2Overexpression of a dominant-negative mutant of SIRT1 in mouse heart causes cardiomyocyte apoptosis and early-onset heart failure显示文摘SIRT1,a mammalian ortholog of yeast silent information regulator 2(Sir2),is an NAD+-dependent protein deacetylase that plays a critical role in the regulation of vascular function.The current study aims to investigate the functional significance of deacetylase activity of SIRT1 in heart.Here we show that the early postnatal hearts expressed the highest level of SIRT1deacetylase activity compared to adult and aged hearts.We generated transgenic mice with cardiac-specific expression of a dominant-negative form of the human SIRT1(SIRT1H363Y),which represses endogenous SIRT1 activity.The transgenic mice displayed dilated atrial and ventricular chambers,and died early in the postnatal period.Pathological,echocardiographic and molecular phenotype confirmed the presence of dilated cardiomyopathy.Terminal deoxynucleotidyl transferase-mediated dUTP nick-end-labeling analysis revealed a greater abundance of apoptotic nuclei in the hearts of transgenic mice.Furthermore,we show that cardiomyocyte apoptosis caused by suppression of SIRT1 activity is,at least in part,due to increased p53acetylation and upregulated Bax expression.These results indicate that dominant negative form of SIRT1(SIRT1H363Y)overexpression in mouse hearts causes cardiomyocyte apoptosis and early-onset heart failure,suggesting a critical role of SIRT1 in preserving normal cardiac development during the early postnatal period.MU WenLi ZHANG QingJun TANG XiaoQiang FU WenYan ZHENG Wei LU YunBiao LI HongLiang WEI YuSheng LI Li SHE ZhiGang CHEN HouZao LIU DePei 2014Science China(Life Sciences)2014,57,9:12
3Neural Stem Cells Transfected with Leukemia Inhibitory Factor Promote Neuroprotection in a Rat Model of Cerebral Ischemia显示文摘Leukemia inhibitory factor(LIF) contributes to the neuroprotection by neural stem cells(NSCs) after ischemic stroke. Our aim was to explore whether LIFtransfected NSCs(LIF-NSCs) can ameliorate brain injury and promote neuroprotection in a rat model of cerebral ischemia. To accomplish this goal, we transfected NSCs with a lentivirus carrying the LIF gene to stably overexpress LIF. The LIF-NSCs reduced caspase 3 activation under conditions of oxygen-glucose deprivation in vitro.Transient cerebral ischemia was induced in rats by 2 h of middle cerebral artery occlusion(MCAo), and LIF-NSCs were intravenously injected at 6 h post-ischemia. LIF-NSC treatment reduced the infarction volume and improved neurological recovery. Moreover, LIF-NSCs improved glial cell regeneration and ameliorated white matter injuryin the MCAo rats. 4Pure drug nano-assemblies: A facile carrier-free nanoplatform for efficient cancer therapy显示文摘Nanoparticulate drug delivery systems(Nano-DDSs) have emerged as possible solution to the obstacles of anticancer drug delivery. However, the clinical outcomes and translation are restricted by several drawbacks, such as low drug loading, premature drug leakage and carrier-related toxicity.Recently, pure drug nano-assemblies(PDNAs), fabricated by the self-assembly or co-assembly of pure drug molecules, have attracted considerable attention. Their facile and reproducible preparation technique helps to remove the bottleneck of nanomedicines including quality control, scale-up production and clinical translation. Acting as both carriers and cargos, the carrier-free PDNAs have an ultra-high or even100% drug loading. In addition, combination therapies based on PDNAs could possibly address the most intractable problems in cancer treatment, such as tumor metastasis and drug resistance. In the present review, the latest development of PDNAs for cancer treatment is overviewed. First, PDNAs are classified according to the composition of drug molecules, and the assembly mechanisms are discussed. Furthermore, the co-delivery of PDNAs for combination therapies is summarized, with special focus on the improvement of therapeutic outcomes. 5ZrO2 nanoparticles anchored on nitrogen-doped carbon nanosheets as efficient catalyst for electrochemical CO2 reduction显示文摘Electrochemical reduction of CO2 to produce value-added feedstock chemicals using high-performance electrocatalysts is a promising protocol to address the excessive CO2 in the atmosphere and the energy crisis. However, the high overpotential, low current density, and poor product selectivity for CO2 electroreduction greatly impede their practical applications. In this work, we develop an efficient catalyst for CO2 reduction to CO consisting of well-dispersed ZrO2 nanoparticles tightly anchored on nitrogendoped carbon nanosheets(ZrO2/N-C) for the first time. Importantly, the ZrO2 nanoparticles possess oxygen vacancies and defects, which regulate the electronic structure of catalyst and thus greatly enhance the electrocatalytic activity. Specifically, ZrO2/N-C demonstrates a high CO Faradaic efficiency(FE) of 64% at-0.4 V vs. the reversible hydrogen electrode(RHE) and a respectable current density of ~2.6 m A cm-2 in CO2-saturated 0.5 M KHCO 3 solution. 6Amorphous NiO_(n)coupled with trace PtO_(x)toward superior electrocatalytic overall water splitting in alkaline seawater media显示文摘Developing corrosion resistance bifunctional electrocatalysts with high activity and stability toward both hydrogen evolution reaction(HER)and oxygen evolution reaction(OER),especially electrolysis in seawater,is of prime significance but still pressingly challenging.Herein,in-situ introduced PtO_(x)on the derivative amorphous NiO_(n)is prepared via heat treatment of Ni ZIFL nanosheets on nickel foam under low temperature(PtO_(x)-NiO_(n)/NF).The synthesized PtO_(x)-NiO_(n)/NF possesses suprahydrophilic and aerophilic surface,then in favor of intimate contact between the electrode and electrolyte and release of the generated gas bubbles during the electrocatalysis.As a result,the in-situ PtO_(x)-NiO_(n)/NF electrode presents outstanding bifunctional activity,which only requires extremely low overpotentials of 32 and 240 mV to reach a current density of 10 mA·cm^(-2)for HER and OER,respectively,which exceeds most of the electrocatalysts previously developed and even suppresses commercial Pt/C and RuO_(2)electrodes.As for two-electrode cell organized by PtO_(x)-NiO_(n)/NF,the voltages down to 1.57 and 1.58 V are necessary to drive 10 mA·cm^(-2)with remarkable durability in 1 M KOH and alkaline seawater,respectively,along with remarkable stability.Moreover,a low cell voltage of 1.88 V is needed to achieve 1,000 mA·cm^(-2)toward water-splitting under industrial conditions.This study provides a new idea for designing in-situ amorphous metal oxide bifunctional electrocatalyst with strong Pt–support interaction for overall water splitting.Wenli Yu Hongru Liu Ying Zhao Yunlei Fu Weiping Xiao Bin Dong Zexing Wu Yongming Chai Lei Wang 2023Nano Research2023,16,5:2

12FBFN-based adaptive repetitive control of nonlinearly parameterized systems显示文摘An adaptive repetitive control scheme is presented for a class of nonlinearly parameterized systems based on the fuzzy basis function network (FBFN).The parameters of the fuzzy rules are tuned with adaptive schemes.To attenuate chattering effectively,the discontinuous control term is approximated by an adaptive PI control structure.The bound of the discontinuous control term is assumed to be unknown and estimated by an adaptive mechanism.Based on the Lyapunov stability theory,an adaptive repetitive control law is proposed to guarantee the closed-loop stability and the tracking performance.By means of FBFNs,which avoid the nonlinear parameterization from entering into the adaptive repetitive control,the controller singularity problem is solved.The proposed approach does not require an exact structure of the system dynamics,and the proposed controller is utilized to control a model of permanent-magnet linear synchronous motor subject to significant disturbances and parameter uncertainties.The simulation results demonstrate the effectiveness of the proposed method.Wenli Sun Hong Cai Fu Zhao 2013Journal of Systems Engineering and Electronics2013,24,6:0

14A study of function mechanism of hemxamethyl tetra-amine in gelation process of uranium显示文摘The UO2 ceramic microspheres are the most important materials in the spherical fuel elements for high temperature reactor (HTR). A process for preparation of UO2 kernels known as total gelation process of uranium (TGU) was developed as the production process of 10 mW HTR at Tsinghua University. The TGU process is based on the traditional sol-gel process, external gelation process and internal gelation process of uranium (EGU and IGU), which implies that the gelation action is initiated both by ammonia out of the gel particles and hemxamethyl tetra-amine (HMTA) inside the gel particles. The gelation behavior and the properties of uranium microspheres were investigated of the solution with and without HMTA. It is observed that good spherical particles can be obtained without HMTA in the sol, which indicates a more controllable and industrialized route will be set up. 15An Improved Rate Control Method Based on Modification at Scene Change for H.264显示文摘This paper presents an improved rate control method for H.264. First, the scene changes are detected by the average absolute difference of the brightness histograms between the adja-cent frames. Then, the bit allocation and quantization parameters are adjusted, using a certain threshold. In addition, the calculation of the mean absolute difference (MAD) is modified in an alterna-tive way, which makes the rate distortion optimization (RDO) more accurate.
