| 1 | Human placenta-derived mesenchymal stem cells loaded on linear ordered collagen scaffold improves functional recovery after completely transected spinal cord injury in canine显示文摘Traumatic spinal cord injury(SCI) is a major challenge in the clinic. In this study, we sought to examine the synergistic effects of linear ordered collagen scaffold(LOCS) and human placenta-derived mesenchymal stem cells(hPMSCs) when transplanted into completely transected beagle dogs. After 36 weeks observation, we found that LOCS+hPMSCs implants promoted better hindlimb locomotor recovery than was observed in the non-treatment(control) group and LOCS group. Histological analysis showed that the regenerated tissue after treatment was well integrated with the host tissue, and dramatically reduced the volume of cystic and chondroitin sulfate proteoglycans(CSPGs) expression. Furthermore, the LOCS+hPMSCs group also showed more neuron-specific βIII-tubulin(Tuj-1)-and NeuN-positive neurons in the lesion area, as well as axonal regeneration, remyelination and synapse formation in the lesion site. Additionally, dogs in the LOCS+hPMSCs group experienced enhanced sprouting of both ascending(CGRP-positive) sensory fibers and descending(5-HT-and TH-positive) motor fibers at the lesion area. All these data together suggested that the combined treatment had beneficial effects on neuronal regeneration and functional improvement in a canine complete transection model. | 3 | Effects of dissolved organic matter from sewage sludge on the atrazine sorption by soils显示文摘The effects of dissolved organic matter (DOM), water soluble organic matter derived from sewage sludge, on the sorption of atrazine (2-chloro-4-ethylamino-6-isopropylamino-1,3,5-trazine) by soils were studied using a batch equilibrium technique. Six paddy soils, chosen so as to have different organic carbon contents, were experimented in this investigation. Atrazine sorption isotherms on soils were described by the linear equation, and the distribution coefficients without DOM (Kd) or with DOM (Kd*) were obtained. Generally, the values of Kd*/Kd initially insoil-solution system form. Critical concentrations of DOM (DOMnp) were obtained where the value of Kd* was equal to Kd. The presence of DOM with concentrations lower than DOMnp promoted atrazine sorption on soils (Kd* > Kd), whereas the presence of DOM with concentrations higher than DOMnp tended to inhibit atrazine sorption (Kd* < Kd). Interestingly, DOMnp for tested soils was negatively correlated to the soil organic carbon content, and the maximum of Kd*/Kd (i.e.Kmax) correlated positively with the maximum of DOM sorption on soil (Xmax). Further investigations showed that the presence of hydrophobic fraction of DOM evidently promoted the atrazine sorption on soils, whereas the presence of hydrophilic DOM fraction obviously tended to inhibit the atrazine sorption. Interactions of soil surfaces with DOM and its fractions were suggested to be the major processes determining atrazine sorption on soils. | 4 | The Inhibitory Effect of Quercetin on IL-6 Production by LPS-Stimulated Neutrophils显示文摘Quercetin is a herbal flavonoid derived from various foods of plant origin and plays a role in anti-inflammation. Although a number of researches in the field have been done, the mechanism of anti-inflammatory effect of quercetin should be further clarified. In the present study, we investigated the effects of quercetin on IL-6 production by LPS-stimulated neutrophils in human. Neutrophils were were pre-treated with quercetin at the final concentrations of ranging from 0-80 μM for 30 min, or not treated, and then incubated in the presence or absence of lipopolysaccharide (LPS) at a final concentration of 100 ng/ml for indicated time. The secretion level of IL-6 in the culture supernatants was assayed by ELISA, the intracellular level of IL-6 was detected by flow cytometry and the expression of IL-6 mRNA was analyzed by RT-PCR. The experiment results showed that neutrophils cultured with medium or quercetin alone did not express IL-6, but LPS (100 ng/ml) induced IL-6 expression of neutrophils. However, after pre-treatment of neutrophils with quercetin (40 μM) for 30 min, the inducible effects of LPS on the increase of IL-6 secretion, intracellular IL-6 level and IL-6 mRNA expression by neutrophils were abrogated. IL-6 is one of the important pro-inflammatory factors, especially in early phage of inflammation. Thus, our data suggested that quercetin might exert its anti-inflammatory effect through negatively modulating pro-inflammatory factors, such as IL-6. | 5 | Analysis of Synonymous Codon Usage Bias in Chlamydia显示文摘Chlamydiae are obligate intracellular bacterial pathogens that cause ocular and sexually transmitted diseases, and are associated with cardiovascular diseases. The analysis of codon usage may improve our understanding of the evolution and pathogenesis of Chlamydia and allow reengineering of target genes to improve their expression for gene therapy. Here, we analyzed the codon usage of C. muridarum, C. trachomatis (here indicating biovar trachoma and LGV), C. pneumoniae, and C. psittaci using the codon usage database and the CUSP (Create a codon usage table) program of EMBOSS (The European Molecular Biology Open Software Suite). The results show that the four genomes have similar codon usage patterns, with a strong bias towards the codons with A and T at the third codon position. Compared with Homo sapiens, the four chlamydial species show discordant seven or eight preferred codons. The ENC (effective number of codons used in a gene)-plot reveals that the genetic heterogeneity in Chlamydia is constrained by the G+C content, while translational selection and gene length exert relatively weaker influences. Moreover, mutational pressure appears to be the major determinant of the codon usage variation among the chlamydial genes. In addition, we compared the codon preferences of C. trachomatis with those of E. coli, yeast, adenovirus and Homo sapiens. There are 23 codons showing distinct usage differences between C. trachomatis and E. coli, 24 between C. trachomatis and adenovirus, 21 between C. trachomatis and Homo sapiens, but only six codons between C. trachomatis and yeast. Therefore, the yeast system may be more suitable for the expression of chlamydial genes. Finally, we compared the codon preferences of C. trachomatis with those of six eukaryotes, eight prokaryotes and 23 viruses. There is a strong positive correlation between the differences in coding GC content and the variations in codon bias (r=0.905, P<0.001). | 6 | Characterization and immunogenicity of bone marrow-derived mesenchymal stem cells under osteoporotic conditions显示文摘Mesenchymal stem cells(MSCs)are characterized by their multilineage potential and low immunogenicity.However,the properties of MSCs under pathological conditions are unclear.The current study investigated the differentiation potential and immunological characteristics of bone marrow-derived MSCs from ovariectomized-osteoporotic rats(OP-BMSCs).Although the expression of cell morphology-and stemness-related surface markers was similar between OP-BMSCs and BMSCs from healthy rats(H-BMSCs),the proliferation rate was significantly decreased compared with that of H-BMSCs.Regarding multilineage potential,osteogenesis and chondrogenesis abilities of OP-BMSCs decreased,but the adipogenesis ability was significantly enhanced compared with that of H-BMSCs.As expected,decreased osteogenesis following osteogenic induction resulted in reduced expression ofβ-catenin,osteocalcin,and runt-related transcription factor 2 in OP-BMSCs.Remarkably,the expression of the co-stimulatory proteins CD40 and CD80 was significantly higher,whereas the expression of the negative costimulatory molecule programmed cell death ligand 1 was significantly lower in the OP-BMSCs than that in H-BMSCs.Consequently,H-BMSCs inhibited the proliferation and secretion of inflammatory cytokines from anti-CD3 antibody-activated T cells,whereas OP-BMSCs did not.These results indicate that decreased osteogenesis and increased immunogenicity of OPBMSCs contribute to bone loss in osteoporosis. | Yingkang Huang Yin Yin Yanzheng Gu Qiaoli Gu Huilin Yang Zhengyu Zhou Qin Shi | 2020 | Science China(Life Sciences)2020,63,3: | 4 |

The use of traditional methods by military experts to evaluate large scale tasks incurs significant time cost and results in low accuracy, and is caused by a variety of factors that cause confusion. Therefore, we developed a system to assist military personnel in improving the efficiency of mission evaluation; the main innovations of our work include the qualitative and quantitative visualization of complex information is realized in a three-pane interface. We also realize the iterative and interactive evaluation modes of large-scale tasks by using the active learning method; moreover, the overall display of large-scale task evaluation results is realized using statistical graphics. | 10 | Interesting effects in harmonic generation by plane elastic waves显示文摘The harmonics of plane longitudinal and transverse waves in nonlinear elastic solids with up to cubic nonlinearity in a one-dimensional setting are investigated in this paper. It is shown that due to quadratic nonlinearity, a transverse wave generates a second longitudinal harmonic.This propagates with the velocity of transverse waves, as well as resonant transverse first and third harmonics due to the cubic and quadratic nonlinearities. A longitudinal wave generates a resonant longitudinal second harmonic, as well as first and third harmonics with amplitudes that increase linearly and quadratically with distance propagated. In a second investigation, incidence from the linear side of a primary wave on an interface between a linear and a nonlinear elastic solid is considered. The incident wave crosses the interface and generates a harmonic with interface conditions that are equilibrated by compensatory waves propagating in two directions away from the interface. The back-propagated compensatory wave provides information on the nonlinear elastic constants of the material behind the interface. | 11 | Integrating a model with remote sensing observations by a data assimilation approach to improve the model simulation accuracy of carbon flux and evapotranspiration at two flux sites显示文摘Model simulation and in situ observations are often used to research water and carbon cycles in terrestrial ecosystems, but each of these methods has its own advantages and limitations. Combining these two methods could improve the accuracy of quantifying the dynamics of the water and carbon fluxes of an ecosystem. Data assimilation is an effective means of integrating modeling with in situ observation. In this study, the ensemble Kalman filter(En KF) and the unscented Kalman filter(UKF) algorithms were used to assimilate remotely sensed leaf area index(LAI) data with the Biome-BGC model to simulate water and carbon fluxes at the Harvard Forest Environmental Monitoring Site(EMS) and the Dinghushan site. After MODIS LAI data from 2000–2004 were assimilated into the improved Biome-BGC model using the En KF algorithm at the Harvard Forest site, the R2 between the simulated and observed results for NEE and evapotranspiration increased by 7.8% and 4.7%, respectively. In addition, the sum of the absolute error(SAE) and the root mean square error(RMSE) of NEE decreased by an average of 21.9% and 26.3%, and the SAE and RMSE of evapotranspiration decreased by 24.5% and 25.5%, respectively. MODIS LAI data of 2003 were assimilated into the Biome-BGC model for the Dinghushan site, and the R2 values between the simulated and observed results for NEE and evapotranspiration were increased by 6.7% and 17.3%, respectively. In addition, the SAE values of NEE and ET were decreased by 11.3% and 30.7%, respectively, and the RMSE values of NEE and ET decreased by 10.1% and 30.9%, respectively. | 14 | Benefits of redundant channels in observer-based H∞ control for discrete-time switched linear systems显示文摘In this paper, the H∞ control problem is investigated for a class of discrete-time switched linear systems with modal persistent dwell-time(MPDT) switching. The redundant channels are considered to use in the data transmission to benefit the capability of overcoming the fragility of networks commonly configured by a single channel in the communication networks subject to random packet losses. In light of a new class of Lyapunov functions, the desired observer-based quasi-time-dependent controllers, which have less conservatism than the time-independent ones, are designed such that the resulting closed-loop system is exponentially mean-square stable with a guaranteed H_∞ disturbance attenuation performance. The MPDT can be minimized while ensuring the existence of such a class of observer-based controllers for a given period of persistence.
