1Baicalin protects neonatal rat brains against hypoxicischemic injury by upregulating glutamate transporter 1 via the phosphoinositide 3-kinase/protein kinase B signaling pathway显示文摘Baicalin is a flavonoid compound extracted from Scutellaria baicalensis root.Recent evidence indicates that baicalin is neuroprotective in models of ischemic stroke.Here,we investigate the neuroprotective effect of baicalin in a neonatal rat model of hypoxic-ischemic encephalopathy.Seven-day-old pups underwent left common carotid artery ligation followed by hypoxia(8% oxygen at 37°C) for 2 hours,before being injected with baicalin(120 mg/kg intraperitoneally) and examined 24 hours later.Baicalin effectively reduced cerebral infarct volume and neuronal loss,inhibited apoptosis,and upregulated the expression of p-Akt and glutamate transporter 1.Intracerebroventricular injection of the phosphoinositide 3-kinase/protein kinase B(PI3 K/Akt) inhibitor LY294002 30 minutes before injury blocked the effect of baicalin on p-Akt and glutamate transporter 1,and weakened the associated neuroprotective effect.Our findings provide the first evidence,to our knowledge that baicalin can protect neonatal rat brains against hypoxic-ischemic injury by upregulating glutamate transporter 1 via the PI3 K/Akt signaling pathway.Zhi-qing Zhou Yong-liang Li Zhen-bo Ao Zhi-li Wen Qi-wen Chen Zheng-gang Huang Bing Xiao Xiao-hua Yan 2017Neural Regeneration Research2017,12,10:15
2Effects of inorganic arsenic on growth and microcystin production of a Microcystis strain isolated from an algal bloom in Dianchi Lake,China显示文摘Our previous data have shown that inorganic arsenic concentrations were high in Dianchi Lake,China,where Microcystis blooms often occur.To explore the relationship between arsenic and the growth of Microcystis,the effects of arsenite[As(III)]and arsenate[As(V)]on the growth and toxin production of M.aeruginosa strain FACHB 905 were tested.Results showed that M.aeruginosa FACHB 905 was tolerant to inorganic arsenic and its growth was not inhibited when the concentration of As(III)was below 10-5 mol L-1 or that of As(V)below 10-3 mol L-1.Total microcystin production was stimulated in the presence of 10-7 mol L-1 As(III)and the response of this M.aeruginosa strain to As(III)seemed to be a typical inverted U-shaped hormesis.The content increase of microcystin-LR per cell indicated that the toxicity was enhanced as M.aeruginosa FACHB 905 was exposed to As(V).Considering the relatively high concentration of inorganic arsenic in Dianchi Lake(139μg L-1 in epilimnetic water),the origin of the M.aeruginosa strain,inorganic arsenic favors survival of M.aeruginosa FACHB 905 and may stimulate its microcystin production and cellular toxicity.GONG Yan AO HongYi LIU BiBo WEN Sheng WANG Zhi HU DingJing ZHANG XingZhong SONG LiRong LIU JianTong 2011Chinese Science Bulletin2011,56,22:6
3Memristive Artificial Synapses for Neuromorphic Computing显示文摘Neuromorphic computing simulates the operation of biological brain function for information processing and can potentially solve the bottleneck of the von Neumann architecture.This computing is realized based on memristive hardware neural networks in which synaptic devices that mimic biological synapses of the brain are the primary units.Mimicking synaptic functions with these devices is critical in neuromorphic systems.In the last decade,electrical and optical signals have been incorporated into the synaptic devices and promoted the simulation of various synaptic functions.In this review,these devices are discussed by categorizing them into electrically stimulated,optically stimulated,and photoelectric synergetic synaptic devices based on stimulation of electrical and optical signals.The working mechanisms of the devices are analyzed in detail.This is followed by a discussion of the progress in mimicking synaptic functions.In addition,existing application scenarios of various synaptic devices are outlined.Furthermore,the performances and future development of the synaptic devices that could be significant for building efficient neuromorphic systems are prospected.Wen Huang Xuwen Xia Chen Zhu Parker Steichen Weidong Quan Weiwei Mao Jianping Yang Liang Chu Xing’ao Li 2021Nano-Micro Letters2021,13,5:4
4High-order mode lasing in all-FMF laser cavities显示文摘We experimentally demonstrate two kinds of all few-mode fiber(FMF) ring lasers with high-order mode(HOM)oscillation in the laser cavity. One kind is a switchable-wavelength all-FMF HOM laser with an output of tunable optical vortex beams(OVBs); the other is a Q-switched all-FMF HOM laser with an output of pulsed cylindrical vector beams(CVBs). The lasers are composed of all-FMF components and few-mode erbium-doped fiber. A Sagnac interferometer made of a 3 dB FMF coupler functions as the wavelength selector, and switchable multiwavelength tunable OVBs are experimentally realized. Carbon nanotube-based saturable absorbers and the nonlinear polarization rotation technique are used to achieve Q-switched CVB lasers. 6Effects of ketamine on mouse hippocampal inflammatory cytokines IL-6,IL-1β and TNF-α levels in acute and chronic administration models显示文摘OBJECTIVE Ketamine is an injectable anesthetic and recreational drug of abuse commonly used worldwide. Many experimental studies have shown that ketamine can impair cognitive function and induce psychotic states. Neuroinflammation has been suggested to play an important role in neurodegeneration. Meanwhile,ketamine has been showed to modulate the levels of inflammatory cytokines.Therefore,we sought to investigate whether the effects of ketamine on the central nervous system is associated with the inflammatory cytokines. METHODS We established acute(single or multiple intraperitoneal injection) and chronic(six months daily intraperitoneal injection) ketamine administration models in C57BL/6 mice,evaluated the spatial recognition memory and emotional response by applying the Y maze test and open field test. We analyzed the changes of inflammatory cytokines interleukin-6(IL-6),interleukin-1β(IL-1β) and tumor necrosis factor-α(TNF-α) levels in mouse hippocampus,employing Western blot,quantitative reverse transcriptase-polymerase chain reaction(qR T-PCR) and immunohistochemistry. RESULTS Ketamine induced spatial recognition memory deficit,reduced anxiety-like behaviors in mice after chronic administration,and it was dose-dependent. Moreover,we found that ketamine could increase the levels of mouse hippocampal inflammatory cytokines IL-6 and IL-1β after single,multiple and long-term administration in a dose-dependent manner. However,the level of TNF-α expressed differently in mouse hippocampus under different conditions. Single administration of ketamine increased the level of TNF-α,whereas multiple and long-term administration decreased it significantly. We considered that TNF-α might exist bi-directional regulatory pathway,which was associated with the dose and duration of ketamine administration. 7Ag@Au core/shell triangular nanoplates with dual enzyme-like properties for the colorimetric sensing of glucose显示文摘Due to the serious harm of diabetes to human health,development of sensitive assays for glucose level is of high significance for early prevention and treatment of diabetes.Currently,most conventional enzyme-based glucose sensors suffer from high cost and low stability due to the inherent defects of natural enzymes.Herein,we develop a pure nanozyme-based glucose detection method using Ag@Au core/shell triangular nanoplates(TNPs),which combines glucose oxidase(GOD)-and horseradish peroxidase(HRP)-like activities of the Au shell and inherent plasmonic properties of Ag TNPs.The sensing mechanism is based on the fact that the Au shell possessed GOD-like activity,enabling the oxidation of glucose to produce H2O2,which can further etch the silver core,leading to the decrease of absorbance at 800 nm and the color change from blue to colorless.Compared with the previous nanozymes-based glucose sensors,our method avoids the use of enzymes and organic chromogenic agent.Moreover,the stability of the Ag@Au core/shell TNPs is much better than that of Ag TNPs due to the protection by the coating of the Au shell.This method was successfully applied to the detection of urine samples from patients with diabetes,indicating its practical applicability for real sample analysis.Ao Liu Mengmeng Li Jingxiang Wang Fan Feng Yu Zhang Zhiwei Qiu Yuzhu Chen Benjamin Edem Meteku Congying Wen Zifeng Yan Jingbin Zeng 2020Chinese Chemical Letters2020,31,5:3

13Decoration of C_(60) nanorods with nickel and their magnetic properties显示文摘We studied the coating of C60 nanorods with nickel by electroless plating method and investigated their magnetic properties. The morphology and structure of the nickel layer coated on C60 nanorods were characterized by transmission electron microscopy (TEM), energy-dispersive X-ray spectroscopy (EDX), X-ray diffraction (XRD) and Raman spectroscopy. The coated nickel is in the form of nano-sized crystals and becomes a continuous layer as reaction time increases. The hysteresis loop shows a super paramagnetic characteristic similar to that of nanometer-sized nickel particle. These results suggest that the average size of nickel particles on C60 rods is below 10 nm. 18Decarboxylated osteocalcin,a possible drug for type 2 diabetes,triggers glucose uptake in MG63 cells显示文摘BACKGROUND Uncarboxylated osteocalcin(GluOC)has been reported to improve glucose metabolism,prevent type 2 diabetes,and decrease the severity of obesity in mice with type 2 diabetes.GluOC can increase glucose uptake in a variety of cells.Glucose metabolism is the main source of energy for osteoblast proliferation and differentiation.We hypothesized that decarboxylated osteocalcin(dcOC),a kind of GluOC,can increase glucose uptake in MG63 cells(osteoblast-like osteosarcoma cells)and influence their proliferation and differentiation.AIM To investigate the effects of dcOC on glucose uptake in human osteoblast-like osteosarcoma cells and the possible signaling pathways involved.METHODS MG63 cells(human osteoblast-like osteosarcoma cells)were treated with dcOC(0,0.3,3,10,or 30 ng/mL)for 1 and 72 h,and glucose uptake was measured by flow cytometry.The effect of dcOC on cell proliferation was measured with a CCK-8 assay,and alkaline phosphatase(ALP)enzyme activity was measured.PI3K was inhibited with LY294002,and hypoxia-inducible factor 1 alpha(HIF-1α)was silenced with siRNA.Then,GPRC6A(G protein-coupled receptor family C group 6 subtype A),total Akt,phosphorylated Akt,HIF-1α,and glucose transporter 1(GLUT1)levels were measured by Western blot to elucidate the possible pathways by which dcOC modulates glucose uptake.RESULTS The glucose uptake of MG63 cells was significantly increased compared with that of the paired control cells after short-term(1 h)treatment with dcOC at different concentrations(0.3,3,and 10 ng/mL groups,P<0.01;30 ng/mL group,P<0.05).Glucose uptake of MG63 cells was significantly increased compared with that of the paired control cells after long-term(72 h)treatment with dcOC at different concentrations(0.3,3,and 10 ng/mL groups,P<0.01;30 ng/mL group,P<0.05).DcOC triggered Akt phosphorylation in a dose-dependent manner,and the most effective stimulatory concentration of dcOC for short-term(1 h)was 3 ng/mL(P<0.01).LY294002 abolished the dcOC-mediated(1 h)promotion of Akt phosphorylation and glucose uptake without affecting GLUT1 protein expression.Long-term dcOC stimulation triggered Akt phosphorylation and increased the protein levels of HIF-1α,GLUT1,and Runx2 in a dose-dependent manner.Inhibition of HIF-1αwith siRNA abolished the dcOC-mediated glucose uptake and substantially decreased GLUT1 protein expression.DcOC interven-tion promoted cell proliferation in a time-and dose-dependent manner as determined by the CCK-8 assay.Treatment with both 3 ng/mL and 10 ng/mL dcOC affected the ALP activity in MG63 cells after 72 h(P<0.01).CONCLUSION Short-and long-term dcOC treatment can increase glucose uptake and affect proliferation and ALP activity in MG63 cells.This effect may occur through the PI3K/Akt,HIF-1α,and GLUT1 signaling factors.Shi Jin Xiao-Cen Chang Jing Wen Jing Yang Na Ao Ke-Ying Zhang Lin-Na Suo Jian Du 2021World Journal of Diabetes2021,12,7:1
