| 1 | Preparation and cytocompatibility of polylactic acid/hydroxyapatite/graphene oxide nanocomposite fibrous membrane显示文摘A series of polylactic acid (PLA) based nanocomposite fibrous membranes,including neat PLA,PLA/hydroxyapatite (HA) and PLA/HA/graphene oxide (GO),were fabricated via electrospinning method.The morphology and composition were investigated by scanning electron microscopy (SEM),X-ray diffraction (XRD) and Fourier transform infrared spectroscopy (FTIR) respectively.The thermal stability was determined by thermogravimetric analysis (TGA).To estimate the cytocompatibility of asprepared PLA/HA/GO fibrous membrane,MC3T3-E1 cells were cultured,and the corresponding cell adhesion and differentiation capability were investigated by fluorescence microscopy,SEM and MTT test.The electrospun ternary PLA/HA/GO membrane exhibited three-dimensional fibrous structure with relatively rough surface morphology,which made itself ideal for cell attachment and proliferation in bone tissue regeneration.The fluorescence microscopy,SEM and MTT test confirmed that the PLA/HA/GO nanocomposite fibrous membrane created a proper environment for the seeding and proliferation of MC3T3-E1 cells. | MA HaiBin SU WenXin TAI ZhiXin SUN DongFei YAN XingBin LIU Bin XUE QunJi | 2012 | Chinese Science Bulletin2012,57,23: | 11 |

| 3 | Suberoylanilide hydroxamic acid overcomes erlotinib-acquired resistance via phosphatase and tensin homolog deleted on chromosome 10-mediated apoptosis in non-small cell lung cancer显示文摘Background::Epidermal growth factor receptor(EGFR)tyrosine kinase inhibitors(TKIs),such as erlotinib and gefitinib,are widely used to treat non-small cell lung cancer(NSCLC).However,acquired resistance is unavoidable,impairing the anti-tumor effects of EGFR-TKIs.It is reported that histone deacetylase(HDAC)inhibitors could enhance the anti-tumor effects of other antineoplastic agents and radiotherapy.However,whether the HDAC inhibitor suberoylanilide hydroxamic acid(SAHA)can overcome erlotinib-acquired resistance is not fully clear.Methods::An erlotinib-resistant PC-9/ER cell line was established through cell maintenance in a series of erlotinib-containing cultures.NSCLC cells were co-cultured with SAHA,erlotinib,or their combination,and then the viability of cells was measured by the 3-(4,5-Dimethyl thiazol-2-yl)-2,5-diphenyl tetrazolium bromide assay and apoptosis was determined by flow cytometry and western blotting.Finally,the expression of phosphatase and tensin homolog deleted on chromosome 10(PTEN)was assessed by western blotting.Results::The half-maximal inhibitory concentration of parental PC-9 cells was significantly lower than the established erlotinib-acquired resistant PC-9/ER cell line.PC-9/ER cells demonstrated reduced expression of PTEN compared with PC-9 and H1975 cells,and the combination of SAHA and erlotinib significantly inhibited cell growth and increased apoptosis in both PC-9/ER and H1975 cells.Furthermore,treating PC-9/ER cells with SAHA or SAHA combined with erlotinib significantly upregulated the expression of PTEN mRNA and protein compared with erlotinib treatment alone.Conclusions::PTEN deletion is closely related to acquired resistance to EGFR-TKIs,and treatment with the combination of SAHA and erlotinib showed a greater inhibitory effect on NSCLC cells than single-drug therapy.SAHA enhances the suppressive effects of erlotinib in lung cancer cells,increasing cellular apoptosis and PTEN expression.SAHA can be a potential adjuvant to erlotinib treatment,and thus,can improve the efficacy of NSCLC therapy. | Peng-Fei Wu Wei-Wei Gao Cui-Lan Sun Tai Ma Ji-Qing Hao | 2020 | Chinese Medical Journal2020,,11: | 7 |

Although high-speed railways are characterized by being fast, comfortable and safe, higher standards for deformation of the railways' frozen subgrade are required. Meanwhile, changes in subgrade soil temperatures are the main factors affecting the deformation of frozen subgrade. Therefore, this paper selected typical test subgrade sections of the Harbin–Qiqihar Line, a special line for passenger transport built in the deep seasonal frozen soil regions of China, to monitor field temperatures. | 6 | Steroid elimination within 24 hours after orthotopic liver transplantation:effectiveness and tolerability显示文摘BACKGROUND:Steroids have been the mainstay of immunosuppressive regimen in liver transplantation.However,the use of steroids is associated with various post-transplant complications.This study evaluated the efficacy and safety of reduced immunosuppressive regimen with steroids(steroid elimination within 24 hours post-transplant) in a cohort of Chinese liver transplant recipients.METHODS:Seventy-six patients in line with the selection criteria were enrolled in this prospective study.All patients received anti-IL-2 receptor antibody induction and tacrolimusbased maintenance therapy.The recipients were divided into two groups according to the duration of steroid use:40 transplant in a 3-month withdrawal group and the remaining 36 in a 24-hour elimination group.Recipient survival,postoperative infections,biopsy-proven acute rejection and steroidresistant acute rejection,non-healing wound,recurrence of hepatitis B virus(HBV) and hepatocellular carcinoma(HCC),de novo diabetes,hyperlipidemia and hypertension were assessed in the two groups.RESULTS:There was no significant difference in patient survival,incidence of acute rejection episodes and hyperlipidemia,and recurrence of HBV and HCC between the two groups.However,the incidence rates of post-transplant infection,non-healing wound,de novo diabetes and hypertension were significantly lower in the 24-hour elimination group than in the 3-month withdrawal group(all P values <0.05).CONCLUSION:Under anti-IL-2 receptor antibody induction and tacrolimus-based maintainance,steroid elimination within 24 hours post-transplant is associated with reduced steroidrelated complications without increasing the risk of rejection. | Lin-Wei Wu,Zhi-Yong Guo,Qiang Tai,Wei-Qiang Ju,Dong-Ping Wang,An-Bin Hu,Xiao-Feng Zhu and Xiao-Shun He Organ Transplantation Center, First Affiliated Hospital, Sun Yat-Sen University, Guangzhou 510080, China | 2012 | Hepatobiliary & Pancreatic Diseases International2012,11,2: | 4 |

The beamline covered a wide energy range from 130 to 18 ke V with both a high photon flux([ 10^(12) phs/s with exit silt 30 lm in soft X-ray and [ 5 9 10^(12) phs/s in hard X-ray within 0.1%BW bandwidth) and promising resolving power(maximum E/DE [ 15,000 in soft X-ray with exit silt 30 lm and [6000 in hard X-ray). | 8 | Greenhouse gas emissions from a constructed wetland for municipal sewage treatment显示文摘The fluxes of greenhouse gases (methane and nitrous oxide) emission from a constructed wetland in the Eastern China as municipal sewage treatment were measured from June 1999 to August 2000 by the closed chamber method. The constructed wetland for municipal sewage treatment is a significant source of methane, up to 976 6×10 6 g CH 4/a, which was emitted from the constructed wetland with the area of 495000 m 2 and wastewater loading rate of 12000 m 3/d. Its daily mean methane flux reached 5 22 g CH 4/(m 2·d), 250 times as much as that in natural wetland in the same latitude region. 227 8 mg CH 4 was produced from the treatment of 1 liter wastewater, up to 700—1000 times as much as that in the secondary treatment. | 9 | Alteration of oncogenic IGF-II gene methylation status associates with hepatocyte malignant transformation显示文摘Background: Oncogenic insulin-like growth factor-II(IGF-II) is overexpressed in hepatocellular carcinoma(HCC). The present study aimed to analyze the dynamic alteration of IGF-II CpG site methylation status and its molecular mechanism in HCC progression. Methods: IGF-II alterations were observed in rat hepatocarcinogenesis models induced by 2-acetylaminofluorene. Liver IGF-II expression was compared by immunohistochemistry or tissue IGF-II specific concentration(nmol/mg protein). Status of human IGF-II promoter 3(P3) or rat IGF-II P2 CpG site methylation was amplified by methylation-specific polymerase chain reaction(MSP). Serum IGF-II levels were quantitatively detected by an enzyme-linked immunosorbent assay. Results: The levels of hepatic IGF-II expression were significantly elevated in the HCC group( P < 0.001). The unmethylation rate of IGF-II P3 CpG sites was 100% in the HCC-, 52.5% in the paracancerous-, and none(0%) in the distal noncancerous-tissues. Abnormal IGF-II expression was related to differentiation degree, tumor invasion, and positive HBV-DNA(all P < 0.001), with a negative correlation between P3 methylation degree and IGF-II expression. There was a positive correlation between liver IGF-II specific concentration and circulating IGF-II level( r = 0.97, P < 0.001). Significantly negative correlation was found between IGF-II P2 CpG site methylation and circulating IGF-II( r s =-0.89, P < 0.001) or liver IGF-II level( r s =-0.84, P < 0.001). | 10 | Prognostic significance of regional lymphadenectomy in T1b gallbladder cancer:Results from 24 hospitals in China显示文摘BACKGROUND Whether regional lymphadenectomy(RL)should be routinely performed in patients with T1b gallbladder cancer(GBC)remains a subject of debate.AIM To investigate whether RL can improve the prognosis of patients with T1b GBC.METHODS We studied a multicenter cohort of patients with T1b GBC who underwent surgery between 2008 and 2016 at 24 hospitals in 13 provinces in China.The logrank test and Cox proportional hazards model were used to compare the overall survival(OS)of patients who underwent cholecystectomy(Ch)+RL and those who underwent Ch only.To investigate whether combined hepatectomy(Hep)improved OS in T1b patients,we studied patients who underwent Ch+RL to compare the OS of patients who underwent combined Hep and patients who did not.RESULTS Of the 121 patients(aged 61.9±10.1 years),77(63.6%)underwent Ch+RL,and 44(36.4%)underwent Ch only.Seven(9.1%)patients in the Ch+RL group had lymph node metastasis.The 5-year OS rate was significantly higher in the Ch+RL group than in the Ch group(76.3%vs 56.8%,P=0.036).Multivariate analysis showed that Ch+RL was significantly associated with improved OS(hazard ratio:0.51;95%confidence interval:0.26-0.99).Among the 77 patients who underwent Ch+RL,no survival improvement was found in patients who underwent combined Hep(5-year OS rate:79.5%for combined Hep and 76.1%for no Hep;P=0.50).CONCLUSION T1b GBC patients who underwent Ch+RL had a better prognosis than those who underwent Ch.Hep+Ch showed no improvement in prognosis in T1b GBC patients.Although recommended by both the National Comprehensive Cancer Network and Chinese Medical Association guidelines,RL was only performed in 63.6%of T1b GBC patients.Routine Ch+RL should be advised in T1b GBC. | Tai Ren Yong-Sheng Li Xue-Yi Dang Yang Li Zi-Yu Shao Run-Fa Bao Yi-Jun Shu Xu-An Wang Wen-Guang Wu Xiang-Song Wu Mao-Lan Li Hong Cao Kun-Hua Wang Hong-Yu Cai Chong Jin Hui-Han Jin Bo Yang Xiao-Qing Jiang Jian-Feng Gu Yun-Fu Cui Zai-Yang Zhang Chun-Fu Zhu Bei Sun Chao-Liu Dai Lin-Hui Zheng Jing-Yu Cao Zhe-Wei Fei Chang-Jun Liu Bing Li Jun Liu Ye-Ben Qian Yi Wang Ya-Wei Hua Xi Zhang Chang Liu Wan-Yee Lau Ying-Bin Liu | 2021 | World Journal of Gastrointestinal Surgery2021,13,2: | 3 |
| 11 | Cholesterol homeostasis regulated by ABCA1 is critical for retinal ganglion cell survival显示文摘Genome-wide association studies have suggested a link between primary open-angle glaucoma and the function of ABCA1.ABCA1 is a key regulator of cholesterol efflux and the biogenesis of high-density lipoprotein(HDL) particles. Here, we showed that the POAG risk allele near ABCA1 attenuated ABCA1 expression in cultured cells. Consistently, POAG patients exhibited lower ABCA1 expression, reduced HDL, and higher cholesterol in white blood cells. Ablation of Abca1 in mice failed to form HDL, leading to elevated cholesterol levels in the retina. Counting retinal ganglion cells(RGCs) by using an artificial intelligence(AI) program revealed that Abca1-deficient mice progressively lost RGCs with age. Single-cell RNA sequencing(scRNA-seq) revealed aberrant oxidative phosphorylation in the Abca1-/-retina, as well as activation of the mTORC1 signaling pathway and suppression of autophagy. Treatment of Abca1-/-mice using atorvastatin reduced the cholesterol level in the retina,thereby improving metabolism and protecting RGCs from death. Collectively, we show that lower ABCA1 expression and lower HDL are risk factors for POAG. Accumulated cholesterol in the Abca1-/-retina causes profound aberrant metabolism, leading to a POAG-like phenotype that can be prevented by atorvastatin.
