1Review and discussion on fire behavior of bridge girders显示文摘This paper presents an overview on fire behavior of bridge girders mainly including prestressed concrete(PC) bridge girders and steel bridge girders. The typical fire accidents occurred on bridges are illustrated and, the seriousness of posing threats to bridge structures resulted from increasing traffic fires, specially intense hydrocarbon fires generated from petrol-chemicals, is highlighted. The current researches, embracing high-temperature properties of constituent materials, prestress state, measurement in fire tests, numerical methods, structural fire resistance, and so forth, taken on coping with problems existing in fire behavior and structural fire behavior in bridge girders are reviewed and discussed. Further, strategies for enhancing fire resistance of bridge girders followed with failure criterion and mode in types of bridge structures are provided. Future research area along with emerging trends in structural fire behavior of bridge girders is also recommended for mitigating fire hazards occurred on bridge girders. Herein, it can be attained a conclusion from review and discussion that prestressed concrete bridge girders with thin webs, specially T-shaped bridge girder, are prone to unstable under fire exposure conditions. High-strength concrete utilized in prestressed concrete bridge girders is vulnerable to spalling at elevated temperature. 2Red and Blue Lights Significantly Affect Photosynthetic Properties and Ultrastructure of Mesophyll Cells in Senescing Grape Leaves显示文摘Light quality significantly affects photosynthetic efficiency in plants. The mechanisms for how light quality affects photosynthesis in grape is poorly understood. Therefore, to investigate the effects of different light qualities on chloroplast ultrastructure and photosynthesis efficiency, two grape cultivars ‘Italia'(slower speed of leaf senescence) and ‘Centennial Seedless'(faster speed of leaf senescence) grown under protected and delayed conditions were used. The three treatments, replicated three times, were control(no supplemental lighting), red light and blue light. Chlorophyll content, net photosynthetic rate, and the ratio of F_v/F_m significantly increased in red light relative to the control. The opposite trend was observed in blue light in the early phase of leaf senescence. At later stages, physiological indexes were gradually higher than that of control, resulting in a delay in leaf senescence. Compared to the control, red and blue light both significantly increased the chlorophyll a/b ratio. Electron microscopy showed that blue light caused severe damage to the fine structure of chloroplasts at early stages of leaf senescence, but effects at later stages of leaf senescence became less severe compared to the control. The degradation of chloroplast ultrastructure was apparently delayed in red light throughout the experimental timeframe compared to other treatments. 3Influence of environmental factors on land-surface water and heat exchange during dry and wet periods in the growing season of semiarid grassland on the Loess Plateau显示文摘On the basis of information from the project 'Land-surface Processes and their Experimental Study on the Chinese Loess Plateau', we analyzed differences in land-surface water and heat processes during the main dry and wet periods of the semiarid grassland growing season in Yuzhong County, as well as the influences of these environmental factors. Studies have shown that there are significant differences in changes of land-surface temperature and humidity during dry and wet periods. Daily average normalized temperature has an overall vertical distribution of 'forward tilting' and 'backward tilting' during dry and wet periods, respectively. During the dry period, shallow soil above 20-cm depth is the active temperature layer. The heat transfer rate in soil is obviously different during dry and wet periods. During the dry period, the ratio of sensible heat flux to net radiation(H/Rn) and the value of latent heat flux to net radiation(LE/Rn) have a linear relationship with 5-cm soil temperature; during the wet period, these have a nonlinear relationship with 5-cm soil temperature, and soil temperature of 16°C is the critical temperature for changes in the land-surface water and heat exchange trend on a daily scale. During the dry period, H/Rn and LE/Rn have a linear relationship with soil water content. During the wet period, these have a nonlinear relationship with 5-cm soil water content, and 0.21 m3 m?3 is the critical point for changes in the land-surface water and heat exchange trend at daily scale. During the dry period, for vapor pressure deficit less than 0.7 k Pa, H/Rn rises with increased vapor pressure deficit, whereas LE/Rn decreases with that increase. When that deficit is greater than 0.7 k Pa, both H/Rn and LE/Rn tend to be constant. During the wet period, H/Rn increases with the vapor pressure deficit, whereas LE/Rn decreases. 4Genome wide abnormal DNA methylome of human blastocyst in assisted reproductive technology显示文摘Proper reprogramming of parental DNA methylomes is essential for mammalian embryonic development.However,it is unknown whether abnormal methylome reprogramming occurs and is associated with the failure of embryonic development.Here we analyzed the DNA methylomes of 57 blastocysts and 29 trophectoderm samples with different morphological grades during assisted reproductive technology(ART) practices.Our data reveal that the global methylation levels of high-quality blastocysts are similar(0.30 ± 0.02,mean ± SD).while the methylation levels of low-quality blastocysts are divergent and away from those of high-quality blastocysts.The proportion of blastocysts with a methylation level falling within the range of 0.30 ± 0.02 in different grades correlates with the live birth rate for that grade.Moreover,abnormal methylated regions are associated with the failure of embryonic development.Furthermore,we can use the methylation data of cells biopsied from trophectoderm to predict the blastocyst methylation level as well as to detect the aneuploidy of the blastocysts.Our data indicate that global abnormal methylome reprogramming often occurs in human embryos,and suggest that DNA methylome is a potential biomarker in blastocyst selection in ART.Guoqiang Li Yang Yu Yong Fan Congru Li Xiaocui Xu Jialei Duan Rong Li Xiangjin Kang Xin Ma Xuepeng Chen Yuwen Ke Jie Yan Ying Lian Ping Liu Yue Zhao Hongcui Zhao Yaoyong Chen Xiaofang Sun Jianqiao Liu Jie Qiao Jiang Liu 2017Journal of Genetics and Genomics2017,44,10:5
5Bioaccumulation and the expression of hepatic cytochrome P450 genes in marine medaka(Oryzias melastigma)exposed to difenoconazole显示文摘This study was conducted to assess the effects of difenoconazole(DFZ), a triazole fungicide,on the hepatic biotransformation system and its bioaccumulation in marine medaka(Oryzias melastigma). Fish were exposed to DFZ(1, 10, 100, 1000 ng/L) for 180 days. The results showed that:(1) The m RNA levels of hepatic CYP1A1, CYP1 B, CYP1C1, CYP27 B and CYP3A40 were up-regulated, but those of CYP3A38 and CYP27A1 were down-regulated.(2) The activity of ethoxyresorufin-O-deethylase(EROD) and the content of reduced glutathione(GSH) in the liver were increased in the DFZ-treated groups, and glutathione S-transferase(GST) activity was increased in the 100 and 1000 ng/L groups.(3) DFZ was accumulated in the muscle and the biological concentration factors in the 10, 100, and1000 ng/L groups were respectively 149, 81 and 25. 6Glycyrrhetinic acid-modified nanoparticles for drug delivery: Preparation and characterization显示文摘In this work, glycyrrhetinic acid-modified chitosan (mGA-suc-CTS) used as liver targeted carrier for drug delivery, was prepared via hemisuccinate as a bridged group. The structure of the product was confirmed by IR and NMR methods and the degree of substitution (DS) of glycyrrhetinic acid groups was estimated via elemental analysis. Nanoparticles were formed by ionic gelation methold. The drug-loading and release behavior of the nanoparticles were investigated using BSA as the model drug. The results indicated that the carrier with a highest DS of 5.19% could be got and the DS was controlled by changing reaction temperature or feed ratio. BSA could be entrapped into the nanoparticles with the drug-loading ratio of 26.3% and the encapsulation efficiency of 81.5%. 8Exposure to difenoconazole inhibits reproductive ability in male marine medaka(Oryzias melastigma)显示文摘Difenoconazole(DFZ) is a triazole fungicide which has been detected in the aquatic environment, including estuaries and embayments. However, few studies addressing the reproductive toxicity and transgenerational effects of DFZ on marine fishes are available.The present study was conducted to investigate the effects of DFZ on male marine medaka(Oryzias melastigma). After exposure of the embryo to 1, 10, 100 and 1000 ng/L DFZ for180 days, the gonadosomatic index was significantly decreased in the 1000 ng/L treatment.The number of sperm was reduced while the abundances of spermatocytes and spermatogonia in the testes were increased in all the treatments. The m RNA levels of salmontype gnrh(sgnrh), the luteinizing hormone(lhβ) and the follicle-stimulating hormone(fshβ)genes in the brain all exhibited a significant down-regulation, the expression of androgen receptors(arα and arβ) was decreased and that of estrogen receptor β and cytochrome P450 aromatase(cyp19 B) was increased in the testes. The expression levels of cyp19 A and cyp19 B were increased in the liver. The decrease of ars m RNA levels might be one of the reasons causing the reduction of sperm. The down-regulation of sgnrh, lhβ and fshβ m RNA levels suggested that DFZ might impact the spermatogenesis via the brain–pituitary–gonad pathway. 11Clinical and radiological features of medullary infarction caused by spontaneous vertebral artery dissection显示文摘Background and purpose Medullary infarction(MI)caused by spontaneous vertebral artery dissection(sVAD)is an important type of stroke.It is important to distinguish sVAD from other causes of stroke since the treatment strategies and prognosis were different between them.In this study,we aimed to explore the clinical and radiological features of MI in patients with acute MI caused by sVAD.Methods Patients with acute MI caused by sVAD and non-sVAD in a single tertiary hospital were enrolled from 2010 to 2020.Epidemiologic,clinical and image features were collected and analysed.MI lesions were categorised into three levels rostrocaudally and four arterial groups:anteromedial,anterolateral,lateral and posterior.Results A total of 128 patients with MI were enrolled with 47 cases of sVAD and 81 cases of non-sVAD.Patients with sVAD were younger than those with non-sVAD(med 44 years old vs 58 years old).The sVAD group was less likely to have hypertension(44.68%vs 67.90%;p=0.010)and diabetes(19.15%vs 45.69%;p=0.003),but more likely to have non-sudden onset(27.66%vs 9.87%,p=0.009),minor neck injury(19.15%vs 1.23%;p=0.001)and headache(46.81%vs 7.41%;p=0.000).Vertically,sVAD became more common in caudal medulla than in rostral medulla.Horizontally,the sVAD group was more likely to have lateral MI(91.48%vs 2.96%,p=0.000).In multivariable logistic regression analysis,age,non-sudden onset and headache were independently associated with sVAD with ORs of 0.935(95%CI 0.892 to 0.981,p=0.006),3.507(95%CI 1.060 to 11.599,p=0.040)and 5.426(95%CI 1.673 to 17.599,p=0.005).Conclusion sVAD was not uncommon in patients with MI,especially in patients with lateral MI.Young patients with headache and non-sudden onset should remind clinician the possibility of sVAD.Chun Yu Zhu Zhu Siying Li Yi Xu Wei Yan Xiaocui Kang Yao Li Qiang Dong Weijun Tang Xiang Han 2022Stroke & Vascular Neurology2022,7,3:3
12Formation process and mechanism of humic acid-kaolin complex determined by carbamazepine sorption experiments and various characterization methods显示文摘To explore the formation process and mechanism of organic matter and organic-mineral complex under humification and mineralization conditions, a series of samples including humic acid, kaolin, and humic acid-kaolin complex were prepared using a subcritical water treatment method(SWT) under specific temperature, pressure and reaction time conditions. HA was used as a surrogate for natural organic matter because it has a similar abundant pore structure,variety of carbon types, and chemical components. These samples were used in carbamazepine(CBZ) sorption experiments and characterized by a variety of techniques. The polymerization of humic acid under the conditions of increased temperature and pressure resulted in an increase in specific surface area and molecular quantity. In addition, the degree of aromaticity rose from59.52% to 70.90%. These changes were consistent with the transformation from ‘soft carbon' to‘hard carbon' that occurs in nature. The results of sorption experiments confirmed the interaction between humic acid and kaolin from the difference between the predicted and actual Qevalues. The conceptual model of humic acid-kaolin complex could be deduced and described as follows. Firstly, the aromatic components of humic acid preferentially combine with kaolin through the intercalation effect, which protects them from the treatment effects.Next, the free carboxyl groups and small aliphatic components of humic acid interact on the surface of kaolin, and these soft species transform into dense carbon through cyclization and polymerization. 13High-density SNP genetic linkage map construction and quantitative trait locus mapping for resistance to cucumber mosaic virus in tobacco(Nicotiana tabacum L.)显示文摘Genetic linkage maps are essential for studies of genetics, genomic structure, and genomic evolution, and for mapping quantitative trait loci (QTL). Identification of molecular markers and construction of genetic linkage maps in tobacco (Nicotiana tabacum L.), a classical model plant and important economic crop, have remained limited. In the present study we identified a large number of single nucleotide polymorphism (SNP) markers and constructed a high-density SNP genetic map for tobacco using restriction site-associated DNA sequencing. In 1216.30 Gb of clean sequence obtained using the Illumina HiSeq 2000 sequencing platform, 99,647,735 SNPs were identified that differed between 203 sequenced plant genomes and the tobacco reference genome. Finally, 13,273 SNP markers were mapped on 24 high-density tobacco genetic linkage groups. The entire linkage map spanned 3421.80 cM, with a mean distance of 0.26 cM between adjacent markers. Compared with genetic linkage maps published previously, this version represents a considerable improvement in the number and density of markers. Seven QTL for resistance to cucumber mosaic virus (CMV) in tobacco were mapped to groups 5 and 8. 14Effect of dietary protein sources and storage temperatures on egg internal quality of stored shell eggs显示文摘This study was conducted to evaluate the effects of various protein sources(soybean meal, SBM; cottonseed protein, CSP; double-zero rapeseed meal, DRM) on the internal quality(Haugh unit, yolk index,albumen pH,yolk hardness and yolk springiness) of eggs when stored at either 4 or 28℃ for 28 d. A total of 288 laying hens(32 wk of age) were randomly allotted to 6 treatment groups(4 replicates per treatment) and fed diets containing SBM, CSP, or DRM individually or in combination with equal crude protein content(SBM-CSP, SBM-DRM, and CSP-DRM) as the protein ingredient(s). A 6 x 2 factorial arrangement was employed with dietary types and storage temperatures(4 and 28℃) as the main effects. After 12 wk of diet feeding, a total of 216 eggs was collected for egg internal quality determination.The results showed as follows: 1) lower egg quality was observed in the DRM group compared with the other groups when stored at 4 and 28℃ for 28 d(P < 0.05), while there was no difference in egg internal quality among the other groups. 2) The CSP diet resulted in higher yolk hardness compared with the other diets when eggs were stored at 4℃ for 28 d(P < 0.05). Lower Haugh unit was observed in the DRM and SBM-DRM groups compared with the other groups when eggs were stored for 28 d at 4℃(P < 0.05).3) Yolk breakage occurred in the DRM group and eggs could not be analyzed for egg internal quality when stored at 28℃ for 28 d. 15Evolutionary transition between invertebrates and vertebrates via methylation reprogramming in embryogenesis显示文摘Major evolutionary transitions are enigmas,and the most notable enigma is between invertebrates and vertebrates,with numerous spectacular innovations.To search for the molecular connections involved,we asked whether global epigenetic changes may offer a clue by surveying the inheritance and reprogramming of parental DNA methylation across metazoans.We focused on gametes and early embryos,where the methylomes are known to evolve divergently between fish and mammals.Here,we find that methylome reprogramming during embryogenesis occurs neither in pre-bilaterians such as cnidarians nor in protostomes such as insects,but clearly presents in deuterostomes such as echinoderms and invertebrate chordates,and then becomes more evident in vertebrates.Functional association analysis suggests that DNA methylation reprogramming is associated with development,reproduction and adaptive immunity for vertebrates,but not for invertebrates.Interestingly,the single HOX cluster of invertebrates maintains unmethylated status in all stages examined.In contrast,the multiple HOX clusters show dramatic dynamics of DNA methylation during vertebrate embryogenesis.Notably,the methylation dynamics of HOX clusters are associated with their spatiotemporal expression in mammals.Our study reveals that DNA methylation reprogramming has evolved dramatically during animal evolution,especially after the evolutionary transitions from invertebrates to vertebrates,and then to mammals.Xiaocui Xu Guoqiang Li Congru Li Jing Zhang Qiang Wang David KSimmons Xuepeng Chen Naveen Wijesena Wei Zhu Zhanyang Wang Zhenhua Wang Bao Ju Weimin Ci Xuemei Lu Daqi Yu Qian-fei Wang Neelakanteswar Aluru Paola Oliveri Yong EZhang Mark QMartindale Jiang Liu 2019National Science Review2019,6,5:2
16Coupling between the Grain for Green Program and a household level-based agricultural eco-economic system in Ansai,Shaanxi Province of China显示文摘The implementation of the Grain for Green Program(GGP)has changed the development track of the agricultural eco-economic system in China.In response to the results of a lag study that investigated the coupling between the GGP and the agricultural eco-economic system in a loess hilly region,we used a structural equation model to analyze the survey data from 494 households in Ansai,a district of Yan’an City in Shaanxi Province of China in 2015.The model clarified the direction and intensity of the coupling between the GGP and the agricultural eco-economic system.The coupling benefits were derived through linkages between the program and various chains in the agricultural eco-economic system.The GGP,the agroecosystem of Ansai and their potential coupling effects were in a state of general coordination.The agroecosystem directly affected the coupling effect,with the standardized path coefficient of 0.87,indicating that the agroecosystem in Ansai at this stage provided basic material support for the coupling between the GGP and the agricultural eco-economic system.The direct path coefficient of agroeconomic system impacted on the coupling effect was-0.76,indicating that partial contradictions occurred between the agroeconomic system and the coupling effect.Therefore,although the current agroecosystem in Ansai should be provided sufficient agroecological resources for the benign coupling between the program and the agricultural eco-economic system,agricultural development failed to effectively transform agroecological resources into agricultural economic advantages in this region,which resulted in a relative lag in the development of the agricultural economic system.Thus,the coupling between the GGP and the agricultural eco-economic system was poor.To improve the coupling and the sustainable development of the agricultural eco-economic system in cropland retirement areas,the industrial structure needs to be diversified,the agricultural resources(including agroecological resources,agricultural economic resources and agricultural social resources)need to be rationally allocated,and the chain structure of the agricultural eco-economic system needs to be continuously improved.LI Yue WANG Jijun HAN Xiaojia GUO Mancai CHENG Simin QIAO Mei ZHAO Xiaocui 2020Journal of Arid Land2020,12,2:2
17Crustal structure beneath Beijing and its surrounding regions derived from gravity data显示文摘In this paper we use gravity data to study fine crustal structure and seismogenic environment beneath Beijing and its surrounding regions.Multi-scale wavelet analysis method is applied to separating gravity fields.Logarithmic power spectrum method is also used to calculate depth of gravity field source.The results show that the crustal structure is very complicated beneath Beijing and its surrounding areas.The crustal density exhibits laterally inhomogeneous.There are three large scale tectonic zones in North China,i.e.,WNW-striking Zhangjiakou-Bohai tectonic zone(ZBTZ),NE-striking Taihang piedmont tectonic zone(TPTZ)and Cangxian tectonic zone(CTZ).ZBTZ and TPTZ intersect with each other beneath Beijing area and both of them cut through the lithosphere.The upper and middle crusts consist of many small-scale faults,uplifts and depressions. In the lower crust,these small-scale tectonic units disappear gradually,and they are replaced by large-scale tectonic units.In surrounding regions of Beijing,ZBTZ intersects with several other NE-striking tectonic units, such as Cangxian uplift,Jizhong depression and Shanxi Graben System(SGS).In west of Taihangshan uplift, gravity anomalies in upper and middle crusts are correlated with geological and topographic features on the surface.Compared with the crust,the structure is comparatively simple in uppermost mantle.Earthquakes mainly occurred in upper and middle crusts,especially in transitional regions between high gravity anomaly and low gravity anomaly.Occurrence of large earthquakes may be related to the upwelling of upper mantle and asthenosphere heat flow materials,such as Sanhe earthquake(M_S8.0)and Tangshan earthquake(M_S7.8).Wenliang Jiang Jingfa Zhang Xiaocui Lu Jing Lu 2011Earthquake Science2011,24,3:2
18Inhibition of p53 and/or AKT as a new therapeutic approach specifically targeting ALT cancers显示文摘While the majority of all human cancers court teract telomere shortening by expressing telomerase,-15%of all cancers maintain telomere length by a telomerase?independent mechanism known as alternative lengthening of telomeres(ALT).Here,we show that high load of intrinsic DNA damage is present in ALT cancer cells,leading to apoptosis stress by activating p53-independent,but JNK/c-IVIyc-dependent apoptotic pathway.Notably,ALT cells expressing wild-type p53 show much lower apoptosis than p53-deficient ALT cells.Mechanistically,we find that intrinsic DNA damage in ALT cells induces low level of p53 that is insufficient to initiate the transcription of apoptosis-related genes,but is sufficient to stimulate the expression of key components of mTORC2(mTOR and Rictor),which in turn leads to phosphorylation of AKT.Activated AKT(p-AKT)thereby stimulates downstream anti-apoptotic events.Therefore,p53 and AKT are the key factors that suppress sponta?neous apoptosis in ALT cells.Indeed,inhibition of p53 or AKT selectively induces rapid death of ALT cells in vitro,and p53 inhibitor severely suppresses the growth of ALT-cell xenograft tumors in mice.These findings reveal a previously unrecognized function of p53 in antiapoptosis and identify that the inhibition of p53 or AKT has a potential as therapeutics for specifically targeting ALT cancers.Yuanlong Ge Shu Wu Zepeng Zhang Xiaocui Li Feng Li Siyu Yan Haiying Liu Junjiu Huang Yong Zhao 2019Protein & Cell2019,10,11:2

20High-temperature wheat leaf rust resistance gene Lr13 exhibits pleiotropic effects on hybrid necrosis显示文摘Dear Editor,Wheat is one of the most important staple food crops.Leaf rust caused by Puccinia triticina Eriks.(Pt)is a destructive foliar disease that threatens world wheat production.Complementary wheat genes Necrosis 1(Ne1)and Necrosis 2(Ne2)on chromosome arms 5BL and 2BS,respectively,cause hybrid necrosis(Caldwell and Compton,1943;Chu et al.,2006).Ne2 shows extremely tight genetic linkage with leaf rust resistance gene Lr13(Zhang et al.,2016).We previously identified a temperature-sensitive leaf rust resistance gene LrZH22 in Chinese wheat cultivar Zhoumai 22(ZM,Figure 1A)that was effective against most Chinese Pt pathotypes(Wang et al.,2016)and shared an overlapping genetic interval with Lr13.In a separate study,a recessive early leaf senescence 1(els1)gene(Figure 1B)shared an overlapping interval with LrZH22/Lr13/Ne2(Chu et al.,2006;Wang et al.,2016;Zhang et al.,2016;Li et al.,2018).However,the function and relationship of LrZH22/Lr13/Ne2/els1 remained unknown for decades.Here,we report the map-based cloning of LrZH22 and els1 and characterize their relationship with Lr13 and Ne2 in conjunction with a companion paper(Hewitt et al.t 2021)studying the same gene.Xiaocui Yan Miaomiao Li Peipei Zhang Guihong Yin Huaizhi Zhang Takele Weldu Gebrewahid Jianping Zhang Lingli Dong Daqun Liu Zhiyong Liu Zaifeng Li 2021Molecular Plant2021,14,7:2
