1Effect of Bushenwenyanghuayu decoction on nerve growth factor and bradykinin/bradykinin B_1 receptor in a endometriosis dysmenorrhea mouse model显示文摘OBJECTIVE:To observe the effects of Bushenwenyanghuayu decoction(BD),a Traditional Chinese Medicine(TCM),on the serum concentration of nerve growth factor(NGF) and bradykinin(BK),and protein and mRNA levels of NGF and bradykinin B_1receptor(BKB1R) in a mouse model of endometriosis dysmenorrhea.METHODS:Seventy-five experimental female BALB/c mice were randomly divided into five groups,15 mice each:sham,model,BD high dose(61.67 g/kg),BD low dose(15.42 g/kg),and gestrinone(0.4 mg/kg) groups.All the mice except for those in the sham group underwent auto-transplantation surgery and were gavaged estradiol valerate(0.5 mg/kg,daily for 12 days) after surgery.On the 12 th day,1 h after administration,writhing response was induced by intraperitoneal injection of oxytocin at 2 U/mouse.The writhing frequency and latency were recorded and the volume of the ectopic foci was measured.The concentration of serum NGF and BK was detected by enzyme-linked immunosorbent assay,the protein expression of NGF and BKB1 R was tested by immunohistochemistry and western blotting,and NGF and BKB1 R mRNAs were detected by real-time PCR.RESULTS:Compared with the model group,the volume of the ectopic foci in the treatment groups was significantly lower(P < 0.01),the writhing frequency was decreased(P < 0.05),and the writhing latency was prolonged(P < 0.01).Compared with the sham group,serum NGF and BK levels in the model group were significantly increased(P <0.01).There were positive correlations for writhing frequency among the NGF and BK groups(P <0.01).The serum NGF and BK levels were significantly lower in the treatment groups than the model group(P < 0.05).The protein expression of NGF,BKB1 R was significantly decreased in the treatment groups compared with the model group(P < 0.01).NGF and BKB1 R mRNA expression was significantly decreased in the treatment groups compared with the model group(P < 0.01).CONCLUSION:NGF and BK/BKB1 R may play an important role in the development of endometriosis-associated dysmenorrhea,and BD was found to inhibit the development of endometriosis and relieve dysmenorrhea by influencing NGF and BK/BKB1 R mRNA and protein levels.Chen Jingwei Du Huilan Tong Ruixiao Yang Hua Ma Huirong 2015Journal of Traditional Chinese Medicine2015,35,2:12
2Enhanced vasoconstriction to α_1 adrenoceptor autoantibody in spontaneously hypertensive rats显示文摘Autoimmune activities have been implicated in the pathogenesis of hypertension.High levels of autoantibodies against the second extracellular loop of α1-adrenoceptor(α1-AR autoantibody,α1-AA) are found in patients with hypertension,and α1-AA could exert a α1-AR agonist-like vasoconstrictive effect.However,whether the vasoconstrictive effect of α1-AA is enhanced in hypertension is unknown.Using aortic rings of spontaneously hypertensive rats(SHR) and normotensive Wistar-Kyoto(WKY) rats,we observed the vasoconstrictive responses to α1-AA with phenylephrine(α1-AR agonist) as a positive control drug.Aortic nitrotyrosine levels were also measured by ELISA and immunohistochemistry.The results showed that the aortic constrictive responses to α1-AA and phenylephrine(both 1 nmol L-1-10 μmol L-1) were greater in SHR than in WKY rats.Endothelial denudation or L-NAME(a non-selective NOS inhibitor)(100 μmol L-1) increased α1-AA- or phenylephrine-induced vasoconstrictions both in SHR and WKY.However,selective iNOS inhibitor 1400W(10 μmol L-1) enhanced the α1-AA-induced aortic constriction in WKY,but not in SHR.The aortic nitrotyrosine level was significantly higher in SHR than WKY,as shown by both ELISA and immunohistochemistry.These results indicate that the vasoconstrictive response to α1-AA is enhanced in SHR,and this altered responsiveness is due to endothelial dysfunction and decreased NO bioavailability.The study suggests an important role of α1-AR autoimmunity in the pathogenesis and management of hypertension especially in those harboring high α1-AA levels.YAN Li TAN XiaoQiu CHEN WenXuan ZHU Hong CAO JiMin LIU HuiRong 2014Science China(Life Sciences)2014,57,7:10

4Altered regional homogeneity in post-traumatic stress disorder: a resting-state functional magnetic resonance imaging study显示文摘Objective Little is known about the brain systems that contribute to vulnerability to post-traumatic stress disorder (PTSD). Comparison of the resting-state patterns of intrinsic functional synchronization, as measured by functional magnetic resonance imaging (fMRI), between groups with and without PTSD following a traumatic event can help identify the neural mechanisms of the disorder and targets for intervention. Methods Fifty-four PTSD patients and 72 matched traumatized subjects who experienced the 2008 Sichuan earthquake were imaged with blood oxygen level-dependent (BOLD) fMRI and analyzed using the measure of regional homogeneity (ReHo) during the resting state. Results PTSD patients presented enhanced ReHo in the left inferior parietal lobule and right superior frontal gyrus, and reduced ReHo in the right middle temporal gyrus and lingual gyrus, relative to traumatized individuals without PTSD. Conclusion Our findings showed that abnormal brain activity exists under resting conditions in PTSD patients who had been exposed to a major earthquake. Alterations in the local functional connectivity of cortical regions are likely to contribute to the neural mechanisms underlying PTSD.Yan Yin Changfeng Jin Lisa T. 7Mild moxibustion at Tianshu (ST 25) decreases expression of prokineticin-1 and prokineticin receptor-1 in colon tissue of rats with chronic visceral hyperalgesia显示文摘Prokineticin-1 and prokineticin receptor-1 play important roles in visceral hypersensitivity and in-flammatory pain. Visceral hypersensitivity is closely associated with irritable bowel syndrome. Mild moxibustion can relieve chronic visceral hyperalgesia in rats with irritable bowel syndrome. We hypothesized that prokineticin-1 and prokineticin receptor-1 is the key target in the mechanism. This study established chronic visceral hyperalgesia rat models by colorectal distention. Protein and mRNA expression of prokineticin-1 and prokineticin receptor-1 were determined by immunohisto-chemical method and fluorescence quantitative-PCR, respectively, and were found to be signifi-cantly increased in visceral hyperalgesic rats. 8Analgesic action of suspended moxibustion in rats with chronic visceral hyperalgesia correlates with enkephalins in the spinal cord显示文摘Rats that modeled chronic visceral hyperalgesia received suspended moxibustion at bilateral Tianshu (ST25) and Shangjuxu (ST37) once daily over a period of 7 days. Results show that suspended moxibustion significantly depressed abdominal withdrawal reflex scores and increased enkephalin concentration in the spinal cord. 9Isolation and phylogenetic assignation of actinomycetes in the marine sediments from the Arctic Ocean显示文摘Actinomycetes in five marine sediments collected from the Arctic Ocean at depths of 43 to 3 050 m were cultivated using a variety of media. A total of 61 actinomycete colonies with substrate mycelia only were observed, and no colonies with aerial mycelia were observed under aerobic conditions at 15 ℃. From these colonies, 28 were selected to represent different morphological types.Denaturing gradient gel electrophoresis(DGGE) was used to check the purity of isolates and select representatives for subsequent sequencing. Phylogentic analyses based on nearly full-length 16S ribosomal RNA gene (rDNA) sequences indicated that the actinomycetes isolated were accommodated within genus Rhodococcus of family Nocardiaceae, genus Dietzia of family Dietziaceae, genera Janibacter and Terrabacter of family Instrasporangiaceae and genera Kocuria and Arthrobacter of family Micrococcaceae. One of the strains(P27-24) from the deep-sea sediment at depth of 3 050 m was found to be identical in 16S rDNA sequence(1474/1474)with the radiation-resistant Kocuria rosea ATCC 187T isolated from air. More than half of the isolates showed the similarities ranging from 99.5% to 99.9% in 16S rDNA sequence to dibenzofran-degrading, butyl 2-ethylhexanoate-hydrolysising and nitrile-metabolizing actinomycetes. All the strains isolated were psychrotolerant bacteria and grew better on the media prepared with natural seawater than on the media prepared with deionized water. Three of them (Dietzia sp. P27-10, Rhodococcus sp. 11Construction of multiform scFv antibodies using linker peptide显示文摘Multiform single chain variable fragments (scFvs) including different length linker scFvs and bispecific scFv were constructed. The linker lengths of 0, 3, 5, 8, 12, and 15 amino acids between VH and VL of antideoxynivalenol (anti-DON) scFv were used to analyze the affinities of scFvs. The affinity constants of these scFvs increased when the linker was lower than 12 amino acids. The affinity constant would not change when the linker was longer than 12 amino acids. Fusion gene of anti-DON scFv and antizearalenone (anti-ZEN) scFv was also constructed through connection by a short peptide linker DNA to express a bispecific scFv. The affinity constants assay showed that the two scFvs of fusion bispecific scFv remained their own affinity compared to their parental scFvs. Competitive direct enzyme linked immunosorbent assay was used to detect DON and ZEN in contaminated wheat (Triticum aestivum L.) samples, and the results indicated that this bispecific scFv was applicable in DON and ZEN detection. 12Lignin biosynthesis by suppression of two O-methyl-transferases显示文摘Caffeic acid O-methyltransferase (COMT) and caffeoyl-CoA-3-O-methyltransferase (CCoAOMT) genes encode two methyltransferases at different substrate levels in lignin biosynthesis. We constructed two single antisense expression vectors containing the cDNA of COMT or CCoAOMT genes and one dual antisense expression vector containing cDNAs of both OMTs genes. The antisense constructs were transferred into tobacco mediated by Agrobac-terium tumefacience. PCR-Southern analysis indicated that antisense cDNAs had been integrated into the genome of the transgenic tobacco. The antisense genes were also expressed at transcriptional level displayed by Northern dot analysis. Klason lignin assay of 3-month-old transgenic tobaccos showed that repression in COMT or CCoAOMT alone could result in reduction of lignin content, more reduction was caused by suppression in CCoAOMT than in COMT. 15Molecular genetic diversity of bacteria in the bottom section of arctic sea ice from the Canada Basin显示文摘PCR-DGGE approach was used to analyze bacterial diversity in the bottom section of seven arctic sea ice samples colleted from the Canada Basin. Thirty-two 16S rDNA sequences were obtained from prominent DGGE bands. The closest relatives of these sequences are found to be those of cultivated or uncultured bacteria from antarctic or arctic sea ice. Phylogenetic analysis clustered these sequences or phylotypes within α- proteobacteria, γ-proteobacteria and CFB (cytophaga-flexibacter-bacteroides) group. Sequences belonging to γ-proteobacteria were dominant and members of the CFB group were highly abundant. 16Effects of calcination temperature on physicochemical property and activity of CuSO4/TiO2 ammonia-selective catalytic reduction catalysts显示文摘CuSO4/TiO2 catalysts with high catalytic activity and excellent resistant to SO2 and H2 O,were thought to be promising catalysts used in Selective catalytic reduction of nitrogen oxides by NH3.The performance of catalysts is largely affected by calcination temperature.Here,effects of calcination temperature on physicochemical property and catalytic activity of CuSO4/TiO2 catalysts were investigated in depth.Catalyst samples calcined at different temperatures were prepared first and then physicochemical properties of the catalyst were characterized by N2 adsorption-desorption,X-ray diffraction,thermogravimetric analysis,Raman spectra,Fourier-transform infrared spectroscopy,X-ray photoelectron spectroscopy,temperature-pro grammed desorption of NH3,temperature-programmed reduction of H2 and in situ diffuse reflectance infrared Fourier transform spectroscopy.Results revealed that high calcination temperature had three main effects on the catalyst.First,sintering and anatase transform into rutile with increase of calcination temperature,causing a decrement of specific surface area.Second,decomposition of CuSO4 under higher calcination temperature,resulting in disappears of Br(?)nsted acid sites(S-OH),which had an adverse effect on surface acidity.Third,CuO from the decomposition of CuSO4 changed surface reducibility of the catalyst and favored the process of NH3 oxidation to nitrogen oxides(NOx).Thus,catalytic activity of the catalyst calcined under high temperatures(≥600℃)decreased largely.Yanke Yu Jiali Zhang Changwei Chen Chi He Jifa Miao Huirong Li Jinsheng Chen 2020Journal of Environmental Sciences2020,32,5:6
17Electroacupuncture at Tianshu (ST 25) for diarrhea-predominant irritable bowel syndrome using positron emission tomography Changes in visceral sensation center显示文摘Previous studies have demonstrated that electroacupuncture therapy is effective in the treatment of irritable bowel syndrome. However, the precise mechanism of this therapy is unknown. The present study served to investigate the effects of electroacupuncture therapy on treatment of patients with diarrhea-predominant irritable bowel syndrome (IBS). We compared brain activation maps based on the changes of cerebral glucose metabolism obtained by 18-fluorodeoxyglucose positron emission tomography scanning under three conditions: resting, rectal balloon distension and rectal balloon distension plus electroacupuncture. Under the resting condition, compared with healthy controls, IBS patients displayed an increasing regional cerebral metabolic rate of glucose over a wide range: bilateral superior temporal gyrus, right middle occipital gyrus, superior frontal gyrus and bilateral middle frontal gyrus. However, there was no significant activity in the visceral pain center. Compared with the resting condition, under the rectal balloon distension condition, patients with IBS had a greater regional cerebral metabolic rate of glucose in the prefrontal cortex, left anterior cingulate cortex, postcentral gyrus, precentral gyrus and temporal gyrus. Under the rectal balloon distension plus electroacupuncture condition, stimulation by electroacupuncture at Tianshu (ST 25) manifested a decreased regional cerebral metabolic rate of glucose in the left cingulate gyrus, right insula, right caudate nucleus, fusiform gyrus and hippocampal gyrus. 18Synthesis and characterization of tungsten and barium co-doped La2Mo2O9 by sol-gel process for solid oxide fuel cells显示文摘Herein,we demonstrate the synthesis of W and Ba co-doped La2Mo2O9(LBMWO)nanocrystalline powder by a sol-gel process.In all the compositions have general formulae La1.9Ba0.1Mo2-xWxO8.95(x=0-0.40).The crystal structure,microstructure and conductivity of LBMWO were characterized by X-ray diffraction,scanning electron microscopy and electrical impedance spectroscopy.In addition,the thermal and decomposition properties of the LBMWO gel were analyzed by differential scanning calorimetry-thermogravimetric.The results reveal that all LBMWO powders calcined at 700℃have a cubic structure;the average crystallite size is about 48 nm.The unit cell parameter of LBMWO powders increases with increase in W content.The as-synthesized nanocrystalline LBMWO samples exhibit excellent sinterability and a relatively lower sintering temperature of 900℃.A high relative density of-96%is achieved after sintering at 900℃which is in good agreeme nt with the re sults of the SEM.Moreover,W and Ba codoping suppresses the phase transition and effectively stabilizes theβ-phase at low temperature.At the same time,La1.9Ba0.1Mo1.85W0.15O8.95 exhibits high ionic conductivity,3.07 x 10^-2 S/cm at 800℃.It is therefore concluded that co-doping can improve the properties of La2MO2O9 electrolytes.Chang′an Tian Lingbo Shao Dongdong Ji Jie Yang Jinsong Xie Qiyi Yin Huirong Le 2019Journal of Rare Earths2019,37,9:5

20Influence of herb-partitioned moxibustion at Qihai (CV6) and bilateral Tianshu (ST25) and Shangjuxu (ST37) acupoints on toll-like receptors 4 signaling pathways in patients with ulcerative colitis显示文摘OBJECTIVE: To evaluate the efficacy of herb-partitioned moxibustion(HPM) at Qihai(CV6), Tianshu(ST25) and Shangjuxu(ST37) acupoints in relieving symptoms and the immune regulation of HPM on the toll-like receptors 4(TLR4) signaling pathway in ulcerative colitis(UC) patients.METHODS: A randomized, single-blind study was conducted 63 patients to receive HPM or sham HPM treatment. The efficacy outcomes included scores of the Mayo, Baron, inflammatory bowel disease questionnaire(IBDQ), self-rating depression scale(SDS), self-rating anxiety scale(SAS). HE staining was used to observe the histopathological changes of the colon. The expression of inflammatory cytokines and TLR4 signaling pathway related molecules were determined by enzyme-linked immunosorbent assay and immunohistochemistry.RESULTS: Baron, SDS, SAS scores were significantly decreased in moxibustion group(P < 0.05), IBDQ score was significantly greater in the moxibustion group than in the sham moxibustion group(P < 0.05). Histopathology of mucosal biopsies showed that both two groups improved in mucosa after treatment. The expression levels of tumor necrosis factor-α, interleukin-2, interleukin-12, interferon-γ, and TLR4, lipopolysaccharide, myeloid differentiation factor 88, interleukin receptor associated kinase, tumor necrosis factor receptor associated factor 6 and nuclear factor kappa-B p65 were significantly lower in the moxibustion group than in the sham moxibustion group(P < 0.05).CONCLUSION: This study showed that HPM at Qihai(CV6), Tianshu(ST25) and Shangjuxu(ST37) acupoints is effective to relieve symptoms, anxiety, depression and improving life quality in UC patients,which may be related to the immune regulation of HPM on TLR4 signaling pathway.QI Qin Hyoyoung Im LI Kunshan GU Muen WU Huangan YANG Ling HUANG Yan ZHAO Jimeng CUI Yunhua LIU Huirong WU Luyi 2021Journal of Traditional Chinese Medicine2021,41,3:5
