| 2 | miR-503-3p promotes epithelialemesenchymal transition in breast cancer by directly targeting SMAD2 and E-cadherin显示文摘Although progress in clinical and basic research has significantly increased our understanding of breast cancer, little is known about the molecular mechanism underlying breast cancer metastasis. Identification of effective therapeutic targets to prevent breast cancer metastasis is urgently needed. The function of mi R-503-3p has been investigated in other cancers, but its role in breast cancer remains undefined.Here, we found that mi R-503-3p was overexpressed in breast cancer tissue and plasma compared with adjacent normal breast tissue and with plasma from healthy individuals. Moreover, we identified mi R-503-3p to be an oncogene of breast cancer cell proliferation, migration and invasion. Upregulation of mi R-503-3p in breast cancer cells inhibited expression of epithelialemesenchymal transition(EMT)-related protein SMAD2 and the epithelial marker protein E-cadherin by directly binding to their m RNA30 untranslated region, whereas increased expression of mesenchymal marker proteins, including vimentin and N-cadherin. | 4 | Polysaccharides in Sipunculus nudus: Extraction Condition Optimization and Antioxidant Activities显示文摘Marine organisms constitute unlimited resource of bioactive substances due to their high biodiversity and represent a valuable source of new compounds. This study optimized the alkali-extraction conditions and antioxidant activities of soluble polysaccharides from the body wall of Sipunculus nudus. The effects of solid-liquid ratio, extraction duration, extraction temperature, and alkali concentration on the yield of S. nudus polysaccharides(SNP) were examined, according to which the optimal combination of extraction parameters was obtained by an orthogonal test. The relative influencing importance of different extraction parameters on the yield of SNP followed the order as solid-liquid ratio > extraction temperature > alkali concentration > extraction duration. The highest extraction yield, 1.98%, was achieved under an optimal extraction condition: temperature 60℃, solid-liquid ratio 1:6 g mL^(-1), duration 5 h, and alkali(Na OH) mass fraction 6%. The in vitro antioxidant activities examination showed that extracted SNP under this optimized condition had strong power in reducing certain hydroxyl and superoxide radical scavenging abilities. | 6 | Distribution and origin of high magnetic anomalies at Luobusa Ophiolite in Southern Tibet显示文摘The Luobusa Ophiolite,Southern Tibet,lies in the eastern portion of Indus–Yarlung Zangbo suture zone that separates Eurasia from the Indian continent.An aeromagnetic reconnaissance survey has revealed an EWtrending Yarlung Zangbo River aeromagnetic anomaly zone,and it is considered to be caused mainly by the Indus–Yarlung Zangbo Ophiolite.The Luobusa Ophiolite reflects the eastern portion of the Yarlung Zangbo River aeromagnetic anomaly zone.Conventionally,the ultramafic rock in the Luobusa Ophiolite is considered as the origin of the high magnetic anomalies.However,results from the surface magnetic survey and the magnetic susceptibility measurements from drill cores indicate that the high magnetic anomalies are distributed inhomogeneously in the Luobusa Ophiolite.In some cases,the susceptibility exhibits more than 30 times difference between two sides of the same sample.A fact emerged that the susceptibility of dunite with serpentinization is higher than that of fresh dunite,harzburgite and chromite when we analyzed the measurement results.In order to understand the origin of the high magnetic anomalies,we measured the density and susceptibility of 17 samples,microscopic and electron probe analyses have been performed as well.The result indicates the presence of dunite with serpentinization containing an abundant of micro-fissures filled with magnetite.Olivine has a susceptibility of about 2.7–351(910-5SI),pyroxene about 16–320,and chromite about200–800.All these units feature relatively low susceptibility in ultramafic rock,and only the magnetite is characterized by a high susceptibility of about 200,000(910-5SI).Based on these observations,we conclude that the precipitation of magnetite in the process of serpentinization of the olivine caused by the geological process in the Luobusa Ophiolite is the origin of high magnetic anomalies. | Lanfang He Xiumian Hu Yabing Zha Ligui Xu Yaohui Wang | 2014 | Chinese Science Bulletin2014,59,23: | 2 |

[Methods]Juvenile peanut worm with average body weight of( 16. 77 ± 0. 40) mg were fed with five isonitrogenous and isoenergetic diets formulated to contain different carbohydrate / lipid( C / L) ratios( 0. 68,1. 02,1. 62,2. 61 or 4. 35) for 8 weeks. [Results] The dietary C / L ratios had no significant effects on survival of juvenile peanut worm( P > 0. 05). With the increasing of dietary C / L ratios,weight growth rate( WGR)and specific growth rate( SGR) increased when dietary C / L ratios were between 0. 68 and 2. 61,and then decreased significantly with dietary C / L ratio further increased. When the dietary C / L ratio was 2. 61( carbohydrate level was 20. 64%,lipid level was 7. 92%) both WGR and SGR produced the maximum value,they were 451. 78 ± 16. 85% and 3. 41 ± 0. 06 % / d,respectively. The dietary C / L ratios had significant effects on body moisture,body protein content and body lipid content( P < 0. 05),but no significant effects on body ash content of juvenile peanut worm( P > 0. 05). Different C / L ratios had significant effects on amylase activity and lipases activity of juvenile peanut worm( P < 0. 05),but no significant effects on protease activity( P > 0. 05). | 14 | Lipid Biomarkers and Their Stable Carbon Isotopes in Ancient Seep Carbonates from SW Taiwan, China显示文摘Four massive brecciated, chimney-like, and slender pipe network carbonate samples(JA-4, JA-5, JX-8 and BG-12) were collected from southwestern Taiwan, which were suggested to have formed as a result of anaerobic oxidization of methane(AOM). Considering that the environmental conditions of the carbonates precipitation and the sources of carbon and organic matter need to be further declared, molecular fossils and compound-specific carbon isotopic investigations of the carbonates were conducted in this study. According to lipid biomarkers of 2,6,10,15,19-pentamethyleicosane(PMI) and squalane diagnostic to methanotrophic archaea, as well as the extremely low δ^(13)C values(as low as -1^(13).4‰) detected in samples JA-4, JA-5 and JX-8, these carbonates were revealed to be a result of AOM. Based on the varied δ^(13)C values of characteristic archaea biomarkers in specific samples, biogenic methane was proposed to be responsible for the formation of samples JA-4 and JA-5, whereas a mixed carbon source of ^(13)C-depleted methane and ^(13)C-enriched residual CO_2 from methanogenesis was suggested for the carbonate of JX-8 due to the co-occurrence of a highly positive δ^(13) C_(carb) value(+8‰) and a moderate ^(13)C depletion of PMI. The low content of AOM-related biomarkers and the absence of indicators for ANME-2 suggested that these carbonates were formed in weak seep settings. By comparison, no typical lipid biomarkers for methanotrophic archaea was detected in carbonate BG-12. The short-chain and long-chain n-alkanes accounted for 30% and 45% of all hydrocarbons, respectively, with a CPI value of 1.2, suggesting that the n-alkanes were derived from both marine organisms and terrestrial inputs. Results showed that diets with different carbohydrate sources had significant influences on daily weight gain and digestive enzyme activities of S. nudus( P < 0. 05). Daily weight gain in gelatinized corn starch group was significantly higher than that in other groups( P < 0. 05); daily weight gain in glucose group was significantly lower than that in other groups( P < 0. 05); daily weight gain in the three ungelatinized starch groups was significantly higher than that in glucose,sucrose and dextrin groups( P < 0. 05). Analysis of digestive enzyme activities showed that dietary carbohydrate sources had significant influences on digestive enzyme activities of S. nudus( P < 0. 05). Protease activities and amylase activities of S. nudus in sucrose group were the highest,which were significantly higher than that in the other groups( P < 0. 05); lipase activities of S. nudus in glucose group were the lowest,which were significantly lower than that in other groups( P < 0. 05).
