| 2 | miR-4458 inhibits the epithelial-mesenchymal transition of hepatocellular carcinoma cells by suppressing the TGF-β signaling pathway via targeting TGFBR1显示文摘Hepatocellular carcinoma(HCC)is one of the most lethal cancers in the world.MicroRNAs play a pivotal role in the progression of various cancers.To date,very lttle attention has beerl paid to miR-4458.Therefore,the aim of our study was to explore the function and underlying molecular mech-anism of miR-4458 in HCC.We found that the expression of miR-4458 was reduced in HCC tissues and cell lines.Forced overexpression of miR-4458 inhibited the migration,invasion,and epithelial-mesenchymal transition(EMT)of HCC cells,while downregulation of miR-4458 promoted the aggressive phenotype.Furthermore,transforming growth factor beta receptor 1(TGFBR1),the modulator of the TGF-βsignaling pathway,was verified to be a novel target gene of miR-4458 by dual-luciferase reporter gene assay.Upregulated miR-4458 dramatically abolished TGFBR1 and p-Smad2/3 expression,thus blocking the TGF-βsig naling pathway.Moreover,restoration of TGFBR1 partially rescued the miR-4458-mediated suppressive effect on the migration,invasion,and EMT and reactivated the TGFβsignaling pathway in HCC cells.In summary,our findings first demonstrated a mechanism of miR 4458 in HCC cell migration,invasion,and EMT by regulating the TGFβsignaling pathway via directly targeting TGFBR1. | Yuke Zhang Kun Shi Hang Liu Wei Chen Yunhai Luo Xufu Wei Zhongjun Wu | 2020 | Acta Biochimica et Biophysica Sinica2020,52,5: | 4 |

| 16 | Lethal Consequences in an Infant with Myelomeningocele Following an Inadvertent Treatment显示文摘Myelomeningocele(MMC)is a protrusion of spinal cord contents and meninges through a vertebral defect.Iatrogenic deaths of patients with MMC are rarely encountered in forensic practice.In our case,a 3‑month‑old female was born with a lumbosacral cyst,the size of which had been increasing gradually over age.There was no neurological,orthopedic,or urologic dysfunction.On the day of her death,she received a repetitive and rapid lumbosacral cyst puncture drainage procedure,performed by an illegal medical practitioner.Postmortem autopsy findings confirmed a diagnosis of MMC and the cause of death to be cerebellar tonsillar herniation.This is a pathetic case of preventable infant death.This report suggests that the possibility of MMC should be considered in infants born with a lumbosacral cyst,and aspiration is inadvisable.Besides,forensic autopsy has a valuable role in determining the exact cause of death,identifying,or excluding iatrogenic factors that may be relevant to death following a medical procedure.A final point is that prevention programs should be developed,especially by the health care sectors to reduce such tragedy. | Xuemei Wu Libing Yun Min Liu Xufu Yi | 2016 | Journal of Forensic Science and Medicine2016,2,3: | 0 |
| 17 | 前列腺癌骨显像诊断骨转移与病理分级及血清PSA的关系探讨(英文)显示文摘Objective:The aim of the study was to investigate the correlations between pathological grade,serum prostatespecific antigen(PSA) and bone scintigraphy in the diagnosis of metastasis diseases for prostate cancers,and to explore the characteristics of bone metastases for prostate cancer.Methods:Seventy-seven newly diagnosed prostate cancers were reviewed in the study.All the cases underwent bone scintigraphy,total serum PSA measurement by luminescent immunoassay before operation and were classified according to post-operative pathology diagnosis.We analyzed the correlations of the bone metastasis incidences and different pathological grades or different PSA levels.Results:Bone scans were indicative of metastases in 33 cases(42.86%).Significantly higher incidence of bone metastasis was observed in patients with poorly differentiated prostate cancer compared with patients with well(χ2 = 10.880,P = 0.001 < 0.05) and moderately(χ2 = 6.166,P = 0.013 < 0.05) differentiated prostate cancers.No significant difference between the well differentiated and moderately differentiated prostate cancers was found(χ2 = 0.612,P = 0.434 > 0.05).The serum PSA concentration had significant correlation with the incidence of bone metastasis.In 26 patients with PSA < 20 ng/mL,5 cases(19.23%) had bone metastasis while 28 of 51 cases(54.90%) with PSA > 20 ng/mL had bone metastasis.The serum PSA concentration had positive correlation with pathological grades of prostate cancer(r = 0.535,P = 0.01).Conclusion:Bone scintigraphy plays a great role in the diagnosis of bone metastasis for prostate cancer patients currently.The poorly differentiated prostate cancer and PSA > 20 ng/mL most likely suggested the possibility of bone metastasis. | Guifeng Yang Shuyao Zuo Chao Ma Bin Liu Guoming Wang Xufu Wang Hongyu Wu | 2009 | The Chinese-German Journal of Clinical Oncology2009,8,12: | 0 |
| 18 | Long noncoding RNA TMEM147-AS1 serves as a microRNA-326 sponge to aggravate the malignancy of gastric cancer by upregulating SMAD5显示文摘The abnormal expression of long noncoding RNAs(lncRNAs)is frequently observed in gastric cancer(GC)and considered an important driving force in GC progression.However,little is known regarding the involvement of TMEM147-AS1 in GC.Therefore,we examined TMEM147-AS1 expression in GC and determined its prognostic value.In addition,TMEM147-AS1 expression was depleted to identify the functional changes in response to TMEM147-AS1 deficiency.Using the cancer genome atlas dataset and our own cohort,we identified a strong expression of TMEM147-AS1 in GC.Increased TMEM147-AS1 levels in GC showed a significant association with poor prognosis.TMEM147-AS1 interference resulted in the inhibition of GC cell proliferation,colony-forming,migration,and invasion in vitro.Additionally,depletion of TMEM147-AS1 restricted the growth of GC cells in vivo.Mechanistically,TMEM147-AS1 functioned as a microRNA-326(miR-326)sponge.Furthermore,SMAD family member 5(SMAD5)was experimentally validated as the functional effector of miR-326.TMEM147-AS1 was demonstrated to sequester miR-326 away from SMAD5;consequently,knocking down TMEM147-AS1 downregulated SMAD5 levels in GC cells.The functional suppression of miR-326 or reintroduction of SMAD5 effectively reversed the attenuated behavior of GC cells caused by TMEM147-AS1 downregulation.In summary,TMEM147-AS1 exhibits tumorigenic activities in GC,which is likely the result of an altered miR-326/SMAD5 axis.Therefore,targeting TMEM147-AS1/miR-326/SMAD5 may represent a target for the treatment of GC. | XUFU QIN ZIYE JIANG YONGCUI ZHU HONGPENG XUE CHENGQUN WEI | 2021 | Oncology Research2021,29,4: | 0 |
