1Expression and Role of Voltage-Gated Sodium Channels in Human Dorsal Root Ganglion Neurons with Special Focus on Nav1.7,Species Differences, and Regulation by Paclitaxel显示文摘Voltage-gated sodium channels(Navs) play an important role in human pain sensation. However, the expression and role of Nav subtypes in native human sensory neurons are unclear. To address this issue, we obtained human dorsal root ganglion(hDRG) tissues from healthy donors. PCR analysis of seven DRG-expressed Nav subtypes revealed that the hDRG has higher expression of Nav1.7(~50% of total Nav expression) and lower expression of Nav1.8(~12%), whereas the mouse DRG has higher expression of Nav1.8(~45%) and lower expression of Nav1.7(~18%). To mimic Nav regulation in chronic pain, we treated hDRG neurons in primary cultures with paclitaxel(0.1-1 μmol/L) for 24 h. Paclitaxel increased the Nav 1.7 but not Nav1.8 expression and also increased the transient Na^+ currents and action potential firing frequency in small-diameter(<50 μm) hDRG neurons. 2Non-invasive prediction of forthcoming cirrhosis-related complications显示文摘In patients with chronic liver diseases,identification of significant liver fibrosis and cirrhosis is essential for determining treatment strategies,assessing therapeutic response,and stratifying long-term prognosis.Although liver biopsy remains the reference standard for evaluating the extent of liver fibrosis in patients with chronic liver diseases,several non-invasive methods have been developed as alternatives to liver biopsies.Some of these non-invasive methods have demonstrated clinical accuracy for diagnosing significant fibrosis or cirrhosis in many cross-sectional studies with the histological fibrosis stage as a reference standard.However,non-invasive methods cannot be fully validated through cross-sectional studies since liver biopsy is not a perfect surrogate endpoint marker.Accordingly,recent studies have focused on assessing the performance of non-invasive methods through longterm,longitudinal,follow-up studies with solid clinical endpoints related to advanced stages of liver fibrosis and cirrhosis.As a result,current view is that these alternative methods can independently predict future cirrhosis-related complications,such as hepatic decompensation,liver failure,hepatocellular carcinoma,or liver-related death.The clinical role of non-invasive models seems to be shifting from a simple tool for predicting the extent of fibrosis to a surveillance tool for predicting future liver-related events.In this article,we will summarize recent longitudinal studies of non-invasive methods for predicting forthcoming complications related to liver cirrhosis and discuss the clinical value of currently available non-invasive methods based on evidence from the literature.Wonseok Kang Seung Up Kim Sang Hoon Ahn 2014World Journal of Gastroenterology2014,20,10:4
3The change of anterior segment parameters after cataract surgery in normal-tension glaucoma显示文摘AIM:To investigate the change of anterior chamber angle morphology and intraocular pressure(IOP) reduction after cataract surgery in patients with normal-tension glaucoma(NTG) using swept-source optical coherence tomography(SS-OCT).METHODS:This prospective,comparative,observational study recruited patients into two groups. Group 1 was the control group including normal subjects except those with cataracts(cataract group,n=67 eyes of 67 patients),and group 2 was NTG group including patients who were diagnosed with NTG and cataracts(n=43 eyes of 43 patients),which were treated with phacoemulsification and intraocular lens implantation. Before surgery,and at postoperative 1 and 6mo,anterior chamber angles were evaluated by SS-OCT under dark conditions using threedimensional angle analysis scan protocol. Angle opening distance(AOD),angle recess area(ARA),and trabeculariris surface area(TISA) at four quadrants(temporal,nasal,superior,and inferior) were calculated automatically by SS-OCT,after the observer marked scleral spurs.RESULTS: A total of 106 patients(54 males and 52 females) were enrolled in the study. Angle parameters,AOD,ARA,and TISA were increased after surgery in both groups. However,changes of angle parameters were only significant in group 2.In group 2,preoperative IOP was 13.2±2.9 mm Hg,and postoperative IOP at 1 and 6mo were 10.5±3.0 and 10.7±2.8 mm Hg,respectively. In group 1,preoperative IOP was 12.4±2.8 mm Hg,and postoperative IOP at 1 and 6mo were 11.6±2.5 and 12.0±2.8 mm Hg,respectively. After cataract surgery,angle parameters changed significantly while IOP significantly reduced and was maintained in group 2(P<0.001). 4When to stop nucleos(t)ide analogues treatment for chronic hepatitis B? Durability of antiviral response显示文摘Introduction of nucleos(t)ide analogues(NAs)for oral antiviral therapy has dramatically improved the clinical outcome in patients with chronic hepatitis B(CHB).Although current international guidelines for the management of CHB provide information regarding when to begin the antiviral therapy with NAs,there is no clear consensus on when to stop the treatment,especially for those who respond to the therapy.Hepatitis B surface antigen loss has been regarded as an ideal endpoint of oral antiviral therapy with NAs,however since this is rarely achieved,practical endpoints have been suggested by the international guidelines.Despite the stopping rules recommended by the international guidelines,whether oral antiviral therapy with NAs can be safely discontinued is of major concern.While attention has been drawn to whether antiviral treatment with NAs can be a finite therapy,there is lack of sufficient data on off-treatment durability of highly potent NAs.Based on the available evidences,current guidelines for stopping NA therapy seems to be inadequate in terms of off-treatment durability,with relapse rates of more than 40%for both hepatitis Be antigen(HBeAg)-positive and HBeAg-negative patients.Therefore,further studies are required to accumulate data on off-treatment durability of highly potent NAs,and future studies are warranted to identify adequate predictive markers that could provide supplementary information to guide the timing of stopping NA therapy.Wonseok Kang Jun Yong Park 2014World Journal of Gastroenterology2014,20,23:3
5Recognition of walking environments and gait period by surface electromyography显示文摘Recognizing and predicting the movement and intention of the wearer in control of an exoskeleton robot is very challenging. It is difficult for exoskeleton robots, which measure and drive human movements, to interact with humans. Therefore, many different types of sensors are needed. When using various sensors, a data design is needed for effective sensing. An electromyographic(EMG) signal can be used to identify intended motion before the actual movement, and the delay time can be shortened via control of the exoskeleton robot. Before using a lower limb exoskeleton to help in walking, the aim of this work is to distinguish the walking environment and gait period using various sensors, including the surface electromyography(sE MG) sensor. For this purpose, a gait experiment was performed on four subjects using the ground reaction force, human–robot interaction force, and position sensors with sEMG sensors. The purpose of this paper is to show progress with the use of sEMG when recognizing walking environments and the gait period with other sensors. For effective data design, we used a combination of sensor types, sEMG sensor locations, and sEMG features. The results obtained using an individual mechanical sensor together with sEMG showed improvement compared to the case of using an individual sensor, and the combination of sEMG and position information showed the best performance in the same number of combinations of three sensors. When four sensor combinations were used, the environment classification accuracy was 96.1%, and the gait period classification accuracy was 97.8%. Vastus medialis(VM) and gastrocnemius(GAS) were the most effective combinations of two muscle types among the five sEMG sensor locations on the legs, and the results were 74.4% in pre-heel contact(preHC) and 71.7% in pre-toe-off(preTO) for environment classification, and 68.0% for gait period classification, when using only the sE MG sensor. The two effective sE MG feature combinations were 'mean absolute value(MAV), zero crossings(ZC)' and 'MAV, waveform length(WL)', and the 'MAV, ZC' results were 80.0%, 77.1%, and 75.5%. 6Differential Inhibition of Nav1.7 and Neuropathic Pain by Hybridoma-Produced and Recombinant Monoclonal Antibodies that Target Nav1.7显示文摘The voltage-gated Na^+ channel subtype Nav1.7 is important for pain and itch in rodents and humans. We previously showed that a Nav1.7-targeting monoclonal antibody(SVmab) reduces Na+ currents and pain and itch responses in mice. Here, we investigated whether recombinant SVmab(rSVmab) binds to and blocks Nav1.7 similar to SVmab. ELISA tests revealed that SVmab was capable of binding to Nav1.7-expressing HEK293 cells,mouse DRG neurons, human nerve tissue, and the voltagesensor domain Ⅱ of Nav1.7. In contrast, rSVmab showed no or weak binding to Nav1.7 in these tests. Patch-clamp recordings showed that SVmab, but not rSVmab, markedly inhibited Na+ currents in Nav1.7-expressing HEK293 cells. Notably, electrical field stimulation increased the blocking activity of SVmab and rSVmab in Nav1.7-expressing HEK293 cells. SVmab was more effective than rSVmab in inhibiting paclitaxel-induced mechanical allodynia. SVmab also bound to human DRG neurons and inhibited their Na^+ currents. 7Metamaterial band theory:fundamentals & applications显示文摘Remarkable progress has been made over the past decade in controlling light propagation and absorption in compact devices using nanophotonic structures and metamaterials.From sensing and modulation,to on-chip communication and light trapping for solar cells,new device applications and opportunities motivate the need for a rigorous understanding of the modal properties of metamaterials over a broad range of frequencies.In this review,we provide an overview of a metamaterial band theory we have developed that rigorously models the behavior of metamaterials made of dispersive materials such as metals.The theory extends traditional photonic band theory for periodic dielectric structures by coupling the mechanical motion of electrons in the metal directly to Maxwell’s equations.The solution for the band structures of metamaterials is then reduced to a standard matrix eigenvalue problem that nevertheless fully takes into account the dispersive properties of the constituent materials.As an application of the metamaterial band theory,we show that one can develop a perturbation formalism based on this theory to physically explain and predict the efect of dielectric refractive index modulation or metallic plasma frequency variation in metamaterials.Furthermore,the metamaterial band theory also provides an intuitive physical picture of the source of modal material loss,as well as a rigorous upper bound on the modal material loss rate of any plasmonic,metamaterial structure.This in turn places fundamental limits on the broadband operation of such devices for applications such as photodetection and absorption.RAMAN Aaswath Pattabhi SHIN Wonseok FAN ShanHui 2013Science China(Information Sciences)2013,56,12:2

13Effect of Ca addition on the microstructure and mechanical properties of heat-treated Mg-6.0Zn-1.2Y-0.7Zr alloy显示文摘In this study,the effects of Ca addition on the microstructure,thermal properties,and mechanical properties of a Mg-6.0Zn-1.2Y-0.7Zr(ZWK611)alloy at room temperature and 150℃ were investigated.With an increase in the Ca content,the ignition resistance of the ZWK611 alloy improved and the grains became finer.The as-cast ZWK611 alloy consisted mainly of the dendriticα-Mg matrix and I-phase(Mg_(3)Zn_(6)Y)at the grain boundaries.On the other hand,theτ-(Ca_(2)Mg_(6)Zn_(3))and I-phases were formed in the Ca-added ZWK611 alloy.The fraction of theτ-phase increased with an increase in the Ca content.After the solid-solution treatment,these phases remained partially at the grain boundaries of the Ca-added ZWK611 alloys and an additional W-phase(Mg_(3)Zn_(3)Y)was observed.The phases remaining at the grain boundaries restricted the grain growth through the grain boundary pinning effect.The aging treatment resulted in the formation of MgZn'precipitates in theα-Mg matrix of the alloys.These precipitates were more uniformly distributed in the Ca-added alloys than in the alloys without Ca.Thus,the heat treatment-induced precipitation improved the tensile and creep properties of the T6-treated alloys.The T6-treated ZWK611+0.7Ca alloy exhibited the best mechanical properties at room temperature and 150℃ among all the tested alloys.Young Gil Jung Wonseok Yang Yong Joo Kim Shae.K.Kim Young Ok Yoon Hyunkyu Lim Do Hyang Kim 2021Journal of Magnesium and Alloys2021,9,5:1
14Effect of Qingxue Dan on obesity and metabolic biomarker: a double-blind randomized-controlled pilot study显示文摘OBJECTIVE: To investigate the clinical effect of Qingxue Dan(QXD) on obesity and metabolic biomarker related to obesity.METHODS: A randomized, double blinded, placebo-controlled trial with a paralleled study design was conducted. Twenty-six obese volunteers aged between 30 and 60 with obesity and more than 2 metabolic risk factors were recruited at the department of oriental rehabilitation medicine, Kyunghee university oriental medical hospital, Seoul, Korea. Subjects were randomly assigned to an intervention(QXD) group or a placebo group, and treated with 900 mg/d of QXD or placebo medicine for 8 weeks. Primary endpoint was the change of body mass index(BMI) at 8 week from baseline. Secondary outcomes included the change of body composition, abdominal fat mass measured with Dual energy X-ray absorptiometry(DXA), blood pressure,lipid profiles and the homeostasis model assessment for insulin resistance(HOMA-IR).RESULTS: BMI was decreased in the QXD group sig-nificantly. Total body fat, abdominal fat mass measured with DXA Region of Interest and waist circumference(WC) showed a trend toward decreasing in the QXD group, but fat free mass was decreased in all groups. Triglyceride(TG) was decreased in QXD group significantly, but WC, total cholesterol and high-density lipoprotein cholesterol were increased in both group. BP didn't change during the study period. HOMA-IR is decreased in both groups without group effect.CONCLUSION: 8-weeks of oral administrations of QXD(900 mg/d) reduces BMI, with a tendency of lose of total body fat mass, especially abdominal fat. It also significantly reduced serum TG level.These results suggest QXD could be used to treat obesity and metabolic risk factors.
