1Inhibition of Mcl-1 enhances cell death induced by the Bcl-2-selective inhibitor ABT-199 in acute myeloid leukemia cells显示文摘Acute myeloid leukemia(AML)is a serious disease.The 5-year survival rates remain frustratingly low(65%for children and 26%for adults).Resistance to frontline chemotherapy(usually cytarabine)often develops;therefore a new treatment modality is needed.Bcl-2 family proteins play an important role in balancing cell survival and apoptosis.The antiapoptotic Bcl-2 family proteins have been found to be dysregulated in AML.ABT-199,a BH3 mimetic,was developed to target antiapoptotic protein Bcl-2.Although ABT-199 has demonstrated promising results,resistance occurs.Previous studies in AML show that ABT-199 alone decreases the association of proapoptotic protein Bim with Bcl-2,but this is compensated by increased association of Bim with prosurvival protein Mcl-1,stabilizing Mcl-1,resulting in resistance to ABT-199.In this study,we investigated the antileukemic activity of the Mcl-1-selective inhibitor A-1210477 in combination with ABT-199 in AML cells.We found that A-1210477 synergistically induced apoptosis with ABT-199 in AML cell lines and primary patient samples.The synergistic induction of apoptosis was decreased upon Bak,Bax and Bim knockdown.While A-1210477 treatment alone also increased Mcl-1 protein levels,combination with ABT-199 reduced binding of Bim to Mcl-1.Our results demonstrate that sequestration of Bim by Mcl-1,a mechanism of ABT-199 resistance,can be abrogated by combined treatment with the Mcl-1 inhibitor A-1201477.Daniel A Luedtke Xiaojia Niu Yihang Pan Jianyun Zhao Shuang Liu Holly Edwards Kang Chen Hai Lin Jeffrey W Taub Yubin Ge 2017Signal Transduction and Targeted Therapy2017,2,1:11
2Temperature-induced deformation of CRTS II slab track and its effect on track dynamical properties显示文摘Two finite-element models of the CRTS II slab track are presented to simulate temperature-induced deformations of the concrete track slab with no deterioration or with a deteriorated cement asphalt mortar(CAM).One model,which considers the fully bonding interface between the slab and the CAM layer,could applied to a track that is in good condition;the other model uses cohesive zone elements to simulate the deteriorated CAM with some possible interfacial separation and slip.Utilizing both of the models,temperature-induced warp deformations of track under various temperature loads are investigated.The influence of temperature deformation on the dynamic properties of the track is analyzed based on the train-track coupled dynamics.Numerical results show that the deteriorated CAM layer can significantly increase temperature deformations of a CRTS II track slab,which would produce tiny rail irregularities.There are clear differences between the deformation shapes of the track slabs that have an inseparable mortar layer and those have a separable mortar layer.The track slab with a deteriorated mortar layer showed more open curl distortion than the track slab in good condition.The dynamical response index of the slab track is intensified to a certain level due to the temperature deformation;with an increase of the train speed,the track dynamical responses increased linearly.However,rail irregularities due to the temperature deformations are very tiny.Even if a track is exposed to extreme temperature loads and the mortar layer is deteriorated,temperature deformation can have a negligible effect on the track’s dynamical properties.SONG XiaoLin ZHAO ChunFa ZHU XiaoJia 2014Science China(Technological Sciences)2014,57,10:8
3Ultrasensitive and stable X-ray detection using zero-dimensional lead-free perovskites显示文摘Sensitive and reliable X-ray detectors are essential for medical radiography,industrial inspection and security screening.Lowering the radiation dose allows reduced health risks and increased frequency and fidelity of diagnostic technologies for earlier detection of disease and its recurrence.Three-dimensional(3 D)organic-inorganic hybrid lead halide perovskites are promising for direct X-ray detection-they show improved sensitivity compared to conventional X-ray detectors.However,their high and unstable dark current,caused by ion migration and high dark carrier concentration in the 3 D hybrid perovskites,limits their performance and long-term operation stability.Here we report ultrasensitive,stable X-ray detectors made using zero-dimensional(0 D)methylammonium bismuth iodide perovskite(MA3Bi2I9)single crystals.The 0 D crystal structure leads to a high activation energy(Ea)for ion migration(0.46 e V)and is also accompanied by a low dark carrier concentration(~10^6 cm^-3).The X-ray detectors exhibit sensitivity of 10,620μC Gy-1 air cm-2,a limit of detection(Lo D)of 0.62 nG yairs-1,and stable operation even under high applied biases;no deterioration in detection performance was observed following sensing of an integrated X-ray irradiation dose of^23,800 m Gyair,equivalent to>200,000 times the dose required for a single commercial X-ray chest radiograph.Regulating the ion migration channels and decreasing the dark carrier concentration in perovskites provide routes for stable and ultrasensitive X-ray detectors.Xiaojia Zheng Wei Zhao Peng Wang Hairen Tan Makhsud I.Saidaminov Shujie Tie Ligao Chen Yufei Peng Jidong Long Wen-HuZhang 2020Journal of Energy Chemistry2020,29,10:7
4Effect of chitooligosaccharides on cyclin D1,bcl-xl and bcl-2 mRNA expression in A549 cells using quantitative PCR显示文摘Chitooligosaccharide (COS) is derived from the chemical and enzymatic hydrolysis of chitosan and has been reported to have potent antitumor activity.Our study investigated the effects of five chitooligomers ranging from the dimer form to the hexamer form (chitobiose,chitotriose,chitotetraose,chitopentaose,chitohexaose) on the expression of cyclin D1,bcl-2 and bcl-xl mRNA in A549 cells using reverse transcription quantitative real-time PCR.We demonstrated that,of the five chitooligomers used,chitohexaose (COS6) had the most potent inhibitory effect on A549 cell proliferation.COS6 also significantly down regulated cyclin D1 and bcl-xl mRNA expression levels.Our data suggested that COS6 exerts its antitumor activity by two different mechanisms:(1) COS6-mediated inhibition of Cyclin D1 levels leads to suppression of tumor cell proliferation;and (2) COS6-mediated down-regulation of the pro-survival protein,Bcl-xl,promotes the apoptosis of tumor cells.LI Xian WANG Ju CHEN XiaoJia TIAN JinHuan LI LiHua ZHAO MingYan JIAO YanPeng ZHOU ChangRen 2011Chinese Science Bulletin2011,56,15:5
5Overexpression CPT1A reduces lipid accumulation via PPARα/CD36 axis to suppress the cell proliferation in ccRCC显示文摘Clear cell renal carcinoma(ccRCC)is histologically defined by its cytoplasmic lipid deposits.Lipid metabolism disorder largely increases the risk of ccRCC.In this study,we aimed to investigate the biological functions and molecular mechanisms of carnitine palmitoyl transferase 1A(CPT1A)in ccRCC.Our results showed that CPT1A is decreased in ccRCC clinical samples and cell lines compared with that in normal samples.Lentivirus overexpressing CPT1A was used to investigate the neoplastic phenotypes of ccRCC,and the results showed that lipid accumulation and tumor growth are attenuated both in vitro and in vivo.In addition,CPT1A prevents cholesterol uptake and lipid accumulation by increasing the peroxisome proliferator-activated receptorα(PPARα)level through regulation of Class B scavenger receptor type 1(SRB1)and cluster of differentiation 36(CD36).Furthermore,PI3K/Akt signaling pathway promotes tumor cell proliferation in ccRCC,which is related to the enhanced expression of CD36.Functionally,weakened CPT1A expression is critical for lipid accumulation to promote ccRCC development.Collectively,our research unveiled a novel function of CPT1A in lipid metabolism via PPARα/CD36 axis,which provides a new theoretical explanation for the pathogenesis of ccRCC.Targeting CPT1A may be a potential therapeutic strategy to treat ccRCC.Hui Yang Hongbo Zhao Zhongkun Ren Xiaojia Yi Qiao Zhang Zhe Yang Yingmin Kuang Yuechun Zhu 2022Acta Biochimica et Biophysica Sinica2022,54,2:4
6Ultralight Magnetic and Dielectric Aerogels Achieved by Metal-Organic Framework Initiated Gelation of Graphene Oxide for Enhanced Microwave Absorption显示文摘The development of a convenient methodology for synthesizing the hierarchically porous aerogels comprising metal–organic frameworks(MOFs)and graphene oxide(GO)building blocks that exhibit an ultralow density and uniformly distributed MOFs on GO sheets is important for various applications.Herein,we report a facile route for synthesizing MOF/reduced GO(rGO)aerogels based on the gelation of GO,which is directly initiated using MOF crystals.Free metal ions exposed on the surface of MIL-88A nanorods act as linkers that bind GO nanosheets to a three-dimensional porous network via metal–oxygen covalent or electrostatic interactions.The MOF/rGOderived magnetic and dielectric aerogels Fe_(3)O_(4)@C/rGO and Ni-doped Fe_(3)O_(4)@C/rGO show notable microwave absorption(MA)performance,simultaneously achieving strong absorption and broad bandwidth at low thickness of 2.5(-58.1 dB and 6.48 GHz)and 2.8 mm(-46.2 dB and 7.92 GHz)with ultralow filling contents of 0.7 and 0.6 wt%,respectively.The microwave attenuation ability of the prepared aerogels is further confirmed via a radar cross-sectional simulation,which is attributed to the synergistic effects of their hierarchically porous structures and heterointerface engineering.This work provides an effective pathway for fabricating hierarchically porous MOF/rGO hybrid aerogels and offers magnetic and dielectric aerogels for ultralight MA.Xiaogu Huang Jiawen Wei Yunke Zhang BinBin Qian Qi Jia Jun Liu Xiaojia Zhao Gaofeng Shao 2022Nano-Micro Letters2022,14,7:4

8Hepatitis B virus evades immune recognition via RNA adenosine deaminase ADAR1-mediated viral RNA editing in hepatocytes显示文摘HBV is considered as a“stealth”virus that does not invoke interferon(IFN)responses;however,the mechanisms by which HBV bypasses innate immune recognition are poorly understood.In this study,we identified adenosine deaminases acting on RNA 1(ADAR1),which is a key factor in HBV evasion from IFN responses in hepatocytes.Mechanically,ADAR1 interacted with HBV RNAs and deaminated adenosine(A)to generate inosine(I),which disrupted host immune recognition and thus promoted HBV replication.Loss of ADAR1 or its deficient deaminase activity promoted IFN responses and inhibited HBV replication in hepatocytes,and blocking the IFN signaling pathways released the inhibition of HBV replication caused by ADAR1 deficiency.Notably,the HBV X protein(HBx)transcriptionally promoted ADAR1 expression to increase the threshold required to trigger intrinsic immune activation,which in turn enhanced HBV escape from immune recognition,leading to persistent infection.Supplementation with 8-azaadenosine,an ADAR1 inhibitor,efficiently enhanced liver immune activation to promote HBV clearance in vivo and in vitro.Taken together,our results delineate a molecular mechanism by which HBx promotes ADAR1-derived HBV immune escape and suggest a targeted therapeutic intervention for HBV infection.Liyuan Wang Yang Sun Xiaojia Song Zehua Wang Yankun Zhang Ying Zhao Xueqi Peng Xiaodong Zhang Chunyang Li Chengjiang Gao Nailin Li Lifen Gao Xiaohong Liang Zhuanchang Wu Chunhong Ma 2021Cellular & Molecular Immunology2021,18,8:2
9Coupling between the Grain for Green Program and a household level-based agricultural eco-economic system in Ansai,Shaanxi Province of China显示文摘The implementation of the Grain for Green Program(GGP)has changed the development track of the agricultural eco-economic system in China.In response to the results of a lag study that investigated the coupling between the GGP and the agricultural eco-economic system in a loess hilly region,we used a structural equation model to analyze the survey data from 494 households in Ansai,a district of Yan’an City in Shaanxi Province of China in 2015.The model clarified the direction and intensity of the coupling between the GGP and the agricultural eco-economic system.The coupling benefits were derived through linkages between the program and various chains in the agricultural eco-economic system.The GGP,the agroecosystem of Ansai and their potential coupling effects were in a state of general coordination.The agroecosystem directly affected the coupling effect,with the standardized path coefficient of 0.87,indicating that the agroecosystem in Ansai at this stage provided basic material support for the coupling between the GGP and the agricultural eco-economic system.The direct path coefficient of agroeconomic system impacted on the coupling effect was-0.76,indicating that partial contradictions occurred between the agroeconomic system and the coupling effect.Therefore,although the current agroecosystem in Ansai should be provided sufficient agroecological resources for the benign coupling between the program and the agricultural eco-economic system,agricultural development failed to effectively transform agroecological resources into agricultural economic advantages in this region,which resulted in a relative lag in the development of the agricultural economic system.Thus,the coupling between the GGP and the agricultural eco-economic system was poor.To improve the coupling and the sustainable development of the agricultural eco-economic system in cropland retirement areas,the industrial structure needs to be diversified,the agricultural resources(including agroecological resources,agricultural economic resources and agricultural social resources)need to be rationally allocated,and the chain structure of the agricultural eco-economic system needs to be continuously improved.LI Yue WANG Jijun HAN Xiaojia GUO Mancai CHENG Simin QIAO Mei ZHAO Xiaocui 2020Journal of Arid Land2020,12,2:2
10Development of a Novel Feedforward Neural Network Model Based on Controllable Parameters for Predicting Effluent Total Nitrogen显示文摘The problem of effluent total nitrogen(TN)at most of the wastewater treatment plants(WWTPs)in China is important for meeting the related water quality standards,even under the condition of high energy consumption.To achieve better prediction and control of effluent TN concentration,an efficient prediction model,based on controllable operation parameters,was constructed in a sequencing batch reactor process.Compared with previous models,this model has two main characteristics:①Superficial gas velocity and anoxic time are controllable operation parameters and are selected as the main input parameters instead of dissolved oxygen to improve the model controllability,and②the model prediction accuracy is improved on the basis of a feedforward neural network(FFNN)with algorithm optimization.The results demonstrated that the FFNN model was efficiently optimized by scaled conjugate gradient,and the performance was excellent compared with other models in terms of the correlation coefficient(R).The optimized FFNN model could provide an accurate prediction of effluent TN based on influent water parameters and key control parameters.This study revealed the possible application of the optimized FFNN model for the efficient removal of pollutants and lower energy consumption at most of the WWTPs.Zihao Zhao Zihao Wang Jialuo Yuan Jun Ma Zheling He Yilan Xu Xiaojia Shen Liang Zhu 2021Engineering2021,7,2:2

17Phase Regulation and Defect Passivation Enabled by Phosphoryl Chloride Molecules for Efficient Quasi‑2D Perovskite Light‑Emitting Diodes显示文摘Quasi-2D perovskites have attracted tremendous interest for application as lightemission layers in light-emitting diodes(LEDs).However,the heterogeneous n phase and non-uniform distribution still severely limit the further development of quasi-2D perovskite LEDs(Pero-LEDs).Meanwhile,the increased defect density caused by the reduced dimension and grain size induces non-radiative recombination and further deteriorates the device performance.Here,we found that a series of molecules containing phosphoryl chloride functional groups have noticeable enhancement effects on the device performance of quasi-2D Pero-LEDs.Then,we studied the modification mechanism by focusing on the bis(2-oxo-3-oxazolidinyl)phosphinic chloride(BOPCl).It is concluded that the BOPCl can not only regulate the phase distribution by decreasing the crystallization rate but also remain in the grain boundaries and passivate the defects.As a result,the corresponding quasi-2D Pero-LEDs obtained a maximum external quantum efficiency(EQE_(max))of 20.82%and an average EQE(EQE_(ave))of around 20%on the optimal 50 devices,proving excellent reproducibility.Our work provides a new selection of molecular types for regulating the crystallization and passivating the defects of quasi-2D perovskite films.Mingliang Li Yaping Zhao Jia Guo Xiangqian Qin Qin Zhang Chengbo Tian Peng Xu Yuqing Li Wanjia Tian Xiaojia Zheng Guichuan Xing Wen‑Hua Zhang Zhanhua Wei 2023Nano-Micro Letters2023,15,8:1

19Nanoparticle enhanced combination therapy for stem-like progenitors defined by single-cell transcriptomics in chemotherapy-resistant osteosarcoma显示文摘The adaptation of osteosarcoma cells to therapeutic pressure impedes the efficacy of chemotherapy for osteosarcoma.However,the characteristics and cellular organization of therapy-resistant cells in osteosarcoma tumors remain elusive.Here,we utilized single-cell transcriptomics to systematically map the cell-type-specific gene expression in a chemotherapy-resistant osteosarcoma tumor.Our data demonstrated the VEGFR2-JMJD3-abundant subsets as quiescent stem-like cells,thereby establishing the hierarchy of therapy-resistant actively cycling progenitor pools(JMJD3-abundant)in osteosarcoma.VEGFR2 inhibitor and JMJD3 inhibitor synergistically impeded osteosarcoma cell propagation and tumor growth.Although osteosarcoma cells are predisposed to apoptosis induced by the synergistic therapy through activation of the CHOP pro-apoptotic factor via the endoplasmic reticulum(ER)stress,the stem-like/progenitor cells exhibit an adaptive response,leading to their survival.Reduction in cellular glutathione levels in stem-like/progenitor cells caused by the treatment with a glutathione synthesis inhibitor increases ER stress-induced apoptosis.Importantly,the marked therapeutic improvement of synergistic therapy against stem-like/progenitor cells was achieved by using glutathione-scavenging nanoparticles,which can load and release the drug pair effectively.Overall,our study provides a framework for understanding glutathione signaling as one of the therapeutic vulnerabilities of stem-like/progenitor cells.Broadly,these findings revealed a promising arsenal by encapsulating glutathione-scavenging nanoparticles with co-targeting VEGFR2 and JMJD3 to eradicate chemotherapy-resistant osteosarcoma.Li Wang Xiaojia Huang Xinru You Tianqi Yi Bing Lu Jiali Liu Guohao Lu Minglin Ma Changye Zou Jun Wu Wei Zhao 2020Signal Transduction and Targeted Therapy2020,5,1:1
20Robust boron nanoplatform provokes potent tumoricidal activities via inhibiting heat shock protein显示文摘Near-infrared(NIR)-light-triggered photothermal therapy(PTT)is a promising treatment for breast cancer.However,its therapeutic efficiency is often compromised due to the heatinduced up-regulation of heat shock proteins,which confer photothermal resistance.To solve this urgent problem,PEGylated two-dimensional boron nanosheets(B-PEG)-which allow both multimodal imaging and photothermal conversion-were loaded with gambogic acid(GA),which can inhibit heat shock protein 90(Hsp90).Experimental findings indicated that this combination of B-PEG and GA could serve as an integrated drug delivery system for cancer diagnosis and treatment.It could be used to administer mild PTT as well as chemotherapy for breast cancer,provide improved anti-tumor effects,and reduce the toxicity of PTT,all while inhibiting breast cancer growth.This drug delivery system could offer a novel tool for administering chemotherapy combined with PTT while avoiding the adverse effects of traditional PTT.Yuying Zhao Ning Liu Piaoxue Liu Taojian Fan Rui Ma Huijie Hong Xiaojia Chen Zhongjian Xie Han Zhang Qi Wang Tongkai Chen 2022Asian Journal of Pharmaceutical Sciences2022,17,5:0
