1Swelling and thermal properties of porous PNIPAM/PEG hydrogels prepared by radiation polymerization显示文摘In this study,a series of porous intelligent hydrogels were synthesized by radiation exhibiting the lower critical solution temperature(LCST) and fast response involving a combination of A’-isopropyl acrylamideas monomer, polyethylene glycol(PEG) as pore-forming agent and N,N-methylene-bis-acrylamide as crosslinking agent.The hydrogels were analyzed by Fourier transform infrared spectroscopy,and the influence of radiation doses on their swelling and thermal behaviors were studied.Their surface morphologies were examined by scanning electron microscopy.The results showed that PEG molecules only acted as pore-forming agent in the cross-linked polymerization.Their swelling ratios reduced with increasing radiation doses.The LCST was around 37℃,and varied little with the radiation doses.The frozen water content of PNIPAM/PEG6000 hydrogel reduced with increasing the radiation dose,and was greater than that of PN1PAM hydrogel at 15 kGy.Hydrogel macropores were prepared by PEG agent,and the hydrogels without PEG had a dense surface.The porous hydrogels are expected to be applied in the field of artificial intelligence material.LI Zhihui LIU Wentao LI Zhongyuan YANG Mingcheng GAO Xujing CUI Haitao HE Suqin ZHU Chengshen 2013Nuclear Science and Techniques2013,24,2:3
2Effects of cultivation of Osr HSA transgenic rice on functional diversity of microbial communities in the soil rhizosphere显示文摘With the widespread cultivation of transgenic crops, there is increasing concern about unintended effects of these crops on soil environmental quality. In this study, we used the Biolog method and ELISA to evaluate the possible effects of Osr HSA transgenic rice on soil microbial utilization of carbon substrates under field conditions. There were no significant differences in average well-color development(AWCD) values, Shannon–Wiener diversity index(H), Simpson dominance indices(D) and Shannon–Wiener evenness indices(E) of microbial communities in rhizosphere soils at eight samplings between Osr HSA transgenic rice and its non-transgenic counterpart. The main carbon sources utilized by soil microbes were carbohydrates, carboxylic acids, amino acids and polymers. The types,capacities and patterns of carbon source utilization by microbial communities in rhizosphere soils were similar throughout the detection period. We detected no Osr HSA protein in the roots of Osr HSA transgenic rice. 7In-situ construction of amorphous/crystalline contact Bi_(2)S_(3)/Bi_(4)O_(7) heterostructures for enhanced visible-light photocatalysis显示文摘Constructing a heterojunction photocatalyst is a significant method to enhance photocatalytic activity because it can promote the separation of photogene rated carriers.Herein,amorphous/crystalline contact Bi_(2)S_(3)/Bi_(4)0_(7) heterostructure was successfully synthesized by in-situ sulfidation of Bi407.The amorphous Bi_(2)S_(3) is diffused on the surface of Bi_(4)0_(7) rod,enhancing the visible light response and improving the transport of photogene rated carriers.Various characterizations confirm that the rapid separation of photogene rated carriers leads to increased photocatalytic performance.The optimized Bi_(2)S_(3)/Bi_(4)O_(7) heterostructure photocatalyst(BiS-0.15) exhibits the highest Cr(Ⅵ) reduction(0.01350 min^(-1)) and RhB oxidation(0.08011 min^(-1)) activity,which is much higher than that of pure Bi_(4)0_(7) and mixture under visible light irradiation.This work provides new insights into the construction of efficient novel photocatalysts.Feihu Mu Benlin Dai Wei Zhao Xiaofan Yang Xiaolong Zhao Xujing Guo 2021Chinese Chemical Letters2021,32,8:0

9SARS-CoV-2 N protein enhances the anti-apoptotic activity of MCL-1 to promote viral replication显示文摘Viral infection in respiratory tract usually leads to cell death,impairing respiratory function to cause severe disease.However,the diversity of clinical manifestations of SARS-CoV-2 infection increases the complexity and difficulty of viral infection prevention,and especially the high-frequency asymptomatic infection increases the risk of virus transmission.Studying how SARS-CoV-2 affects apoptotic pathway may help to understand the pathological process of its infection.Here,we uncovered SARS-CoV-2 imployed a distinct anti-apoptotic mechanism via its N protein.We found SARS-CoV-2 virus-like particles(trVLP)suppressed cell apoptosis,but the trVLP lacking N protein didn’t.Further study verified that N protein repressed cell apoptosis in cultured cells,human lung organoids and mice.Mechanistically,N protein specifically interacted with anti-apoptotic protein MCL-1,and recruited a deubiquitinating enzyme USP15 to remove the K63-linked ubiquitination of MCL-1,which stabilized this protein and promoted it to hijack Bak in mitochondria.Importantly,N protein promoted the replications of IAV,DENV and ZIKV,and exacerbated death of IAV-infected mice,all of which could be blocked by a MCL-1 specific inhibitor,S63845.Altogether,we identifed a distinct anti-apoptotic function of the N protein,through which it promoted viral replication.These may explain how SARS-CoV-2 effectively replicates in asymptomatic individuals without cuasing respiratory dysfunction,and indicate a risk of enhanced coinfection with other viruses.We anticipate that abrogating the N/MCL-1-dominated apoptosis repression is conducive to the treatments of SARS-CoV-2 infection as well as coinfections with other viruses.Pan Pan Weiwei Ge Zhiwei Lei Wei luo Yuqing Liu Zhanwen Guan Lumiao Chen Zhenyang Yu Miaomiao Shen Dingwen Hu Qi Xiang Wenbiao Wang Pin Wan Mingfu Tian Yang Yu Zhen Luo Xulin Chen Heng Xiao Qiwei Zhang Xujing Liang Xin Chen Yongkui Li Jianguo Wu 2023Signal Transduction and Targeted Therapy2023,8,6:0
10Effects of sgRNA length and number on gene editing efficiency and predicted mutations generated in rice显示文摘CRISPR-Cas9 is a common tool for gene editing, and appropriate sg RNAs are the key factor for successful editing. In this study, the effect of sg RNA length and number on editing efficiency was analyzed in rice using CYP81 A6 as the target gene. A series of CRISPR-Cas9 plant expression vectors containing single sg RNAs with different lengths(17, 18, 19, 20, 21, 22, 23 nt) or two sg RNAs were constructed and introduced into rice cultivar Zhonghua11 by Agrobacterium-mediated transformation. Analysis of the editing status of 1283 transgenic rice plants showed that 371 were successfully edited with base preference.Single A or T insertions were the most frequent among the six edited types. The editing efficiency of transgenic rice with two sg RNAs was higher than that with a single sg RNA. Editing efficiency and sg RNA length showed a normal distribution with 20 nt sg RNA(25%) being the most efficient. The editing efficiency decreased slightly with decreases of 1–2 bases(19 nt 20%, 18 nt 21%), but decreased significantly with a decrease of 3 bases(17 nt 4.5%). Editing efficiency was significantly reduced by adding 1 to 3 bases(21 nt 16.8%, 22 nt 13%, 23 nt 13%) to the sg RNA.
