Alcohol, inflammation, and gut-liver-brain interactions in tissue damage and disease development显示文摘Chronic inflammation is often associated with alcoholrelated medical conditions. The key inducer of such inflammation, and also the best understood, is gut microflora-derived lipopolysaccharide (LPS). Alcohol can significantly increase the translocation of LPS from the gut. In healthy individuals, the adverse effects of LPS are kept in check by the actions and interactions of multiple organs. The liver plays a central role in detoxifying LPS and producing a balanced cytokine milieu. The central nervous system contributes to anti-inflammatory regulation through neuroimmunoendocrine actions. Chronic alcohol use impairs not only gut and liver functions, but also multi-organ interactions, leading to persistent systemic inflammation and ultimately, to organ damage. The study of these interactions may provide potential new targets for therapeutic intervention.H Joe Wang Samir Zakhari M Katherine Jung 2010World Journal of Gastroenterology2010,16,11:26
Nature-inspired chiral metasurfaces for circular polarization detection and full-Stokes polarimetric measurements显示文摘The manipulation and characterization of light polarization states are essential for many applications in quantum communication and computing, spectroscopy, bioinspired navigation, and imaging.Chiral metamaterials and metasurfaces facilitate ultracompact devices for circularly polarized light generation, manipulation, and detection.Herein,we report bioinspired chiral metasurfaces with both strong chiral optical effects and low insertion loss.We experimentally demonstrated submicron-thick circularly polarized light filters with peak extinction ratios up to 35 and maximum transmission efficiencies close to 80% at near-infrared wavelengths(the best operational wavelengths can be engineered in the range of 1.3-1.6μm).We also monolithically integrated the microscale circular polarization filters with linear polarization filters to perform full-Stokes polarimetric measurements of light with arbitrary polarization states. With the advantages of easy on-chip integration, ultracompact footprints, scalability, and broad wavelength coverage, our designs hold great promise for facilitating chip-integrated polarimeters and polarimetric imaging systems for quantum-based optical computing and information processing, circular dichroism spectroscopy, biomedical diagnosis, and remote sensing applications.Ali Basiri Xiahui Chen Jing Bai Pouya Amrollahi Joe Carpenter Zachary Holman Chao Wang Yu Yao 2019Light(Science & Applications)2019,8,1:12
On the mechanism and control for the ultra-low frequency oscillation in NY Power Grid with large-scale hydropower显示文摘The power grid with large proportion of hydropower may engage the ultra-low frequency oscillation (ULFO),which threatening the operation of system. To the NY Power Grid system, whose dimension is over thousands, this paper reveals the cause of ULFO with the phase portrait method,i.e.,lies in the saturation of the different limits of the governor system of the generators,in other words, the occurrence of limit cycle is due to the non-smooth bifurcation. And furthermore,it proposes two methods to suppress ULFO with the philosophy of destroying the condition of non-smooth bifurcation. In detail, first, the phenomenon of ULFO in NY Power Grid, the damping characteristics and the limits in the governor system of the hydropower units are presented.Second, based on the non-smooth bifurcation theory, the mathematical basis of the oscillation in the non-smooth system caused by the saturation of limits is introduced. And then, the non-oscillation and oscillation dynamic of the system corresponding to without/with limits in the governor system are analyzed.Furthermore,it discusses the influence of the saturation of single limit on the characteristics of ULFO with the phase portraits. Moreover, the phase portraits of the ULFO under different faults is analyzed, and it shows that the limiting saturation occurs with a large enough disturbance. Thus, the ULFO is resulting from the negative damping of generation and the saturation of multiple limits under a large enough disturbance. In addition, according to the above mechanism, two strategies to suppress the ULFO,i.e.,the governor parameters tuning and the fault strength relief are designed. The simulation shows the effectiveness of proposed strategies.Ancheng XUE Jiawei WANG Chao ZHENG Joe H.CHOW Tianshu BI 2019Control Theory and Technology2019,17,3:11
Magnetic resonance imaging in personalized medicine显示文摘Continued biomedical advances and increased demands on quality health care have led to a new era of personalized medicine—a concept of medicine that uses specific information to an individual to help diagnose disease, plan treatment, assess treatment efficacy,and/or predict prognosis. This concept has evolved from the idea of'patient-centered care',which intends to shift the focus of health care from diseases to patients(Abujudeh et al.,2016).Medical imaging is essential in the practice of modern medicine, and its role in personalized medicine has never been greater. In particular, magnetic resonance(MR)Xiaoying Wang Xiaohong Joe Zhou 2017Science China(Life Sciences)2017,60,1:6

Temporal changes in inflammatory mitochondria-enriched microRNAs following traumatic brain injury and effects of miR-146a nanoparticle delivery显示文摘MicroRNAs(miRNAs)are small non-coding RNA molecules that regulate post-transcriptional gene expression and contribute to all aspects of cellular function. We previously reported that the activities of several mitochondria-enriched miRNAs regulating inflammation(i.e.,miR-142-3p,miR-142-5p,and miR-146a)are altered in the hippocampus at 3–12 hours following a severe traumatic brain injury. In the present study,we investigated the temporal expression profile of these inflammatory miRNAs in mitochondria and cytosol fractions at more chronic post-injury times following severe controlled cortical impact injury in rats.In addition, several inflammatory genes were analyzed in the cytosol fractions. The analysis showed that while elevated levels were observed in cytoplasm, the mitochondria-enriched miRNAs,miR-142-3p and miR-142-5p continued to be significantly reduced in mitochondria from injured hippocampi for at least 3 days and returned to near normal levels at 7 days post-injury.Although not statistically significant,miR-146a also remained at reduced levels for up to 3 days following controlled cortical impact injury, and recovered by 7 days.In contrast,miRNAs that are not enriched in mitochondria, including miR-124a,miR-150,miR-19b,miR-155,and miR-223 were either increased or demonstrated no change in their levels in mitochondrial fractions for 7 days. The one exception was that miR-223 levels were reduced in mitochondria at 1 day following injury. No major alterations were observed in sham operated animals. This temporal pattern was unique to mitochondria-enriched miRNAs and correlated with injury-induced changes in mitochondrial bioenergetics as well as expression levels of several inflammatory markers. These observations suggested a potential compartmental re-distribution of the mitochondria-enriched inflammatory miRNAs and may reflect an intracellular mechanism by which specific miRNAs regulate injury-induced inflammatory signaling. To test this,we utilized a novel peptide-based nanoparticle strategy for in vitro and in vivo delivery of a miR-146a mimic as a potential therapeutic strategy for targeting nuclear factor-kappa B inflammatory modulators in the injured brain.Nanoparticle delivery of miR-146a to BV-2 or SH-SY5Y cells significantly reduced expression of TNF receptor-associated factor 6(TRAF6)and interleukin-1 receptor-associated kinase 1(IRAK1),two important modulators of the nuclear factor-kappa B(NF-κB)pro-inflammatory pathway. Moreover, injections of miR-146a containing nanoparticles into the brain immediately following controlled cortical impact injury significantly reduced hippocampal TNF receptor-associated factor 6 and interleukin-1 receptor-associated kinase 1 levels.Taken together, our studies demonstrate the subcellular alteration of inflammatory miRNAs after traumatic brain injury and establish proof of principle that nanoparticle delivery of miR-146a has therapeutic potential for modulating pro-inflammatory effectors in the injured brain. All of the studies performed were approved by the University of Kentucky Institutional Animal Care and Usage Committee(IACUC protocol#2014-1300)on August 17,2017.Wang-Xia Wang Paresh Prajapati Hemendra J.Vekaria Malinda Spry Amber L.Cloud Patrick G.Sullivan Joe E.Springer 2021Neural Regeneration Research2021,16,3:3

Application and evaluation of Stanghellini model in the determination of crop evapotranspiration in a naturally ventilated greenhouse显示文摘Stanghellini model is one of the few models primarily developed to predict the evapotranspiration of crops(ET_(c))in naturally ventilated greenhouses. However, there are insufficient data on the model regarding its use, particularly in China where solar greenhouses without heating systems are fast spreading for vegetable growth and production. The application of Stanghellini model and the evaluation of its performance using meteorological and tomato plant data generated inside an unheated and naturally ventilated multi-span Venlo-type greenhouse is exploited in this study.Model capability was evaluated by utilizing data from sap flow measurements, meteorological and crop data.Measured meteorological data included solar radiation(R_(s)),air temperature(T_(a)),relative humidity(RH)and net radiation(Rn).Average leaf area index(LAI)values measured during the experimental period were 1.00,3.30,4.05 and 2.93;while determined crop coefficients(K_(c))changed from 0.40,0.62,1.12 to 0.83 for the initial stage, development stage,mid-season stage and late-season stage, respectively. Results from the study indicated that the average hourly ET_(c) values of tomato plants using sap flow measurements were 0.165 mm/h,0.148 mm/h,0.192 mm/h and 0.154 mm/h for the initial stage, development stage,mid-season stage and late-season stage, respectively.Meanwhile, the ET_(c) values obtained from calculation using Stanghellini model were 0.158 mm/h,0.152 mm/h,0.202 mm/h and 0.162 mm/h for the initial stage, development stage,mid-season stage and late-season stage, respectively. These ET_(c) values calculated by the Stanghellini model were close to the measured values within the same period. The coefficients of correlation(R^(2))based on hourly ET_(c) for the calibration data was 0.94 and that of the validation dataset was 0.90.Scatter plots of the estimated and measured hourly ET_(c) revealed that the R^(2) and the slope of the regression line for May,June and July were 0.94,0.90,0.96 and 1.15,0.97,1.10 respectively. These data were well represented around the 1:1 regression line.A model sensitivity analysis carried out illustrates how the changes in R_(s) and T_(a) affect greenhouse ET_(c).Stanghellini model was therefore proven to be suitable for ET_(c) estimation with acceptable accuracy in unheated and naturally ventilated greenhouses in the Northeast region of China.Samuel Joe Acquah Haofang Yan Chuan Zhang Guoqing Wang Baoshan Zhao Haimei Wu Hengnian Zhang 2018International Journal of Agricultural and Biological Engineering2018,11,6:3

Role of mitochondria in regulating micro RNA activity and its relevance to the central nervous system显示文摘Mitochondria serve as the powerhouse of cells, respond to cellular demands and stressors, and play an essential role in cell signaling, differentiation, and survival.Aberrant mitochondria function has been linked to diverse and complex human diseases such as neurodegenerative diseases, cancers, myopathies, premature aging, and metabolic syndromes(Nunnari and Suomalainen,2012).For example, there is clear evidence of a rapid and sustained loss of mitochondrial function following traumatic injury to the central nervous system(CNS)(Sullivan et al.,2005;Mc Ewen et al.,2011).However,our understanding of how compromised mitochondrial activity impacts overall cellular function in the CNS is far from complete.Wang-Xia Wang Joe E.Springer 2015Neural Regeneration Research2015,10,7:2

On brain activity mapping: Insights and lessons from Brain Decoding Project to map memory patterns in the hippocampus显示文摘The BRAIN project recently announced by the president Obama is the reflection of unrelenting human quest for cracking the brain code, the patterns of neuronal activity that define who we are and what we are. While the Brain Activity Mapping proposal has rightly emphasized on the need to develop new technologies for measuring every spike from every neuron, it might be helpful to consider both the theoretical and experimental aspects that would accelerate our search for the organizing principles of the brain code. Here we share several insights and lessons from the similar proposal, namely, Brain Decoding Project that we initiated since 2007. We provide a specific example in our initial mapping of real-time memory traces from one part of the memory circuit, namely, the CA1 region of the mouse hippocampus. We show how innovative behavioral tasks and appropriate mathematical analyses of large datasets can play equally, if not more, important roles in uncovering the specific-to-general feature-coding cell assembly mechanism by which episodic memory, semantic knowledge, and imagination are generated and organized. Our own experiences suggest that the bottleneck of the Brain Project is not only at merely developing additional new technologies, but also the lack of efficient avenues to disseminate cutting edge platforms and decoding expertise to neuroscience community. Therefore, we propose that in order to harness unique insights and extensive knowledge from various investigators working in diverse neuroscience subfields, ranging from perception and emotion to memory and social behaviors, the BRAIN project should create a set of International and National Brain Decoding Centers at which cutting-edge recording technologies and expertise on analyzing large datasets analyses can be made readily available to the entire community of neuroscientists who can apply and schedule to perform cutting-edge research.TSIEN Joe Z.
