| 1 | Study on determining of reinforced concrete false roof strength and design of reinforcement based on reliability theory显示文摘Study on efficient mining of the steep incline and fractured ore-bodies in Yongshaba mine of Guizhou Kailin Group shows that ore-body is fractured and difficult to support the roadways in-vein.After research of the actual conditions about the ore-bodies,we have made the initial decision to adopt reconstruction of roof downward sublevel cut-and-fill mining.The men work safely under the false roof supporting the top plate.However,the difficult problem is how to determine the strength of the false roof.In this case,the method based on reliability theory has been put forward.Combined with elastic mechanics and field practice,when practical value of reliable probability is 90 %,the value of the false roof strength has been calculated,and the study shows that stope span greatly influences the false roof strength.With the strength of artificial roof,the reasonable reinforcement design ensures the false roof which can supply the demand of strength under large span and load. | Fan Wenlu Li Xibing Hu Guohong | 2012 | Engineering Sciences2012,10,5: | 2 |
| 2 | The pattern of plasma BCAA concentration and liver Bckdha gene expression in GK rats during T2D progression显示文摘Background: This study was conducted to measure the concentration of branched chain amino acid(BCAA) in different species and detect the expression pattern of the liver Bckdha gene in Goto-Kakizaki(GK) rats during type 2 diabetes(T2D)progression.Methods: We measured the concentration of BCAA in GK rats, induced T2D cynomolgus monkeys and T2D humans by liquid chromatography tandem mass spectrometry, and used real-time quantitative PCR to analyze the gene expression of Bckdha and Bckdk, which encode the rate-limiting enzymes in catabolism of,respectively, branched chain amino acids and branched chain α-keto acid dehydrogenase kinase.Results: In this study, we showed that GK rat BCAA concentrations were significantly reduced at 4 and 8 weeks(P < 0.05 and P < 0.01, respectively), while the expression of Bckdha in GK rat liver was increased at 4 and 8 weeks(1.62-fold and1.93-fold, respectively). The BCAA concentrations were significantly reduced in diet-induced T2D cynomolgus monkeys(P < 0.01), but significantly increased in T2D humans(P < 0.001).Conclusions: Our results showed that BCAA concentrations changed at different times and by different amounts in different species and during different periods of T2D progress, and the significant changes of BCAA concentration in the three species indicated that BCAA might participate in the progress of T2D. The results suggested that the increased expression of Bckdha in GK rat liver might partially explain the reduced plasma BCAA concentration at 4 and 8 weeks. | 4 | Exploration and Validation of the Potential Downstream Genes Underlying ipa1-2D Locus for Rice Panicle Branching显示文摘In recent years,some super hybrid rice varieties were bred with strong culms and large panicles,which are mainly contributed by the ipa1-2D locus.A gain-of-function allele of OsSPL14 is the ipa1-2D and it can greatly increase the panicle primary branch number.However,the key downstream genes mediating this trait variation are not fully explored.In this study,we developed high-quality near-isogenic lines(NILs)with a difference of only 30 kb chromosomal segment covering the ipa1-2D locus.Using the NILs,we explored the impact of ipa1-2D on five sequential stages of early inflorescence development,and found that the locus can greatly enhance the initiation of primary branch meristems.A transcriptomic analysis was performed to unveil the downstream molecular network of ipa1-2D,and 87 genes were found differentially expressed,many of which are involved in metabolism and catalysis processes.In addition,transgenic lines of overexpression and RNA interference were generated to shape different levels of OsSPL14.They were also used to validate the expression variation explored by transcriptome.Based on the gene annotation,twelve potential downstream targets of ipa1-2D were selected,and their expression variation was confirmed by qRT-PCR analysis both in NILs and transgenic lines.This research expands the molecular network underlying ipa1-2D and provides novel gene information which might be involved in the control of panicle branching.We discussed the potential function of identified genes and highlighted their values for future function exploration and breeding application. | Lin Zhang Dong Xie Zhong Bian Yiting Zou Han Zhou Wenlu Cai Hadi Yeilaghi Xiaolei Fan Changquan Zhang Qiaoquan Liu | 2021 | Phyton-International Journal of Experimental Botany2021,90,3: | 0 |
| 5 | Regulation of serotonin production by specific microbes from piglet gut显示文摘Background Serotonin is an important signaling molecule that regulates secretory and sensory functions in the gut.Gut microbiota has been demonstrated to affect serotonin synthesis in rodent models.However,how gut microbes regulate intestinal serotonin production in piglets remains vague.To investigate the relationship between microbiota and serotonin specifically in the colon,microbial composition and serotonin concentration were analyzed in ileum-cannulated piglets subjected to antibiotic infusion from the ileum when comparing with saline infusion.Microbes that correlated positively with serotonin production were isolated from piglet colon and were further used to investi-gate the regulation mechanisms on serotonin production in IPEC-J2 and a putative enterochromaffin cell line RIN-14B cells.Results Antibiotic infusion increased quantities of Lactobacillus amylovorus(LA)that positively correlated with increased serotonin concentrations in the colon,while no effects observed for Limosilactobacillus reuteri(LR).To understand how microbes regulate serotonin,representative strains of LA,LR,and Streptococcus alactolyticus(SA,enriched in feces from prior observation)were selected for cell culture studies.Compared to the control group,LA,LR and SA supernatants significantly up-regulated tryptophan hydroxylase 1(TPH1)expression and promoted serotonin production in IPEC-J2 cells,while in RIN-14B cells only LA exerted similar action.To investigate potential mechanisms mediated by microbe-derived molecules,microbial metabolites including lactate,acetate,glutamine,andγ-aminobutyric acid were selected for cell treatment based on computational and metabolite profiling in bacte-rial supernatant.Among these metabolites,acetate upregulated the expression of free fatty acid receptor 3 and TPH1 while downregulated indoleamine 2,3-dioxygenase 1.Similar effects were also recapitulated when treating the cells with AR420626,an agonist targeting free fatty acid receptor 3.Conclusions Overall,these results suggest that Lactobacillus amylovorus showed a positive correlation with sero-tonin production in the pig gut and exhibited a remarkable ability to regulate serotonin production in cell cultures.These findings provide evidence that microbial metabolites mediate the dialogue between microbes and host,which reveals a potential approach using microbial manipulation to regulate intestinal serotonin biosynthesis. | Ziyu Liu Yidan Ling Yu Peng Shuibing Han Yuting Ren Yujia Jing Wenlu Fan Yong Su Chunlong Mu Weiyun Zhu | 2023 | Journal of Animal Science and Biotechnology2023,14,6: | 0 |
| 6 | 血管内皮生长因子-D在胃癌中表达的意义(英文)显示文摘Objective: To investigate the expression of vascular endothelial growth factor D (VEGF-D) in gastric cancer and its relationship with lymph node metastasis. Methods: 100 cases of gastric carcinoma tissues (50 cases with lymph node metastasis, 50 cases negative) and 30 cases of normal gastric tissues were gathered to detect the expressions of VEGF-C and D proteins by immunohistochemistry. Results: VEGF-C and D were revealed in cytoplasm of gastric carcinoma tissues and normal gastric tissues. The positive rates of VEGF-C and D expressions were significantly higher in gastric cancer tissues than those in the normal ones (51%, 60% vs 10%, 20% respectively; both P < 0.05). There were significant correlations be- tween the positive expression of VEGF-D and lymph node metastasis, lymphatic invasion, positive expression of VEGF-C, but not with tumour size, tissue differentiation, and venous invasion.
