| 1 | Soil Microbial Community Structure in Diverse Land Use Systems:A Comparative Study Using Biolog,DGGE,and PLFA Analyses显示文摘Biolog,16S rRNA gene denaturing gradient gel electrophoresis(DGGE) ,and phospholipid fatty acid(PLFA) analyses were used to assess soil microbial community characteristics in a chronosequence of tea garden systems(8-,50-,and 90-year-old tea gardens) ,an adjacent wasteland,and a 90-year-old forest. Biolog analysis showed that the average well color development(AWCD) of all carbon sources and the functional diversity based on the Shannon index decreased(P < 0.05) in the following order:wasteland > forest > tea garden. For the DGGE analysis,the genetic diversity based on the Shannon index was significantly lower in the tea garden soils than in the wasteland. However,compared to the 90-year-old forest,the tea garden soils showed significantly higher genetic diversity. PLFA analysis showed that the ratio of Gram positive bacteria to Gram negative bacteria was significantly higher in the tea garden soils than in the wasteland,and the highest value was found in the 90-year-old forest. Both the fungal PLFA and the ratio of fungi to bacteria were significantly higher in the three tea garden soils than in the wasteland and forest,indicating that fungal PLFA was significantly affected by land-use change. | 2 | Deferoxamine promotes recovery of traumatic spinal cord injury by inhibiting ferroptosis显示文摘Ferroptosis is an iron-dependent novel cell death pathway. Deferoxamine, a ferroptosis inhibitor, has been reported to promote spinal cord injury repair. It has yet to be clarified whether ferroptosis inhibition represents the mechanism of action of Deferoxamine on spinal cord injury recovery. A rat model of Deferoxamine at thoracic 10 segment was established using a modified Allen's method. Ninety 8-week-old female Wistar rats were used. Rats in the Deferoxamine group were intraperitoneally injected with 100 mg/kg Deferoxamine 30 minutes before injury. Simultaneously, the Sham and Deferoxamine groups served as controls. Drug administration was conducted for 7 consecutive days. The results were as follows:(1) Electron microscopy revealed shrunken mitochondria in the spinal cord injury group.(2) The Basso, Beattie and Bresnahan locomotor rating score showed that recovery of the hindlimb was remarkably better in the Deferoxamine group than in the spinal cord injury group.(3) The iron concentration was lower in the Deferoxamine group than in the spinal cord injury group after injury.(4) Western blot assay revealed that, compared with the spinal cord injury group, GPX4, xCT, and glutathione expression was markedly increased in the Deferoxamine group.(5) Real-time polymerase chain reaction revealed that, compared with the Deferoxamine group, mRNA levels of ferroptosis-related genes Acyl-CoA synthetase family member 2(ACSF2) and iron-responsive element-binding protein 2(IREB2) were up-regulated in the Deferoxamine group.(6) Deferoxamine increased survival of neurons and inhibited gliosis. These findings confirm that Deferoxamine can repair spinal cord injury by inhibiting ferroptosis. | 6 | Effect of lime application on microbial community in acidic tea orchard soils in comparison with those in wasteland and forest soils显示文摘Lime application is a conventional technology to control acidification in tea orchard soils. We investigated the effect of lime application on soil microbial community diversity in the soils of three tea orchards, wasteland and forest. The BIOLOG data showed that both the average well color development of all carbon sources and the functional diversity index increased with the liming rate in the tea orchards and the forest, but decreased in the wasteland. The phospholipid fatty acid (PLFA) analysis showed that the structural diversity index of soil microbial community increased with the liming rate in all the tea orchards, the wasteland and the forest. Lime application also increased the soil-bacterial PLFA content in all the soils. Soil fungal and actinomycete PLFAs in the tea orchards showed an increasing trend from 0 to 3.2 g CaCO 3 /kg application and then a decreasing trend from 3.2 to 6.4 g CaCO 3 /kg application. | 9 | Prognosis-related classifi cation and dynamic monitoring of immune status in patients with sepsis:A prospective observational study显示文摘BACKGROUND:The dynamic monitoring of immune status is crucial to the precise and individualized treatment of sepsis.In this study,we aim to introduce a model to describe and monitor the immune status of sepsis and to explore its prognostic value.METHODS:A prospective observational study was carried out in Zhongshan Hospital,Fudan University,enrolling septic patients admitted between July 2016 and December 2018.Blood samples were collected at days 1 and 3.Serum cytokine levels(e.g.,tumor necrosis factor-α[TNF-α],interleukin-10[IL-10])and CD14+monocyte human leukocyte antigen-D-related(HLA-DR)expression were measured to serve as immune markers.Classifi cation of each immune status,namely systemic inflammatory response syndrome(SIRS),compensatory anti-inflammatory response syndrome(CARS),and mixed antagonistic response syndrome(MARS),was defined based on levels of immune markers.Changes of immune status were classifi ed into four groups which were stabilization(SB),deterioration(DT),remission(RM),and non-remission(NR).RESULTS:A total of 174 septic patients were enrolled including 50 non-survivors.Multivariate analysis discovered that IL-10 and HLA-DR expression levels at day 3 were independent prognostic factors.Patients with MARS had the highest mortality rate.Immune status of 46.1%patients changed from day 1 to day 3.Among four groups of immune status changes,DT had the highest mortality rate,followed by NR,RM,and SB with mortality rates of 64.7%,42.9%,and 11.2%,respectively.CONCLUSIONS:Severe immune disorder defi ned as MARS or deterioration of immune status defi ned as DT lead to the worst outcomes.The preliminary model of the classifi cation and dynamic monitoring of immune status based on immune markers has prognostic values and is worthy of further investigation. | Jun Yin Yao Chen Jun-ling Huang Lei Yan Zhong-shu Kuang Ming-ming Xue Si Sun Hao Xiang Yan-yan Hu Zhi-min Dong Chao-yang Tong Chun-xue Bai Zhen-ju Song | 2021 | World Journal of Emergency Medicine2021,12,3: | 16 |

| 17 | Comparative analysis of the complete chloroplast genome among Prunus mume,P.armeniaca,and P.salicina显示文摘Prunus mume Sieb.et Zucc.,P.armeniaca L.,and P.salicina L.are economically important fruit trees in temperate regions.These species are taxonomically perplexing because of shared interspecific morphological traits and variation,which are mainly attributed to hybridization.The chloroplast is cytoplasmically inherited and often used for evolutionary studies.We sequenced the complete chloroplast genomes of P.mume,P.armeniaca,and P.salicina using Illumina sequencing followed by de novo assembly.The three chloroplast genomes exhibit a typical quadripartite structure with conserved genome arrangement,structure,and moderate divergence.The lengths of the genomes are 157,815,157,797,and 157,916 bp,respectively.The length of the large single-copy region(LSC)region is 86,113,86,283,and 86,122 bp,and the length of the SSC region is 18,916,18,734,and 19,028 bp;the IR region is 26,393,26,390,and 26,383 bp,respectively.Each of the three chloroplast genomes encodes 133 genes,including 94 protein-coding,31 tRNA,and eight rRNA genes.Differential gene analysis for the three species revealed that trnY-ATA is a unique gene in P.armeniaca;in contrast,the gene trnI-TAT is only present in P.mume and P.salicina,though the position of the gene in these chloroplast genomes differs.Further comparative analysis of the complete chloroplast genome sequences revealed that the ORF genes and the sequences of linked regions rps16 and atpA,atpH and atpI,trnc-GCA and psbD,ycf3 and atpB,and rpL32 and ndhD are significantly different and may be used as molecular markers in taxonomic studies.Phylogenetic evolution analysis of the three species suggests that P.mume has a closer genetic relationship to P.armeniaca than to P.salicina. | Song Xue Ting Shi Wenjie Luo Xiaopeng Ni Shahid Iqbal Zhaojun Ni Xiao Huang Dan Yao Zhijun Shen Zhihong Gao | 2019 | Horticulture Research2019,6,1: | 10 |

| 19 | Investigation into the Formation, Structure, and Evolution of an EF4 Tornado in East China Using a High-Resolution Numerical Simulation显示文摘Devastating tornadoes in China have received growing attention in recent years, but little is known about their formation, structure, and evolution on the tornadic scale. Most of these tornadoes develop within the East Asian monsoon regime, in an environment quite different from tornadoes in the U.S. In this study, we used an idealized, highresolution(25-m grid spacing) numerical simulation to investigate the deadly EF4(Enhanced Fujita scale category 4)tornado that occurred on 23 June 2016 and claimed 99 lives in Yancheng, Jiangsu Province. A tornadic supercell developed in the simulation that had striking similarities to radar observations. The violent tornado in Funing County was reproduced, exceeding EF4(74 ms–1), consistent with the on-site damage survey. It was accompanied by a funnel cloud that extended to the surface, and exhibited a double-helix vorticity structure. The signal of tornado genesis was found first at the cloud base in the pressure perturbation field, and then developed both upward and downward in terms of maximum vertical velocity overlapping with the intense vertical vorticity centers. The tornado's demise was found to accompany strong downdrafts overlapping with the intense vorticity centers. One of the interesting findings of this work is that a violent surface vortex was able to be generated and maintained, even though the simulation employed a free-slip lower boundary condition. | 20 | Neuroprotective effect of deferoxamine on erastin-induced ferroptosis in primary cortical neurons显示文摘The iron chelator deferoxamine has been shown to inhibit ferroptosis in spinal cord injury.However,it is unclear whether deferoxamine directly protects neurons from ferroptotic cell death.By comparing the survival rate and morphology of primary neurons and SH-SY5Y cells exposed to erastin,it was found that these cell types respond differentially to the duration and concentration of erastin treatment.Therefore,we studied the mechanisms of ferroptosis using primary cortical neurons from E16 mouse embryos.After treatment with 50μM erastin for 48 hours,reactive oxygen species levels increased,and the expression of the cystine/glutamate antiporter system light chain and glutathione peroxidase 4 decreased.Pretreatment with deferoxamine for 12 hours inhibited these changes,reduced cell death,and ameliorated cellular morphology.Pretreatment with the apoptosis inhibitor Z-DEVD-FMK or the necroptosis inhibitor necrostain-1 for 12 hours did not protect against erastin-induced ferroptosis.Only deferoxamine protected the primary cortical neurons from ferroptosis induced by erastin,confirming the specificity of the in vitro ferroptosis model.This study was approved by the Animal Ethics Committee at the Institute of Radiation Medicine of the Chinese Academy of Medical Sciences,China(approval No.DWLL-20180913)on September 13,2018. | Yan Zhang Bao-You Fan Yi-Lin Pang Wen-Yuan Shen Xu Wang Chen-Xi Zhao Wen-Xiang Li Chang Liu Xiao-Hong Kong Guang-Zhi Ning Shi-Qing Feng Xue Yao | 2020 | Neural Regeneration Research2020,15,8: | 9 |
