| 1 | Regulation of sister chromatid cohesion by nuclear PD-L1显示文摘Programmed death ligand-1(PD-L1 or B7-H1)is well known for its role in immune checkpoint regulation,but its function inside the tumor cells has rarely been explored.Here we report that nuclear PD-L1 is important for cancer cell sister chromatid cohesion.We found that depletion of PD-L1 suppresses cancer cell proliferation,colony formation in vitro,and tumor growth in vivo in immune-deficient NSG mice independent of its role in immune checkpoint.Specifically,PD-L1 functions as a subunit of the cohesin complex,and its deficiency leads to formation of multinucleated cells and causes a defect in sister chromatid cohesion.Mechanistically,PD-L1 compensates for the loss of Sororin,whose expression is suppressed in cancer cells overexpressing PD-L1.PD-L1 competes with Wing Apart-Like(WAPL)for binding to PDS5B,and secures proper sister chromatid cohesion and segregation.Our findings suggest an important role for nuclear PD-L1 in cancer cells independent of its function in immune checkpoint. | Jia Yu Bo Qin Ann MMoyer Somaira Nowsheen Xinyi Tu Haidong Dong Judy CBoughey Matthew PGoetz Richard Weinshilboum Zhenkun Lou Liewei Wang | 2020 | Cell Research2020,30,7: | 3 |
| 2 | The transcription factor KLF14 regulates macrophage glycolysis and immune function by inhibiting HK2 in sepsis显示文摘Sepsis is a heterogeneous syndrome induced by a dysregulated host response to infection.Glycolysis plays a role in maintaining the immune function of macrophages,which is crucial for severely septic patients.However,how the pathways that link glycolysis and macrophages are regulated is still largely unknown.Here,we provide evidence to support the function of KLF14,a novel Krüppel-like transcription factor,in the regulation of glycolysis and the immune function of macrophages during sepsis.KLF14 deletion led to significantly increased mortality in lethal models of murine endotoxemia and sepsis.Mechanistically,KLF14 decreased glycolysis and the secretion of inflammatory cytokines by macrophages by inhibiting the transcription of HK2.In addition,we confirmed that the expression of KLF14 was upregulated in septic patients.Furthermore,pharmacological activation of KLF14 conferred protection against sepsis in mice.These findings uncover a key role of KLF14 in modulating the inflammatory signaling pathway and shed light on the development of KLF14-targeted therapeutics for sepsis. | Yuan Yuan Guangjian Fan Yuqi Liu Lu Liu Tong Zhang Pengfei Liu Qing Tu Xinyi Zhang Shiyuan Luo Liangfang Yao Feng Chen Jingbao Li | 2022 | Cellular & Molecular Immunology2022,19,4: | 1 |
| 3 | Transcriptomic analysis of interstockinduced dwarfism in Sweet Persimmon(Diospyros kaki Thunb.)显示文摘Growth monitoring indicated that the height of‘Kanshu’plants with‘Nantong-xiaofangshi’as an interstock was significantly shorter than that of‘Kanshu’plants with no interstock.A transcriptome analysis of the two graft combinations(‘Kanshu’/Diospyros lotus and‘Kanshu’/‘Nantong-xiaofangshi’/Diospyros lotus)was conducted to explore the dwarfing genes related to the use of the‘Nantong-xiaofangshi’interstock.Hormone levels and water conductance were also measured in these two graft combinations.The results indicated that the levels of both IAA and GA were lower in‘Kanshu’that had been grafted onto the‘Nantong-xiaofangshi’interstock than in‘Kanshu’with no interstock;additionally,the water conductance was lower in grafts with interstocks than in grafts without interstocks.The expression of AUX/IAA and auxin-responsive GH3 genes was enhanced in scions grafted on the interstock and was negatively correlated with the IAA content and growth of scions.The expression of GA2ox,DELLA,and SPINDLY genes were also upregulated and associated with a decrease in the level of GA in scions grafted on the interstock.Since one of the GA2ox unigenes was annotated as DkGA2ox1 in Diospyros kaki,but was not functionally validated,a functional analysis was conducted in transgenic tobacco.Overexpression of DkGA2ox1 in transgenic plants resulted in a dwarf phenotype that could be recovered by the exogenous application of GA3.We conclude that the‘Nantong-xiaofangshi’interstock affects the water conductance and expression of genes related to the metabolism and transduction of IAA and GA in the grafted scion and thus regulates phytohormone levels,producing dwarfing. | Yanying Shen Weibing Zhuang Xutong Tu Zhihong Gao Aisheng Xiong Xinyi Yu Xuehan Li Feihong Li Shenchun Qu | 2019 | Horticulture Research2019,6,1: | 1 |
| 4 | On-tissue chemical derivatization enables spatiotemporal heterogeneity visualization of oxylipins in esophageal cancer xenograft via ambient mass spectrometry imaging显示文摘On-tissue chemical derivatization(OTCD)effectively enhances ionization efficiency of low abundant and poorly ionized functional molecules to improve detection sensitivity and coverage of mass spectrometry imaging(MSI).Combination OTCD and MSI provides a novel strategy for visualizing previously undisclosed metabolic heterogeneity in tumor.Herein,we present a method to visualize heterogeneous metabolism of oxylipins within tumor by coupling OTCD with airflow-assisted desorption electrospray ionization(AFADESI)-MSI.Taking Girard’s P as a derivatization reagent,easily ionized hydrazide and quaternary amine groups were introduced into the structure of carbonyl metabolites via condensation reaction.Oxylipins,including 127 fatty aldehydes(FALs)and 71 oxo fatty acids(FAs),were detected and imaged in esophageal cancer xenograft with AFADESI-MSI after OTCD.Then t-distributed stochastic neighbor embedding and random forest were exploited to precisely locate the distribution of oxylipins in heterogeneous tumor tissue.With this method,we surprisingly found almost all FALs and oxo FAs significantly accumulated in the core region of tumor,and exhibited a gradual increase trend in tumor over time.These results reveal spatiotemporal heterogeneity of oxylipins in tumor progression,highlighting the value of OTCD combined with MSI to gain deeper insights into understanding tumor metabolism. | Jialin Liu Qingce Zang Xinzhu Li Xinyi Tu Ying Zhu Lingzhi Wang Zitong Zhao Yongmei Song Ruiping Zhang Zeper Abliz | 2023 | Chinese Chemical Letters2023,34,11: | 0 |

| 6 | Reduced Firing of Nucleus Accumbens Parvalbumin Interneurons Impairs Risk Avoidance in DISC1 Transgenic Mice显示文摘A strong animal survival instinct is to approach objects and situations that are of benefit and to avoid risk.In humans,a large proportion of mental disorders are accompanied by impairments in risk avoidance.One of the most important genes involved in mental disorders is disrupted-in-schizophrenia-1(DISC1),and animal models in which this gene has some level of dysfunction show emotion-related impairments.However,it is not known whether DISC1 mouse models have an impairment in avoiding potential risks.In the present study,we used DISC1-N terminal truncation(DISC1-N^(TM))mice to investigate risk avoidance and found that these mice were impaired in risk avoidance on the elevated plus maze(EPM)and showed reduced social preference in a three-chamber social interaction test.Following EPM tests,c-Fos expression levels indicated that the nucleus accumbens(NAc)was associated with risk-avoidance behavior in DISC1-N^(TM)mice.In addition,in vivo electrophysiological recordings following tamoxifen administration showed that the firing rates of fast-spiking neurons(FS)in the NAc were significantly lower in DISC1-N^(TM)mice than in wild-type(WT)mice.In addition,in vitro patch clamp recording revealed that the frequency of action potentials stimulated by current injection was lower in parvalbumin(PV)neurons in the NAc of DISC1-N^(TM)mice than in WT controls.The impairment of risk avoidance in DISC1-N^(TM)mice was rescued using optogenetic tools that activated NAcPV neurons.Finally,inhibition of the activity of NAcPV neurons in PV-Cre mice mimicked the risk-avoidance impairment found in DISC1-N^(TM)mice during tests on the elevated zero maze.Taken together,our findings confirm an impairment in risk avoidance in DISC1-N^(TM)mice and suggest that reduced excitability of NAc^(PV) neurons is responsible. | Xinyi Zhou Bifeng Wu Wenhao Liu Qian Xiao Wei He Ying Zhou Pengfei Wei Xu Zhang Yue Liu Jie Wang Jufang He Zhigang Zhang Weidong Li Liping Wang Jie Tu | 2021 | Neuroscience Bulletin2021,37,9: | 0 |

| 8 | Survey on performance optimization for database systems显示文摘The performance optimization of database systems has been widely studied for years. From the perspective of the operation and maintenance personnel, it mainly includes three topics: prediction, diagnosis,and tuning. The prediction of future performance can guide the adjustment of configurations and resources.The diagnosis of anomalies can determine the root cause of performance regression. Tuning operations improve performance by adjusting influencing factors, e.g., knobs, indexes, views, resources, and structured query language(SQL) design. In this review, we focus on the performance optimization of database systems and review notable research work on the topics of prediction, diagnosis, and tuning. For prediction, we summarize the techniques, strengths, and limitations of several proposed systems for single and concurrent queries. For diagnosis, we categorize the techniques by the input data, i.e., monitoring metrics, logs, or time metrics, and analyze their abilities. For tuning, we focus on the approaches commonly adopted by the operation and maintenance personnel, i.e., knob tuning, index selection, view materialization, elastic resource, storage management, and SQL antipattern detection. | 9 | Isolation,identification,and virulence gene analysis of pathogenic Aeromonas dhakensis in Macrobrachium rosenbergii and histopathological observation显示文摘To identify the cause of mass mortality of adult Macrobrachium rosenbergii in a farm in Gaoyou City,Jiangsu Province,China,a dominant strain named DKQ-1 was isolated from the hepatopancreas of dying M.rosenbergii and identified as Aeromonas dhakensis by purification culture,biochemical characterization,and 16S rRNA and gyrB gene sequence analysis.The results of the challenge test revealed that the strain was highly pathogenic and the 50%lethal dose(LD_(50))in 72 h to M.rosenbergii was 1.54×10^(5)CFU/mL.The amplification results of virulence genes show that strain DKQ-1 carried 9 virulence genes,including ascV,aexT,aer,act,lip,ompAI,gcaT,acg,and exu,supporting the strong virulence of strain DKQ-1 to M.rosenbergii.Histopathological observation of the hepatopancreas,gills,and intestines indicated that DKQ-1 injection into M.rosenbergii could cause serious tissue damage,which further supported the strong virulence of this strain.In addition,a drug susceptibility test revealed that strain DKQ-1 was sensitive to 16 kinds of antibiotics,resistant to 9 kinds of antibiotics,and had intermediate resistance to spectinomycin and kanamycin.This study is the first report of A.dhakensis isolated from M.rosenbergii and provided a reference for the pathogen identification of bacterial diseases in M.rosenbergii,and for the prevention and treatment caused by A.dhakensis. | Xin PENG Haihui TU Xinyi YAO Xuan LAN Zhenxiao ZHONG Jinping LUO Qiongying TANG Shaokui YI Zhenglong XIA Guoliang YANG | 2024 | Journal of Oceanology and Limnology2024,42,2: | 0 |
