| 1 | Humification degrees of peat in Qinghai-Xizang Plateau and palaeoclimate change显示文摘In this article we report a new and sensitive palaeoclimate proxy indieator-humification degrees of peat. Based on the comparison of humification degrees with other climate proxy records, such as δ^13C time series of the C. mulieensis remains cellulose in the same peat profile, we suggest that humifieation degrees of peat in Qinghai-Xizang Plateau can served as a palaeoclimate proxy indicator. The higher the hnmification degrees of peat, the warmer-wetter the climate; on the contrary, the lower the humification degrees, the colder-drier the climate. | 2 | Effects of transforming growth factor beta 1 on the growth of rhabdomyosarcoma cell line RD显示文摘Backgroud Transforming growth factor-β (TGF-β) can inhibit the growth of most epithelial and endothelial cells. The growth regulative role of TGF-β on soft tissue sarcoma was seldom reported. Here we examined TGF-β1 effects on the growth of human rhabdomyosarcoma cell RD and searched the relative molecular mechanism. Methods The viability of RD was examined by [ 3H]-thymidine incorporation and [3-(4,5-dimethylthiazol-z-yl)-2,5-diphenyltetraz olium bromide] (MTT) assay. RD cell cycle was analysed by flow cytometry. The protein and mRNA of cell cycle regulative factors in RD were detected by Western blot and reverse transcription-polymerase chain reation (RT-PCR), respectively. The kinase activity of cdk2 or cdk4 was examined by immunoprecipitation and kinase assay. Immunofluorescent staining was used to detect the location of cell cycle regulative factors in RD by laser scanning confocal microscope.Results TGF-β1 inhibits RD proliferation by G1-arrest in cell cycle progression. TGF-β1 can prominently up-regulate P27 of RD, then augment P27 to bind cyclinE-cdk2 complexes, which effectively suppress cdk2 kinase activity. P21 increased and c-myc decreased in RD due to TGF-β1. Both P15 and cdk4 have not been involved in the growth inhibitory event. TGF-β1 treatment induced P27 to congregate around nucleus. P21 pervaded from nucleus to both nucleus and cytoplasm by TGF-β1 treatment.Conclusion TGF-β1 inhibits the proliferation of human rhabdomyosarcoma cell line RD and induces RD G1-arrest. This course is accomplished by TGF-β1 up-regulating P27 to suppress cdk2 kinase activity. | 7 | Development and Validation of a Novel Reverse Phase HPLC for Determination of Aloperine in Aloperine Tablets显示文摘Aim To develop a reverse phase HPLC method for the determination of aloperine, an alkaloid that is newly extracted from Sophora alopecuraides and has shown wide pharmacological effects including antibacterial and antimflammatory actions. Methods The samples were analyzed on a ODS column with methanol-water-triethylamine (3:97:0.1 V/V) as a mobile phase, The flow rate was 1.0 mL·min^-1, and UV detection wavelength 205 nm. Results Linear regression equation was A = 1.6920C + 1.7455 (r^2 = 0.9999, n = 5) in concentratim ranging from 20 to 120μg·mL^-1. The recoveries were 101.2±1.46 % at 80 μg·mL^-1, 100.5±0.75% at 100μg·mL^-1, and 100.7±1.10% at 120 μg·mL^-1, respectively, and the precisions of aloperine within or between run were from 0.80% to 1.98% ( n = 5). | 8 | Notch signaling stimulates osteogenic differentiation of human bone marrow-derived mesenchymal stem cells显示文摘Notch signaling is one of the most important pathways mediating cell determination and differentiation. In this study, the roles of Notch signal in the regulation of osteogenic differentiation of human bone marrow mesen-chymal stem cells (hMSCs) were investigated. The expression of Notch1, Jagged1 and DTX1 detected by reverse transcrip-tion polymerase chain reaction (RT-PCR) suggested that Notch signal might exhibit a physiological regulatory role in the differentiation of MSCs. Constitutive expression of the intracellular domain of Notch1 (ICN), the active form of Notch1 protein, can activate Notch signal in cells without ligands binding. hMSCs were isolated, expanded, and in-fected with retrovirus carrying green fluorescent protein (GFP) gene or ICN. Overexpression of ICN in hMSCs re-sulted in enhanced osteogenic differentiation induced by dexamethasone (Dex), which was characterized by an in-crease of cellular alkaline phosphatase (ALP) activity and calcium deposition. | 9 | Thermodynamic Analysis and Experimental Investigation into Nonflame Combustion Technology(NFCT) with Thermal Cyclic Carrier显示文摘The utilization of fossil fuels causes serious negative impacts on the environment and human life. To mitigate greenhouse gases and other pollutants, a novel combustion process-the nonflame combustion technology with a thermal cyclic carrier of molten salt is introduced. In this technology, a whole combustion is divided into two steps, i.e., the section of producing oxide and the section of combustion. In the first step, oxygen is separated from air, and pure N2 is simultaneously formed which is easily recovered. In the other step, the fuels react with lattice oxygen in the oxides formed in the first step, and at the same time, thermal energy, CO2 and H2O vapor are produced. It is noted that the CO2 is easily separated from water vapor and ultimately captured. Theoretically, there are no environmental-unfriendly gases such as CO2, NOx and SO2 discharged in the whole combustion process. Some metal oxides scattered into molten salts play the roles of oxygen carriers in the combustion system, and they can constantly charge and discharge oxygen element from air to fuels during the combustion process. A nonflame combustion system with Li2CO3+K2CO3+Na2SO4 as the molten salt system, CH4 as the fuel and CuO as the catalyst was experimentally investigated. The experimental results show that the combustion process proceeded as it was theoretically analyzed, and CO2 with a high volume fraction of 77.0%95.0% and N2 with a high volume fraction of 91.9%99.3% were obtained. The high concentration of CO2 is favorable for capturing and storing subsequently. | 14 | Electrochemical behavior of Cu-Zn-Al shape memory alloy after surface modification by electroless plated Ni-P显示文摘The electrochemical behavior of Cu-Zn-Al shape memory alloy (SMA) with and without electroless plated Ni-P was investigated by electrochemical methods in artificial Tyrode's solution. The results showed that Cu-Zn-Al SMA engendered dezincification corrosion in Tyrode's solution. The anodic active current densities as well as electrochemical dissolution sensitivity of the electroless plated Ni-P Cu-Zn-Al SMA increased with NaCl concentration rising, pH of solution decreasing and environmental temperature uprising. X-ray diffraction analysis indicated that after surface modification by electroless plated Ni-P, an amorphous plated film formed on the surface of Cu-Zn-Al SMA.
