| 1 | Cluster analysis for syndromes of real-world coronary heartdisease with angina pectoris显示文摘Syndromes of coronary heart disease with angina pectoris were analyzed to provide guidance forclinical practice and to improve accuracy of traditional Chinese medicine (TCM) diagnoses and efficacy of TCMtreatment. A total of 860 cases with coronary heart disease with angina pectoris were selected from TCM ClinicalResearch Information Sharing System for TCM clinics and research. Syndromes were automatically extractedwith the cluster method and were analyzed to provide objective evidence for clinical studies. Final syndromeclassifications were recognized and confirmed by clinical experts. Popular syndromes included Qi and blooddeficiency, blood stasis and obstruction collaterals, liver depression and spleen deficiency, and Qi stagnation andblood stasis. Syndromes Qi and blood deficiency and blood stasis and obstruction collaterals accounted for 28.61%of total syndromes, whereas liver depression and spleen deficiency and Qi stagnation and blood stasis accountedfor 26.44%. | 2 | Au nanoclusters suppress chronic lymphocytic leukaemia cells by inhibiting thioredoxin reductase 1 to induce intracellular oxidative stress and apoptosis显示文摘Chronic lymphocytic leukaemia(CLL) is a rare blood cancer that always relapses as refractory disease and eventually leads to death. To date, therapeutic options for CLL patients are scarce and there is an urgent need to develop novel chemotherapeutics that are both effective and safe. Gold-containing compounds induce a lethal oxidative and endoplasmic reticulum stress response in cultured and primary CLL cells via inhibition of thioredoxin reductase(TrxR). However, traditional gold-containing medicines have revealed side effects during clinical applications. Therefore, safer gold-containing drugs are needed to overcome this challenge. In this study, a novel peptide templated gold cluster Au_(25)Sv_9 was synthesized and its therapeutic effect on CLL cells was evaluated. This nanocluster could induce cell apoptosis in MEC-1 cells in a dose-dependent manner which correlated with the uptake amount of clusters in cells.As expected, increasing intracellular reactive oxidative species(ROS) in MEC-1 cells was exhibited with the increase of cluster dosage. Further analyses demonstrated the underlying mechanism that the nanoclusters suppress the activity of TrxR1, increase the level of intracellular ROS, destroy the mitochondrial membrane potential and finally trigger the mitochondrial apoptotic pathway in MEC-1 cells.Furthermore, the direct interaction between Au_(25)Sv_9 clusters and TrxR1 was confirmed for the first time by isothermal titration calorimetry. | 4 | Intratumoural microbiota:a new frontier in cancer development and therapy显示文摘Human microorganisms,including bacteria,fungi,and viruses,play key roles in several physiological and pathological processes.Some studies discovered that tumour tissues once considered sterile actually host a variety of microorganisms,which have been confirmed to be closely related to oncogenesis.The concept of intratumoural microbiota was subsequently proposed.Microbiota could colonise tumour tissues through mucosal destruction,adjacent tissue migration,and hematogenic invasion and affect the biological behaviour of tumours as an important part of the tumour microenvironment.Mechanistic studies have demonstrated that intratumoural microbiota potentially promote the initiation and progression of tumours by inducing genomic instability and mutations,affecting epigenetic modifications,promoting inflammation response,avoiding immune destruction,regulating metabolism,and activating invasion and metastasis.Since more comprehensive and profound insights about intratumoral microbiota are continuously emerging,new methods for the early diagnosis and prognostic assessment of cancer patients have been under examination.In addition,interventions based on intratumoural microbiota show great potential to open a new chapter in antitumour therapy,especially immunotherapy,although there are some inevitable challenges.Here,we aim to provide an extensive review of the concept,development history,potential sources,heterogeneity,and carcinogenic mechanisms of intratumoural microorganisms,explore the potential role of microorganisms in tumour prognosis,and discuss current antitumour treatment regimens that target intratumoural microorganisms and the research prospects and limitations in this field. | Yaqi Cao Hui Xia Xueyun Tan Chunwei Shi Yanling Ma Daquan Meng Mengmeng Zhou Zhilei Lv Sufei Wang Yang Jin | 2024 | Signal Transduction and Targeted Therapy2024,9,2: | 0 |
