1Two gap-free reference genomes and a global view of the centromere architecture in rice显示文摘Rice(Oryza sativa),a major staple throughout the world and a model system for plant genomics and breeding,was the first crop genome sequenced almost two decades ago.However,reference genomes for all higher organisms to date contain gaps and missing sequences.Here,we report the assembly and analysis of gap-free reference genome sequences for two elite O.sativa xian/indica rice varieties,Zhenshan 97 and Minghui 63,which are being used as a model system for studying heterosis and yield.Gap-free reference genomes provide the opportunity for a global view of the structure and function of centromeres.We show that all rice centromeric regions share conserved centromere-specific satellite motifs with different copy numbers and structures.In addition,the similarity of CentO repeats in the same chromosome is higher than across chromosomes,supporting a model of local expansion and homogenization.Both genomes have over 395 non-TE genes located in centromere regions,of which∼41%are actively transcribed.Two large structural variants at the end of chromosome 11 affect the copy number of resistance genes between the two genomes.The availability of the two gap-free genomes lays a solid foundation for further understanding genome structure and function in plants and breeding climate-resilient varieties.Jia-Ming Song Wen-Zhao Xie Shuo Wang Yi-Xiong Guo Dal-Hoe Koo Dave Kudrna Chenbo Gong Yicheng Huang Jia-Wu Feng Wenhui Zhang Yong Zhou Andrea Zuccolo Evan Long Seunghee Lee Jayson Talag Run Zhou Xi-Tong Zhu Daojun Yuan Joshua Udall Weibo Xie Rod AWing Qifa Zhang Jesse Poland Jianwei Zhang Ling-Ling Chen 2021Molecular Plant2021,14,10:9
2Biochemical sensing in graphene-enhanced microfiber resonators with individual molecule sensitivity and selectivity显示文摘Photonic sensors that are able to detect and track biochemical molecules offer powerful tools for information acquisition in applications ranging from environmental analysis to medical diagnosis.The ultimate aim of biochemical sensing is to achieve both quantitative sensitivity and selectivity.As atomically thick films with remarkable optoelectronic tunability,graphene and its derived materials have shown unique potential as a chemically tunable platform for sensing,thus enabling significant performance enhancement,versatile functionalization and flexible device integration.Here,we demonstrate a partially reduced graphene oxide(prGO)inner-coated and fiber-calibrated Fabry-Perot dye resonator for biochemical detection.Versatile functionalization in the prGO film enables the intracavity fluorescent resonance energy transfer(FRET)to be chemically selective in the visible band.Moreover,by measuring the intermode interference via noise canceled beat notes and locked-in heterodyne detection with Hz-level precision,we achieved individual molecule sensitivity for dopamine,nicotine and single-strand DNA detection.This work combines atomic-layer nanoscience and high-resolution optoelectronics,providing a way toward high-performance biochemical sensors and systems.Zhongxu Cao Baicheng Yao Chenye Qin Run Yang Yanhong Guo Yufeng Zhang Yu Wu Lei Bi Yuanfu Chen Zhenda Xie Gangding Peng Shu-Wei Huang Chee Wei Wong Yunjiang Rao 2019Light(Science & Applications)2019,8,1:8
3Human adenovirus(HAdV)infection in children with acute respiratory tract infections in Guangzhou,China,2010–2021:a molecular epidemiology study显示文摘Background Human adenovirus(HAdV)infection can cause a variety of diseases.It is a major pathogen of pediatric acute respiratory tract infections(ARIs)and can be life-threatening in younger children.We described the epidemiology and subtypes shifting of HAdV among children with ARI in Guangzhou,China.Methods We conducted a retrospective study of 161,079 children diagnosed with acute respiratory illness at the Guangzhou Women and Children’s Medical Center between 2010 and 2021.HAdV specimens were detected by real-time PCR and the hexon gene was used for phylogenetic analysis.Results Before the COVID-19 outbreak in Guangzhou,the annual frequency of adenovirus infection detected during this period ranged from 3.92%to 13.58%,with an epidemic peak every four to fve years.HAdV demonstrated a clear seasonal distribution,with the lowest positivity in March and peaking during summer(July or August)every year.A signifcant increase in HAdV cases was recorded for 2018 and 2019,which coincided with a shift in the dominant HAdV subtype from HAdV-3 to HAdV-7.The latter was associated with a more severe disease compared to HAdV-3.The average mortality proportion for children infected with HAdV from 2016 to 2019 was 0.38%but increased to 20%in severe cases.After COVID-19 emerged,HAdV cases dropped to 2.68%,suggesting that non-pharmaceutical interventions probably reduced the transmission of HAdV in the community.Conclusion Our study provides the foundation for the understanding of the epidemiology of HAdV and its associated risks in children in Southern China.Yi Chen Tao Lin Chang‑Bing Wang Wan‑Li Liang Guang‑Wan Lian Mark Zanin Sook‑San Wong Xin‑Gui Tian Jia‑Yu Zhong Ying‑Ying Zhang Jia‑Hui Xie Ling‑Ling Zheng Fei‑Yan Chen Run Dang Ming‑Qi Zhao Yi‑Yu Yang Rong Zhou Bing Zhu 2022World Journal of Pediatrics2022,18,8:5
4Binding between Prion Protein and Aβ Oligomers Contributes to the Pathogenesis of Alzheimer’s Disease显示文摘A plethora of evidence suggests that protein misfolding and aggregation are underlying mechanisms of various neurodegenerative diseases,such as prion diseases and Alzheimer's disease(AD).Like prion diseases,AD has been considered as an infectious disease in the past decades as it shows strain specificity and transmission potential.Although it remains elusive how protein aggregation leads to AD,it is becoming clear that cellular prion protein(PrP^c)plays an important role in AD pathogenesis.Here,we briefly reviewed AD pathogenesis and focused on recent progresses how PrP^c contributed to AD development.In addition,we proposed a potential mechanism to explain why infectious agents,such as viruses,conduce AD pathogenesis.Microbe infections cause AD deposition and upregulation of PrP^c,which lead to high affinity binding between AD oligomers and PrP^c.The interaction between PrP^c and AP oligomers in turn activates the Fyn signaling cascade,resulting in neuron death in the central nervous system(CNS).Thus,silencing PrP^c expression may turn out be an effective treatment for PrP^c dependent AD.Chang Kong Hao Xie Zhenxing Gao Ming Shao Huan Li Run Shi Lili Cai Shanshan Gao Taolei Sun Chaoyang Li 2019Virologica Sinica2019,34,5:4
5A novel approach for fabricating a CNT/AlSi composite with the self-aligned nacre-like architecture by cold spraying显示文摘A fully dense carbon nanotubes (CNTs) reinforced AlSi matrix composite with the multiscale nacre-like architecture was designed and successfully realized by flake powder metallurgy followed by cold spraying (CS). The nanolaminated and ultrafine-grained structure initially created in the CNT/AlSi flaky powder was perfectly conserved, due to the typical ‘cold’ feature of CS. As discussed based on finite element analysis and single splat observation, self-alignment behavior of the flaky powders during impact also allowed the formation of the microlaminated structure. 9Designated Cloud Server Public Key Encryption with Keyword Search from Lattice in the Standard Model显示文摘Recently, how to retrieve the encrypted data efficiently from a cloud storage system becomes a hot topic. Public key encryption with keyword search(PEKS)can allow one to search the encrypted data with a keyword efficiently. Due to the booming of post-quantum cryptography, we propose public key encryption with keyword search from lattice assumption, which can resist quantum computer attacks. We delegate a particular cloud server to search the results, thus only the designated cloud server can finish the test process. Our scheme has been proved ciphertext indistinguishability in the standard model, which can reflect its security in the real world.
