1Quality Control and Nonclinical Research on CAR-T Cell Products: General Principles and Key Issues显示文摘Adoptive cell therapy using chimeric antigen receptor T (CAR-T) cells, which is a promising cancer immunotherapy strategy, has been developing very rapidly in recent years. CAR-T cells are genetically modified T cells that can specifically recognize tumor specific antigens on the surface of tumor cells, and then effectively kill tumor cells. At present, exciting results are being achieved in clinical applications of CAR-T cells for patients with hematological malignancies. The research and development of CAR-T cells for various targets and for the treatment of solid tumors have become a hot topic worldwide, so an increasing number of investigational new drug applications (INDAs) and new drug applications (NDAs) of CAR-T cell products are expected to be submitted in future. The quality control and nonclinical research of these products are of great significance in ensuring the safety and effectiveness of these products;however, they also present great challenges and difficulties. 3Immunogenicity and protective efficacy of a recombinant protein subunit vaccine and an inactivated vaccine against SARS-CoV-2 variants in non-human primates显示文摘Emerging SARS-CoV-2 variants and the gradually decreasing neutralizing antibodies over time post vaccination have led to an increase in incidents of breakthrough infection across the world.To investigate the potential protective effect of the recombinant protein subunit COVID-19 vaccine targeting receptor-binding domain(RBD)(PS-RBD)and whole inactivated virus particle vaccine(IV)against the variant strains,in this study,rhesus macaques were immunized with PS-RBD or IV vaccine,followed by a Beta variant(B.1.351)challenge.Although neutralizing activity against the Beta variant was reduced compared with that against the prototype,the decreased viral load in both upper and lower respiratory tracts,milder pathological changes,and downregulated inflammatory cytokine levels in lung tissues after challenge demonstrated that PS-RBD and IV still provided effective protection against the Beta variant in the macaque model.Furthermore,PS-RBD-induced macaque sera possessed general binding and neutralizing activity to Alpha,Beta,Delta,and Omicron variants in our study,though the neutralizing antibody(NAb)titers declined by varying degrees,demonstrating potential protection of PS-RBD against current circulating variants of concern(VOCs).Interestingly,although the IV vaccine-induced extremely low neutralizing antibody titers against the Beta variant,it still showed reduction for viral load and significantly alleviated pathological change.Other correlates of vaccine-induced protection(CoP)like antibody-dependent cellular cytotoxicity(ADCC)and immune memory were both confirmed to be existing in IV vaccinated group and possibly be involved in the protective mechanism.Qian He Qunying Mao Xiaozhong Peng Zhanlong He Shuaiyao Lu Jialu Zhang Fan Gao Lianlian Bian Chaoqiang An Wenhai Yu Fengmei Yang Yanan Zhou Yun Yang Yanyan Li Yadi Yuan Xujia Yan Jinghuan Yang Xing Wu Weijin Huang Changgui Li Junzhi Wang Zhenglun Liang Miao Xu 2022Signal Transduction and Targeted Therapy2022,7,4:2

5Ufl1 deficiency causes kidney atrophy associated with disruption of endoplasmic reticulum homeostasis显示文摘The UFMylation modification is a novel ubiquitin-like conjugation system,consisting of UBA5(E1),UFC1(E2),UFL1(E3),and the conjugating molecule UFM1.Deficiency in this modification leads to embryonic lethality in mice and diseases in humans.However,the function of UFL1 is poorly characterized.Studies on Ufl1 conditional knockout mice have demonstrated that the deletion of Ufl1 in cardiomyocytes and in intestinal epithelial cells causes heart failure and increases susceptibility to experimentally induced colitis,respectively,suggesting an essential role of UFL1 in the maintenance of the homeostasis in these organs.Yet,its physiological function in other tissues and organs remains completely unknown.In this study,we generate the nephron tubules specific Ufl1 knockout mice and find that the absence of Ufl1 in renal tubular results in kidney atrophy and interstitial fibrosis.In addition,Ufl1 deficiency causes the activation of unfolded protein response and cell apoptosis,which may be responsible for the kidney atrophy and interstitial fibrosis.Collectively,our results have demonstrated the crucial role of UFL1 in regulating kidney function and maintenance of endoplasmic reticulum homeostasis,providing another layer of understanding kidney atrophy.You Zhou Xifu Ye Chenlu Zhang Jiabao Wang Zeyuan Guan Juzhen Yan Lu Xu Ke Wang Di Guan Qian Liang Jian Mao Junzhi Zhou Qian Zhang Xiaoying Wu Miao Wang Yu-Sheng Cong Jiang Liu 2021Journal of Genetics and Genomics2021,48,5:1

7Effect of Polyisobutylene Succinimide on the Physical Stability of an Environmentally Friendly Pesticide Oil Dispersion Suspension显示文摘Oil dispersible suspension concentrates are safe,green,and environmentally friendly formulations.Problems such as layering,pasting,and bottoming are frequently encountered during the production,storage,and transportation process.Polyisobutylene succinimide functions as a dispersant and exhibits great potential to improve the physical stability of the oil dispersible suspension concentrate.From a microscopic perspective,the sorption characteristics of the polyisobutylene succinimide dispersant T151 on penoxsulam particle surfaces were comprehensively evaluated with XPS,FTIR,and SEM.The T151 adsorption procedure complied with a pseudo-second-order kinetic adsorption model,and it was a kind of physical sorption with an Ea of 22.57 kJ⋅mol^(−1).The T151 sorption model was consistent with the Langmuir isotherm.The adsorption process was spontaneous and followed by an entropy increase.TheΔH^(θ)of dispersant T151 on the surface of penoxsulam particles was 31.59 kJ⋅mol^(−1).The adsorption procedure was endothermic,and the primary force was hydrogen bonding.The XPS results showed that the F and S electronic peaks at the penoxsulam interface decreased,and that the C electronic peak increased significantly after the adsorption of dispersant T151,indicating the adsorption on the surface of penoxsulam particles.The results of this study provide a vital theoretical basis for the application of polyisobutylene succinimide dispersants in oil dispersible suspension systems.Liying Wang Junzhi Liu Chong Gao Xinxin Yan 2023Journal of Renewable Materials2023,11,6:0
8The TMRT K band observations towards 26 infrared dark clouds:NH_3, CCS, and HC_3N显示文摘We present one of the first Shanghai Tian Ma Radio Telescope (TMRT) K Band observations towards a sample of 26 infrared dark clouds (IRDCs).We observed the (1,1),(2,2),(3,3),and (4,4) transitions of NH_(3) together with CCS (2_(1)–>1_(0)) and HC_(3)NJ=2-1,simultaneously.The survey dramatically increases the existing CCS-detected IRDC sample from 8 to 23,enabling a better statistical study of the ratios of carbon-chain molecules (CCM) to N-bearing molecules in IRDCs.With the newly developed hyperfine group ratio (HFGR) method of fitting NH_(3) inversion lines,we found the gas temperature to be between 10 and18 K.The column density ratios of CCS to NH_(3) for most of the IRDCs are less than 10^(-2),distinguishing IRDCs from low-mass star-forming regions.We carried out chemical evolution simulations based on a three-phase chemical model NAUTILUS.Our measurements of the column density ratios between CCM and NH_(3) are consistent with chemical evolutionary ages of 10^(5) yr in the models.Comparisons of the data and chemical models suggest that CCS,HC_(3)N,and NH_(3) are sensitive to the chemical evolutionary stages of the sources.Jinjin Xie Gary A.Fuller Di Li Longfei Chen Zhiyuan Ren Jingwen Wu Yan Duan Junzhi Wang Juan Li Nicolas Peretto Tie Liu Zhiqiang Shen 2021Science China(Physics,Mechanics & Astronomy)2021,64,7:0
