| 2 | Gastrin attenuates sepsis-induced myocardial dysfunction by down-regulation of TLR4 expression in macrophages显示文摘Myocardial dysfunction is the most serious complication of sepsis.Sepsis-induced myocardial dysfunction(SMD)is often associated with gastrointestinal dysfunction,but its pathophysiological significance remains unclear.The present study found that patients with SMD had higher plasma gastrin concentrations than those without SMD.In mice,knockdown of the gastrin receptor,cholecystokinin B receptor(Cckbr),aggravated lipopolysaccharide(LPS)-induced cardiac dysfunction and increased inflammation in the heart,whereas the intravenous administration of gastrin ameliorated SMD and cardiac injury.Macrophage infiltration plays a significant role in SMD because depletion of macrophages by the intravenous injection of clodronate liposomes,48 h prior to LPS administration,alleviated LPSinduced cardiac injury in Cckbr-deficient mice.The intravenous injection of bone marrow macrophages(BMMs)overexpressing Cckbr reduced LPS-induced myocardial dysfunction.Furthermore,gastrin treatment inhibited toll-like receptor 4(TLR4)expression through the peroxisome proliferator-activated receptor a(PPAR-a)signaling pathway in BMMs.Thus,our findings provide insights into the mechanism of the protective role of gastrin/CCKBR in SMD,which could be used to develop new treatment modalities for SMD. | Dandong Fang Yu Li Bo He Daqian Gu Mingming Zhang Jingwen Guo Hongmei Ren Xinyue Li Ziyue Zhang Ming Tang Xingbing Li Donghai Yang Chunmei Xu Yijie Hu Hongyong Wang Pedro AJose Yu Han Chunyu Zeng | 2023 | Acta Pharmaceutica Sinica B2023,13,9: | 1 |

| 4 | Fixation of hybrid sterility genes and favorable alleles of key yield-related genes with dominance contribute to the high yield of the Yongyou series of intersubspecific hybrid rice显示文摘In rice,the Yongyou series of Xian-Geng intersubspecific hybrids have excellent production performance,as shown by their extremely high yield.However,the mechanisms underlying the success of these rice hybrids are unclear.In this study,three F2 populations are generated from three Yongyou hybrids to determine the genetic basis of the extremely high yield of intersubspecific hybrids.Genome constitution analysis reveals that the female and male parental lines belong to the Geng and Xian subspecies,respectively,although introgression of 20%of the Xian ancestry and 14%of the Geng ancestry are observed.Twenty-five percent of the hybrid genomes carries homozygous Xian or Geng fragments,which harbors hybrid sterility genes such as Sd,Sc,f5,and q S12 and favorable alleles of key yield-related genes,including NAL1,Ghd7,and Ghd8.None of the parents carries the S5+killer of the S5 killer-protector system.Compatible allele combinations of hybrid sterility genes ensure the fertility of these intersubspecific hybrids and overcome the bottleneck in applying intersubspecific hybrids.Additive effects of favorable alleles of yield-related genes fixed in both parents enhances midparent values.Many QTLs for yield and its key component spikelets per panicle shows dominance and the net positive dominant effects lead to heterosis.These factors result in an extremely high yield of the hybrids.These findings will aid in the development of new intersubspecific rice hybrids with diverse genetic backgrounds. | Pengfei Wang Feixiang Qi Honglin Yao Xingbing Xu Wenjun Li Jianghu Meng Qinglu Zhang Weibo Xie Yongzhong Xing | 2022 | Journal of Genetics and Genomics2022,49,5: | 1 |
| 5 | Acute myeloid leukemia cells inhibit the differentiation and maturation of dendritic cells and induce the generation of regulatory T cells显示文摘Objective:To investigate the effects of soluble factors secreted by acute myeloid leukemia(AML) cells on the phenotypical and functional properties of DCs derived from normal mononuclear cells. Methods:Mononuclear cells were cul-tured with interleukin-4(IL-4) and granulocyte-macrophage colony-stimulating factor(GM-CSF),in the presence or absence of 24 h culture supernatants from fresh primary AML cells,to generate immature DCs. The maturation of DCs was induced by cytokines IL-1beta,IL-6,tumor necrosis factor-alpha(TNF-alpha),and prostaglandin-2(PGE-2). The phenotypic alterations of DCs and DCs-primed CD4+ T cells were evaluated using flow cytometry. Precursor frequency(PF) was calculated to monitor the allostimulatory effects of DCs on CD4+ and CD8+ T cells. Results:AML cell supernatant-treated DCs showed significantly lower expression of co-stimulatory molecules CD80 and CD86,and reduced response to cytokines IL-1beta,IL-6,TNF-alpha,and PGE-2. The allostimulatory effects of AML cell supernatant-treated DCs on CD4+ and CD8+ T cells were significantly lower than those of normal mature DCs [PF:(1.8 ± 0.5)% vs.(5.2 ± 1.6)% for CD4+ T cells,(2.1 ± 0.6)% vs.(6.5 ± 2.0)% for CD8+ T cells,P < 0.01]. These AML supernatant-induced DCs could also induce allogeneic CD4+ T cells to differentiate into CD4+CD25high T cells,which had immunophenotyping characteristics of regulatory T cells,i.e. they expressed Foxp3 but not active maker CD69. Conclusion:This study demonstrates that soluble factors secreted by AML cells can inhibit development and functions of DCs.
