2The role of protein arginine methyltransferase 7 in human developmentally arrested embryos cultured in vitro显示文摘Human embryos of in vitro fertilization(IVF)are often susceptible to developmental arrest,which greatly reduces the efficiency of IVF treatment.In recent years,it has been found that protein arginine methyltransferase 7(PRMT7)plays an important role in the process of early embryonic development.However,not much is known about the relationship between PRMT7 and developmentally arrested embryos.The role of PRMT7 in developmentally arrested embryos was thus investigated in this study.Discarded human embryos from IVF were collected for experimental materials.Quantitative real-time polymerase chain reaction(qRT-PCR)and confocal analyses were used to identify PRMT7 mRNA and protein levels in early embryos at different developmental stages,as well as changes in the methylation levels of H4R3me2s.Additionally,PRMT7 was knocked down in the developmentally arrested embryos to observe the further development of these embryos.Our results demonstrated that PRMT7 mRNA and protein levels in arrested embryos were significantly increased compared with those in control embryos;meanwhile,the methylation levels of H4R3me2s in arrested embryos were also increased significantly.Knockdown of PRMT7 could rescue partially developmentally arrested embryos,and even individual developmentally arrested embryos could develop into blastocysts.In conclusion,over-expression of PRMT7 disrupts the early embryo development process,leading to early embryos developmental arrest,but these developmentally arrested defects could be partially rescued by knockdown of the PRMT7 protein.Wuwen Zhang Shifeng Li Kai Li Li Li Ping Yin Guoqing Tong 2021Acta Biochimica et Biophysica Sinica2021,53,7:2
3Interactions between gut microbiota and berberine,a necessary procedure to understand the mechanisms of berberine显示文摘Berberine(BBR),an isoquinoline alkaloid,has been found in many plants,such as Coptis chinensis Franch and Phellodendron chinense Schneid.Although BBR has a wide spectrum of pharmacological effects,its oral bioavailability is extremely low.In recent years,gut microbiota has emerged as a cynosure to understand the mechanisms of action of herbal compounds.Numerous studies have demonstrated that due to its low bioavailability,BBR can interact with the gut microbiota,thereby exhibiting altered pharmacological effects.However,no systematic and comprehensive review has summarized these interactions and their corresponding influences on pharmacological effects.Here,we describe the direct interactive relationships between BBR and gut microbiota,including regulation of gut microbiota composition and metabolism by BBR and metabolization of BBR by gut microbiota.In addition,the complex interactions between gut microbiota and BBR as well as the side effects and personalized use of BBR are discussed.Furthermore,we provide our viewpoint on future research directions regarding BBR and gut microbiota.This review not only helps to explain the mechanisms underlying BBR activity but also provides support for the rational use of BBR in clinical practice.Hao Cheng Juan Liu Yuzhu Tan Wuwen Feng Cheng Peng 2022Journal of Pharmaceutical Analysis2022,12,4:1

14CDX2 is essential for human IVF early embryonic development显示文摘Human in vitro fertilization (IVF)embryos usually have developmental block at 4-8 cell stages,in which only 30%-50% percent of IVF embryos develop to blastocyst stage.This is the main cause for low efficiency and repeated treatment failure in human fertility therapy.Transcription factor CDX2 is one of the key genes in human early embryonic development,which decides the differentiation from blastomere to trophoblastic lineage.However,the relationship between the developmental block of human IVF embryos and CDX2 is poorly understood.In this study,we showed that CDX2 is crucial for human IVF embryonic development,q-PCR analysis confirmed that the level of CDX2 in developmental block embryos was dramatically decreased.Immunofluorescence analysis further demonstrated that CDX2 protein level in developmental block embryos was significantly decreased compared with that in control embryos.We also addressed whether reexpression of CDX2 could rescue developmental block defects in human IVF developmental block embryos,As expected,these developmental block defects could be partially rescued by overexpression of CDX2 protein.In conclusion,these findings suggest that CDX2 plays an important role in the process of human early embryonic development.Wuwen Zhang Kai Li Xiufang Zhong Hua Yan Guoqing Tong 2018Acta Biochimica et Biophysica Sinica2018,50,12:1

17An Investigation into the Compressive Strength,Permeability and Microstructure of Quartzite-Rock-Sand Mortar显示文摘River sand is an essential component used as a fine aggregate in mortar and concrete.Due to unrestrained exploitation,river sand resources are gradually being exhausted.This requires alternative solutions.This study deals with the properties of cement mortar containing different levels of manufactured sand(MS)based on quartzite,used to replace river sand.The river sand was replaced at 20%,40%,60%and 80%with MS(by weight or volume).The mechanical properties,transfer properties,and microstructure were examined and compared to a control group to study the impact of the replacement level.The results indicate that the compressive strength can be improved by increasing such a level.The strength was improved by 35.1%and 45.5%over that of the control mortar at replacement levels of 60%and 80%,respectively.Although there was a weak link between porosity and gas permeability in the mortars with manufactured sand,the gas permeability decreased with growing the replacement level.The microstructure of the MS mortar was denser,and the cement paste had fewer microcracks with increasing the replacement level.Wei Chen Wuwen Liu Yue Liang 2024Fluid Dynamics & Materials Processing2024,20,4:0

19Prmt7 regulates epiboly and gastrulation ce movements by facilitating syntenin显示文摘Epiboly spreads and thins the blastoderm over the yolk cell during zebrafish gastrulation.Despite of its fundamental function,little is known about the molecular mechanisms that control this coordinated cell movement.In this study,we investigated protein arginine methyltransferase 7 (PrmtT)morphants with an epibolic delay defect in zebrafish.The ratio of morphants with epiboly delay phenotypes increased as the dose of the injected morpholino (MO)increased.Here,syntenin transcripts are maternally deposited and ubiquitously expressed from the oocyte period to the early larva stage.Furthermore,we demonstrated that Prmt7 modulates epibolic movements of the enveloping layer by regulating F-actin organization.These defects can be partially rescued by re-expression of Prmt7 or syntenin protein. Analysis of the earliest cellular defects suggested a role of Prmt7 in the autonomous vegetal expansion of the yolk syncytial layer and the rearrangement of the actin cytoskeleton in extra-embryonic tissues. 20Measurement of serum granulocyte colonystimulating factor levels in patients with different phase of infection显示文摘Objective: To detect the serum granulocyte colony-stimulating factor (G-CSF) levels between the patients with frequently repeated infection (repeaters) and others (non-repeaters) in different phase of infection. Methods: An enzyme-linked immunosorbent assay (ELISA) method was used to detect serum G-CSF levels in 50 cases (32 non-repeaters and 18 repeaters) with acute phase of infection. Serum G-CSF levels were detected in recovery phase in 10 cases. Results: Serum G-CSF levels were significantly higher (1429.97±506.43ng/L) in 32 non-repeaters with acute infection. There was a positive correlation between white blood cell count (WBC) and serum G-CSF level (r =0. 396, P <0.05). There was also a positive correlation between absolute neutrophil count (ANC) and serum G-CSF level (r=0.346,P<0,05). Serum G-CSF levels were higher (98.62 -+56.40ng/L) in 18 repeaters with acute infection, It was showed that serum G-CSF levels were significantly higher in non-repeaters than in repeaters with acute phase of infection ( P <0. 001 ). In the meanwhile, the body temperature was significantly higher in non-repeaters than in repeaters with acute infection (37.95±0.14℃ rs 36.91±0.13℃ ,P <0.001), There were no significant differences in age, WBC, ANC, type of bacterial, liver function and renal function (P > 0.05).
