| 1 | Rapid, automated, and reliable antimicrobial susceptibility test from positive blood culture by CAST‐R显示文摘Antimicrobial susceptibility tests(ASTs)are pivotal in combating multidrug resistant pathogens,yet they can be time‐consuming,labor‐intensive,and unstable.Using the AST of tigecycline for sepsis as the main model,here we establish an automated system of Clinical Antimicrobials Susceptibility Test Ramanometry(CAST‐R),based on D2O‐probed Raman microspectroscopy.Featuring a liquid robot for sample pretreatment and a machine learning‐based control scheme for data acquisition and quality control,the 3‐h,automated CAST‐R process accelerates AST by>10‐fold,processes 96 paralleled antibiotic‐exposure reactions,and produces high‐quality Raman spectra.The Expedited Minimal Inhibitory Concentration via Metabolic Activity is proposed as a quantitative and broadly applicable parameter for metabolism‐based AST,which shows 99%essential agreement and 93%categorical agreement with the broth microdilution method(BMD)when tested on 100 Acinetobacter baumannii isolates.Further tests on 26 clinically positive blood samples for eight antimicrobials,including tigecycline,meropenem,ceftazidime,ampicillin/sulbactam,oxacillin,clindamycin,vancomycin,and levofloxacin reveal 93%categorical agreement with BMD‐based results.The automation,speed,reliability,and general applicability of CAST‐R suggest its potential utility for guiding the clinical administration of antimicrobials. | Pengfei Zhu Lihui Ren Ying Zhu Jing Dai Huijie Liu Yuli Mao Yuandong Li Yuehui He Xiaoshan Zheng Rongze Chen Xiaoting Fu Lili Zhang Lijun Sun Yuanqi Zhu Yuetong Ji Bo Ma Yingchun Xu Jian Xu Qiwen Yang | 2022 | mLife2022,1,3: | 3 |
| 2 | New Algorithm Rules Out Acute-on-chronic Liver Failure Development within 28 Days from Acute Decompensation of Cirrhosis显示文摘Background and Aims:Approximately 10%of patients with acute decompensated(AD)cirrhosis develop acute-on-chronic liver failure(ACLF)within 28 days.Such cases have high mortality and are difficult to predict.Therefore,we aimed to establish and validate an algorithm to identify these patients on hospitalization.Methods:Hospitalized patients with AD who developed ACLF within 28 days were considered pre-ACLF.Organ dysfunction was defined accord-ing to the chronic liver failure-sequential organ failure as-sessment(CLIF-SOFA)criteria,and proven bacterial infec-tion was taken to indicate immune system dysfunction.A retrospective multicenter cohort and prospective one were used to derive and to validate the potential algorithm,re-spectively.A miss rate of<5%was acceptable for the calcu-lating algorithm to rule out pre-ACLF.Results:In the deri-vation cohort(n=673),46 patients developed ACLF within 28 days.Serum total bilirubin,creatinine,international normalized ratio,and present proven bacterial infection at admission were associated with the development of ACLF.AD patients with≥2 organ dysfunctions had a higher risk for pre-ACLF patients[odds ratio=16.58195%confidence interval:(4.271-64.363),p<0.001].In the derivation co-hort,67.5%of patients(454/673)had≤1 organ dysfunction and two patients(0.4%)were pre-ACLF,with a miss rate of 4.3%(missed/total,2/46).In the validation cohort,65.9%of patients(914/1388)had≤1 organ dysfunction,and four(0.3%)of them were pre-ACLF,with a miss rate of 3.4%(missed/total,4/117).Conclusions:AD patients with≤1 organ dysfunction had a significantly lower risk of developing ACLF within 28 days of admission and could be safely ruled out with a pre-ACLF miss rate of<5%. | Xiaoting Tang Hai Li Guohong Deng Xin Zheng Xianbo Wang Yan Huang Yanhang Gao Zhongji Meng Zhiping Qian Feng Liu Xiaobo Lu Yu Shi Beiling Li Wenyi Gu Xiaomei Xiang Yan Xiong Yixin Hou Jun Chen Na Gao Sen Luo Liujuan Ji Jing Li Rongjiong Zheng Haotang Ren Jinjun Chen | 2023 | Journal of Clinical and Translational Hepatology2023,11,3: | 1 |
| 3 | Phylotranscriptomics of Swertiinae(Gentianaceae)reveals that key floral traits are not phylogenetically correlated显示文摘Establishing how lineages with similar traits are phylogenetically related remains critical for understanding the origin of biodiversity on Earth.Floral traits in plants are widely used to explore phylogenetic relationships and to delineate taxonomic groups.The subtribe Swertiinae(Gentianaceae)comprises more than 350 species with high floral diversity ranging from rotate to tubular corollas and possessing diverse nectaries.Here we performed phylogenetic analysis of 60 species from all 15 genera of the subtribe Swertiinae sensu Ho and Liu,representing the range of floral diversity,using data from the nuclear and plastid genomes.Extensive topological conflicts were present between the nuclear and plastome trees.Three of the 15 genera represented by multiple species are polyphyletic in both trees.Key floral traits including corolla type,absence or presence of lobe scales,nectary type,nectary position,and stigma type are randomly distributed in the nuclear and plastome trees without phylogenetic correlation.We also revealed the likely ancient hybrid origin of one large clade comprising 10 genera with diverse floral traits.These results highlight the complex evolutionary history of this subtribe.The phylogenies constructed here provide a basic framework for further exploring the ecological and genetic mechanisms underlying both species diversification and floral diversity. | Chunlin Chen Brad R.Ruhfel Jialiang Li Zefu Wang Lushui Zhang Lei Zhang Xingxing Mao Ji Wang Dashan He Yue Luo Quanjun Hu Yuanwen Duan Xiaoting Xu Zhenxiang Xi Jianquan Liu | 2023 | Journal of Integrative Plant Biology2023,65,6: | 1 |
| 4 | Atmospheric oxidation capacity and O_(3) formation in a coastal city of southeast China:Results from simulation based on four-season observation显示文摘The pollution of atmospheric ozone in China shows an obvious upward trend in the past decade.However,the studies on the atmospheric oxidation capacity and O_(3)formation in four seasons in the southeastern coastal region of China with the rapid urbanization remain limited.Here,a four-season field observation was carried out in a coastal city of southeast China,using an observation-based model combining with the Master Chemical Mechanism,to explore the atmospheric oxidation capacity(AOC),radical chemistry,O_(3)formation pathways and sensitivity.The results showed that the average net O_(3)production rate(14.55 ppbv/hr)in summer was the strongest,but the average O_(3)concentrations in autumn was higher.The AOC and ROx levels presented an obvious seasonal pattern with the maximum value in summer,while the OH reactivity in winter was the highest with an average value of 22.75 sec^(-1).The OH reactivity was dominated by oxygenated VOCs(OVOCs)(30.6%-42.8%),CO(23.2%-26.8%),NO_(2)(13.6%-22.0%),and alkenes(8.4%-12.5%)in different seasons.HONO photolysis dominated OH primary source on daytime in winter,while in other seasons,HONO photolysis in the morning and ozone photolysis in the afternoon contributed mostly.Sensitivity analysis indicated that O_(3)production was controlled by VOCs in spring,autumn and winter,but a VOC-limited and NOx-limited regime in summer,and alkene and aromatic species were the major controlling factors to O_(3)formation.Overall,the study characterized the atmospheric oxidation capacity and elucidated the controlling factors for O_(3)production in the coastal area with the rapid urbanization in China. | Gaojie Chen Taotao Liu Jinsheng Chen Lingling Xu Baoye Hu Chen Yang Xiaolong Fan Mengren Li Youwei Hong Xiaoting Ji Jinfang Chen Fuwang Zhang | 2024 | Journal of Environmental Sciences2024,,2: | 0 |

| 7 | Correlation between imaging features on computed tomography and combined positive score of PD-L1 expression in patients with gastric cancer显示文摘Objective:To explore the correlation between computed tomography(CT)features and combined positive score(CPS)of programmed cell death ligand 1(PD-L1)expression in patients with gastric cancer(GC).Methods:This study reviewed an institutional database of patients who underwent GC operation without neoadjuvant chemotherapy between December 2019 and September 2020.The CPS results of PD-L1 expression of postoperative histological examination were recorded by pathology.Baseline CT features were measured,and their correlation with CPS 5 or 10 score groups of PD-L1 expression was analyzed.Results:Data for 153 patients with GC were collected.Among them,124 were advanced GC patients,and 29were early GC patients.None of the CT features significantly differed between CPS groups with a cutoff score of 5and a score of 10 in patients with early GC.In advanced GC,the presence of lymph nodes with short diameters>10mm was significantly different(P=0.024)between the CPS<5 and CPS≥5 groups.CT features such as tumor attenuation in the arterial phase,long and short diameter of the largest lymph node,the sum of long diameter of the two largest lymph nodes,the sum of short diameter of the two largest lymph nodes,and the presence of lymph nodes with short diameters>10 mm significantly differed between the CPS<10 and CPS≥10 groups in advanced GC.The sensitivity,specificity and area under receiver operating characteristic(ROC)curve of logistic regression model for predicting CPS≥10 was 71.7%,50.0%and 0.671,respectively.Microsatellite instability(MSI)status was significantly different in CPS groups with cutoff score of 5 and 10 in advanced GC patients.Conclusions:CT findings of advanced GC patients with CPS≥10 showed greater arterial phase enhancement and larger lymph nodes.CT has the potential to help screen patients suitable for immunotherapy. | Zhilong Wang Yinkui Wang Xiaoting Li Yanling Li Zhaode Bu Zhongwu Li Yingshi Sun Jiafu Ji | 2022 | Chinese Journal of Cancer Research2022,34,5: | 0 |
