3Comparative analysis of antioxidant activities of essential oils and extracts of fennel(Foeniculum vulgare Mill.)seeds from Egypt and China显示文摘The present study was conducted to assay the antioxidant activities of essential oils and ethanol extracts of fennel(Foeniculum vulgare Mill.)seeds from Egypt and China.The major components of the Egyptian fennel essential oil were estragole(51.04%),limonene(11.45%),l-fenchone(8.19%)and trans-anethole(3.62%)by GC–MS analysis.Whereas,the major constituents of Chinese fennel essential oil were transanethole(54.26%),estragole(20.25%),l-fenchone(7.36%)and limonene(2.41%).The fennel seed extracts from Egypt and China contained appreciable levels of total phenolic contents(42.24 and 30.94 mg PE/g,respectively).The extracts exhibited good DPPH radical scavenging capacity with IC50(6.34 and 7.17 mg/g),respectively.A high variation in free radical scavenging activities of essential oils was observed.The Chinese fennel essential oil showed high activity in DPPH radical scavenging with IC50(15.66 mg/g).The Egyptian fennel essential oil showed very low activity with IC50(141.82 mg/g).The results of the present investigation demonstrated significant variations in the antioxidant activities of fennel essential oils and extracts from Egypt and China.Adel F.Ahmed Mengjin Shi Cunyu Liu Wenyi Kang 2019Food Science and Human Wellness2019,8,1:6
4Spectrum-effect relationship of immunologic activity of Ganoderma lucidum by UPLC-MS/MS and component knock-out method显示文摘This study was designed to elucidate the immunoregulation of Ganoderma lucidum.HPLC fingerprint and spectrum-effect relationship of G.lucidum were established to predict the active compounds and BP neural network model was established to predict the efficacy.Then the target compounds were identified by high resolution mass spectrometry.The results indicated that there are both enhanced immunity and immunosuppressive components in G.lucidum.BP neural network was trained with the common peak area and immune effi cacy index of G.lucidum fi ngerprint as samples,and a combined evaluation system of G.lucidum fi ngerprint effi cacy was established.The correlation coeffi cient R of BP network model was 0.98643,and the error of pharmacodynamic prediction results was in the ideal range.Eight compounds were identifi ed by high resolution mass spectrometry.The compounds related to immune activity in G.lucidum were determined in this study.Changqin Li Yiping Cui Jie Lu Lijun Meng Changyang Ma Zhenhua Liu Yan Zhang Wenyi Kang 2021Food Science and Human Wellness2021,10,3:6
5Antrodia Cinnamomea ameliorates neointimal formation by inhibiting infl ammatory cell infi ltration through downregulation of adhesion molecule expression in vitro and in vivo显示文摘The increased vascular infl ammation is a key event in the development of atherosclerotic lesions.Antrodia cinnamomea has been shown to promote anticancerogenic activity through decreasing infl ammation.However,the potential role of A.cinnamomea in cardiovascular diseases remains unexplored.Herein,using carotid arterial ligation models,we found that ethanol extract from A.cinnamomea(EEAC)signifi cantly inhibited neointimal hyperplasia in a dose-dependent manner,accompanied with the reduced expression of activated p65 and infl ammatory cytokines.We also show that EEAC ameliorated TNF-α-induced phosphorylation of p65 and pro-infl ammatory cytokine expression in both vascular smooth muscle cells(VSMCs)and macrophages in vitro.Mechanistically,EEAC suppressed expression levels of intercellular adhesion molecule-1(ICAM-1)and vascular cell adhesion molecule(VCAM-1)in VSMCs,which attenuates the ability of monocytes/macrophages adhesion to VSMCs.Furthermore,the expression level of these adhesion molecules and infi ltration of monocytes/macrophages were also decreased in neointimal VSMCs of arteries pretreated with EEAC.Altogether,our results reveal a novel function of A.cinnamomea in suppressing vascular infl ammation upon ligation injury during neointimal formation,likely through inhibition of infl ammatory cell infi ltration via downregulating the adhesion molecules in VSMCs.Thus,A.cinnamomea may offer a pharmacological therapy to slow down disease progression in patients with vascular injury.Yan Zhang Aijin Ma Hao Xi Ning Chen Rong Wang Chenhui Yang Jinbang Chen Pin Lü Fuping Zheng Wenyi Kang 2021Food Science and Human Wellness2021,10,4:4
6Antioxidant activity and total phenolic content of essential oils and extracts of sweet basil(Ocimum basilicum L.)plants显示文摘The present study was conducted to evaluate the chemical composition and antioxidant activity of the essential oils and ethanol extracts of Ocimum basilicum L.obtained from Assiut,Minia and BeniSuef of Egypt.The major constituents of sweet basil essential oils from three locations were linalool,estragole,methyl cinnamate,bicyclosesquiphellandrene,eucalyptol,-bergamotene,eugenol,-cadinene and germacrene D by the method of GC–MS.The relative concentration of these compounds varies.The basil extracts contained appreciable levels of total phenolic contents and exhibited good DPPH radical scavenging capacity higher than that of essential oils.The highest TPC(82.45 mg PE/g)and maximum radical scavenging activity with IC50 value(1.29 mg/mL)was obtained from Minia basil extract.High correlation between antioxidant activity and total phenolic contents of basil extracts was observed.High variation in free radical scavenging activity of essential oils was found.The essential oil from Minia basil showed high activity in DPPH radical scavenging with IC50(11.23 mg/mL)and contained the highest content of phenolic(41.3 mg PE/g).On contrary,low correlation between antioxidant activity and total phenolic contents of basil essential oils from different locations.The results of the present investigation demonstrated significant variations in the antioxidant activities of sweet basil essential oils and extracts from Egypt.Adel F.Ahmed Fatma A.K.Attia Zhenhua Liu Changqin Li Jinfeng Wei Wenyi Kang 2019Food Science and Human Wellness2019,8,3:4
7Thermo-Mechanically Coupled Thermo-Elasto-Visco-Plastic Modeling of Thermo-Induced Shape Memory Polyurethane at Finite Deformation显示文摘A series of mono tonic tensile experiments of thermo-induced shape memory polyurethane(TSMPU) at different loading rates were carried out to investigate the interaction between the internal heat production and the mechanical deformation. It is shown that the temperature variation on the surfaces of the specimens due to the internal heat production affects the mechanical properties of TSMPU remarkably. Then, based on irreversible thermodynamics, the Helmholtz free energy was decomposed into three parts, i.e., the instantaneous elastic free energy,visco-plastic free energy and heat free energy. The total deformation gradient was decomposed into the mechanical and thermal parts, and the mechanical deformation gradient was further divided into the elastic and visco-plastic components. The Hencky's logarithmic strain was used in the current configuration. The heat equilibrium equation of internal heat production and heat exchange was derived in accordance with the first and second thermodynamics laws. The temperature of specimens was contributed by the internal heat, production and the ambient temperature simultaneously, and a thermo-mechanic ally coupled thermo-elas to-visco-plastic model was established. The effect of temperature variation of specimens on the mechanical properties of the material was considered in this work. 9α-Glucosidase inhibitors isolated from medicinal plants显示文摘Objective:α-Glucosidase inhibitors can be used as a new class of antidiabetic drug.By competitively inhibiting glycosidase activity,these inhibitors help to prevent the fast breakdown of sugars and thereby control the blood sugar level.This study provides a wealth of information aboutα-glucosidase inhibitors isolated from medicinal plants;this knowledge will be useful in finding more potent antidiabetic candidates from the natural resources for the clinical development of antidiabetic therapeutics.Results:411 compounds exhibitingα-glucosidase inhibitory activity were summarized and isolated them from medicinal plants.The compound classes isolated include:terpenes(61)from 14 genus,alkaloids(37)from 11 genus,quinines(49)from 4 genus,flavonoids(103)from 24 genus,phenols(37)from 9 genus,phenylpropanoids(73)from 20 genus,sterides(8)from 5 genus,and other types of compounds(43).Conclusion:Compounds withα-glucosidase inhibitory activity are abundant in nature and can be obtained from several sources.They have highα-glucosidase inhibitory potential,and can be clinically developed for treating diabetes mellitus.Zhenhua Yin Wei Zhang Fajin Feng Yong Zhang Wenyi Kang 2014Food Science and Human Wellness2014,3,3:3
10Antrodia cinnamomea exerts an anti-hepatoma effect by targeting PI3K/AKT-mediated cell cycle progression in vitro and in vivo显示文摘Antrodia cinnamomea is extensively used as a traditional medicine to prevention and treatment of liver cancer.However,its comprehensive chemical fingerprint is uncertain,and the mechanisms,especially the potential therapeutic target for anti-hepatocellular carcinoma(HCC)are still unclear.Using UPLC-Q-TOF/MS,139 chemical components were identified in A.cinnamomea dropping pills(ACDPs).Based on these chemical components,network pharmacology demonstrated that the targets of active components were significantly enriched in the pathways in cancer,which were closely related with cell proliferation regulation.Next,HCC data was downloaded from Gene Expression Omnibus database(GEO).The Cancer Genome Atlas(TCGA)and Dis Ge NET were analyzed by bioinformatics,and 79 biomarkers were obtained.Furtherly,nine targets of ACDP active components were revealed,and they were significantly enriched in PI3 K/AKT and cell cycle signaling pathways.The affinity between these targets and their corresponding active ingredients was predicted by molecular docking.Finally,in vivo and in vitro experiments showed that ACDPs could reduce the activity of PI3 K/AKT signaling pathway and downregulate the expression of cell cycle-related proteins,contributing to the decreased growth of liver cancer.Altogether,PI3 K/AKT-cell cycle appears as the significant central node in anti-liver cancer of A.Cinnamomea.Yan Zhang Pin Lv Junmei Ma Ning Chen Huishan Guo Yan Chen Xiaoruo Gan Rong Wang Xuqiang Liu Sufang Fan Bin Cong Wenyi Kang 2022Acta Pharmaceutica Sinica B2022,12,2:2
11Effects of edpetiline from Fritillaria on inflammation and oxidative stress induced by LPS stimulation in RAW264.7 macrophages显示文摘The dry bulbs of Fritillaria cirrhosa species can help resolve phlegm,soothe cough,clear heat,and moisten the lung,and the main active components responsible for these effect are its alkaloids.However,it is unclear whether or how edpetiline in Fritillaria can inhibit the excessive inflammatory response and oxidative stress.In this research,we aimed to examine this aspect using lipopolysaccharide(LPS)-stimulated RAW264.7 macrophages as an inflammatory model.The quantitative real-time polymerase chain reaction and western blot analysis results showed that edpetiline significantly inhibited the content and mRNA expression levels of proinflammatory cytokines(TNF-αand IL-6)in LPS-induced RAW264.7 cells,significantly increased the mRNA expression of IL-4(anti-inflammatory cytokine),and markedly downregulated the inflammatory mediators inductible nitric oxide synthase(iNOS)and cyclooxygenase-2(COX-2)mRNA and protein expression levels.The oxidative stress induced by LPS was also inhibited by edpetiline,as the level of intracellular reactive oxygen species decreased notably.Edpetiline may exert anti-inflammatory and antioxidant effects through inhibiting the phosphorylation of IκB and the nuclear transcription of nuclear transcription factor-κB p65 and decreasing the phosphorylation of p38 and ERK in the mitogen-activated protein kinase signaling pathway,without activating the JNK/mitogen-activated protein kinase signaling pathway.These findings suggest that edpetiline may be a potential therapeutic agent for the prevention or treatment of inflammation-and oxidative stress-related pathophysiological processes and diseases.Hongli Zhang Xiaoqing Xu Zhenhua Liu Dongxiao Sun-Waterhouse Jinmei Wang Changyang Ma Geoffrey I.N.Waterhouse Wenyi Kang 2021Acta Biochimica et Biophysica Sinica2021,53,2:2
12The CRL3^(BTBD9) E3 ubiquitin ligase complex targets TNFAIP1 for degradation to suppress cancer cell migration显示文摘Tumor necrosis factor alpha-induced protein 1(TNFAIP1)modulates a plethora of important biological processes,including tumorigenesis and cancer cell migration.However,the regulatory mechanism of TNFAIP1 degradation remains largely elusive.In the present study,with a label-free quantitative proteomic approach,TNFAIP1 was identified as a novel ubiquitin target of the Cullin-RING E3 ubiquitin ligase(CRL)complex.More importantly,Cul3-ROC1(CRL3),a subfamily of CRLs,was identified to specifically interact with TNFAIP1 and promote its polyubiquitination and degradation.Mechanistically,BTBD9,a specific adaptor component of CRL3 complex,was further defined to bind and promote the ubiquitination and degradation of TNFAIP1 in cells.As such,downregulation of BTBD9 promoted lung cancer cell migration by upregulating the expression of TNFAIP1,whereas TNFAIP1 deletion abrogated this effect.Finally,bioinformatics and clinical sample analyses revealed that BTBD9 was downregulated while TNFAIP1 was overexpressed in human lung cancer,which was associated with poor overall survival of patients.Taken together,these findings reveal a previously unrecognized mechanism by which the CRL3^(BTBD9) ubiquitin ligase controls TNFAIP1 degradation to regulate cancer cell migration.Lihui Li Wenjuan Zhang Yue Liu Xiaojun Liu Lili Cai Jihui Kang Yunjing Zhang Wenlian Chen Changsheng Dong Yanmei Zhang Mingsong Wang Wenyi Wei Lijun Jia 2020Signal Transduction and Targeted Therapy2020,5,1:2
13Chemical composition and glucose uptake effect on 3T3-L1 adipocytes of Ligustrum lucidum Ait. flowers显示文摘Ligustri lucidi Fructus is a traditional Chinese medicine and possesses various bioactivities,including hypoglycemic effect.Ligustrum lucidum Ait flowers are poorly investigated.Thus,we hypothesized that L.lucidum flowers also could have hypoglycemic effect.Chemical composition and glucose uptake effect of L.lucidum flowers on 3T3-L1 adipocytes were investigated.In this study,the components of L.lucidum flowers were investigated by various chromatographic and spectroscopic methods and the effects of L.lucidum flowers on the induction of glucose uptake were investigated by 3T3-L1 adipocytes.Seven compounds were isolated and identified from L.lucidum flowers,including ursolic acid(1),kaempferol-7-O-α-Lrhamnoside(2,KR),β-sitosterol(3),β-daucosterol(4),kaempferitrin(5,KF),10-hydroxy oleuropein(6,HO),and kaempferol-3-O-β-D-glucopyranoside-7-α-L-rhamnopyranoside(7,KGR).The results of glucose uptake showed that total extract(TE),KR,KF and HO from L.lucidum flowers significantly enhanced glucose uptake.This study demonstrated that compounds 2,5-7 were isolated for the first time from this plant and compounds 2 and 7 were isolated for the first time in genus Ligustrum,L.lucidum flowers,KR,KF and HO may possess potential hypoglycemic effect.Lili Cui Jinmei Wang Mengke Wang Baoguang Wang Zhenhua Liu Jinfeng Wei Wenyi Kang 2020Food Science and Human Wellness2020,9,2:2

17A systematic study on mycochemical profiles, antioxidant, and anti-inflammatory activities of 30 varieties of Jew’s ear(Auricularia auricula-judae)显示文摘The health-promoting properties and chemical profiles of 30 Jew’s ear mushroom varieties were investigated. The antioxidant properties were determined by ferric reducing antioxidant power(FRAP), 1,1-diphenyl-2-picrylhydrazyl(DPPH) free radical scavenging, 2,2’-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid)(ABTS) free radical scavenging, and metal chelating ability(MCA) assays, while phenolic profiles were determined by total phenol content(TPC) and total flavonoid content(TFC) colorimetric assays. Total carbohydrate, β-glucan, and melanin contents were determined by colorimetric methods. 5’-Nucleotides, vitamin D_(2), ergosterol, and ergothioneine contents were determined by high performance liquid chromatography(HPLC). Anti-inflammation activities of Jew’s ear were evaluated by the colorimetric protease inhibitory method. The results showed that Jew’s ear mushrooms possessed substantial phenolics and antioxidant properties. All the Jew’s ear varieties contain high amount of total carbohydrate, β-glucan, reducing sugar, melanin, pectin, vitamin D2, ergosterol, and ergothioneine. 18Immunoregulatory polysaccharides from Apocynum venetum L.flowers stimulate phagocytosis and cytokine expression via activating the NF-κB/MAPK signaling pathways in RAW264.7 cells显示文摘Two immunomodulatory polysaccharides(Vp2a-Ⅱ and Vp3) were isolated and identified from Apocynum venetum L. flowers, and their innate immune-stimulating functions and working mechanisms were evaluated in RAW264.7 cells. Both the level of released nitric oxide(NO) and expression of inducible nitric oxide synthase(iNOS) m RNA were significantly enhanced in the RAW264.7 macrophages cells treated by Vp2a-Ⅱ and Vp3. Vp2a-Ⅱ(100–800 μg/m L) and Vp3(400 μg/mL) could significantly increase the phagocytic activity of RAW264.7 cells and the secretion and m RNA expression of TNF-α and IL-6 in a concentrationdependent manner through affecting mitogen-activated protein kinase(MAPK) activity and nuclear factor κB(NF-κB) nuclear translocation.
