1Radiofrequency ablation or microwave ablation combined with transcatheter arterial chemoembolization in treatment of hepatocellular carcinoma by comparing with radiofrequency ablation alone显示文摘Objective:To compare radiofrequency ablation(RFA) or microwave ablation(MWA) and transcatheter arterial chemoembolization(TACE) with RFA or MWA monotherapy in hepatocellular carcinoma(HCC).Methods:A prospective,randomized,controlled trial was conducted on 94 patients with HCC ≤7 cm at a single tertiary referral center from June 2008 to June 2010 at the Department of Hepatobiliary Surgery,the Second Affiliated Hospital of Southeast University.The patients were randomly assigned into the TACERFA or TACE-MWA(combined treatment group) and the RFA-alone or MWA-alone groups(control group).The primary end point was overall survival.The secondary end point was recurrence-free survival,and the tertiary end point was adverse effects.Results:Until the time of censor,17 patients in the TACE-RFA or TACE-MWA group had died.The median follow-up time of the patients who were still alive for the TACE-RFA or TACE-MWA group was 47.5±11.3 months(range,29 to 62 months).The 1-,3- and 5-year overall survival for the TACE-RFA or TACE-MWA group was 93.6%,68.1% and 61.7%,respectively.Twenty-five patients in the RFA or MWA group had died.The median follow-up time of the patients who were still alive for the RFA or MWA group was 47.0±12.9 months(range,28 to 62 months).The 1-,3- and 5-year overall survival for the RFA or MWA group was 85.1%,59.6% and 44.7%,respectively.The patients in the TACE-RFA or TACE-MWA group had better overall survival than the RFA or MWA group [hazard ratio(HR),0.526;95% confidence interval(95% CI),0.334-0.823;P=0.002],and showed better recurrence-free survival than the RFA or MWA group(HR,0.582;95% CI,0.368-0.895;P=0.008).Conclusions:RFA or MWA combined with TACE in the treatment of HCC ≤7 cm was superior to RFA or MWA alone in improving survival by reducing arterial and portal blood flow due to TACE with iodized oil before RFA.Yongxiang Yi Yufeng Zhang Qiang Wei Liang Zhao Jianbo Han Yan Song Ying Ding Guilan Lu Junmao Liu Huaiying Ding Feng Dai Xiaojun Tang 2014Chinese Journal of Cancer Research2014,26,1:29
2Microbial response to CaCO_3 application in an acid soil in southern China显示文摘Calcium carbonate(CaCO_3) application is widely used to ameliorate soil acidification. To counteract soil and bacterial community response to CaCO_3 application in an acidic paddy soil in southern China, a field experiment was conducted with four different dosages of CaCO_3 addition, 0, 2.25, 4.5 and 7.5 tons/ha, respectively. After one seasonal growth of rice, soil physicochemical properties, soil respiration and bacterial communities were investigated. Results showed that soil p H increased accordingly with increasing dose of CaCO_3 addition, and 7.5 tons/ha addition increased soil p H to neutral condition. Moderate dose of CaCO_3 application(4.5 tons/ha) significantly increased soil dissolved organic carbon(DOC) and dissolved organic nitrogen(DON) content, enhanced soil respiration, while the excessive CaCO_3 application(7.5 tons/ha) decreased these soil properties. High-throughput sequencing results illustrated that moderate dose of CaCO_3 application increased the richness and alpha diversity of soil bacterial community. Compared with control, the relative abundance of Anaerolineaceae family belonging to Chloroflexi phylum increased by 38.7%, 35.4% and 24.5% under 2.25, 4.5 and 7.5 tons/ha treatments, respectively.Redundancy analysis(RDA) showed that soil p H was the most important factor shaping soil bacterial community. 3Combined pollution of arsenic and Polymyxin B enhanced arsenic toxicity and enriched ARG abundance in soil and earthworm gut microbiotas显示文摘Polymyxin B(PMB)is considered as the last line of antibiotic defense available to humans.The environmental effects of the combined pollution with PMB and heavy metals and their interaction mechanisms are unclear.We explored the effects of the combined pollution with PMB and arsenic(As)on the microbial composition of the soil and in the earthworm gut,as well as the spread and transmission of antibiotic resistance genes(ARGs).The results showed that,compared with As alone,the combined addition of PMB and As could significantly increase the bioaccumulation factor and toxicity of As in earthworm tissues by 12.1%and 16.0%,respectively.PMB treatment could significantly increase the abundance of Actinobacteria in the earthworm gut(from 35.6%to 45.2%),and As stress could significantly increase the abundance of Proteobacteria(from 19.8%to 56.9%).PMB and As stress both could significantly increase the abundance of ARGs and mobile genetic elements(MGEs),which were positively correlated,indicating that ARGs might be horizontally transferred.The inactivation of antibiotics was the main resistance mechanism that microbes use to resist PMB and As stress.Network analysis showed that PMB and As might have antagonistic effects through competition with multi-drug resistant ARGs.The combined pollution by PMB and As significantly promoted the relative abundance of microbes carrying multi-drug resistant ARGs and MGEs,thereby increasing the risk of transmission of ARGs.This research advances the understanding of the interaction mechanism between antibiotics and heavy metals and provides new theoretical guidance for the environmental risk assessment and combined pollution management.Lyu Li Dong Zhu Xingyun Yi Jianqiang Su Guilan Duan Xianjin Tang Yongguan Zhu 2021Journal of Environmental Sciences2021,33,11:2
4Root exudates increased arsenic mobility and altered microbial community in paddy soils显示文摘Root exudates are crucial for plants returning organic matter to soils,which is assumed to be a major source of carbon for the soil microbial community.This study investigated the influence of root exudates on the fate of arsenic(As)with a lab simulation experiment.Our findings suggested that root exudates had a dose effect on the soil physicochemical properties,As speciation transformation and the microbial community structure at different concentrations.The addition of root exudates increased the soil pH while decreased the soil redox potential(Eh).These changes in the soil pH and Eh increased As and ferrous(Fe(Ⅱ))concentrations in soil porewater.Results showed that 40 mg/L exudates addition significantly increased arsenite(As(Ⅲ))and arsenate(As(Ⅴ))by 541 and 10 times respectively within 30 days in soil porewater.The relative abundance of Fe(Ⅲ)-reducing bacteria Geobacter and Anaeromyxobacter increased with the addition of root exudates,which enhanced microbial Fe reduction.Together these results suggest that investigating how root exudates affect the mobility and transformation of As in paddy soils is helpful to systematically understand the biogeochemical cycle of As in soil-rice system,which is of great significance for reducing the health risk of soil As contamination.Ouyuan Jiang Lvyao Li Guilan Duan Williamson Gustave Weiwei Zhai Lina Zou Xia An Xianjin Tang Jianming Xu 2023Journal of Environmental Sciences2023,,5:2

10Role of autoreactive Tc17 cells in the pathogenesis of experimental autoimmune encephalomyelitis显示文摘Background:The pathogenesis of multiple sclerosis(MS)and experimental autoimmune encephalomyelitis(EAE-an animal model of MS)is primarily mediated by T cells.However,recent studies have only focused on interleukin(IL)-17-secreting CD4^(+)T-helper cells,also known as Th17 cells.This study aimed to compare Th17 cells and IL-17-secreting CD8^(+)T-cytotoxic cells(Tc17)in the context of MS/EAE.Methods:Female C57BL/6 mice were immunized with myelin oligodendrocyte glycoprotein peptides 35-55(MOG35-55),pertussis toxin,and complete Freund's adjuvant to establish the EAE animal model.T cells were isolated from the spleen(12-14 days postimmunization).CD4^(+)and CD8^(+)T cells were purified using isolation kit and then differentiated into Th17 and Tc17,respectively,using MOG35-55 and IL-23.The secretion levels of interferon-(IFN-γ)and IL-17 were measured via enzyme-linked immunosorbent assay using cultured CD4^(+)and CD8^(+)T cell supernatants.The pathogenicity of Tc17 and Th17 cells was assessed through adoptive transfer(tEAE),with the clinical course assessed using an EAE score(0-5).Hematoxylin and eosin as well as Luxol fast blue staining were used to examine the spinal cord.Purified CD8^(+)CD3^(+)and CD4^(+)CD3^(+)cells differentiated into Tc17 and Th17 cells,respectively,were stimulated with MOG35-55 peptide for proliferation assays.Results:The results showed that Tc17 cells(15,951±19855VS.55,709±4196cpm;p<0.050)exhibited a weaker response to highest dose(20μg/mL)MOG35-55 than Th17 cells.However,this response was not dependent on Th17 cells.After the 48h stimulation,at the highest dose(20μg/mL)of MOG35-55.Tc17 cells secreted lower levels of IFN-(280.00±15.00vs.556.67±15.28pg/mL,p<0.050)and IL-17(102.67±5.86 pg/mL vs.288.33±12.58 pg/mL;p<0.050)than Th17 cells.Similar patterns were observed for IFN-γsecretion at 96 and 144h.Furthermore,Tc17 cell-induced tEAE mice exhibited similar EAE scores to Th17 cell-induced tEAE mice and also showed similar inflammation and demyelination.Conclusion:The degree of pathogenicity of Tc17 cells in EAE is lower than that of Th17 cells.Future investigation on different immune cells and EAE models is warranted to determine the mechanisms underlying MS.Yong Peng Xiuli Zhang Yandan Tang Shunqing He Guilan Rao Quan Chen Yahui Xue Hong Jin Shu Liu Ziyang Zhou Yun Xiang 2024Neuroprotection2024,2,1:0
