.3%were men.Circulatory CSF3R reduction was noted with the advance-ment of cirrhosis,and confirmed by qRT-PCR and IHC in BM.CSF3R decline was related to decreased hematopoietic stem cells(HSCs)and downregulation of CSF3R in the re-maining HSCs.Cocultures confirmed that CEACAM1 led to CSF3R downregulation in BM cells by possible lysosomal degradation.Baseline low peripheral blood-(PB)-CSF3R also predisposed development of infections on follow-up.Decreased CSF3R was also associated with nonresponse to exogenous G-CSF treatment of neutropenia.Conclu-sions:Advanced liver cirrhosis was associated with low CSF3R and high CEACAM1 levels in the BM and circula-tion,making patients prone to infection and inadequate response to exogenous G-CSF. | Chhagan Bihari Sukriti Baweja Seggere Murlaikrishna Shasthry Deepika Lal Preeti Negi Swati Thangariyal Dinesh Mani Tripathi Shiv Kumar Sarin | 2022 | Journal of Clinical and Translational Hepatology2022,10,1: | 2 |

| 7 | Feasibility and efficacy of delayed pharmacoinvasive therapy for STelevation myocardial infarction显示文摘BACKGROUND ST-elevation myocardial infarction(STEMI)refers to a clinical syndrome that features symptoms of myocardial ischemia with consequent ST-elevation on electrocardiography and an associated rise in cardiac biomarkers.Rapid restoration of brisk flow in the coronary vasculature is critical in reducing mortality and morbidity.In patients with STEMI who could not receive primary percutaneous coronary intervention(PCI)on time,pharmacoinvasive strategy(thrombolysis followed by timely PCI within 3-24 h of its initiation)is an effective option.AIM To analyze the role of delayed pharmacoinvasive strategy in the window period of 24-72 h after thrombolysis.METHODS This was a physician-initiated,single-center prospective registry between January 2017 and July 2017 which enrolled 337 acute STEMI patients with partially occluded coronary arteries.Patients received routine pharmacoinvasive therapy(PCI within 3-24 h of thrombolysis)in one group and delayed pharmacoinvasive therapy(PCI within 24-72 h of thrombolysis)in another group.The primary endpoint was major adverse cardiac and cerebrovascular events(MACCE)within 30 d of the procedure.The secondary endpoints included major bleeding as defined by Bleeding Academic Research Consortium classification,angina,and dyspnea within 30 d.RESULTS The mean age in the two groups was comparable(55.1±10.1 years vs 54.2±10.5 years,P=0.426).Diabetes was present among 20.2%and 22.1%of patients in the routine and delayed groups,respectively.Smoking rate was 54.6%and 55.8%in the routine and delayed groups,respectively.Thrombolysis was initiated within 6 h of onset of symptoms in both groups(P=0.125).The mean time from thrombolysis to PCI in the routine and delayed groups was 16.9±5.3 h and 44.1±14.7 h,respectively.No significant difference was found for the occurrence of measured clinical outcomes in the two groups within 30 d(8.7%vs 12.9%,P=0.152).Univariate analysis of demographic characteristics and risk factors for patients who reported MACCE in the two groups did not demonstrate any significant correlation.Secondary endpoints such as angina,dyspnea,and major bleeding were non-significantly different between the two groups.CONCLUSION Delayed PCI pharmacoinvasive strategy in a critical diseased but not completely occluded artery beyond 24 h in patients who have been timely thrombolyzed seems a reasonable strategy. | Rishi Sethi Lalit Mohan Pravesh Vishwakarma Abhishek Singh Swati Sharma Monika Bhandari Ayush Shukla Akhil Sharma Gaurav Chaudhary Akshyaya Pradhan Sharad Chandra Varun Shankar Narain Sudhanshu Kumar Dwivedi | 2023 | World Journal of Cardiology2023,15,1: | 1 |

| 12 | Comparative cytotoxicity of nanoparticles and ions to Escherichia coli in binary mixtures显示文摘The objective of this study was to understand toxicity of mixture of nanoparticles(NPs)(ZnO and TiO_2)and their ions to Escherichia coli. Results indicated the decrease in percentage growth of E. coli with the increase in concentration of NPs both in single and mixture setups.Even a small concentration of 1 mg/L was observed to be significantly toxic to E. coli in binary mixture setup(exposure concentration:1 mg/L ZnO and 1 mg/L TiO_2; 21.15% decrease in plate count concentration with respect to control). Exposure of E.coli to mixture of NPs at 1000 mg/L(i.e.,1000 mg/L ZnO and 1000 mg/L TiO_2)resulted in 99.63% decrease in plate count concentration with respect to control. Toxic effects of ions to E. coli were found to be lesser than their corresponding NPs. The percentage growth reduction was found to be 36% for binary mixture of zinc and titanium ions at the highest concentration(i.e.,803.0 mg/L Zn and593.3 mg/L Ti where ion concentrations are equal to the Zn ions present in 1000 mg/L ZnO NP solution and Ti^(+4) ions present in 1000 mg/L TiO_2 NP solution). Nature of mixture toxicity of the two NPs to E. coli was found to be antagonistic. The alkaline phosphatase(Alp)assay indicated that the maximum damage was observed when E. coli was exposed to 1000 mg/L of mixture of NPs. | 16 | Andrographolide inhibits chikungunya virus infection by up-regulating host innate immune pathways显示文摘Objective: To investigate the therapeutic efficacy of andrographolide, a plant derived compound, against chikungunya virus(CHIKV) infection. Methods: Using flow cytometry and immunoblotting assay, in vitro viral protein expression was studied in THP-1 cells line. In Balb/c mouse neonates, viral RNA copy number was determined by real time PCR. Results:The results showed reduced CHIKV protein expression on andrographolide treatment in CHIKV-infected human peripheral blood mononuclear cells, Vero cells and THP-I cell line.In vivo, andrographolide treatment to CHIKV-infected neonates reduced viral RNA copy number. Further. andrographolide also increased cytotoxic T lymphocytes both in vitro and in vivo. Andrographolide also activated host innate immune pathways, viz., protein kinase R.phosphorylated eukaryotic initiation factor 2 α, retinoic acid inducible gene-Ⅰ and interferon regulatory factor 3/7, thereby increasing IFN-a secretion. CHIKV-induced nuclear factor κlight chain enhancer of activated B cells and tumor necrosis factor-a was also reduced on andrographolide treatment. | 19 | Expression of multidrug resistance proteins in retinoblastoma显示文摘AIM: To elucidate the mechanism of multidrug resistance in retinoblastoma, and to acquire more insights into in vivo drug resistance.METHODS: Three anticancer drug resistant Y79 human RB cells were generated against vincristine, etoposide or carboplatin, which are used for conventional chemotherapy in RB. Primary cultures from enucleated eyes after chemotherapy(PCNC) were also prepared. Their chemosensitivity to chemotherapeutic agents(vincristine, etoposide and carboplatin) were measured using MTT assay. Western blot analysis was performed to evaluate the expression of p53, Bcl-2 and various multidrug resistant proteins in retinoblastoma cells.RESULTS: Following exposure to chemotherapeutic drugs, PCNC showed less sensitivity to drugs. No significant changes observed in the p53 expression, whereas Bcl-2 expression was found to be increased in the drug resistant cells as well as in PCNC. Increased expression of P-glycoprotein(P-gp) was observed in drug resistant Y79 cells; however there was no significant change in the expression of P-gp found between primary cultures of primarily enucleated eyes and PCNC. Multidrug resistance protein 1(Mrp-1) expression was found to be elevated in the drug resistant Y79 cells as well as in PCNC. | 20 | Consequences of forced migration during early childhood on cognitive well-being in later childhood in Andhra Pradesh,India显示文摘Unlike its short-term impact on consumption and income,forced migration is expected to deliver a permanent shock to the overall well-being of households,specifically children in the stage of infancy.Studies on the effect of forced migration on child cognitive well-being are few in number.Therefore,the present study is intended to examine the consequences of forced migration during infancy on child cognition at later age.We hypothesized that the effect of forced migration on child cognitive wellbeing can be mitigated by social support.The study used longitudinal data from three waves of the Young Lives Study(YLS)conducted in 2002,2006–2007,and 2009 in the state of Andhra Pradesh,India.We used bivariate and multivariate regression models to analyze the consequences of forced migration in early childhood on the cognitive wellbeing in later childhood.The information on forced migration was collected in Wave 1(at age 1),whereas the information on the cognitive well-being of the children was collected in Wave 3(at age 8).Child cognitive well-being was measured using scores obtained by the children on the Peabody Picture Vocabulary Test(PPVT),math,Early Grade Reading Assessment(EGRA),and memory tests.The results of the bivariate analysis show that the mean PPVT,math,EGRA,and memory scores obtained by children from the migrated households were lower than those obtained by children from the non-migrated households.Results of the multivariate linear regression models also show that children from the migrated households were statistically less likely to achieve higher scores on math(coefficient:-2.008,95%C.I.-3.108,-0.908),EGRA(coefficient:-0.746,95%C.I.-1.366,-0.126),and memory(coefficient:-0.503,95%C.I.-0.834,-0.173)as compared to children from the non-migrated households.Our findings also indicate that the effect of forced migration on child cognitive wellbeing was not mitigated by social support.Findings of this study conclude that forced migration during infancy has a significant effect on child cognitive well-being at later age.Therefore,interventions should be made,paying attention to the most vulnerable children who were displaced during critical development ages. | Ashish Kumar Upadhyay Swati Srivastava Chhavi Paul | 2017 | International Journal of Population Studies2017,3,2: | 0 |
