| 1 | Multi-omics analyses of 398 foxtail millet accessions reveal genomic regions associated with domestication,metabolite traits,and antiinflammatory effects显示文摘Foxtail millet(Setaria italica),which was domesticatedfromthewild speciesgreenfoxtail(Setaria viridis),isa richsource of phytonutrientsfor humans.To evaluate how breeding changed themetabolome offoxtail millet grains,we generated and analyzed the datasets encompassing the genomes,transcriptomes,metabolomes,and anti-inflammatory indices from 398 foxtail millet accessions.We identified hundreds of common variants that influence numerous secondary metabolites.We observed tremendous differences in natural variations of the metabolites and their underlying genetic architectures between distinct sub-groups of foxtail millet.Furthermore,we found that the selection of the gene alleles associated with yellow grains led to altered profiles of metabolites such as carotenoids and endogenous phytohormones.Using CRiSPR-mediated genome editing wevalidated the function of PHYTOENE SYNTHASE1(PSY1)gene in affecting milletgrain colorand quality.Interestingly,our in vitro cell inflammation assays showed that 83 metabolites in millet grains have anti-inflammatory effects.Taken together,ourmulti-omics study illustrates how the breeding history of foxtail millet has shaped its metabolite profile.The datasets we generated in this study also provide important resources for further understanding how millet grain quality is affected by different metabolites,laying the foundations for future millet genetic research and metabolome-assisted improvement. | Xukai Li Jianhua Gao Jingyi Song Kai Guo Siyu Hou Xingchun Wang Qiang He Yanyan Zhang Yakun Zhang Yulu Yang Jiaoyan Tang Hailang Wang Staffan Persson Mingquan Huang Lishuai Xu Linlin Zhong Dongqin Li Yongming Liu Hua Wu Xianmin Diao Peng Chen Xiaowen Wang Yuanhuai Han | 2022 | Molecular Plant2022,15,8: | 8 |
| 2 | A benchmark test of boson sampling on Tianhe-2 supercomputer显示文摘Boson sampling, thought to be intractable classically, can be solved by a quantum machine composed of merely generation, linear evolution and detection of single photons. Such an analog quantum computer for this specific problem provides a shortcut to boost the absolute computing power of quantum computers to beat classical ones. However, the capacity bound of classical computers for simulating boson sampling has not yet been identified. Here we simulate boson sampling on the Tianhe-2 supercomputer, which occupied the first place in the world ranking six times from 2013 to 2016. We computed the permanent of the largest matrix using up to 312 000 CPU cores of Tianhe-2, and inferred from the current most efficient permanent-computing algorithms that an upper bound on the performance of Tianhe-2 is one 50-photon sample per ~100 min. | 3 | Tuning charge density of chimeric antigen receptor optimizes tonic signaling and CAR-T cell fitness显示文摘Tonic signaling of chimeric antigen receptor(CAR),i.e.,the spontaneous CAR activation in the absence of tumor antigen stimulation,is considered to be a pivotal event controlling CAR-T efficacy.However,the molecular mechanism underlying the spontaneous CAR signals remains elusive.Here,we unveil that positively charged patches(PCPs)on the surface of the CAR antigenbinding domain mediate CAR clustering and result in CAR tonic signaling.For CARs with high tonic signaling(e.g.,GD2.CAR and CSPG4.CAR),reducing PCPs on CARs or boosting ionic strength in the culture medium during ex vivo CAR-T cell expansion minimizes spontaneous CAR activation and alleviates CAR-T cell exhaustion.In contrast,introducing PCPs into the CAR with weak tonic signaling,such as CD19.CAR,results in improved in vivo persistence and superior antitumor function.These results demonstrate that CAR tonic signaling is induced and maintained by PCP-mediated CAR clustering.Notably,the mutations we generated to alter the PCPs maintain the antigen-binding affinity and specificity of the CAR.Therefore,our findings suggest that the rational tuning of PCPs to optimize tonic signaling and in vivo fitness of CAR-T cells is a promising design strategy for the nextgeneration CAR. | Jian Chen Shizhen Qiu Wentao Li Kun Wang Yu Zhang Han Yang Baichuan Liu Guangfei Li Li Li Min Chen Junjie Lan Jiahua Niu Peijie He Lei Cheng Gaofeng Fan Xin Liu Xianmin Song Chenqi Xu Haitao Wu Haopeng Wang | 2023 | Cell Research2023,33,5: | 2 |

| 10 | Resistive switching and artificial synaptic performances of memristor based on low-dimensional bismuth halide perovskites显示文摘Zero-dimensional(0D)-Cs_(3)Bi_(2)I_(9),two-dimensional(2D)-Cs_(3)Bi_(2)Br_(9),and one-dimensional(1D)-Cs3Bi2Cl9 perovskite films have been successfully grown on indium tin oxide(ITO)glass substrates,which were used to fabricate memristors with the structure of Al/Cs_(3)Bi_(2)X_(9)(X=I,Br,and Cl)/ITO glass.The current three types of memristors exhibited bipolar resistive switching behaviors.Both the endurance and retention time tests clearly demonstrated the excellent stability of present devices.Especially,the ON/OFF ratio of the 0D-Cs_(3)Bi_(2)I_(9)device is close to 104 at the reading voltage of 0.1 V,which is nearly 100 and 1000 times larger than those of the 1D-Cs3Bi2Cl9 device and the 2D-Cs_(3)Bi_(2)Br_(9)device,respectively.The activation energy of halide vacancies in the Cs_(3)Bi_(2)X_(9)(X=I,Br,and Cl)films was calculated using the density functional theory by considering a minimum migration path,demonstrating the dimensionality of the Cs_(3)Bi_(2)X_(9)(X=I,Br,and Cl)film affected the formation and rupture of conductive filaments.Moreover,the short-term plasticity and long-term plasticity of biological synapse were simulated by evaluating the conductance responses of Al/Cs_(3)Bi_(2)X_(9)(X=I,Br,and Cl)/ITO devices under various voltage pulses in detail.The duration time of long-term plasticity in all the present devices can last for up to 250 s.The 0D-Cs_(3)Bi_(2)I_(9)device showed both the highest spikeduration-dependent plasticity and paired-pulse facilitation indexes compared to the other two devices.Additionally,the 0DCs_(3)Bi_(2)I_(9)device successfully established the associative learning behavior by simulating the Pavlov’s dog experiment. | Feifei Luo Yanzhao Wu Junwei Tong Fubo Tian Xianmin Zhang | 2023 | Nano Research2023,16,7: | 1 |

| 18 | Identification of no pollen 1 provides a candidate gene for heterosis utilization in foxtail millet(Setaria italica L.)显示文摘Male sterility is a common biological phenomenon in plant kingdom and has been used to generate male-sterile lines, which are important genetic resources for commercial hybrid seed production. Although increasing numbers of male-sterility genes have been identified in rice(Oryza sativa) and Arabidopsis(Arabidopsis thaliana), few male-sterility-related genes have been characterized in foxtail millet(Setaria italica). In this study, we isolated a male-sterile ethyl methanesulfonate-generated mutant in foxtail millet, no pollen 1(sinp1), which displayed abnormal Ubisch bodies, defective pollen exine and complete male sterility. Using bulk segregation analysis, we cloned SiNP1 and confirmed its function with CRISPR/Cas9 genome editing. SiNP1 encoded a putative glucose-methanol-choline oxidoreductase.Subcellular localization showed that the SiNP1 protein was preferentially localized to the endoplasmic reticulum and was predominantly expressed in panicle. Transcriptome analysis revealed that many genes were differentially expressed in the sinp1 mutant, some of which encoded proteins putatively involved in carbohydrate metabolism, fatty acid biosynthesis, and lipid transport and metabolism, which were closely associated with pollen wall development. Metabolome analysis revealed the disturbance of flavonoids metabolism and fatty acid biosynthesis in the mutant. | 20 | A straight-forward seed production technology system for foxtail millet(Setaria italica)显示文摘For autogamous crops, a precondition for using heterosis is to produce sufficient pure male-sterile female parents that can be used to produce hybrid seeds. To date, cytoplasmic male sterility(CMS)and environment-sensitive genic male sterility(EGMS) have been used commercially to exploit heterosis for autogamous species. However, neither CMS nor EGMS has been established for foxtail millet(Setaria italica). Here, we report on the establishment and application of a seed production technology(SPT) system for this crop.First, we established a Ds Red-based SPT system,but found that it was unsuitable because it required the use of a fluorescent device for seed sorting. Instead, we constructed an SPT system with de novo betalain biosynthesis as the selection marker. This allowed us to distinguish transgenic seeds with the naked eye, thereby facilitating the identification of SPT maintainer line seeds. In this system, a seed sorter was not required to obtain sufficient seeds. The key point of the strategy is that the seed pool of the SPT maintainer line is propagated by artificial identification and harvesting of male-fertile individuals in the field, and the male-sterile line seed pool for hybrid production is produced and propagated by free pollination of male-sterile plants with the SPT maintainer line. In a field experiment, we obtained 423.96 kg male-sterile line seeds per acre,which is sufficient to plant 700.18 acres of farmland for hybrid seed production or male-sterile line reproduction.
