| 1 | Fundamental CRISPR-Cas9 tools and current applications in microbial systems显示文摘Derived from the bacterial adaptive immune system,CRISPR technology has revolutionized conventional genetic engineering methods and unprecedentedly facilitated strain engineering.In this review,we outline the fundamental CRISPR tools that have been employed for strain optimization.These tools include CRISPR editing,CRISPR interference,CRISPR activation and protein imaging.To further characterize the CRISPR technology,we present current applications of these tools in microbial systems,including model-and non-model industrial microorganisms.Specially,we point out the major challenges of the CRISPR tools when utilized for multiplex genome editing and sophisticated expression regulation.To address these challenges,we came up with strategies that place emphasis on the amelioration of DNA repair efficiency through CRISPR-Cas9-assisted recombineering.Lastly,multiple promising research directions were proposed,mainly focusing on CRISPR-based construction of microbial ecosystems toward high production of desired chemicals. | Pingfang Tian Jia Wang Xiaolin Shen Justin Forrest Rey Qipeng Yuan Yajun Yan | 2017 | Synthetic and Systems Biotechnology2017,2,3: | 4 |
| 2 | A Recommendation Mechanism for Web Publishing Based on Sentiment Analysis of Microblog显示文摘Microblog is a social platform with huge user community and mass data. We propose a semantic recommendation mechanism based on sentiment analysis for microblog. Firstly, the keywords and sensibility words in this mechanism are extracted by natural language processing including segmentation, lexical analysis and strategy selection. Then, we query the background knowledge base based on linked open data(LOD) with the basic information of users. The experiment result shows that the accuracy of recommendation is within the range of 70%-89% with sentiment analysis and semantic query. | 4 | Cloud detection from visual band of satellite image based on variance of fractal dimension显示文摘Cover ratio of cloud is a very important factor which affects the quality of a satellite image, therefore cloud detection from satellite images is a necessary step in assessing the image quality. The study on cloud detection from the visual band of a satellite image is developed. Firstly, we consider the differences between the cloud and ground including high grey level, good continuity of grey level, area of cloud region, and the variance of local fractal dimension (VLFD) of the cloud region. A single cloud region detection method is proposed. Secondly, by introducing a reference satellite image and by comparing the variance in the dimensions corresponding to the reference and the tested images, a method that detects multiple cloud regions and determines whether or not the cloud exists in an image is described. | 5 | Overview of Rim2/Hipa transposon superfamily:Structure,distribution,transposition and utilization显示文摘The rice Rim2/Hipa is a unique transposon superfamily in Oryza genomes, which can be transcriptionally activated by the infection of the rice blast pathogen Magnaporthe grisea and its elicitors. Rim2/Hipa elements belong to En/Spm family, with perfect terminal inverted repeats in both ends, direct and inverted sub-terminal repeats, and 3-bp target site duplication upon insertion. Intriguingly, the typical 'CACTG' end sequences in the terminal inverted repeats of most Rim2/Hipa elements distinguish them from previously reported CACTA elements, such as En/Spm, Tam1, and Tgm1, which harbor 'CACTA' sequence in their terminal inverted repeats. In particular, the plentiful Rim2/Hipa elements unevenly distribute over all 12 chromosomes and make up at least 1% of the entire genome. Southern hybridization revealed the existence of Rim2/Hipa elements in all rice varieties examined and the Rim2/Hipa core sequence is unique to rice genomes. Evidence derived from sequence analysis indicated the occurrence of transposition events during the long evolution. The abundance and insertion polymorphisms suggested their applications as novel molecular marker in rice fingerprinting and fine chromo- some mapping.
