| 1 | A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. | 2 | A preliminary study on measurements of black carbon in the atmosphere of northwest Qilian Shan显示文摘Black carbon (BC) concentration and meteorological data are measured discontinuously from May 2009 to March 2011,at the Qilian Shan Station of Glaciology and Ecologic Environment (hereafter 'QSSGEE'),located near the terminal of the Laohugou No.12 Glacier in northwestern Qilian Shan,China.We measured the daily,monthly and seasonal variations of BC concentration in the atmosphere and discussed the possible emission sources.Black carbon background concentration in this region varied in the range of 18-72 ng/m 3 with the highest in summer and the lowest in autumn.The relations between BC concentration and surface wind direction indicated that BC concentration was higher when northwest wind prevails while lower when southeast wind prevails.Air masses backward trajectories showed the potential emission sources in the northwest.Significant positive correlations between daily mean BC concentration and relative humidity indicated that BC might be one of important cloud condensation nuclei.This hypothesis needs to be confirmed further through cloud microphysical features in this region. | Shuyu Zhao Jing Ming Cunde Xiao Weijun Sun Xiang Qin | 2012 | Journal of Environmental Sciences2012,24,1: | 9 |
| 3 | Endothelial CDS2 deficiency causes VEGFA-mediated vascular regression and tumor inhibition显示文摘The response of endothelial cells to signaling stimulation is critical for vascular morphogenesis,homeostasis and function.Vascular endothelial growth factor-a(VEGFA)has been commonly recognized as a pro-angiogenic factor in vertebrate developmental,physiological and pathological conditions for decades.Here we report a novel finding that genetic ablation of CDP-diacylglycerol synthetase-2(CDS2),a metabolic enzyme that controls phosphoinositide recycling,switches the output of VEGFA signaling from promoting angiogenesis to unexpectedly inducing vessel regression.Live imaging analysis uncovered the presence of reverse migration of the angiogenic endothelium in cds2 mutant zebrafish upon VEGFA stimulation,and endothelium regression also occurred in postnatal retina and implanted tumor models in mice.In tumor models,CDS2 deficiency enhanced the level of tumorsecreted VEGFA,which in-turn trapped tumors into a VEGFA-induced vessel regression situation,leading to suppression of tumor growth.Mechanistically,VEGFA stimulation reduced phosphatidylinositol(4,5)-bisphosphate(PIP2)availability in the absence of CDS2-controlled-phosphoinositide metabolism,subsequently causing phosphatidylinositol(3,4,5)-triphosphate(PIP3)deficiency and F0X01 activation to trigger regression of CDS2-null endothelium.Thus,our data indicate that the effect of VEGFA on vasculature is context-dependent and can be converted from angiogenesis to vascular regression. | Wencao Zhao Le Cao Hanru Ying Wenjuan Zhang Dantong Li Xiaolong Ziiu Wenzhi Xue Shuang Wu Mengye Cao Cong Fu Haonan Qi Yimei Hao Yun-Chi Tang Jun Qin Tao PZhong Xiaoxi Lin Luyang Yu Xuri Li Lin Li Dianqing Wu Weijun Pan | 2019 | Cell Research2019,29,11: | 9 |
| 4 | Cystathionine-γ-lyase ameliorates the histone demethylase JMJD3-mediated autoimmune response in rheumatoid arthritis显示文摘Cystathionine-γ-lyase(CSE),an enzyme associated with hydrogen sulfide(H2S)production,is an important endogenous regulator of inflammation.Jumonji domain-containing protein 3(JMJD3)is implicated in the immune response and inflammation.Here,we investigated the potential contribution of JMJD3 to endogenous CSE-mediated inflammation in rheumatoid arthritis(RA).Upregulated CSE and JMJD3 were identified in synovial fibroblasts(SFs)from RA patients as well as in the joints of arthritic mice.Knocking down CSE augmented inflammation in IL-1β-induced SFs by increasing JMJD3 expression.In addition,CSE−/−mice with collagen-induced arthritis(CIA)developed severe joint inflammation and bone erosion.Conversely,overexpressing CSE inhibited JMJD3 expression by the transcription factor Sp-1 and was accompanied by reduced inflammation in IL-1β-treated SFs.Furthermore,JMJD3 silencing or the administration of the JMJD3 inhibitor GSK-J4 significantly decreased the inflammatory response in IL-1β-treated SFs,mainly by controlling the methylation status of H3K27me3 at the promoter of its target genes.GSK-J4 markedly attenuated the severity of arthritis in CIA mice.In conclusion,suppressing JMJD3 expression by the transcription factor Sp-1 is likely responsible for the ability of CSE to negatively modulate the inflammatory response and reduce the progression of RA. | Weijun Wu Ming Qin Wanwan Jia Zheng Huang Zhongzheng Li Di Yang Mengwei Huang Chenxi Xiao Fen Long Jianchun Mao Philip K.Moore Xinhua Liu Yi Zhun Zhu | 2019 | Cellular & Molecular Immunology2019,16,8: | 9 |
| 5 | The mass-balance characteristics and sensitivities to climate variables of Laohugou Glacier No.12,western Qilian Mountains,China显示文摘Due to global warming, glaciers on the Tibetan Plateau(TP) are experiencing widespread shrinkage; however, the mechanisms controlling glacier variations across the TP are still rather unclear, especially on the northeastern TP. In this study, a physically based, distributed surface-energy and mass-balance model was used to simulate glacier mass balance forced by meteorological data. The model was applied to Laohugou No. 12 Glacier, western Qilian Mountains, China, during2010~2012. The simulated albedo and mass balance were validated and calibrated by in situ measurements. The simulated annual glacier-wide mass balances were-385 mm water equivalent(w.e.) in 2010/2011 and-232 mm w.e. in 2011/2012,respectively. The mean equilibrium-line altitude(ELA) was 5,015 m a.s.l., during 2010~2012, which ascended by 215 m compared to that in the 1970 s. The mean accumulation area ratio(AAR) was 39% during the two years. Climatic-sensitivity experiments indicated that the change of glacier mass balance resulting from a 1.5 °C increase in air temperature could be offset by a 30% increase in annual precipitation. | 7 | Chemical Characteristics and Environmental Records of a Snow-Pit at the Glacier No.12 in the Laohugou Valley, Qilian Mountains显示文摘In June 2006, samples were collected from a 90 cm snow-pit at an elevation of 5 040 m a.s.l. in the northern branch firn basin of the Glacier No. 12, Laohugou Valley, in the western part of Qilian Mountain, China. Oxygen isotopes(δ18O) and major soluble ions were measured. Results showed that Ca2+ is the dominant cation, and SO42- is the dominant anion. All major ions have close positive correlations with eachother except NO3- and NH4+. δ18O show positive correlation with air temperature. The method of correlation analysis, sea-salt ion tracer and trend analysis were used to determine the source of the chemical components. | 8 | Potential Effect of Black Carbon on Glacier Mass Balance during the Past 55 Years of Laohugou Glacier No. 12, Western Qilian Mountains显示文摘This study reconstructed the annual mass balance(MB)of Laohugou Glacier No.12 in the western Qilian Mountains during 1961–2015.The annual MB was calculated based on a temperature-index and an accumulation model with inputs of daily air temperature and precipitation recorded by surrounding meteorological stations.The model was calibrated by in-situ MB measurements conducted on the glacier during 2010–2015.Change in constructed annual MB had three phases.During Phase Ⅰ(1961-1984),glacier-wide MB values were slightly positive with an average MB of 24±276 mm w.e.(water equivalent).During Phase Ⅱ(1984-1995),the MB values became slightly negative with an average MB of?50±276 mm w.e..The most negative MB values were found during Phase Ⅲ(1996–2015),with an average MB of?377±276 mm w.e.Climatic analysis showed that the warming led to accelerated glacier mass loss despite a persistent increase of precipitation during the analysis period.However,an increase of black carbon deposited on the glacier surface since the 1980s could have contributed to intensified glacier melt.From simulations and measurements of MB on the Urumqi Glacier No.1,26%of glacier melt caused by black carbon could be identified. | Jizu Chen Xiang Qin Shichang Kang Wentao Du Weijun Sun Yushuo Liu | 2020 | Journal of Earth Science2020,31,2: | 4 |

| 10 | Experimental Gaussian Boson sampling显示文摘Gaussian Boson sampling(GBS) provides a highly efficient approach to make use of squeezed states from parametric down-conversion to solve a classically hard-to-solve sampling problem. The GBS protocol not only significantly enhances the photon generation probability, compared to standard Boson sampling with single photon Fock states, but also links to potential applications such as dense subgraph problems and molecular vibronic spectra. | 14 | Two Ultraviolet Radiation Datasets that Cover China显示文摘Ultraviolet(UV) radiation has significant effects on ecosystems, environments, and human health, as well as atmospheric processes and climate change. Two ultraviolet radiation datasets are described in this paper. One contains hourly observations of UV radiation measured at 40 Chinese Ecosystem Research Network stations from 2005 to 2015. CUV3 broadband radiometers were used to observe the UV radiation, with an accuracy of 5%, which meets the World Meteorology Organization's measurement standards. The extremum method was used to control the quality of the measured datasets. The other dataset contains daily cumulative UV radiation estimates that were calculated using an all-sky estimation model combined with a hybrid model. The reconstructed daily UV radiation data span from 1961 to 2014. The mean absolute bias error and root-mean-square error are smaller than 30% at most stations, and most of the mean bias error values are negative, which indicates underestimation of the UV radiation intensity. These datasets can improve our basic knowledge of the spatial and temporal variations in UV radiation. | 20 | Growth performance,fatty acid composition,and lipid metabolism are altered in groupers(Epinephelus coioides)by dietary fish oil replacement with palm oil显示文摘In this study,we conducted a 56-d feeding trial to investigate the effects of replacing the fish oil(FO)with palm oil(PO)on the performance,tissue fatty acid(FA)composition,and mRNA levels of genes related to hepatic lipid metabolism in grouper(Epinephelus coioides).Five isolipidic(13%crude lipid)and isonitrogenous(48%CP)diets were formulated by incrementally adding PO to the control diet(25%fish meal and 9%added FO)to replace FO in the control diets.Triplicate groups of 30 groupers(initial weight:12.6±0.1 g)were fed one of the diets twice daily,to apparent satiety.The replacement of FO with 50%PO revealed maximum growth without affecting the performance and whole-body proximate compositions,and replacing FO with 100%PO revealed a comparable(P>0.05)growth with that of the control diet,suggesting PO as a suitable alternative to FO.The analysis of FA profiles in the dorsal muscle and liver though reflected the FA profile of the diet,PO substitutions above 50%could compromise(P<0.05)the FA profile in the liver and flesh of the fish species in comparison with the control diet.Furthermore,the mRNA levels of FAS,G6PD,LPL,PPARА,and D6FAD genes in the liver had positive linear and/or quadratic responses,but the SCD,HSL,ATGL,FABP,SREBP-1C and ELOVL5 had the opposite trend,with increasing dietary PO inclusion levels,whereas the mRNA level of ACC was not affected by dietary treatments.The optimal level of PO substitution for FO was estimated to be 47.1%of the feed,based on the regression analysis of percent weight gains against dietary PO inclusion levels;however,it might affect the FA profile in the liver and flesh of the fish species,and further study is required to investigate whether the changes in tissue FA composition will affect the welfare and market value over a production cycle of grouper. | Yingmei Qin Lingyun He Yanfei Wang Dong Li Weijun Chen Jidan Ye | 2022 | Animal Nutrition2022,,1: | 1 |
