1Activation of extrasynaptic GABA_A receptors inhibits cyclothiazideinduced epileptiform activity in hippocampal CA1 neurons显示文摘Extrasynaptic GABA_A receptors(GABA_ARs)-mediated tonic inhibition is reported to involve in the pathogenesis of epilepsy.In this study,we used cyclothiazide(CTZ)-induced in vitro brain slice seizure model to explore the effect of selective activation of extrasynaptic GABA_ARs by 4,5,6,7-tetrahydroisoxazolo[5,4-c]pyridine-3-ol(THIP)on the CTZ-induced epileptiform activity in hippocampal neurons.Perfusion with CTZ dose-dependently induced multiple epileptiform peaks of evoked population spikes(PSs)in CA1 pyramidal neurons,and treatment with THIP(5 umol/L)significantly reduced the multiple PS peaks induced by CTZ stimulation.Western blot showed that the 5-subunit of the GABA_AR,an extrasynaptic specific GABA_AR subunit,was also significantly down-regulated in the cell membrane 2h after CTZ treatment.Our results suggest that the CTZ-induced epileptiform activity in hippocampal CA1neurons is suppressed by the activation of extrasynaptic GABA_ARs,and further support the hypothesis that tonic inhibition mediated by extrasynaptic GABA_ARs plays a prominent role in seizure generation.Li Wan Xu Liu Zheng Wu Wanting Ren Shuzhen Kong Raya Abou Dargham Longzhen Cheng Yun Wang 2014Neuroscience Bulletin2014,30,5:3
2Laser surface treatment-introduced gradient nanostructured TiZrHfTaNb refractory high-entropy alloy with significantly enhanced wear resistance显示文摘Heterogeneous gradient nanostructured metals have been shown to achieve the strength-ductility synergy, thus potentially possessing the enhanced tribological performance in comparison with their homogeneous nanograined counterparts. In this work, a facile laser surface remelting-based surface treatment technique is developed to fabricate a gradient nanostructured layer on a TiZrHfTaNb refractory highentropy alloy. The characterization of the microstructural evolution along the depth direction from the matrix to the topmost surface layer shows that the average grain size in the ~100 μm-thick gradient nanostructured layer is dramatically refined from the original ~200 μm to only ~8 nm in the top surface layer. The microhardness is therefore gradually increased from ~240 HV in matrix to ~650 HV in the topmost surface layer, approximately 2.7 times. Noticeably, the original coarse-grained single-phase bodycentered-cubic TiZrHfTaNb refractory high-entropy alloy is gradually decomposed into TiNb-rich bodycentered-cubic phase, TaNb-rich body-centered-cubic phase, ZrHf-rich hexagonal-close-packed phase and TiZr Hf-rich face-centered-cubic phase with gradient distribution in grain size along the depth direction during the gradient refinement process. As a result, the novel laser surface treatment-introduced gradient nanostructured TiZrHfTaNb refractory high-entropy alloy demonstrates the significantly improved wear resistance, with the wear rate reducing markedly by an order of magnitude, as compared with the as-cast one. The decomposed multi-phases and gradient nanostructures should account for the enhanced wear resistance.
