1Novel SNPs in IL-17F and IL-17A genes associated with somatic cell count in Chinese Holstein and Inner-Mongolia Sanhe cattle显示文摘Background: Bovine mastitis is the most common and costly disease of lactating cattle worldwide. Apart from milk somatic cell count(SCC) and somatic cell score(SCS), serum cytokines such as interleukin-17(IL-17) and interleukin-4(IL-4) may also be potential indicators for bovine mastitis. The present study was designed to investigate the effects of single nucleotide polymorphisms(SNPs) in bovine IL-17 F and IL-17 A genes on SCC, SCS and serum cytokines in Chinese Holstein and Inner-Mongolia Sanhe cattle, and to compare the m RNA expression variations of the cows with different genotypes.Results: A total of 464 lactating cows(337 Holstein and 127 Inner-Mongolia Sanhe cattle) were screened for SNPs identification and the data were analyzed using fixed effects of herd, parity, season and year of calving by general linear model procedure. The results revealed that SNP g.24392436 C > T in IL-17 F and SNP g.24345410 A > G in IL-17 A showed significant effects on SCC and IL-4 in Holstein(n = 337) and on IL-17 and IL-4 in Sanhe cattle(n = 127). 2Reverse vaccinology assisted designing of multiepitope-based subunit vaccine against SARS-CoV-2显示文摘Background:Coronavirus disease 2019(COVID-19)linked with severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)cause severe illness and life-threatening pneumonia in humans.The current COVID-19 pandemic demands an effective vaccine to acquire protection against the infection.Therefore,the present study was aimed to design a multiepitope-based subunit vaccine(MESV)against COVID-19.Methods:Structural proteins(Surface glycoprotein,Envelope protein,and Membrane glycoprotein)of SARS-CoV-2 are responsible for its prime functions.Sequences of proteins were downloaded from GenBank and several immunoinformatics coupled with computational approaches were employed to forecast B-and T-cell epitopes from the SARS-CoV-2 highly antigenic structural proteins to design an effective MESV.Results:Predicted epitopes suggested high antigenicity,conserveness,substantial interactions with the human leukocyte antigen(HLA)binding alleles,and collective global population coverage of 88.40%.Taken together,276 amino acids long MESV was designed by connecting 3 cytotoxic T lymphocytes(CTL),6 helper T lymphocyte(HTL)and 4 B-cell epitopes with suitable adjuvant and linkers.The MESV construct was non-allergenic,stable,and highly antigenic.Molecular docking showed a stable and high binding affinity of MESV with human pathogenic toll-like receptors-3(TLR3).Furthermore,in silico immune simulation revealed significant immunogenic response of MESV.Finally,MEV codons were optimized for its in silico cloning into the Escherichia coli K-12 system,to ensure its increased expression.Conclusion:The MESV developed in this study is capable of generating immune response against COVID-19.Therefore,if designed MESV further investigated experimentally,it would be an effective vaccine candidate against SARS-CoV-2 to control and prevent COVID-19.Muhammad Tahir ul Qamar Farah Shahid Sadia Aslam Usman Ali Ashfaq Sidra Aslam Israr Fatima Muhammad Mazhar Fareed Ali Zohaib Ling-Ling Chen 2020Infectious Diseases of Poverty2020,9,5:2
3Transcriptome sequencing analysis for the identification of stable lncRNAs associated with bovine Staphylococcus aureus mastitis显示文摘Background:Staphylococcus aureus(S.aureus)mastitis is one of the most difficult diseases to treat in lactating dairy cows worldwide.S.aureus with different lineages leads to different host immune responses.Long non-coding RNAs(lncRNAs)are reported to be widely involved in the progress of inflammation.However,no research has identified stable lncRNAs among different S.aureus strain infections.In addition,folic acid(FA)can effectively reduce inflammation,and whether the inflammatory response caused by S.aureus can be reduced by FA remains to be explored.Methods:lncRNA transcripts were identified from Holstein mammary gland tissues infected with different concentrations of S.aureus(in vivo)and mammary alveolar cells(Mac-T cells,in vitro)challenged with different S.aureus strains.Differentially expressed(DE)lncRNAs were evaluated,and stable DE lncRNAs were identified in vivo and in vitro.On the basis of the gene sequence conservation and function conservation across species,key lncRNAs with the function of potentially immune regulation were retained for further analysis.The function of FA on inflammation induced by S.aureus challenge was also investigated.Then,the association analysis between these keys lncRNA transcripts and hematological parameters(HPs)was carried out.Lastly,the knockdown and overexpression of the important lncRNA were performed to validate the gene function on the regulation of cell immune response.Results:Linear regression analysis showed a significant correlation between the expression levels of lncRNA shared by mammary tissue and Mac-T cells(P<0.001,R^(2)=0.3517).lncRNAs PRANCR and TNK2-AS1 could be regarded as stable markers associated with bovine S.aureus mastitis.Several HPs could be influenced by SNPs around lncRNAs PRANCR and TNK2-AS1.The results of gene function validation showed PRANCR regulates the mRNA expression of SELPLG and ITGB2 within the S.aureus infection pathway and the Mac-T cells apoptosis.In addition,FA regulated the expression change of DE lncRNA involved in toxin metabolism and inflammation to fight against S.aureus infection.Conclusions:The remarkable association between SNPs around these two lncRNAs and partial HP indicates the potentially important role of PRANCR and TNK2-AS1 in immune regulation.Stable DE lncRNAs PRANCR and TNK2-AS1 can be regarded as potential targets for the prevention of bovine S.aureus mastitis.FA supplementation can reduce the negative effect of S.aureus challenge by regulating the expression of lncRNAs.Siyuan Mi Yongjie Tang Gerile Dari Yuanjun Shi Jinning Zhang Hailiang Zhang Xueqin Liu Yibing Liu Usman Tahir Ying Yu 2022Journal of Animal Science and Biotechnology2022,13,2:2

5Overview of multi-stage charging strategies for Li-ion batteries显示文摘To reduce the carbon footprint in the transportation sector and improve overall vehicle efficiency,a large number of electric vehicles are being manufactured.This is due to the fact that environmental concerns and the depletion of fossil fuels have become significant global problems.Lithium-ion batteries(LIBs)have been distinguished themselves from alternative energy storage technologies for electric vehicles(EVs) due to superior qualities like high energy and power density,extended cycle life,and low maintenance cost to a competitive price.However,there are still certain challenges to be solved,like EV fast charging,longer lifetime,and reduced weight.For fast charging,the multi-stage constant current(MSCC) charging technique is an emerging solution to improve charging efficiency,reduce temperature rise during charging,increase charging/discharging capacities,shorten charging time,and extend the cycle life.However,there are large variations in the implementation of the number of stages,stage transition criterion,and C-rate selection for each stage.This paper provides a review of these problems by compiling information from the literature.An overview of the impact of different design parameters(number of stages,stage transition,and C-rate) that the MSCC charging techniques have had on the LIB performance and cycle life is described in detail and analyzed.The impact of design parameters on lifetime,charging efficiency,charging and discharging capacity,charging speed,and rising temperature during charging is presented,and this review provides guidelines for designing advanced fast charging strategies and determining future research gaps.Muhammad Usman Tahir Ariya Sangwongwanich Daniel-Ioan Stroe Frede Blaabjerg 2023Journal of Energy Chemistry2023,,9:1

7Countrywide Survey for MERS-Coronavirus Antibodies in Dromedaries and Humans in Pakistan显示文摘Middle East Respiratory Syndrome Coronavirus (MERS-CoV) is a zoonotic pathogen capable of causing severe respiratory disease in humans. Although dromedary camels are considered as a major reservoir host, the MERS-CoV infection dynamics in camels are not fully understood. Through surveillance in Pakistan, nasal (n = 776) and serum (n = 1050)samples were collected from camels between November 2015 and February 2018. Samples were collected from animal markets, free-roaming herds and abattoirs. An in-house ELISA was developed to detect IgG against MERS-CoV. A total of 794 camels were found seropositive for MERS-CoV. Prevalence increased with the age and the highest seroprevalence was recorded in camels aged [ 10 years (81.37%) followed by those aged 3.1–10 years (78.65%) and B 3 years (58.19%).Higher prevalence was observed in female (78.13%) as compared to male (70.70%). Of the camel nasal swabs, 22 were found to be positive by RT-qPCR though with high Ct values. Moreover, 2,409 human serum samples were also collected from four provinces of Pakistan during 2016–2017. 8A 37 GHz Millimeter-Wave Antenna Array for 5G Communication Terminals显示文摘This work presents,design and specific absorption rate(SAR)analysis of a 37GHz antenna,for 5th Generation(5G)applications.The proposed antenna comprises of 4-elements of rectangular patch and an even distribution.The radiating element is composed of copper material supported by Rogers RT5880 substrate of thickness,0.254 mm,dielectric constant(εr),2.2,and loss tangent,0.0009.The 4-elements array antenna is compact in size with a dimension of 8mm×20mm in length and width.The radiating patch is excited with a 50 ohms connector i.e.,K-type.The antenna resonates in the frequency band of 37 GHz,that covers the 5G applications.The antenna behavior is studied both in free space and in the proximity of the human body.Three models of the human body,i.e.,belly,hand,and head(contain skin,fat,muscles,and bone)are considered for on-body simulations.At resonant frequency,the antenna gives a boresight gain of 11.6 dB.The antenna radiates efficiently with a radiated efficiency of more than 90%.Also,it is observed that the antenna detunes to the lowest in the proximity of the human body,but still a good impedance matching is achieved considering the−10 dB criteria.Moreover,SAR is also being presented.The safe limit of 2 W/kg for any 10 g of biological tissue,specified by the European International Electro Technical Commission(IEC)has been considered.The calculated values of SAR for human body models,i.e.,belly,hand and head are 1.82,1.81 and 1.09 W/kg,respectively.The SAR values are less than the international recommendations for the three models.Furthermore,the simulated and measured results of the antenna are in close agreement,which makes it,a potential candidate for the fifth-generation smart phones and other handheld devices.Jalal Khan Sadiq Ullah Usman Ali Ladislau Matekovits Farooq Ahmad Tahir Muhammad Inam Abbasi 2023Computers, Materials & Continua2023,,4:0

10In vitro biological screening of the stem of Desmodium elegans显示文摘Objective:To explore the medicinal importance of the stem of Desmodium elegans,methanolic extract,and its different solvent fractions were evaluated for brine shrimp lethality,insecticidal and phytotoxicity,antifungal,and antibacterial activities.Methods:The methanolic extract and its solvent fractions were tested for cytotoxic,phytotoxic,insecticidal,antifungal,and antibacterial effects using our previous published protocols.Results:The methanolic,DCM,ethyl acetate and n-butanol fractions exhibited insecticidal effect against Callosobruchus analis and Rhyzopertha dominie.The methanolic extract,n-hexane,DCM ethyl acetate and n-butanol showed 75,85,85,65 and 5%phytotoxicity at the tested concentration of 500μg/ml.respectively.The solvent fractions(DCM and ethyl acetate)were effective against F.solani(10%and 20%,inhibition respectively).All the tested samples were devoid of cytotoxic and antibacterial effects.Conclusions:It was concluded diat diis plant can be practiced for control of weeds and insects.Arshad Khan Rabia Usman Ming-Liang Wang Abdur Rauf Naveed Muhammad Akhatar Aman Taha Hussein Musa Tahir 2013Asian Pacific Journal of Tropical Biomedicine2013,3,9:0
11Evaluation of heat stress effects on cellular and transcriptional adaptation of bovine granulosa cells显示文摘Background:Heat stress is known to affect follicular dynamics,oocyte maturation,and fertilization by impairing steroidogenic ability and viability of bovine granulosa cell(bGCs).The present study explored the physiological and molecular response of bGCs to different heat stress intensities in-vitro.We exposed the primary bGCs to heat stress(HS)at 39℃,40℃ and 41℃ along with control samples(38℃)for 2 h.To evaluate the impact of heat stress on bGCs,several in vitro cellular parameters including cell apoptosis,intracellular reactive oxygen species(ROS)accumulation and HSP70 kinetics were assessed by flow cytometry,florescence microscopy and western blot,respectively.Furthermore,the ELISA was performed to confirm the 17β-estradiol(E2)and progesterone(P4)levels.In addition,the RNA sequencing(RNA-Seq)method was used to get the molecular based response of bGCs to different heat treatments.Results:Our findings revealed that the HS significantly decreased the cell viability,E2 and P4 levels in bGCs,whereas,increased the cellular apoptosis and ROS.Moreover,the RNA-Seq experiments showed that all the treatments(39℃,40℃ and 41℃)significantly regulated many differentially expressed genes(DEGs)i.e.BCL2L1,STAR,CYP11A1,CASP3,SOD2,HSPA13,and MAPK8IP1 and pathways associated with heat stress,apoptosis,steroidogenesis,and oxidative stress.Conclusively,our data demonstrated that the impact of 40°C treatment was comparatively detrimental for cell viability,apoptosis and ROS accumulation.Notably,a similar trend of gene expression was reported by RT-qPCR for RNA-seq data.Conclusions:Our study presented a worthy strategy for the first time to characterize the cellular and transcriptomic adaptation of bGCs to heat stress(39,40 and 41°C)in-vitro.The results infer that these genes and pathways reported in present study could be useful candidates/indicators for heat stress research in dairy cattle.Moreover,the established model of bGCs to heat stress in the current study provides an appropriate platform to understand the mechanism of how heat-stressed bGCs can affect the quality of oocytes and developing embryo.Adnan Khan Jinhuan Dou Yachun Wang Xiaolong Jiang Muhammad Zahoor Khan Hanpeng Luo Tahir Usman Huabin Zhu 2020Journal of Animal Science and Biotechnology2020,11,2:0

133D Kronecker Convolutional Feature Pyramid for Brain Tumor Semantic Segmentation in MR Imaging显示文摘Brain tumor significantly impacts the quality of life and changes everything for a patient and their loved ones.Diagnosing a brain tumor usually begins with magnetic resonance imaging(MRI).The manual brain tumor diagnosis from the MRO images always requires an expert radiologist.However,this process is time-consuming and costly.Therefore,a computerized technique is required for brain tumor detection in MRI images.Using the MRI,a novel mechanism of the three-dimensional(3D)Kronecker convolution feature pyramid(KCFP)is used to segment brain tumors,resolving the pixel loss and weak processing of multi-scale lesions.A single dilation rate was replaced with the 3D Kronecker convolution,while local feature learning was performed using the 3D Feature Selection(3DFSC).A 3D KCFP was added at the end of 3DFSC to resolve weak processing of multi-scale lesions,yielding efficient segmentation of brain tumors of different sizes.A 3D connected component analysis with a global threshold was used as a post-processing technique.The standard Multimodal Brain Tumor Segmentation 2020 dataset was used for model validation.Our 3D KCFP model performed exceptionally well compared to other benchmark schemes with a dice similarity coefficient of 0.90,0.80,and 0.84 for the whole tumor,enhancing tumor,and tumor core,respectively.Overall,the proposed model was efficient in brain tumor segmentation,which may facilitate medical practitioners for an appropriate diagnosis for future treatment planning.Kainat Nazir Tahir Mustafa Madni Uzair Iqbal Janjua Umer Javed Muhammad Attique Khan Usman Tariq Jae-Hyuk Cha 2023Computers, Materials & Continua2023,76,9:0
14Discovery of human coronaviruses pan-papain-like protease inhibitors using computational approaches显示文摘The papain-like protease(PLpro)is vital for the replication of coronaviruses(Co Vs),as well as for escaping innate-immune responses of the host.Hence,it has emerged as an attractive antiviral drug-target.In this study,computational approaches were employed,mainly the structure-based virtual screening coupled with all-atom molecular dynamics(MD)simulations to computationally identify specific inhibitors of severe acute respiratory syndrome coronavirus 2(SARS-Co V-2)PLpro,which can be further developed as potential pan-PLprobased broad-spectrum antiviral drugs.The sequence,structure,and functional conserveness of most deadly human Co Vs PLprowere explored,and it was revealed that functionally important catalytic triad residues are well conserved among SARS-Co V,SARS-Co V-2,and middle east respiratory syndrome coronavirus(MERS-Co V).The subsequent screening of a focused protease inhibitors database composed of^7,000 compounds resulted in the identification of three candidate compounds,ADM13083841,LMG15521745,and SYN15517940.These three compounds established conserved interactions which were further explored through MD simulations,free energy calculations,and residual energy contribution estimated by MM-PB(GB)SA method.All these compounds showed stable conformation and interacted well with the active residues of SARS-Co V-2 PLpro,and showed consistent interaction profile with SARS-Co V PLproand MERS-Co V PLproas well.Conclusively,the reported SARS-Co V-2 PLprospecific compounds could serve as seeds for developing potent pan-PLprobased broad-spectrum antiviral drugs against deadly human coronaviruses.Moreover,the presented information related to binding site residual energy contribution could lead to further optimization of these compounds.Mubarak A.Alamri Muhammad Tahir ul Qamar Muhammad Usman Mirza Safar M.Alqahtani Matheus Froeyen Ling-Ling Chen 2020Journal of Pharmaceutical Analysis2020,10,6:0
15Classification of Gastric Lesions Using Gabor Block Local Binary Patterns显示文摘The identification of cancer tissues in Gastroenterology imaging poses novel challenges to the computer vision community in designing generic decision support systems.This generic nature demands the image descriptors to be invariant to illumination gradients,scaling,homogeneous illumination,and rotation.In this article,we devise a novel feature extraction methodology,which explores the effectiveness of Gabor filters coupled with Block Local Binary Patterns in designing such descriptors.We effectively exploit the illumination invariance properties of Block Local Binary Patterns and the inherent capability of convolutional neural networks to construct novel rotation,scale and illumination invariant features.The invariance characteristics of the proposed Gabor Block Local Binary Patterns(GBLBP)are demonstrated using a publicly available texture dataset.We use the proposed feature extraction methodology to extract texture features from Chromoendoscopy(CH)images for the classification of cancer lesions.The proposed feature set is later used in conjuncture with convolutional neural networks to classify the CH images.The proposed convolutional neural network is a shallow network comprising of fewer parameters in contrast to other state-of-the-art networks exhibiting millions of parameters required for effective training.The obtained results reveal that the proposed GBLBP performs favorably to several other state-of-the-art methods including both hand crafted and convolutional neural networks-based features.Muhammad Tahir Farhan Riaz Imran Usman Mohamed Ibrahim Habib 2023Computer Systems Science & Engineering2023,46,9:0
16A Hybrid Attention-Based Residual Unet for Semantic Segmentation of Brain Tumor显示文摘Segmenting brain tumors in Magnetic Resonance Imaging(MRI)volumes is challenging due to their diffuse and irregular shapes.Recently,2D and 3D deep neural networks have become famous for medical image segmentation because of the availability of labelled datasets.However,3D networks can be computationally expensive and require significant training resources.This research proposes a 3D deep learning model for brain tumor segmentation that uses lightweight feature extraction modules to improve performance without compromising contextual information or accuracy.The proposed model,called Hybrid Attention-Based Residual Unet(HA-RUnet),is based on the Unet architecture and utilizes residual blocks to extract low-and high-level features from MRI volumes.Attention and Squeeze-Excitation(SE)modules are also integrated at different levels to learn attention-aware features adaptively within local and global receptive fields.The proposed model was trained on the BraTS-2020 dataset and achieved a dice score of 0.867,0.813,and 0.787,as well as a sensitivity of 0.93,0.88,and 0.83 for Whole Tumor,Tumor Core,and Enhancing Tumor,on test dataset respectively.Experimental results show that the proposed HA-RUnet model outperforms the ResUnet and AResUnet base models while having a smaller number of parameters than other state-of-the-art models.Overall,the proposed HA-RUnet model can improve brain tumor segmentation accuracy and facilitate appropriate diagnosis and treatment planning for medical practitioners.Wajiha Rahim Khan Tahir Mustafa Madni Uzair Iqbal Janjua Umer Javed Muhammad Attique Khan Majed Alhaisoni Usman Tariq Jae-Hyuk Cha 2023Computers, Materials & Continua2023,,7:0
17Tocilizumab in severe COVID-19-A randomized,double-blind,placebo-controlled trial显示文摘Background:The therapeutic effectiveness of interleukin-6 receptor inhibitor in critically ill hospitalized patients with coronavirus disease 2019(COVID-19)is uncertain.Methods:To evaluate the efficacy and safety of the outcome as recovery or death of tocilizumab for severe acute respiratory syndrome-coronavirus-2(SARS-CoV-2)infection,we conducted a randomized,double-blinded,placebo-controlled phase 2 trial in critically ill COVID-19 adult patients.The patients were randomly assigned in a 4:1 ratio to receive standard medical treatment plus the recommended dose of either tocilizumab or the placebo drug.Randomization was stratified.The primary outcome was the recovery or death after administration of tocilizumab or a placebo drug.The secondary outcomes were clinical recovery or worsening of the patients’symptoms and inflammatory markers and discharge from the hospital.Results:Of 190 patients included in this study,152 received tocilizumab,and 38 received a placebo.The duration of hospital stay of the interventional group was 12.9±9.2,while the placebo group had a more extended hospital stay(15.6±8.8).The mortality ratio for the primary outcome,ie,mortality or recovery in the tocilizumab group was 17.8%;p=0.58 by log-rank test).The mortality ratio in the placebo group was 76.3%;p=0.32 by log-rank test).The inflammatory markers in the tocilizumab group significantly declined by day 16 compared to the placebo group.Conclusions:The use of tocilizumab was associated with decreased mortality,earlier improvement of inflamma-tory markers,and reduced hospital stay in patients with severe COVID-19.Muhammad Irfan Malik Sardar Al Fareed Zafar Fabiha Qayyum Muna Malik Muhammad Sohaib Asghar Muhammad Junaid Tahir Ammarah Arshad Fatima Khalil Hafiza Shafia Naz Mudassar Aslam Jodat Saleem Abdul Aziz Mustafa Usman Azhar Muhammad Naqash Zohaib Yousaf 2022Infectious Medicine2022,1,2:0
