| 1 | Long-term ex vivo monitoring of in vivo microRNA activity in liver using a secreted luciferase sensor显示文摘Technology for monitoring in vivo microRNA (miRNA) activity is extremely important for elucidating miRNA biology.However,in vivo studies of miRNA have been hampered by the lack of a convenient approach to reliably reflect real-time functional changes in miRNAs.Sensors for miRNA were developed by adding miRNA target sequences to the 3'-untranslated region of Gaussia princeps luciferase (Gluc) mRNA.These sensors were then evaluated in vitro and in vivo by measuring Gluc activity in cell supernatants and in peripheral blood.Sensors driven by the CMV promoter were effective for monitoring miR-122 in living cells,but not for the long-term monitoring of miR-122 or miR-142 in mouse liver because of CMV-promoter silencing.Replacing the CMV promoter with a CAG promoter rendered these sensors effective for the long-term monitoring of relevant liver miRNA activities.We subsequently used the CAG-promoter-based sensor for the long-term monitoring of endogenous liver miR-122,miR142 and miR-34a activities,as well as for exogenous miR-34a activity.Our study demonstrates that real-time in vivo activities of miRNAs can be continuously and conveniently detected in mouse liver using the sensors that we have developed. | WANG Gang DONG XiaoYan HU JianYang TIAN WenHong YUCHI Jie WANG Yue WU XiaoBing | 2011 | Science China(Life Sciences)2011,54,5: | 6 |
| 2 | Evaluation of miR-122-regulated suicide gene therapy for hepatocellular carcinoma in an orthotopic mouse model显示文摘Objective:Intratumoral administration of adenoviral vector encoding herpes simplex virus(HSV)thymidine kinase(TK)gene(Ad-TK)followed by systemic ganciclovir(GCV)is an effective approach in treating experimental hepatocellular carcinoma(HCC).However,hepatotoxicity due to unwanted vector spread and suicide gene expression limited the application of this therapy.miR-122 is an abundant,liver-specific microRNA whose expression is decreased in human primary HCC and HCC-derived cell lines.These different expression profiles provide an opportunity to induce tumor-specific gene expression by miR-122 regulation.Methods:By inserting miR-122 target sequences(miR-122T)in the 3'untranslated region(UTR)of TK gene,we constructed adenovirus(Ad)vectors expressing miR-122-regulated TK(Ad-TK-122T)and report genes.After intratumoral administration of Ad vectors into an orthotopic miR-122-deficient HCC mouse model,we observed the miR-122-regulated transgene expression and assessed the antitumor activity and safety of Ad-TK-122T.Results:Insertion of miR-122T specifically down-regulated transgene expression in vitro and selectively protected the miR-122-positive cells from killing by TK/GCV treatment.Insertion of miR-122T led to significant reduction of tansgene expression in the liver without inhibition of its expression in tumors in vivo,resulting in an 11-fold improvement of tumor-specific transgene expression.Intratumoral injection of Ad vectors mediated TK/GCV system led to a vector dosage-dependent regression of tumor.The insertion of miR-122T does not influence the antitumor effects of suicide gene therapy.Whereas mice administrated with Ad-TK showed severe lethal hepatotoxicity at the effective therapeutic dose,no liver damage was found in Ad-TK-122T group.Conclusions:miR-122-regulated TK expression achieved effective anti-tumor effects and increased the safety of intratumoral delivery of adenovirus-mediated TK/GCV gene therapy for miR-122-deficient HCC. | Gang Wang Xiaoyan Dong Wenhong Tian Yue Lu Jianyan Hu Yunfan Liu Jie Yuchi Xiaobing Wu | 2013 | Chinese Journal of Cancer Research2013,25,6: | 5 |

| 4 | Cenozoic Sea-land Transition and its Petroleum Geological Significance in the Northern South China Sea显示文摘The process of Cenozoic sea-land changes in the northern South China Sea(SCS)controlled the sedimentary filling pattern and played an important role in the petroleum geological characteristics of the northern marginal sedimentary basins.Under the control of the opening process of the SCS,the northern SCS Cenozoic transgression generally showed the characteristics of early in the east and late in the west,and early in the south and late in the north.The initial transgression occurred in the Eocene in the Taixinan Basin(TXNB)of the eastern SCS,while the transgression occurred until the Pliocene in the Yinggehai Basin(YGHB)of the western SCS.International Ocean Discovery Program(IODP)expeditions(Expeditions 367/368)revealed that the initial transgression of the SCS basin occurred at approximately 34 Ma,which was the initial opening time of the SCS.The period of drastic changes in the sedimentary environment caused by large-scale transgression corresponded to the opening time of the southwestern subbasin(approximately 23 Ma),which also represented the peak of the spreading of the SCS.The sea-land transition process controls the distribution of alternating continent-marine facies,marine facies source rocks and reservoirs in the basins.The marine facies source rocks of the basins in the northern SCS have a trend of gradually becoming younger from east to west,which is consistent with the regional process of gradual transgression from east to west.Regional sea-level changes were comprehensively influenced by SCS opening and global sea-level changes.These processes led to the early development in the east and south and late development in the west and north for the carbonate platform in the SCS.Carbonate platforms form another type of'selfgenerating and self-accumulating'oil-gas reservoir in the northern SCS.The sea-land transition controlled the depositional filling patterns of different basins and laid the foundation of marine deposits for oil and gas resources.The source-reservoircap assemblage in the northern SCS was controlled horizontally by provenance supply and sedimentary environmental changes caused by sea-land transition and vertically by the tectonic evolution of the SCS and regional sea-level changes. | ZHAO Zhigang ZHANG Hao CUI Yuchi TANG Wu QIAO Peijun | 2021 | Acta Geologica Sinica(English Edition)2021,95,1: | 2 |
| 5 | In vitro activity of allicin combined with two antibiotics on intestinal Shigella显示文摘Objective: We aimed to evaluate the combined antibacterial effects of allicin in combination with levofloxacin and ceftriaxone on Shigella isolated from the intestinal tract in vitro. Materials and Methods: Using a checkerboard design, broth microdilution assay was used to test the effects of the compounds on the organism. We also determined the MIC of the two groups of antibacterial drugs against 30 strains of Shigella and calculated the fractional inhibitory concentration(FIC) index, to judge the combination effect. Result: After the combined application of allicin and ceftriaxone the MIC decreased significantly. Distribution of the FIC index was as follows: FIC ≤0.5, accounting for 10%; 0.5< FIC ≤1.0, accounting for 60%; 1 < FIC ≤2, accounting for 30%; FIC >2, percentage is zero. After combined application of allicin and levofloxacin, distribution of FIC index was as follows: FIC≤0.5, ratio is zero; 0.5< FIC ≤1, accounting for 56.7%; 1 < FIC ≤2, accounting for 43.3%; FIC >2, ratio is zero. | 9 | Late Eocene–early Miocene provenance evolution of the Crocker Fan in the southern South China Sea显示文摘There are many large-scale Cenozoic sedimentary basins with plentiful river deltas,deep-water fans and carbonate platforms in the southern South China Sea.The Crocker Fan was deposited as a typical submarine fan during the late Eocene–early Miocene,and stretches extensively across the entire Sarawak–Sabah of the northern Borneo area.However,systematic analyses are still lacking regarding its sediment composition and potential source suppliers.No consensus has been reached yet on the provenance evolution and sedimentary infilling processes,which seriously impeded the oil-and-gas exploration undertakings.By combining with sedimentaryfacies identification,heavy mineral assemblages,elemental geochemistry and detrital zircon U-Pb dating,this paper aims to generalize an integrated analysis on the potential provenance terranes and restore source-to-sink pathways of the Crocker Fan.In general,the Crocker Fan was initially formed over the Cretaceous–lower/middle Eocene Rajang Group by an angular Rajang unconformity.The continual southward subduction of the protoSouth China Sea resulted in magmatic activities and subsequent regional deformation and thrusting along the Lupar Line in the northern Borneo.The lowermost Crocker sequence is featured by a thick conglomerate layer sourced from in-situ or adjacent paleo-uplifts.From the late Eocene to the early Miocene,the Crocker Fan was constantly delivered with voluminous detritus from the Malay Peninsula of the western Sundaland.The Zengmu Basin was widely deposited with delta plain and neritic facies sediments,while the Brunei-Sabah Basin,to the farther east,was ubiquitously characterized by turbiditic sequences.The Crocker Fan successions are overall thick layers of modest-grained sandstones,which formed high-quality reservoirs in the southern South China Sea region. | Yuchi Cui Lei Shao Wu Tang Peijun Qiao Goh Thian Lai Yongjian Yao | 2023 | Acta Oceanologica Sinica2023,42,3: | 1 |
